Pharmacological regulation of tissue fibrosis by targeting the mechanical contraction of myofibroblasts

Fibrosis can occur in almost all tissues and organs and affects normal physiological function,which may have serious consequences,such as organ failure.However,there are currently no effective,broad-spectmm drugs suitable for clinical application.Revealing the process of fibrosis is an important prerequisite for the development of new therapeutic targets and drugs.Studies have shown that the limiting of myofibroblast activation or the promoting of their elimination can ameliorate fibrosis.However,it has not been reported whether a direct decrease in cell contraction can inhibit fibrosis in vivo.here,we have shown that(-)-blebbistatin(Ble),a non-muscle myosin II inhibitor,displayed significant inhibition of liver fibrosis in different chronic injury mouse models in vivo.We found that Ble reduced the stiffness of fibrotic tissues from the early stage,which reduced the extent of myofibroblast activation induced by a stiffer extracellular matrix(ECM).Moreover,Ble also reduced the activation of myofibroblasts induced by TGF-β1,which is the most potent pro-fibrotic cytokine.Mechanistically,Ble reduced mechanical contraction,which inhibited the assembly of stress fibers,decreased the F/G-actin ratio,and led to the exnucleation of YAPJ and MRTF-A.Finally,we verified its broad-spectrum antifibrotic effect in multiple models of organ fibrosis.Our results highlighted the important role of mechanical contraction in myofibroblast activation and maintenance,rather than just a characteristic of activation,suggesting that it may be a potential target to explore broad-spectrum drugs for the treatment of fibrotic diseases.

Numerical simulation of transient characteristics in a bulb turbine during the load rejection process

To evaluate the safety of the bulb tubular turbine,the dynamic hydraulic characteristics of a hydropower station system during the load rejection process are studied through numerical simulations and a prototype test.In the developed model,a dynamic grid technology(DGT)controls the closure of the guide vane and the blade,whilst the moment balance equation and the user-defined function(UDF)provide the runner’s rotation speed.The 3-D transient simulation method can well predict the rotation speed and mass flow curves in the state of load rejection.The simulation outcomes of the system performance are basically consistent with the measurement data of the prototype.As observed,the runner is subjected to the reversely increased torque and axial force,the system is in a braking phase,and the maximum speed peaks at 144.6%of the rated speed.Moreover,the internal flow of the runner is greatly affected by the closure of the guide vane,and the draft tube forms an eccentric spiral vortex rope.It breaks downstream,aggravating the instability of the draft tube.Overall,the transient characteristics span for the first five seconds,demonstrating the importance of establishing an efficient governing controller.The obtained results are useful for designing the turbine’s flow channel with a double regulating function and comprehending the turbine’s transient characteristics.

A survey on facial image deblurring

When a facial image is blurred,it significantly affects high-level vision tasks such as face recognition.The purpose of facial image deblurring is to recover a clear image from a blurry input image,which can improve the recognition accuracy,etc.However,general deblurring methods do not perform well on facial images.Therefore,some face deblurring methods have been proposed to improve performance by adding semantic or structural information as specific priors according to the characteristics of the facial images.In this paper,we survey and summarize recently published methods for facial image deblurring,most of which are based on deep learning.First,we provide a brief introduction to the modeling of image blurring.Next,we summarize face deblurring methods into two categories:model-based methods and deep learning-based methods.Furthermore,we summarize the datasets,loss functions,and performance evaluation metrics commonly used in the neural network training process.We show the performance of classical methods on these datasets and metrics and provide a brief discussion on the differences between model-based and learning-based methods.Finally,we discuss the current challenges and possible future research directions.

Potential transformation of organic matter by microbes in cryoconite,Tibetan Plateau

Microorganisms play an essential role in the glacier carbon cycle;how they transform organic matter in mountain glacial cryoconite remains to be studied.here,we applied ultra-high resolution Fourier transform ion cyclotron resonance mass spectrometry(FT-ICR MS)and deep sequencing of 16S rRNA gene,to investigate the temporal microbial transformation of dissolved organic matter(DOM)of the Tibetan Plateau cryoconite.During the 60-day incubation,we found that DOM in cryoconite underwent a three-stage transformation,with decreasing bioavailability over time.The microbial community did not change much in the first week while degrading DOM molecules that were associated with higher H/C_(wa)and lower O/C_(wa).During days 15-30,DOM composition remained stable while microbial diversity increased.By day 60,the DOM was microbially converted into a higher state of recalcitrance,with higher values of aromatic index,O/C_(wa),and lower H/C_(wa),which contained molecules containing more heteroatoms.Cooperation among various microbial taxa,like Cyanobacteria,Bacteroidota,Gammaproteobacteria,Firmicutes,and Actinobacteriota,drove the DOM transformation in cryoconite.This study sheds light on the in-situ transformation of DOM composition meditated by microbial populations in cryoconite at a temporal scale,providing new insights into understanding the microbial roles in the glacier organic carbon transformation.

The breaking of spin symmetry in the single-particle resonances in deformed nuclei

The exploration of spin symmetry (SS) in nuclear physics has been instrumental in identifying atomic nucleus structures.In this study,we solve the Dirac equation from the relativistic mean field (RMF) in complex momentum representation.We investigated SS and its breaking in single-particle resonant states within deformed nuclei,with a focus on the illustrative nucleus168Er.This was the initial discovery of a resonant spin doublet in a deformed nucleus,with the expectation of the SS approaching the continuum threshold.With increasing single-particle energy,the splitting of the resonant spin doublets widened significantly.This escalating splitting implies diminishing adherence to the SS,indicating a departure from the expected behavior as the energy levels increase.We also analyzed the width of the resonant states,showing that lower orbital angular momentum resonances possess shorter decay times and that SS is preserved within broad resonant doublets,as opposed to narrow resonant doublets.Comparing the radial density of the upper components for the bound-state and resonant-state doublets,it becomes evident that while SS is well-preserved in the bound states,it deteriorates in the resonant states.The impact of nuclear deformation (β_(2)) on SS was examined,demonstrating that an increase in β_(2) resulted in higher energy and width splitting in the resonant spin doublets,which is attributed to increased component mixing.Furthermore,the sensitivity of spin doublets to various potential parameters such as surface diffuseness (a),radius (R),and depth (Σ0) is discussed,emphasizing the role of these parameters in SS.This study provides valuable insights into the behavior of spin doublets in deformed nuclei and their interplay with the nuclear structure,thereby advancing our understanding of SS in the resonance state.

The presence of protein aggregates in numerous human diseases underscores the significance of detecting these aggregates to comprehend disease mechanisms and develop novel therapeutic approaches for combating these disorders.Despite the development of various biosensors and fluorescent probes that selectively target amyloid fibers or amorphous aggregates,there is still a lack of tools capable of simultaneously detecting both types of aggregates.herein,we demonstrate the quantitative discernment of amorphous aggregates by QM-FN-SO3,an aggregationinduced emission(AIE)probe initially designed for detecting amyloid fibers.This probe easily penetrates the membranes of the widely-used prokaryotic model organism Escherichia coli,enabling the visualization of both amorphous aggregates and amyloid fibers through near-infrared fluorescence.Notably,the probe exhibits sensitivity in distinguishing the varying aggregation propensities of proteins,regardless of whether they form amorphous aggregates or amyloid fibers in vivo.These properties contribute to the successful application of the QM-FN-SO3 probe in the subsequent investigation of the antiaggregation activities of two outer membrane protein(OMP)chaperones,both in vitro and in their physiological environment.Overall,our work introduces a near-infrared fluorescent chemical probe that can quantitatively detect amyloid fibers and amorphous aggregates with high sensitivity in vitro and in vivo.Furthermore,it demonstrates the applicability of the probe in chaperone biology and its potential as a high-throughput screening tool for protein aggregation inhibitors and folding factors.

State Estimation of Drive-by-Wire Chassis Vehicle Based on Dual Unscented Particle Filter Algorithm

Accurate vehicle dynamic information plays an important role in vehicle driving safety.However,due to the characteristics of high mobility and multiple controllable degrees of freedom of drive-by-wire chassis vehicles,the current mature application of traditional vehicle state estimation algorithms can not meet the requirements of drive-by-wire chassis vehicle state estimation.This paper proposes a state estimation method for drive-by-wire chassis vehicle based on the dual unscented particle filter algorithm,which make full use of the known advantages of the four-wheel drive torque and steer angle parameters of the drive-by-wire chassis vehicle.In the dual unscented particle filter algorithm,two unscented particle filter transfer information to each other,observe the vehicle state information and the tire force parameter information of the four wheels respectively,which reduce the influence of parameter uncertainty and model parameter changes on the estimation accuracy during driving.The performance with the dual unscented particle filter algorithm,which is analyzed in terms of the time-average square error,is superior of the unscented Kalman filter algorithm.The effectiveness of the algorithm is further verified by driving simulator test.In this paper,a vehicle state estimator based on dual unscented particle filter algorithm was proposed for the first time to improve the estimation accuracy of vehicle parameters and states.

Roles of nanoparticles in arsenic mobility and microbial community composition in arsenic-enriched soils

Environmental effects of nano remediation engineering of arsenic(As)pollution need to be considered.In this study,the roles of Fe_(2)O_(3)and TiO_(2)nanoparticles(NPs)on the microbial mediated As mobilization from As contaminated soil were investigated.The addition of Fe_(2)O_(3)and TiO_(2)NPs restrained As(V)release,and stimulated As(Ⅲ)release.As(V)concentration decreased by 94% and 93% after Fe_(2)O_(3)addition,and decreased by 89% and 45% after Ti O_(2)addition compared to the Biotic and Biotic+Acetate(amended with sodium acetate)controls,respectively.The maximum values of As(Ⅲ)were 20.5 and 27.1μg/L at 48 d after Fe_(2)O_(3)and TiO_(2)NPs addition,respectively,and were higher than that in Biotic+Acetate control(12.9μg/L).The released As co-precipitated with Fe in soils in the presence of Fe_(2)O_(3)NPs,but adsorbed on TiO_(2)NPs in the presence of TiO_(2)NPs.Moreover,the addition of NPs amended with sodium acetate as the electron donor clearly promoted As(V)reduction induced by microbes.The NPs addition changed the relative abundance of soil bacterial community,while Proteobacteria(42.8%-70.4%),Planctomycetes(2.6%-14.3%),and Firmicutes(3.5%-25.4%)were the dominant microorganisms in soils.Several potential As/Fe reducing bacteria were related to Pseudomonas,Geobacter,Desulfuromonas,and Thiobacillus.The addition of Fe_(2)O_(3)and TiO_(2)NPs induced to the decrease of arr A gene.The results indicated that the addition of NPs had a negative impact on soil microbial population in a long term.The findings offer a relatively comprehensive assessment of Fe_(2)O_(3)and TiO_(2)NPs effects on As mobilization and soil bacterial communities.

Predictors of complications after prophylactic ileostomy reversal for rectal cancer:A retrospective study

BACKGROUND Previous studies have analyzed the risk factors for complications after ileostomy reversal for rectal cancer(RC),but there were significant differences in the reported risk factors for complications after stoma reversal.No studies have analyzed the risk factors for stoma-related complications and overall postoperative com-plications separately.AIM To analyze the risk factors for overall complications and stoma-related complications after ileostomy reversal for patients with RC.METHODS This was a retrospective study of 439 patients who underwent ileostomy reversal at a clinical center and were followed up between September 2012 and September 2022.Continuous variables are expressed as the mean±SD and were analyzed with independent-sample t tests,while frequency variables are expressed as n(%),and theχ2 test or Fisher’s exact test was used.Univariate and multivariate logistic regression analyses were used to identify predictors of overall complications and stoma-related complications.RESULTS The overall complication rate after ileostomy reversal was 11.4%.Patients with lower preoperative albumin concentration(Phe incidence of stoma-related complications was 6.4%.Analysis of the study showed that a higher body mass index(BMI)(Phe pre
