ImSpiRE: image feature-aided spatial resolution enhancement method

The resolution of most spatially resolved transcriptomic technologies usually cannot attain the single-cell level, limiting their applications in biological discoveries. Here, we introduce ImSpiRE, an image feature-aided spatial resolution enhancement method for in situ capturing spatial transcriptome. Taking the information stored in histological images, ImSpiRE solves an optimal transport problem to redistribute the expression profiles of spots to construct new transcriptional profiles with enhanced resolution, together with extending the gene expression profiles into unmeasured regions. Applications to multiple datasets confirm that ImSpiRE can enhance spatial resolution to the subspot level while contributing to the discovery of tissue domains, signaling communication patterns, and spatiotemporal characterization.

SSVEP-based brain-computer interfaces are vulnerable to square wave attacks

Electroencephalogram(EEG) based brain-computer interfaces(BCIs) have attracted wide attention in recent years. Steady-state visual evoked potential(SSVEP) is one of the most popular BCI paradigms,which has high information transmission rate and short user calibration time. Recent research has shown that EEG-based BCIs are under the threat of adversarial examples. However, existing attack approaches are very difficult to implement in a real-world system. Considering SSVEP’s dependency on the frequency information, this paper proposes to use square wave signals as adversarial perturbations to attack SSVEP-based BCIs, which are easy to generate and apply in practice. EEG trials contaminated by the square wave perturbation can be classified into any target class specified by the attacker. Compared with previous approaches,our perturbation can be implemented much more easily. Experiments on two SSVEP datasets demonstrated the efficiency and robustness of the proposed approach in attacking two SSVEP classifiers based on canonical correlation analysis, exposing a critical security problem in SSVEP-based BCIs.

Cohort study on the treatment of BRAF V600E mutant metastatic colorectal cancer with integrated Chinese and western medicine

BACKGROUND Patients with BRAF V600E mutant metastatic colorectal cancer(mCRC)have a low incidence rate,poor biological activity,suboptimal response to conventional treatments,and a poor *** the previous cohort study on mCRC conducted by our team,it was observed that integrated Chinese and Western medicine treatment could significantly prolong the overall survival(OS)of patients with colorectal ***,we further explored the survival benefits in the population with BRAF V600E mutant *** To evaluate the efficacy of integrated Chinese and Western medicine in the treatment of BRAF V600E mutant metastatic colorectal *** A cohort study was conducted on patients with BRAF V600E mutant metastatic colorectal cancer admitted to Xiyuan Hospital of China Academy of Chinese Medical Sciences and Traditional Chinese Medicine Hospital of Xinjiang Uygur Autonomous Region from January 2016 to December *** patients were divided into two *** A total of 34 cases were included,with 23 in Chinese-Western medicine cohort(cohort A)and 11 in Western medicine cohort(cohort B).The median overall survival was 19.9 months in cohort A and 14.2 months in cohort B,with a statistically significant difference(P=0.038,hazard ratio=0.46).The 1-3-year survival rates were 95.65%(22/23),39.13%(9/23),and 26.09%(6/23)in cohort A,and 63.64%(7/11),18.18%(2/11),and 9.09%(1/11)in cohort B,*** analysis showed statistically significant differences in median OS between the two cohorts in the right colon,liver metastasis,chemotherapy,and first-line treatment subgroups(P<0.05).CONCLUSION Integrated Chinese and Western medicine can prolong the survival and reduce the risk of death in patients with BRAF V600E mutant metastatic colorectal cancer,with more pronounced benefits observed in patients with right colon involvement,liver metastasis,combined chemotherapy,and first-line treatment.

Comparison of long-term total ozone observations from space-and ground-based methods at Zhongshan Station,Antarctica

Total ozone errors for satellite observations at Zhongshan Station in Antarctica are characterized using their relative difference （RD） from ground-based Brewer observations during 1993-2015. All satellite total ozone observations slightly overestimated ground-based ones （with RD less than 4%）. This is in contrast to conclusions drawn from global-scale validation studies, where main ground-based reference stations are located in middle latitudes. Given multiple total ozone data per day at Zhongshan Station, observed by a sun synchronous orbit satellite, measurements at the lowest solar zenith angle （SZA） show greatest consistency with Brewer ones, having an overall RD of-0.02-1.15%. Algorithm-retrieved total ozone data from the Total Ozone Mapping Spectrometer （TOMS）, including Solar Backscatter Ultra Violet （SBUV）, TOMS-Earth Probe （EP）, Ozone Monitoring Instrument （OMI）-TOMS, show best agreement with ground-based values; followed by the Global Ozone Measurement Experiment-type Direct Fitting （GOD-FIT） algorithm for the GOME-2A, and finally the Differential Optical Absorption Spectroscopy （DOAS） --Algorithm retrieved products for satellites-detectors of Global Ozone Measurement Experiment （GOME）, Scanning Imaging Absorption spectroMetr for Atmospheric CHartographY （SCIAMACHY）, and OMI. Satellite total ozone RD presents some statistical characteristics, but no specific trends. DOAS and GOME-2A algorithms have values that significantly increase, when the SZA is above 60°-70°, whereas values for GOME-2A decrease, when the SZA is 80-85°. Satellite total ozone RD is a minimum, when the Brewer total ozone is 300-350 DU, with an obvious increase in RD values for DOAS- and GOME-2A, when the Brewer total ozone is 150-300 DU. Satellite total ozone RD obviously increases, as the time difference between satellite overpasses and Brewer measurements grows. Specifically, RD rises as the absolute time difference increases to more than 4 h, yielding an OMI-TOMS RD of more than 10%

Cobalamin(Cbl)-dependent radical S-adenosyl-L-methionine(SAM) proteins constitute the largest collection of the radical SAM superfamily that has hundreds of thousands of individual members. Many of these proteins are involved in the biosynthesis of pharmaceutically important natural products to catalyze chemically demanding reactions. In the biosynthetic pathway of chuangxinmycin(CXM), a unique indole alkaloid antibiotic with potent anti-infective activity, functionalization of the characteristic thiopyrano[4,3,2-cd]indole scaffold by regio-and stereoselective C3-methylation is believed to rely on a Cbl-dependent radical process, which, however, remained to be reconstituted biochemically. We here report the dissection of this enzymatic process, which requires the incorporation of Cxm8, a Cbl-dependent radical SAM protein, with Cxm9, a DUF5825family protein that shares no homology to any proteins of known functions. Cxm8 and Cxm9 function together by forming an unexpected heterodimeric complex that selectively catalyzes C3-methylation of the tricyclic indole-S-hetero ring system in a successive manner, achieving CXM and a recently identified, C3-dimethylated congener. Detailed biochemical characterization,isotope labeling, structural simulation and bioinformatics analysis rationalized the catalysis of the Cxm8/Cxm9 complex and particularly the necessity of the DUF5825 protein for C3-methylase activity. This is the first example that a Cbl-dependent protein acts with a partner to exhibit radical SAM activity.

Iron recovery and rare earths enrichment from Bayan Obo tailings using Coal-Ca(OH)2-NaOH roasting followed by magnetic separation

The recovery of iron and enrichment of rare earths from Bayan Obo railings were investigated using Coal Ca（OH） 2 NaOH roasting followed by magnetic separation. The influences of roasting temperature, roasting time, coal content, milling time, Ca（OH）2 dosage and NaOH dosage on the iron and rare earths recovery were explored. The results showed that the magnetic concentrate containing 70.01 wt. % Fe with the iron recovery of 94.34G and the tailings of magnetic separation containing 11.46 wt. % rare earth oxides （REO） with the REO recovery of 98. 19% were obtained under the optimum conditions （i. e. , roasting temperature of 650℃, roasting time of 60 min, coal content of 2.0%, milling time of 5 min, and NaOH dosage of 2.0%）. The Ca（OH）2 dosage had no effect on the separation of iron and rare earths. According to the mineralogical and morphologic analysis, the iron and rare earths of Bayan Obo tailings could be utilized in subsequent ironmaking process and hydrometallurgy process.

High-Efficiency Dynamic Terahertz Deflector Utilizing a Mechanically Tunable Metasurface

Terahertz (THz) wave manipulation, especially the beam deflection, plays an essential role in various applications, such as next-generation communication, space exploration, and high-resolution imaging. Current THz optical components and devices are hampered by their large bulk sizes and passive responses, limiting the development of high-performance, miniaturized THz microsystems. Tunable metasurfaces offer a powerful dynamic optical platform for controlling the propagation of electromagnetic waves. In this article, we presented a mechanically tunable metasurface (MTM), which can achieve terahertz beam deflection and vary the intensity of the anomalous reflected terahertz wave by changing the air gap between the metallic resonator (MR) array with phase discontinuities and Au ground plane. The absence of lossy spacer materials substantially enhances deflection efficiency. The device was fabricated by a combination of the surface and bulk-micromachining processes. The THz beam steering capability was characterized using terahertz time domain spectroscopy. When the air gap is 50 μm, the maximum deflection coefficient reaches 0.60 at 0.61 THz with a deflection angle of ~44.5°, consistent with theoretical predictions. We further established an electrically tunable miniaturized THz device for dynamic beam steering by introducing a micro voice coil motor to control the air gap continuously. It is shown that our designed MTM demonstrates a high modulation depth of deflection coefficient (~ 62.5%) in the target steered angle at the operating frequency. Our results showcase the potential of the proposed MTM as a platform for high-efficiency THz beam manipulation.
