Nanomedicine Disrupts Stromal Barriers to Augment Drug Penetration for Improved Cancer Therapy

Tumor stroma composing diverse extracellular matrixes(ECM)and stromal cells shapes a condensed physical barrier,which severely hampers the efficient accessibility of nanomedicine to tumor cells,especially these deep-seated in the core of *** barrier significantly compromises the antitumor effects of drug-loaded nanomedicine,revealing the remarkable importance of disrupting stromal barrier for improved tumor therapy with deep penetration *** achieve this goal,various nanoparticle-based strategies have been developed,including direct depleting ECM components via delivering anti-fibrotic agents or targeting stromal cells to suppress ECM expression,dynamic regulation of nanoparticles’physicochemical properties(i.e.,size,surface charge,and morphology),mechanical force-driven deep penetration,natural/biomimetic self-driven nanomedicine,and transcytosis-inducing *** these nanostrategies were systemically summarized in this review,and the design principles for obtaining admirable nanomedicine were *** the rapid development of nanotechnology,elaborate design of multifunctional nanomedicine provides new opportunities for overcoming the critical stromal barriers to maximize the therapeutic index of various therapies,such as chemotherapy,photodynamic therapy,and immunotherapy.

Objective Hepatic stellate cells(HSCs)play a crucial role in liver ***-stage liver fibrosis is reversible and intimately associated with the state of ***-like factor 4(KLF4)plays a pivotal role in a wide array of physiological and pathological *** study aimed to investigate the effect of KLF4 on the proliferation,apoptosis and phenotype of quiescent HSCs Methods We designed a KLF4 lentiviral vector and a KLF4 siRNA lentiviral vector,to upregulate and silence KLF4 expression in human HSC LX-2 cells via *** proliferation was assessed using the CCK-8 *** cytometry was used to detect the cell cycle distribution and apoptosis *** blotting was used to determine the levels of some quiescence and activation markers of HSCs Results Overexpression of KLF4 significantly increased the levels of E-cadherin and ZO-1,which are quiescent HSC markers,while significantly decreased the levels of N-cadherin and a-SMA,known activated HSC *** contrast,cell proliferation and apoptosis rates were elevated in LX-2 cells in which KLF4 expression was silenced Conclusion KLF4 inhibits the proliferation and activation of human LX-2 *** might be a key regulatory protein in the maintenance of HSC quiescence and may serve as a target for the inhibition of hepatic fibrosis.

Prevalence and Risk Factors of Stroke in Inpatients with Type 2 Diabetes Mellitus in China

Objective:The prevalence and the cluster characteristics of risk factors of stroke were assessed in a Chinese diabetic ***:Clinical data of 30 693 inpatients who were diagnosed with type 2 diabetes mellitus(T2DM)and admitted between 2013 and 2018 were retrospectively *** age-standardized prevalence of stroke was estimated using the 2010 Chinese population census data,and risk factors were analyzed by multiple imputation and ***:The crude and standardized prevalence rates of stroke in patients with T2DM were 34.4％and 21.5％,respectively,and 85.2％of the stroke patients had ischemic *** half of the patients who experienced stroke had clusters of more than 4 risk *** with no-risk-factor clustering,the risk of stroke significantly increased 3-4 times in the presence of more than 4 risk-factor clusters(P＜0.001).Hypertension was the most common major risk factor for ischemic stroke[odds ratio(OR),2.34;95％confidence interval(CI),2.18-2.50]and hemorrhagic stroke(OR,3.68;95％CI 2.95-4.59;P＜0.001).Moreover,a 1-standard-deviation increase in fasting blood glucose(FBG)was significantly negatively correlated with ischemic stroke risk,and the same change in FBG was significantly associated with an 8％increased risk of hemorrhagic ***:The prevalence of stroke in patients with T2DM is rather high,and the clustering of risk factors is associated with the development of stroke in T2DM *** factors differ in different stroke *** risk factors for a specific high-risk group is necessary.

Disturbance Observer-based Pointing Control of Leighton Chajnantor Telescope

Leighton Chajnantor Telescope(LCT), i.e., the former Caltech Submillimeter Observatory telescope, will be refurbished at the new site in Chajnantor Plateau, Chile in 2023. The environment of LCT will change significantly after its relocation, and the telescope will be exposed to large wind disturbances directly because its enclosure will be completely open during observation. The wind disturbance is expected to be a challenge for LCT's pointing control since the existing control method cannot reject this disturbance very well. Therefore, it is very necessary to develop a new pointing control method with good capability of disturbance rejection. In this research, a disturbance observer—based composite position controller(DOB-CPC) is designed, in which an H∞feedback controller is employed to compress the disturbance, and a feedforward linear quadratic regulator is employed to compensate the disturbance precisely based on the estimated disturbance signal. Moreover, a controller switching policy is adopted, which applies the proportional controller to the transient process to achieve a quick response and applies the DOB-CPC to the steady state to achieve a small position error. Numerical experiments are conducted to verify the good performance of the proposed pointing controller(i.e., DOB-CPC) for rejecting the disturbance acting on LCT.

Numerical Study of Water Transport and Operating Strategy for Fuel Cells with Segmented Water Management Flow Fields

The effective cathode flow field design can realize the internal water balance and higher current density output of proton exchange membrane fuel cells(PEMFC).Therefore,a segmented water management flow field is proposed in this study,i.e.a half separated-half coupled cathode(HSHC)flow field which has two inlets but just one outlet.A 3D numerical PEMFC model is applied to study the effect of the HSHC flow field on PEMFC performance and its operating strategy in terms of operating *** study results are shown as follows:Compared with the two conventional cathode flow fields,the HSHC flow field improves the water balance along the channel and increases the current density by 17.1%at a cathode stoichiometry of *** is because the HSHC flow field can overcome the water loss at channels upstream and the water accumulation at channels *** draw water phenomenon(DWP)in the HSHC flow field is observed,which is mainly affected by the water vapor pressure of *** on the DWP,cooling channel inlet flow rates can be used to adjust water balance,but severe water loss should be *** addition,the inlet temperature control in HSHC flow field should be that cell temperature>cathode channel inlet temperature>cooling channel inlet temperature>ambient temperatures for better water balance.
