c-Jun N-terminal kinase is required for thermotherapyinduced apoptosis in human gastric cancer cells

AIM:To investigate the role of c-Jun N-terminal kinase(JNK) in thermotherapy-induced apoptosis in human gastric cancer SGC-7901 ***:Human gastric cancer SGC-7901 cells were cultured in *** thermotherapy at 43 ℃ for 0,0.5,1,2 or 3 h,the cells were cultured for a further 24 h with or without the JNK specific inhibitor,SP600125 for 2 *** was evaluated by immunohistochemistry [terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)] and flow cytometry(Annexin vs propidium iodide).Cell proliferation was determined by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium *** production of p-JNK,Bcl-2,Bax and caspase-3 proteins was evaluated by Western *** expression of JNK at mRNA level was determined by reverse transcription polymerase chain ***:The proliferation of gastric carcinoma SGC-7901 cells was significantly inhibited following thermotherapy,and was 32.7%,30.6%,43.8% and 52.9% at 0.5,1,2 and 3 h post-thermotherapy,*** cytometry analysis revealed an increased population of SGC790l cells in G0/G1 phase,but a reduced population in S phase following thermotherapy for 1 or 2 h,compared to untreated cells(P < 0.05).The increased number of SGC-790l cells in G0/G1 phase was consistent with induced apoptosis(flow cytometry) following thermotherapy for 0.5,1,2 or 3 h,compared to the untreated group(46.5% ± 0.23%,39.9% ± 0.53%,56.6% ± 0.35% and 50.4% ± 0.29% vs 7.3% ± 0.10%,P < 0.01),*** was supported by the TUNEL assay(48.2% ± 0.4%,40.1% ± 0.2%,61.2% ± 0.29% and 52.0% ± 0.42% vs 12.2% ± 0.22%,P < 0.01) *** importantly,the expression of p-JNK protein and JNK mRNA levels were significantly higher at 0.5 h than at 0 h post-treatment(P < 0.01),and peaked at 2 h.A similar pattern was detected for Bax and caspase-3 ***-2 increased at 0.5 h,peaked at 1 h,and then ***,the JNK specific inhibitor,SP600125,suppressed p-JNK,Bax and caspase-3 at th

Sensitivity of forest phenology in China varies with proximity to forest edges

Background:Shifts in forest phenological events serve as strong indicators of climate ***,the sensitivity of phenology events to climate change in relation to forest origins has received limited ***,it is unknown whether forest phenology changes with the proximity to forest ***:This study examined the green-up dates,dormancy dates,time-integrated NDVI(LiNDVI,a measure of vegetation productivity in growing season),and their sensitivities to climatic factors along the gradients of distance(***)to forest edge(0–2 km)in China's natural forests(NF)and planted forests(PF).For the analysis,field-surveyed data were integrated with Moderate Resolution Imaging Spectroradiometer(MODIS)NDVI from 2000 to ***:Our results reveal that PF had earlier green-up dates,later dormancy dates,and higher LiNDVI than ***,green-up sensitivities to temperature were higher at the edges of NF,whereas no such pattern was observed in ***,the sensitivity of dormancy dates remains relatively stable from the inner to the edge of both NF and PF,except for a quadratic change in dormancy date sensitivity to precipitation found in ***,we found that the green-up sensitivity to temperature increased with decreasing proximity to edge in NF evergreen forests,while it showed the opposite trend in PF evergreen ***,we observed that the precipitation impact on green-up dates shifts from postponing to advancing from the inner to the edge of NF,whereas precipitation dominantly postpones PF's green-up dates regardless of the proximity to *** LiNDVI exhibits higher sensitivity to precipitation at the edge areas,a phenomenon observed in NF but not in ***:These results suggest that the responses of forests to climate change vary with the distance to the *** increasing edge forests,which results from fragmentation caused by global changes,we anticipate that desynchronized phenological events along th

Single-Nucleotide Resolution Mapping of the Gossypium raimondii Transcriptome Reveals a New Mechanism for Alternative Splicing of Introns

Alternative splicing （AS） is a vital genetic mechanism that enhances the diversity of eukaryotic transcriptomes. Here, we generated 8.3 Gb high-quality RNA-sequencing data from cotton （Gossypium raimondii） and performed a systematic, comparative analysis of AS events. We mapped 85% of the RNA-sequencing data onto the reference genome and identified 154 368 splice junctions with 16 437 as events in 10197 genes. I ntron retention constituted the majority （40%） of all AS events in G. raimondii. Comparison across 11 eukaryote species showed that intron retention is the most common AS type in higher plants. Although transposable elements （TEs） were found in only 2.9% of all G. raimondii introns, they are present in 43% of the retained introns, suggesting that TE-insertion may be an important mechanism for intron retention during AS. The majority of the TE insertions are concentrated 0-40 nt upstream of the 3＇-splice site, substantially altering the distribution of branch points from preferred positions and reducing the efficiency of intron splicing by decreasing RNA secondary structure flexibility. Our data suggest that TE-insertion-induced changes in branch point-site distribution are important for intron retention-type AS. Our findings may help explain the vast differences in intron-retention frequencies between vertebrates and higher plants.

Predicting Chaotic Time Series Using Recurrent Neural Network

A new proposed method, i.e. the recurrent neural network (RNN), is introduced to predict chaotic time series. The effectiveness of using RNN for making one-step and multi-step predictions is tested based on remarkable few datum points by computer-generated chaotic time series. Numerical results show that the RNN proposed here is a very powerful tool for making prediction of chaotic time series.

Exact Calculations of Vertex ^-sγb and ,^-sZb in the Unitary Gauge

In this paper, we present the exact calculations for the vertex ^-sγb and ^sZb in the unitary gauge. We find that （a） the divergent- and μ-dependent terms are left in the effective vertex function Г^γμ（p, k） for b → sγ transition even after we sum up the contributions from four related Feynman diagrams; （b） for an on-shell photon, such terms do not contribute et al.; （c） for off-shell photon, these terms will be canceled when the contributions from both vertex ^sγb and ^sZb are taken into account simultaneously, and therefore the finite and gauge-independent function Zo（xt） = Co（xt）＋ Do（xt）/4, which governs the semi-leptonic decay b → sl^- l^＋, is derived in the unitary gauge.

Predicting Hyper-Chaotic Time Series Using Adaptive Higher-Order Nonlinear Filter

A newly proposed method,*** adaptive higher-order nonlinear finite impulse response(HONFIR)filter based on higher-order sparse Volterra series expansions,is introduced to predict hyper-chaotic time *** effectiveness of using adaptive HONFIR filter for making one-step and multi-step predictions is tested based on very few data points by computer-generated hyper-chaotic time series including Mackey-Glass equation and 4-dimensional nonlinear dynamical system.A comparison is made with some neural networks for predicting the Mackey-Glass hyper-chaotic time *** simulation results show that the adaptive HONFIR filter proposed here is a very powerful tool for making prediction of hyper-chaotic time series.

Quasi-birth and death processes with block tridiagonal matrices find many applications in various areas. Neuts gave the necessary and sufficient conditions for the ordinary ergodicity and found an expression of the stationary distribution for a class of quasi-birth and death processes. In this paper we obtain the explicit necessary and sufficient conditions for/-ergodicity and geometric ergodicity for the class of quasi-birth and death processes, and prove that they are not strongly ergodic. Cretaceous magmatic arc in Hainan and the peri-South China Sea asevidenced by geochemical fingerprinting of granitoids in the region

Mesozoic magmatic rocks occur widely in the South China Block and are generally interpreted as the manifestations of the subduction of the Paleo-Pacific oceanic lithosphere beneath *** magmatism in southeast(SE)China continued from the Late Permian through the Late Cretaceous with an inferred lull between 125 Ma and 115 Ma that is known in the literature as the Cretaceous"magmatic quiescence".We report in-situ zircon U-Pb ages,Hf-O and whole-rock Sr-Nd isotopes,and whole-rock geochemistry of Cretaceous granitoids on Hainan Island and discuss their magmatic evolution within the framework of the Late Mesozoic geodynamics of SE *** recognize two main stages of the emplacement of Cretaceous granitoids on Hainan,first around 120 Ma and then around 100-95 Ma,displaying high-K calc-alkaline,I-type geochemical *** in both age groups are enriched in LILE and LREE,but depleted in Nb,Ta,Ba,Sr,and *** 120 Ma granites have zircon ε_(Hf)(t)values of-2.6 to 2.3 corresponding to Hf crustal model ages,ranging from 0.79 Ga to 1.03 Ga,and δ^(18)O values ranging from 6.9‰to 7.7‰.Zircons from 100-95 Ma granites have ε_(Hf)(t)values of-4.2 to 1.1 corresponding to Hf crustal model ages of 1.08 Ga to 1.42 Ga,and δ^(18)O values ranging from 6.7‰to 8.4‰.Increasing ε_(Hf)(t)values of the Cretaceous Hainan granites with younger crystallization ages indicate addition of more juvenile components and reworking of crustal material into their melt *** ε_(Nd)(t)values of the 120 Ma and 100-95 Ma granitoids range between-4.1 to-0.4 and-7.7 to-4.0,*** calculated two-stage model age of the 100-95 Ma granitoids clusters between 1.25 Ga and 1.53 *** isotopic data suggest that magmas of the Cretaceous granitoids were produced by partial melting of Mesoproterozoic metabasaltic rocks,which make up much of the crystalline basement of the southern Cathaysia *** geochemical and isotopic characteristics of the Cretaceous granitoids on H

Atomic Decomposition and Boundedness Criterion of Operators on Multi-parameter Hardy Spaces of Homogeneous Type

The main purpose of this paper is to derive a new （p,q）-atomic decomposition on the multi-parameter Hardy space HP（X1 × X2） for 0 〈 po 〈 P ≤ 1 for some po and all 1 〈 q 〈 ∞, where X1 ×X2 is the product of two spaces of homogeneous type in the sense of Coifman and Weiss. This decomposition converges in both L^q（X1 × X2）（for 1 〈 q 〈 ∞） and Hardy space HP（X1× X2）（for 0 〈 p _〈 1）. As an application, we prove that an operator T, which is bounded on Lq（X1× X2） for some 1 〈 q 〈 ∞, is bounded from H^p（X1 × X2） to L^p（X1 × X2） if and only if T is bounded uniformly on all （p, q）-product atoms in LP（X1 × X2）. The similar boundedness criterion from HP（X1 × X2） to HP（X1 × X2） is also obtained.
