Exosome-Transmitted miR-224-5p Promotes Colorectal Cancer Cell Proliferation via Targeting ULK2 in p53-Dependent Manner

Objective To investigate the role and molecular mechanism of exosomal miR-224-5p in colorectal cancer(CRC).Methods The miR-224-5p expression in CRC patient tissues and cell-derived exosomes was measured by laser capture microdissection and qRT-PCR,***-luciferase reporter gene assay was used to determine the target gene of *** protein expressions of p53 and unc-51 like kinase 2(ULK2)in CRC cells were detected by western *** cytometry was used to detect cell cycle and *** proliferation was measured by CCK8 and EdU *** The miR-224-5p expression was upregulated in CRC tissues and increased progressively with the rise of CRC *** cells secreted extracellular miR-224-5p mainly in an exosome-dependent manner,and then miR-224-5p could be transferred to surrounding tumor cells to regulate cell proliferation in the form of autocrine or ***,ULK2 was characterized as a direct target of miR-224-5p and was downregulated in CRC ***,ULK2 inhibited CRC cell proliferation in a p53-dependent ***,exosome-derived miR-224-5p partially reversed the proliferation regulation of ULK2 on CRC *** Our findings demonstrate that exosome-transmitted miR-224-5p promotes p53-dependent cell proliferation by targeting ULK2 in CRC,which may offer promising targets for CRC prevention and therapy.

Organochlorine pesticides in soils from the Issyk-Kul region in the western Tian Shan Mountains,Kyrgyzstan：Implication for spatial distribution,source apportionment and ecological risk assessment

As organchlorine pesticides(OCPs) may be an ecologic threat to mountain environments due to their tendency to deposit and accumulate in mountain regions undergoing long-range air transport, OCPs were analyzed in soils collected from an intermontane basin of the western Tian Shan Mountains, which is the UNESCO protected natural reserve of Issyk-Kul. Total OCP concentrations in the Issyk-Kul region ranged from 4.63 to 414 ng/g dw, of which two extraordinary high OCP concentrations(414 ng/g dw and 213 ng/g dw, respectively) influenced by an abandoned dumping site and urban sewage, respectively, were found. Principal component analysis(PCA) and correlation analysis inferred that the OCP inputs in the east of the Issyk-Kul region were mainly from local endogenous sources, and exogenous input via LRAT processes were prominent in the west and south. Additionally, the isomeric and parent substance/metabolite ratios revealed most pesticides accumulated in this region were from old usage, while DDTs had fresh input because of possibly illegal regional application and a slow degradation from the dumping site. Furthermore, ecological risk assessment revealed that no frequently adverse ecological effects were observed in the Issyk-Kul region, but potential risks on neighbouring organisms induced by p,p'-DDT and γ-HCH in dumping site and urban sewage should be considered when devising an efficient management plan to prevent secondary pollution.

Effect of Processing Condition on Texture and Drawability of a Ferritic Rolled and Annealed Interstitial-Free Steel

The processing conditions of the texture formation and deep drawability of a Ti-IF steel strip hot-rolled in ferritic region and subsequently annealed were investigated. The r-value increases with the decrease of reheating temperature, and finish rolling temperature and the increase of reductions in ferritic region. For lubricated ferritic rolling and annealing, the r-value is raised up to 1.75, and elongation rate is over 50% at the finish rolling temperature of 650 ℃, which is suitable for DDQ grade products. However, the r-value is below 1.0 in the case of unlubricated rolling. The X-ray diffraction was used to analyze the textural characteristic of samples. For samples subjected to lubricated rolling and annealing, the strong { 111 }//ND recrystallization texture is distributed homogeneously along the thickness direction, and the intensity of { 110} recrystallization texture is very low even in surface. However, for unlubricated samples, the {111} texture is distributed inhomogeneously and is weak along the thickness direction, and （110}//ND recrystallization texture is strong, which deteriorates the formability.

Analysis of Process Control Model of Plate MAS Rolling

Using the identification quantity to identify the long ear disfigurement was brought up, and by calculation, it can be found that identification quantity should be set as the quantity of long ear disfigurement of the position which is about 0. 2 times of plate width to the plate border. Conception of plate head tiny cell was brought up, and the long ear disfigurement was uncoupled into the product of elongation percentage and length of tiny cell. On the basis of prediction of long ear disfigurement, process control model of MAS rolling was built up, and application indicates that it can fit the on-line production and eliminate the long ear disfigurement commendably.

Derivation of Simplified Models of Plan View Pattern Control Function for Plate Mill

The plan view pattern control theoretical models were simplified. Under the condition of constant volume for the plan view pattern compensation, the relation between the thickness and the length can be simplified to the linearity in the segment for the plan view pattern control function. The compensation volume can be dispersed for easy calculation. By comparing the model calculation result with the actual result, it was concluded that the simplified model can be used for the online control process.

Calculation of Taper Rolling Time in Plan View Pattern Control Process

The forward slip model with adhesion was used to derive the formula of calculating taper rolling time. The relation between the rolling time and the taper length and the relation between the rolling time and the taper thickness can be obtained. The numerical solution for this formula was used on site. According to the simulation result, the roll gap value should be changed linearly with rolling time.

Study on the signalling pathway of inhibitory effect of adreno-medullin on the growth of cultured glomerular mesangial cells

Background Adrenomedullin （ADM）, a potent hypotensive small peptide, was recently found to inhibit the proliferation of glomerular mesangial cells （ MsC ） in vitro and to attenuate glomerular lesions in vivo, however the mechanisms remain poorly understood. In this study, we attempted to elucidate them using molecular signal transduetion. Methods Cultured rat MsC were treated with ADM and several inhibitors of signalling molecules. Methyl thiazoleterazolium （MTT） assay and BrdU incorporation method were employed for examining MsC proliferation. Western blot analysis was used for detecting total mitogen activated protein kinases （t-MAPKs） and phosphorylated MAPKs （p-MAPKs） proteins. Results ADM suppressed MsC proliferation in a concentration- and time-dependent fashion. This response was inhibited by ADM receptor antagonist CGRP8-37 and a potent protein kinase-A （PKA） inhibitor, H89. Forskolin, a direct adenylate cyclase activator, also significantly inhibited MsC proliferation. SB203580, a P38MAPK inhibitor, and U0126, a MEK inhibitor, both completely blocked ADM mediated responses in MsC. However, cureumin, a SAPK/JNK inhibitor, and GF109203X, a potent protein kinase-C （PKC） inhibitor, had no effect on MsC growth. Western blot analysis showed that ADM did not change the expression of t-MAPKs but increased p-SAPK/JNK and p-P38MAPK levels and decreased p-ERK level. These responses were inhibited by CGRP8-37. All these kinase phosphorylations, except for the increase in p-SAPK/JNK, could be stimulated using forskolin. In addition, only ADM mediated changes in ERK and P38MAPK phosphorylations were inhibited by H89. GF109203X did not affect ADM induced changes in three p-MAPKs expressions. Conclusions ADM inhibits MsC proliferation possibly through cAMP-PKA pathway. Both phosphorylations of ERK and P38MAPK pathways were necessary in mediating the antiproliferative response of ADM.
