Background Histone deacetylase （HDAC） inhibitors are a group of small chemical molecules that inhibit histone deacetylase. At cell level, HDAC inhibitors have multiple biological effects such as cell cycle arrest, apoptosis, cell differentiation and auotophagy. At molecular level, HDAC inhibitors cause histone and nonhistone acetylation and induce gene expression. HDAC inhibitors are widely used in cancer therapy because of its function of inducing apoptosis. However, the mechanisms of apoptosis effect are not fully understood. TSA is a classical HDAC inhibitor and widely used in epigenetic and anti-cancer research. In this study, we selected Trichostatin A （TSA） to investigate the mechanisms of HDAC inhibitors apoptotic effect on cancer cells. Methods Cervical cancer cell lines such as Hela, Caski and {1. human keratinocyte line HaCaT were treated with various concentrations of TSA. Crystal violent assay and 3-（4,5-Dimethylthiazol-2.yl）-2.5-diphenyltetrazolium bromide （MTT） assay were performed to determine cell number. PARP cleavage and FITC-AnexinV were performed to determine apoptosis. DNA-methyltransferase （DNMT）I, DNMT3A and DNMT3B were determined by regular PCR, qPCR and Western Blotting. Small interfering RNA （SiRNAi） was used to knock down DNMT3B. Results HDAC inhibitors only induce cervical cancer cell apoptosis. At 1.Iumol/L of TSA, 86% of Hela cell and 76% of Caski went apoptosis. For {1. cells, HDAC inhibitors have no cytotoxic effect at therapeutic dosage, （90.0＋8.4）% of {1. cell survive after treated with 1.IJmol/L of TSA. We compared 1.pmol/L group with untreated control with t-test. There was no significance between 1.pmol/L group and untreated control for {1. cell （P 〉0.05）. HDAC inhibitors decreased DNMT3B in cancer cell but not in {1. cell. Manually knock-down of DNMT3B induced Hela and Caski cell apoptosis. PIG3 (p53-inducible gene 3), originally identified as one of a set of genes induced by p53 before the onset of apoptosis, was assumed to contribute to early cellular response to DNA damage. Here, we studied the relation between p53 status and the increased expression of PIG3 by ionizing radiation (IR), and the related clues regarding the involvement of PIG3 in the cellular response to IR-induced DNA damage signaling. We demonstrated that the pentanucleotide microsatellite sequence was responsible for the p53-dependent induction of PIG3 transcription after irradiation, while sequence upstream of PIG3 promoter could maintain the basal level of expression which was not inducible by irradiation. The interaction of PIG3 and the KRAB-ZFP-associated protein 1.(KAP1., a DNA damage response protein, was revealed. PIG3 nucleus foci were formed 1. min after γ-ray irradiation, and which were found to partially colocalize with the phospho-KAP-1.foci as well as γ-H2.X foci. Although the lac operator tagged EGFP based reporter system revealed that PIG3 does not remodel chromatin in large scale in the cells under normal growing condition, it indeed prompted the chromatin relaxation in the cellular response to DNA damage signaling. All these data suggest that PIG3 is involved in IR-induced DNA damage response, and which maybe partially attribute to its interaction with KAP1.

Background:In the face of continued degradation and loss of wetlands in the Yangtze River floodplain(YRF),there is an urgent need to monitor the abundance and distribution of wintering *** understand fully observed annual changes,we need to monitor demographic rates to understand factors affecting global population *** reproduction success contributes to dynamic changes in population size and age structure,so an assessment of the juvenile ratio(*** winter birds as a proportion of total number aged)of overwintering waterbirds can be an important indicator of the reproductive success in the preceding breeding ***:During 2.1.-2.1.,we sampled juvenile ratios among 1. key waterbird species from the wetlands in the *** on these data,we here attempt to establish a simple,efficient,focused and reliable juvenile ratio monitoring scheme,to assess consistently and accurately relative annual breeding success and its contribution to the age structure among these waterbird ***:We compared juvenile ratio data collected throughout the winter and found that the optimal time for undertaking these samples was in the early stages of arrival for migratory waterbirds reaching their wintering area(early to mid-December).We recommend counting consistently at key points(*** where>1.biogeographical flyway population were counted)at sites of major flyway importance(Poyang Lake,East Dongting Lake,Shengjin Lake,Caizi Lake,Longgan Lake and Chen Lake).Based on this,the error rate of the programme(1.5 planned points,the count of 1. waterbird species is 82.-8955)is less than 5%.Conclusions:We established a juvenile ratio monitoring programme for 1. key waterbird species in the wetlands of the YRF,and discuss the feasibility and necessity of implementing such a future programme,and how to use these data in our monitoring and understanding of the population dynamics of these waterbird populations.

BACKGROUND： Subacute combined degeneration of the spinal cord is caused by vitamin B12.deficiency and is a kind of degenerative disease owing the characteristics of nervous system diseases. In addition, different patients have variously clinical manifestations and various prognoses after vitamin B12.therapy. OBJECTIVE： To investigate and analyze diagnosis, clinical manifestations and prognosis of subacute combined degeneration of the spinal cord. DESIGN： Case analysis. SETTING： department of Neurology, the Third Hospital of Peking university. PARTICIPANTS： A total of 2. subacute combined degeneration of the spinal cord patients including 14 males and 7 females aged from 33 to 82.years were selected from department of Neurology, the Third Hospital of Peking university from January 1999 to December 2.05. Duration from onset to final diagnosis lasted for 1.5 - 108 months. All patients had typically clinical manifestations; meanwhile, level of serum vitamin B12.was decreased and/or vitamin B12.therapy was effective. All patients provided the confirmed consent. METHODS： Clinical data of 2. subacute combined degeneration of the spinal cord patients were retrospectively analyzed, while general data and clinical characteristics were recorded at the same time. Levels of blood routine, serum vitamin B12.and homocysteine were measured at the phase of hospitalization. normal value of serum vitamin B12.was 187 - 1 059 ng/L and normal value of serum homocysteine was 5 - 15μ mol/L. All patients received neuroelectrophysiological examination and 15 patients received MRI examinations of spinal cord. After final diagnosis, patients were given vitamin B12.therapy. And follow-up was performed to investigate the prognosis. MAIN OUTCOME MEASURES：（1） Levels of blood routine, serum vitamin B12.and homocysteine; （2. results of neuroelectrophysiological examination; （3） results of MRI examination of spinal cord; （4） prognosis. Dear Editor,Rice dwarf virus(RDV),a member of the Phytoreoviurs genus,is transmitted to rice(Oryza sativa)plants by leaf-hopper(Nephotettix cincticeps)in a propagative *** by RDV results in severe stunting growth pheno-types and a dramatic reduction in grain *** genome of RDV is composed of 12.segmented double-stranded RNAs(S1-S12.based on their migration rates in agarose gel electro-phoresis).The 12.segments encode seven structural proteins(P1,P2.P3,P5,P7,P8,and P9 as the products of Sl,S2.S3,S5,S7,S8,and S9,respectively)and five nonstructural proteins(Pns4,Pns6,Pnsl0,Pns11,and Pns12.as the products of S4,S6,S10,S11,and S12.respectively).The outer capsid protein P2.is essential for RDV infection of insects and thus influences transmission of RDV by the insect vector(Omura et al.,1998;Zhou et al.,2.07).P2.also contributes to the dwarf phenotype of infected rice by interfering with gibberellic acid synthesis(Zhu et al.,2.05).When RDV-infected rice plants were main-tained via vegetative propagation for several years without insect transmission,they regained normal growth height due to loss of RDV P2.and Pnsl0 proteins(Pu et al.,2.11).

Nucleic acids are natural biopolymers of nucleotides that store, encode, transmit and express genetic information, which play central roles in diverse cellular events and diseases in living things. The analysis of nucleic acids and nucleic acids-based analysis have been widely applied in biological studies, clinical diagnosis, environmental analysis, food safety and forensic *** the past decades, the field of nucleic acids analysis has been rapidly advancing with many technological *** this review, we focus on the methods developed for analyzing nucleic acids, nucleic acids-based analysis, device for nucleic acids analysis, and applications of nucleic acids analysis. The representative strategies for the development of new nucleic acids analysis in this field are summarized, and key advantages and possible limitations are discussed. Finally, a brief perspective on existing challenges and further research development is provided.
