The INO80 chromatin remodeling complex promotes thermo-morphogenesis by connecting H2A.Z eviction and active transcription in Arabidopsis.

Global warming imposes a major threat to plant growth and crop production. In some plants including Arabidopsis thaliana, elevated temperatures induce a series of morphological and developmental adjustments, termed thermomorphogenesis to facilitate plant cooling under high-temperature conditions. Plant thermal response is suppressed by histone variant H2A.Z. At warm temperatures, H2A.Z is evicted from nucleosomes at thermo-responsive genes, resulting in their expression changes. However, the mechanisms that regulate H2A.Z eviction and subsequent transcription changes are largely unknown. Here, we show that the INO80 chromatin remodeling complex (INO80-C) promotes thermomorphogenesis and activates the expression of thermo-responsive and auxin-related genes. INO80-C associates with PHYTOCHROME-INTERACTING FACTOR 4 (PIF4), a potent regulator in thermomorphogenesis, and mediates temperature-induced H2A.Z eviction at PIF4 targets. Moreover, INO80-C directly interacts with COMPASS-like and transcription elongation factors to promote active histone modification Histone H3 lysine 4 trimethylation (H3K4me3) and RNA Polymerase II (RNA Pol II) elongation, leading to the thermal induction of transcription. Notably, transcription elongation factors SPT4 and SPT5 are required for the H2A.Z eviction at PIF4 targets, suggesting the cooperation of INO80-C and transcription elongation in H2A.Z removal. Our results demonstrate that the (PIF4)-(INO80-C)-(COMPASS-like)-(transcription elongator) module controls plant thermal response, and establish a link between H2A.Z eviction and active transcription.

Effects of water-salt coordinated regulation technique on ion distribution and antioxidant enzyme activities in tomato

In order to explore the effects of brackish water combined with regulated deficit irrigation of different growth periods,different levels of water,and salt synergistic stress on the characteristics of ion absorption,distribution,transport,and antioxidant enzyme activity of tomatoes,a greenhouse pot experiment was *** influencing factors were set as:irrigation water salinity(local shallow groundwater S01.1 g/L,S12.0 g/L,S24.0 g/L),different water deficit levels(W170%-80%field capacity(FC),W260%-70%FC,W350%-60%FC),and three growth stages(T1 seedling stage,T2 blossoming and bearing fruits stage,T3 mature picking stage).Monitoring and analyzing the dynamic changes of K^(+)and Na^(+)content in tomato root stems and leaves under water-salt stress and antioxidant enzyme activities of leaves at the seedling *** results showed the coordinated regulation of water and salt can significantly change the ion absorption of roots and the transfer between stems and leaves at different growth *** roots and leaves of tomatoes mainly accumulated K^(+),and the stems mainly accumulate Na^(+).The increase of irrigation salinity can reduce the ratio of K^(+)/Na^(+)ratio in *** the coordinated regulation of water and salt,K^(+)became the main osmotic adjustment ion of the leaves *** activities of catalase,superoxide dismutase,and peroxidase in tomato leaves increased with the increase of water-salt *** results have great significance to the response of crops and to meet the requirements for water quality and quantity of brackish water under water-salt stress at different growth stages of tomatoes.

A warm temperature-released negative feedback loop fine-tunes PIF4-mediated thermomorphogenesis in Arabidopsis

Plants can sense temperature changes and adjust their growth *** Arabidopsis,high ambient temperatures stimulate stem elongation by activating a key thermoresponsive regulator,PHYTOCHROME INTERACTING FACTOR 4(PIF4).Here,we show that warmth promotes the nighttime transcription of GI,which is necessary for the high temperature-induced transcription of *** analyses suggest that GI prevents excessive thermoresponsive growth by inhibiting PIF4,with this regulatory mechanism be-ing partially reliant on *** transcription is repressed by ELF3 and HY5,which concurrently inhibit PIF4 expression and *** elevation causes the deactivation or degradation of ELF3 and HY5,leading to PIF4 activation and relief of GI transcriptional repression at high *** allows PIF4 to further activate GI transcription in response to elevated ***,in turn,inhibits PIF4,es-tablishing a negative feedback loop thatfine-tunes PIF4 *** addition,we demonstrate that ELF3,HY5,and PIF4 regulate GI transcription by modulating the enrichment of histone variant H2A.Z at the GI ***,ourfindings suggest that thermal release of a negative feedback loopfinely adjusts plant thermomorphogenesis.

Light is one of the most essential environmental factors that tightly and precisely control various physiological and developmental processes in plants.B-box CONTAINING PROTEINs(BBXs)play central roles in the regulation of light-dependent *** this study,we report that BBX9 is a positive regulator of light ***9 interacts with the red light photoreceptor PHYTOCHROME B(phyB)and transcription factors PHYTOCHROME-INTERACTING FACTORs(PIFs).phyB promotes the stabilization of BBX9 in light,while BBX9 inhibits the transcriptional activation activity of *** turn,PIFs directly bind to the promoter of BBX9 to repress its *** the other hand,BBX9 associates with the positive regulator of light signaling,BBX21,and enhances its biochemical ***21 associates with the promoter regions of BBX9 and transcriptionally up-regulates its ***,this study unveiled that BBX9 forms a negative feedback loop with PIFs and a positive one with BBX21 to ensure that plants adapt to fluctuating light conditions.

Isoflavonoids,secondary metabolites derived from the phenylalanine pathway,are predominantly bio-synthesized in legumes,especially soybean(Glycine max).They are not only essential for plant responses to biotic and abiotic stresses but also beneficial to human *** this study,we report that light signaling controls isoflavonoid biosynthesis in ***-light photoreceptors(GmCRY1s,GmCRY2s,GmPHOT1s,and GmPHOT2s)and the transcription factors GmSTF1 and GmSTF2 promote isoflavonoid accumulation,whereas the E3 ubiquitin ligase GmCOP1b negatively regulates isoflavonoid ***1s and GmPHOT2s stabilize GmSTF1/2,whereas GmCOP1b promotes the degradation of these two proteins in ***1/2 regulate the expression of approximately 27.9%of the genes involved in soybean isoflavonoid biosynthesis,including GmPAL2.1,GmPAL2.3,and *** also repress the expression of GmBBX4,a negative regulator of isoflavonoid biosynthesis in *** addition,GmBBX4 physically interacts with GmSTF1 and GmSTF2 to inhibit their transcriptional activation activity toward target genes related to isoflavonoid ***,GmSTF1/2 and GmBBX4 form a negative feedback loop that acts downstream of photoreceptors in the regulation of isoflavonoid *** study provides novel insights into the control of isoflavonoid biosynthesis by light signaling in soybean and will contribute to the breeding of soybean cultivars with high isoflavonoid content through genetic and metabolic engineering.

The isoquinolones have caused widespread concern because of its specific structural motifs [1]. The method of the transition metal catalyzed C-H functionalization has become quite perfect and use this method can quickly form a variety of isoquinolinones. Recently, Aslam and co-workers[2] was reported a site-selective（C-4/C-8） alkynylation of isoquinolones enabled by gold and rhodium complexes. Although the Aslam group has made a plausible mechanical pathway, the key issues still need further discussion.（1） What is the detailed reaction mechanism?（2） What is the origin of site-selective C-H alkynylation of isoquinolones catalyzed by gold and rhodium catalysts?Here we would like to report our detailed density functional theory（DFT）calculations on the reaction *** expect this work would help understand the detailed mechanisms and design new related reactions.

Allylmetal species are important intermediates in organic synthesis.[1] Recently, Stamatis E. Korkis and co-workers[2] first reported allyl-to-allyl 1,4-Rh（III） migration in the oxidative C-H allylation of benzamides with 1,3-dienes which is different to the characteristic reaction we known that 1,3-transposition of the metal from one end of the allylic fragment to the *** process could serve as a valuable addition to the currently limited number of catalytic C-H functionalizations involving 1,*** a plausible mechanistic pathway has been proposed by Stamatis E. Korkis group,key issues still need to be further discussed. What is the detailed reaction mechanism? Here we would like to report our detailed density functional theory（DFT） calculations on the reaction *** expect this work would help understand the detailed mechanisms and design new related reactions.

【目的】探索协同过滤与内容过滤新的融合方法,提高个性化推荐方法的推荐准确度。【方法】提出项目热度计算方法并对Pearson相关系数算法进行改进,建立当前用户与其邻居的兴趣模型,对邻居用户进行过滤,由最终得到的可信邻居对当前用户进行推荐。【结果】在Movie Lens 1M电影评分数据集上的实验结果表明,提出的混合推荐方法推荐效度要好于现存的两种混合方法。【局限】在为用户建立兴趣模型时,项目的不同标志性特征需要人为抽取,且对于项目的标志性特征个数及其在用户的兴趣中所占的权重分配问题,不同的研究者可能会有不同的见解。【结论】本文提出的混合推荐方法可有效提高个性化推荐的准确度。
