Coxsackievirus B3(CVB3)is the pathogen causing hand,foot and mouth disease(HFMD),which manifests across a spectrum of clinical severity from mild to ***,CVB3-infected mouse models mainly demonstrate viral myocarditis and pancreatitis,failing to replicate human HFMD *** several enteroviruses have been evaluated in Syrian hamsters and rhesus monkeys,there is no comprehensive data on *** this study,we have first tested the susceptibility of Syrian hamsters to CVB3 infection via different *** results showed that Syrian hamsters were successfully infected with CVB3 by intraperitoneal injection or nasal drip,leading to nasopharyngeal colonization,acute severe pathological injury,and typical HFMD ***,the nasal drip group exhibited a longer viral excretion cycle and more severe pathological *** the subsequent study,rhesus monkeys infected with CVB3 through nasal drips also presented signs of HFMD symptoms,viral excretion,serum antibody conversion,viral nucleic acids and antigens,and the specific organ damages,particularly in the ***,there were no significant differences in myocardial enzyme levels,and the clinical symptoms resembled those often associated with common,mild *** summary,the study successfully developed severe Syrian hamsters and mild rhesus monkey models for CVB3-induced *** models could serve as a basis for understanding the disease pathogenesis,conducting pre-trial prevention and evaluation,and implementing post-exposure intervention.

Three-dimensional Model Analysis of Electric Field Excited by Multi-circuit Intersecting Overhead Transmission Lines

This work is carried out to predict the special distribution of electric field induced by multi-circuit intersecting overhead high-voltage (HV) transmission lines (TLs) within a large range without any expensive and time-consuming computation. The two main parts of the presented methodology are 1) setting up a three-dimensional (3D) model to calculate the electric field based on combining ca- tenary equations with charge simulation method and 2) calculating the hybrid electric field excited by multi-circuit intersecting TLs using coordinate transformation and superposition technique. Examples of different TLs configurations, including a 220 kV single-circuit hori- zontally configured TLs, a 500 kV single-circuit triangularly configured TLs and a combination of the 220 kV TLs and the 550 kV TLs, are illustrated to verify the validity of this methodology. A more complicatal configurations, including a 500 kV double-circuit TLs and two 220 kV single-circuit horizontally configured TLs, are also calculated. Conclusions were drawn from the simulation: 1) The presented 3D model outperforms 2D models in describing the electric field distribution generated by practical HV TLs with sag and span. 2) Coordinate trans- formation and superposition technique considerably simplify the electric field computation for multi-circuit TLs configurations, which makes it possible to deal with complex engineering problems. 3) The electric field in the area covered by multiple intersecting overhead TLs is distorted and the hybrid electric field strength in some partial region increases so sharply that it might exceed the admissible value. 4) The configuration parameters of the TLs and the spatial configuration of multi-circuit TLs, for instance, the height of TLs, the length of span and the intersection angle of multiple circuits, influence the strength and the distribution of hybrid electric field. Dear Editor,The unprecedented COVID-19 pandemic caused by SARS-CoV-2 remains ongoing,but there is a lack of fully effective *** plasma-derived hyperimmune globulins have been a safe and effective treatment but restricted by the difficulties in obtaining sufficient plasma with high antibody titers from a large number of recovered *** antibodies,particularly equine antibodies,have been widely used for decades as the therapeutics against some viral infections or as antivenoms.1 Equine antibodies could be rapidly developed and manufactured into therapeutic antibodies in large quantities under WHO standardized *** we explored the development of equine antibody-derived F(ab′)2 as an option to treat COVID-19 by targeting the receptor-binding domain(RBD)of the viral spike protein that is essential for the viral entry into the host cells.2 We observed excellent neutralization titers of the F(ab′)2 in vitro and high potency against SARS-CoV-2 infection in the nonhuman primate rhesus macaques.

Colorectal cancer has a high incidence and mortality rate in China, with the majority of cases being middle and low rectal cancer. Surgical intervention is currently the main treatment modality for locally advanced rectal cancer, with the common goal of improving oncological outcomes while preserving function. The controversy regarding the circumferential resection margin distance in rectal cancer surgery has been resolved. With the promotion of neoadjuvant therapy concepts and advancements in technology, treatment strategies have become more *** tumor downstaging, there is an increasing trend towards extending the safe distance of distal rectal margin. This provides more opportunities for patients with low rectal cancer to preserve their anal ***, there is currently no consensus on the specific distance of distal resection margin.

Coxsackievirus A10(CV-A10)is one of the etiological agents associated with hand,foot and mouth disease(HFMD)and also causes a variety of illnesses in humans,including pneumonia,and *** people,particularly young children,may have different immunological responses to *** CV-A10 infection animal models provide only a rudimentary understanding of the pathogenesis and effects of this *** characteristics of CV-A10 infection,replication,and shedding in humans remain *** this study,rhesus macaques were infected by CV-A10 via respiratory or digestive route to mimic the HFMD in *** clinical symptoms,viral shedding,inflammatory response and pathologic changes were investigated in acute infection(1–11 day post infection)and recovery period(12–180 day post infection).All infected rhesus macaques during acute infection showed obvious viremia and clinical symptoms which were comparable to those observed in *** inflammatory pathological damages were observed in multi-organs,including the lung,heart,liver,and *** the acute period,all rhesus macaques displayed clinical signs,viral shedding,normalization of serum cytokines,and increased serum neutralizing antibodies,whereas inflammatory factors caused some animals to develop severe hyperglycemia during the recovery *** addition,there were no significant differences between respiratory and digestive tract infected ***,all data presented suggest that the rhesus macaques provide the first non-human primate animal model for investigating CV-A10 pathophysiology and assessing the development of potential human therapies.

Immunogenicity and protective efficacy of a recombinant protein subunit vaccine and an inactivated vaccine against SARS-CoV-2 variants in non-human primates

Emerging SARS-CoV-2 variants and the gradually decreasing neutralizing antibodies over time post vaccination have led to an increase in incidents of breakthrough infection across the *** investigate the potential protective effect of the recombinant protein subunit COVID-19 vaccine targeting receptor-binding domain(RBD)(PS-RBD)and whole inactivated virus particle vaccine(IV)against the variant strains,in this study,rhesus macaques were immunized with PS-RBD or IV vaccine,followed by a Beta variant(B.1.351)*** neutralizing activity against the Beta variant was reduced compared with that against the prototype,the decreased viral load in both upper and lower respiratory tracts,milder pathological changes,and downregulated inflammatory cytokine levels in lung tissues after challenge demonstrated that PS-RBD and IV still provided effective protection against the Beta variant in the macaque ***,PS-RBD-induced macaque sera possessed general binding and neutralizing activity to Alpha,Beta,Delta,and Omicron variants in our study,though the neutralizing antibody(NAb)titers declined by varying degrees,demonstrating potential protection of PS-RBD against current circulating variants of concern(VOCs).Interestingly,although the IV vaccine-induced extremely low neutralizing antibody titers against the Beta variant,it still showed reduction for viral load and significantly alleviated pathological *** correlates of vaccine-induced protection(CoP)like antibody-dependent cellular cytotoxicity(ADCC)and immune memory were both confirmed to be existing in IV vaccinated group and possibly be involved in the protective mechanism.

目的采用人胚肺二倍体细胞(human embryonic lung diploid cell)KMB17株对柯萨奇病毒B族(Coxsackievirus B,CVB)1、2、3型进行适应性培养,探索应用KMB17细胞培养CVB1-3型病毒的相关参数,为基于KMB17细胞研发CVB多价疫苗提供参考。方法将CVB 1-3型分别设置8个不同的感染复数(mulitiplicity of infection, MOI)梯度(0.05、0.1、0.15、0.2、0.25、0.3、0.35、0.4),在KMB17细胞上进行培养观察,对病毒培养结果进行统计分析。结果 CVB 1-3型病毒均能KMB17细胞中复制,并使细胞产生明显的细胞病变(cytopathic effect, CPE)。其中CVB 1型在KMB17细胞上适宜的种毒MOI值为0.25~0.3,病毒收获液的平均滴度为7.25 lgCCID50/mL,适宜MOI范围内病毒培养时间的95%置信区间为96.1~97.6 h;CVB 2型在KMB17细胞上适宜的种毒MOI值为0.20~0.25,病毒收获液平均滴度为7.375 lgCCID50/mL,适宜MOI范围内病毒培养时间的95%置信区间为74.8~76.8 h。CVB 3型在KMB17细胞上适宜的种毒MOI值为0.30~0.35,病毒收获液的平均滴度为7.25 lgCCID50/mL,适宜MOI范围内病毒培养时间的95%置信区间为82.7~84.7 h。结论 KMB17为一株理想的用于CVB1、2、3型培养的基质细胞,可为基于KMB17细胞研发CVB多价疫苗提供参考。

Noroviruses are a leading viral cause of epidemic and sporadic acute gastroenteritis in humans of all ages, causing substantial illness and death. Each year, noroviruses cause ≈21 million infections in the United States and≈200,000 deaths worldwide. Among the 6 known norovirus genogroups(GⅠ-Ⅵ), all GⅠ, most GⅡ, and a few GⅣ noroviruses infect humans(humannoroviruses). Each genogroup includes up to 22 genotypes based on the sequences of major capsid protein 1(VP1). Although GⅡ.4 noroviruses were predominant globally for 2 decades, the previously rare GⅡ.17 genotype emerged during the 2014-15 epidemic season in China and other Southeast Asian countries/regions, causing major epidemics of acute gastroenteritis. Infection of domestic pigs, cattle, dogs, and rhesus macaques with human norovirus has been reported. We report the detection and characterization of norovirus GⅡ.17 that extensively and naturally infected farm-raised rhesus monkeys in southwestern China.
