Identifying the critical phosphorus balance for optimizing phosphorus input and regulating soil phosphorus effectiveness in a typical winter wheat-summer maize rotation system in North China

Phosphorus(P)is a nonrenewable resource and a critical element for plant growth that plays an important role in improving crop *** P fertilizer application is widespread in agricultural production,which not only wastes phosphate resources but also causes P accumulation and groundwater ***,we hypothesized that the apparent P balance of a crop system could be used as an indicator for identifying the critical P input in order to obtain a high yield with high phosphorus use efficiency(PUE).A 12-year field experiment with P fertilization rates of 0,45,90,135,180,and 225 kg P_(2)O_(5)ha^(-1)was conducted to determine the crop yield,PUE,and soil Olsen-P value response to P balance,and to optimize the P *** yield stagnation occurred when the P fertilizer application exceeded a certain level,and high yield and PUE levels were achieved with annual P fertilizer application rates of 90-135 kg P_(2)O_(5)ha^(-1).A critical P balance range of 2.15-4.45 kg P ha^(-1)was recommended to achieve optimum yield with minimal environmental *** critical P input range estimated from the P balance was 95.7-101 kg P_(2)O_(5)ha^(-1),which improved relative yield(>90%)and PUE(90.0-94.9%).In addition,the P input-output balance helps in assessing future changes in Olsen-P values,which increased by 4.07 mg kg^(-1)of P for every 100 kg of P ***,the P balance can be used as a critical indicator for P management in agriculture,providing a robust reference for limiting P excess and developing a more productive,efficient and environmentally friendly P fertilizer management strategy.

AIM:To observe the effects of N-acetylserotonin(NAS)administration on retinal ischemia-reperfusion(RIR)injury in rats and explore the underlying mechanisms involving the high mobility group box 1(HMGB1)/receptor for advanced glycation end-products(RAGE)/nuclear factor-kappa B(NF-κB)signaling ***:A rat model of RIR was developed by increasing the pressure of the anterior chamber of the *** male Sprague Dawley were randomly divided into five groups:sham group(n=8),RIR group(n=28),RIR+NAS group(n=28),RIR+FPS-ZM1 group(n=8)and RIR+NAS+FPS-ZM1 group(n=8).The therapeutic effects of NAS were examined by hematoxylin-eosin(H&E)staining,and retinal ganglion cells(RGCs)*** expression of interleukin 1 beta(IL-1β),HMGB1,RAGE,and nod-like receptor 3(NLRP3)proteins and the phosphorylation of nuclear factorkappa B(p-NF-κB)were analyzed by immunohistochemistry staining and Western blot *** expression of HMGB1 protein was also detected by enzyme-linked immunosorbent assay(ELISA).RESULTS:H&E staining results showed that NAS significantly reduced retinal edema and increased the number of RGCs in RIR *** NAS therapy,the HMGB1 and RAGE expression decreased significantly,and the activation of the NF-κB/NLRP3 pathway was antagonized along with the inhibition of p-NF-κB and NLRP3 protein ***,NAS exhibited an anti-inflammatory effect by reducing IL-1β*** inhibitory of RAGE binding to HMGB1 by RAGE inhibitor FPS-ZM1 led to a significant decrease of p-NF-κB and NLRP3 expression,so as to the IL-1βexpression and retinal edema,accompanied by an increase of RGCs in RIR ***:NAS may exhibit a neuroprotective effect against RIR via the HMGB1/RAGE/NF-κB signaling pathway,which may be a useful therapeutic target for retinal disease.

Partners of patients with ulcerative colitis exhibit a biologically relevant dysbiosis in fecal microbial metacommunities

AIM To investigate alterations in the fecal microbiome using 16 S r RNA amplicon sequencing in couples in the same cohabitation *** Fecal samples were collected from eight ulcerative colitis(UC) patients and their healthy partners at Lishui People's Hospital, Zhejiang Province, China. DNA was extracted and the variable regions V3 and V4 of the 16 S r RNA genes were PCR amplified using a two-step protocol. Clear reads were clustered into operational taxonomic units(OTUs) at the 97% sequence similarity level using UCLUST v1.2.22. The Wilcoxon rank-sumtest(R v3.1.2) was used to compare inter-individual differences. Differences with a P value < 0.05 were considered statistically *** Fecal microbial communities were more similar among UC patients than their healthy partners(P = 0.024). UC individuals had a lower relative abundance of bacteria belonging to the Firmicutes, especially Blautia, Clostridium, Coprococcus and Roseburia(P < 0.05). Microbiota dysbiosis was detected in UC patients and their healthy partners. Relevant genera included Akkermansiam, Bacteroides, Escherichia, Lactobacillales, Klebsiella and Parabacteroides. The enriched pathways in fecal samples of UC patients were related to lipid and nucleotide metabolism. Additionally, the pathways involved in membrane transport and metabolism of cofactors and vitamins were more abundant in the healthy *** Our results suggested that the microbial composition might be affected in healthy partners cohabiting with UC patients, especially in terms of microbiota dysbiosis.

Genetic relationships of introduced Colorado potato beetle Leptinotarsa decemlineata populations in Xinjiang, China

The Colorado potato beetle （CPB）, Leptinotarsa decemlineata （Say）, is an infamous invasive species worldwide that aggressively attacks potato and other Solanaceae crops. CPB was first found in China in 1993 and has since spread across 2.77 × 105 km2 in Xinjiang Uygur Autonomous Region. To better understand genetic variation and migration patterns, we used seven polymorphic microsatellite loci to elucidate the genetic relationships and gene flow among 10 CPB populations across Xinjiang. （i） Overall low levels of genetic diversity were detected on the entire population in Xinjiang but most of the diversity was retained among populations during invasion. （ii） The mean pairwise FST was low （0.071 ± 0.043） among populations. The genetic differentiation was little （pairwise FST 0.038 ± 0.016） between the five interior populations （Wusu, Urumqi, Jimsar, Qitai and Mulei） and Tacheng population. The six populations might come from the same genetic group via Bayesian clustering and were closely related on a neighbor-joining tree. Combining the history data, the five interior populations may have originated from Tacheng. （iii） Gene flow was frequent, especially among the five interior populations. Individuals from the interior populations could be assigned to Tacheng at higher probabilities （means 0.518 ± 0.127） than vice versa （means 0.328 ± 0.074）, suggesting that the beetle population has spread from the border to the interior in Xinjiang.

Divergence in flowering time is a major component contributing to reproductive isolation between two wild rice species(Oryza rufipogon and O. nivara)

It is of critical importance for our understanding of speciation process to determine the forms of reproductive isolation and their relative importance in species divergence. Oryza nivara and O. rufipogon are direct ancestors of Asian cultivated rice and a progenitor-daughter species pair. Investigating the reproductive isolation between them provides insights into plant speciation and helps understanding of the rice domestication. Here, we quantitatively measured the major components of reproductive isolation between the two species based on common garden and crossing experiments for three pairs of sympatric populations in Nepal, Cambodia and *** revealed significant differences in the flowering times between species pairs, with O. nivara flowering much earlier than O. rufipogon. A very weak reduction in seed set but no reduction in F1 viability and fertility were detected for the crosses between species relative to those within species. Moreover, we detected asymmetrical compatibility between species and found that emasculation significantly decreased pollination success in O. nivara but not in O. *** study demonstrates that the divergence between O. nivara and O. rufipogon is maintained almost entirely by the difference in flowering times and suggests that differential flowering times contribute to both habitat preferences and reproductive isolation between species.
