Transcriptomic analysis reveals the transcription factors involved in regulating the expression of EPSPS gene,which confers glyphosate resistance of goosegrass (Eleusine indica)

Glyphosate inhibits the enzyme 5-enolpyruvylshikimate-3-phosphate synthase(EPSPS)and overexpression of the EPSPS gene is one of the molecular mechanisms conferring glyphosate resistance in weeds.A regulatory sequence of EPSPS gene was isolated previously,and an alteration in its 5´-untranslated region(UTR)pyrimidine(Py)-rich stretch element is involved in the regulation of EPSPS expression in glyphosate-resistant(GR)Eleusine ***,the transcription factors involved in this regulatory sequence remain to be *** this study,we investigated the regulatory network of EPSPS overexpression associated genes in a GR *** population by *** differentially expressed transcript analyses revealed that glyphosate treatment caused an increase in the expression of 2752 unigenes and a decrease in the expression of 4025 unigenes in the GR ***,compared to the glyphosatesusceptible(GS)*** them,1373 unigenes were identified to be co-expressed with the EPSPS gene in GR *** and KEGG pathway analyses showed that the up-regulated unigenes were mainly enriched in chloroplasts and associated with the shikimate biosynthesis pathway,chlorophy II and peroxisome metabolism ***,the expression of a Shikimate kinase which catalyzed the conversion of Shikimate to Shikimate 3-phosphate(S3P,a substrate of EPSPS),was also *** transcription factors were identified as likely to be involved in the regulation of the EPSPS expression,and three of them(ARF2,ARF8 and BPC6)showed more binding sites because of a(CT)n insertion of the 5´-UTR Py-rich stretch element in ***,the yeast one-hybrid assay illustrated that ARF8 and BPC6 could bind to the 5´-UTR Py-rich stretch element of wild type EPSPS,but could not bind to the mutated *** data suggests that the transcriptional regulation of EPSPS expression is complex and was significantly altered in GR *** discoveries provide new references for further study of the

A microgravity environment has been shown to cause ocular damage and affect visual acuity,but the underlying mechanisms remain ***,we established an animal model of weightlessness via tail suspension to examine the pathological changes and molecular mechanisms of retinal damage under *** 4 weeks of tail suspension,there were no notable alterations in retinal function and morphology,while after 8 weeks of tail suspension,significant reductions in retinal function were observed,and the outer nuclear layer was thinner,with abundant apoptotic *** investigate the mechanism underlying the degenerative changes that occurred in the outer nuclear layer of the retina,proteomics was used to analyze differentially expressed proteins in rat retinas after 8 weeks of tail *** results showed that the expression levels of fibroblast growth factor 2(also known as basic fibroblast growth factor)and glial fibrillary acidic protein,which are closely related to Müller cell activation,were significantly *** addition,Müller cell regeneration and Müller cell gliosis were observed after 4 and 8 weeks,respectively,of simulated *** findings indicate that Müller cells play an important regulatory role in retinal outer nuclear layer degeneration during weightlessness.

The E3 ubiquitin ligase,carboxyl terminus of heat shock protein 70(Hsp70)interacting protein(CHIP),also functions as a co-chaperone and plays a crucial role in the protein quality control *** this study,we aimed to investigate the neuroprotective effect of overexpressed CHIP on Alzheimer’s *** used an adeno-associated virus vector that can cross the blood-brain barrier to mediate CHIP overexpression in APP/PS1 mouse *** overexpression significantly ameliorated the performance of APP/PS1 mice in the Morris water maze and nest building tests,reduced amyloid-βplaques,and decreased the expression of both amyloid-βand phosphorylated *** also alleviated the concentration of microglia and astrocytes around *** APP/PS1 mice of a younger age,CHIP overexpression promoted an increase in ADAM10 expression and inhibitedβ-site APP cleaving enzyme 1,insulin degrading enzyme,and neprilysin *** of HSP70 and HSP40,which have functional relevance to CHIP,were also *** nuclei transcriptome sequencing in the hippocampus of CHIP overexpressed mice showed that the lysosomal pathway and oligodendrocyte-related biological processes were up-regulated,which may also reflect a potential mechanism for the neuroprotective effect of *** research shows that CHIP effectively reduces the behavior and pathological manifestations of APP/PS1 ***,overexpression of CHIP could be a beneficial approach for the treatment of Alzheimer’s disease.

Lithium-sulfur （Li-S） battery,as a promising next-generation high-energy-density battery,suffers from lithium polysulfides（LPSs） shuttle and Li dendrites *** crystalline framework-based membranes as separator can effectively absorb and restrain LPSs but without precise structural design principal exploration and mechanism ***,bio-inspired 1D oriented lithium-ion transport channels with molecular negatively charged PO3（-1） have been developed in hybrid tetrazole frameworks for promoting the transmission of Li+*** theoretical calculations and in situ spectroscopy demonstrated the higher binding energy and inhibited diffusion of LPSs in *** assembled PP@PO3Janus separator in Li-S coin cell delivered a high initial capacity of 1410.9 mAh g-1at 0.1 C and a low attenuation rate of 0.038%per cycle over 500 cycles at 5 ***,the high capacities of 862,542 and 409 mAh g-1based on high-sulfur-loading cathodes of 2.6,3.8 and 5.0 mg cm-2at0.5 C were achieved,***,the punch battery of PO3@PP separator with S-cathode of 1.2 mg cm-2has been developed for demonstrating its potentials commercial application,which displays impressive capacity of 873.4 mAh g-1with the retention of～78.9%over 50 cycles.

Surface Nanocrystallization of Nb-Ti Stabilized 439M Ferritic Stainless Steel

A new nanocrystallization process was designed, which included punching the surface coating to generate dislocation network or cells and recovery treatment to turn the dislocation cells into nano-sized grains. The effort was made to increase the service life of Nb-Ti stabilized 439M stainless steel surface with medium passive ability, particularly to enhance the outmost surface layer by nanocrystallization for improving resistances to corrosion. Electrochemical properties, electron work functions （EWFs）, nano-mechanical properties of the nanocrystallized surfaces were investigated. It was demonstrated that the nanocrystallized 439M stainless steel surface exhibited improved resistance to corrosion in 0.5 mol/L H2 SO4 solution. However, the nanoci＇ystallized 439M stainless steel surface exhibited decreased resistance to corrosion in 3.5% NaCl solution.

Investigation of a non-explosive directional roof cutting technology for self-formed roadway

Traditional explosives have characteristics of high risk,large vibration,and poor directional ***,an instantaneous expander with a single crack surface(IESCS),which is a novel nonexplosive directional rock-breaking technique,has been *** directional roof-cutting mechanism of the IESCS method,driven by high-pressure gas,was theoretically *** experiments and numerical simulations proved the directional slitting effect of the IESCS method to be *** with shaped-charge blasting,the charge of IESCS was reduced by 8.9%,but the crack rate increased by 9%in field *** IESCS pre-splitting,the roof directionally collapsed along the cutting line,and the gangue filled the ***,the directional roof cutting by the IESCS could decrease roadway *** average pressure of hydraulic supports on the cutting side of the roof was 31%lower than that on the non-cutting side of the roof after *** the self-formed roadway constructed by the IESCS was stabilized,the final relative displacement of the roof and floor was 157.3 mm,meeting the required standard of the next working ***,the IESCS was effectively applied to directional roof *** results demonstrate the promising potential of IESCS in the mining and geotechnical fields.

Post-disaster reconstruction is a topic of global concern,and traditional villages have special heritage attributes and need to face more requirements and obstacles in post-disaster *** paper summarizes four concepts based on the research on post-disaster reconstruction both domestically and internationally,as well as the recovery and reconstruction of cultural *** a field survey of traditional villages in the Ms 6.8 Luding earthquake-stricken area,it is found that there are problems such as insufficient awareness of heritage value,misalignment of scientific reconstruction technology,and insufficient protection of reconstruction elements during the reconstruction *** villages face the risk of declining or even loss of heritage *** order to effectively protect traditional villages and inherit the carrier of regional culture,four targeted reconstruction response strategies are proposed,i.e.,to"establish special planning for traditional village preservation","emphasize recovery of the authenticity of village heritage","ensure elements for village heritage recovery"and"promote the activation and utilization of village heritage",based on the problems discovered during the survey and the four concepts summarized in the research on post-disaster reconstruction of traditional *** research results hope to provide useful reference for ancient cultural areas affected by earthquakes on how to protect cultural heritage during the post-disaster reconstruction process.

The collision experiment between rolling stones of different shapes and protective cushion in open-pit mines

Though gravel cushions are used worldwide in open-pit mines and railway slopes to control the impact of rolling stones,no universal technical standards have been put in place to guide engineers in their correct design,and few laboratory test results are available with which to characterize collisions between rolling stones and a gravel *** carried out a large number of experiments in which rolling stones made of the same material but differently shaped were dropped from various heights onto cushions with various particle sizes and *** investigated the characteristics of the resulting collisions,and the relationships between coefficients of restitution(CORs)of blocks with different shape and release height H,cushion thickness h and particle diameter d are obtained through linear fitting *** testing reveals the relative influence of block shape,release height,and the particle size and thickness of the cushion on the collision characteristics,which can assist engineers in designing a gravel cushion suitable to the distribution and weathering characteristics of rolling stones in a specific area.

Deciphering the role of ultra-low-loaded rhodium in NiFe-MIL-53 for superior oxygen evolution reaction

Designing highly active and stable electrocatalysts of oxygen evolution reaction （OER） is one of the crucial *** this study,a novel OER electrocatalyst,NiFe-MIL-53 modified with ultra-low rhodium（Rh@NiFe-MIL-53）,is successfully prepared via the hydrothermal ***-situ Raman spectroscopy and electrochemical impedance spectroscopy reveal that the doped Rh accelerates the phase transformation of NiFe-MIL-53 and the in-situ formed Rh@NiFeOOH is the actual active *** importantly,the enhanced reversibility of electrochemical reconstruction between NiFeOOH and NiFe（OH）2after doping Rh is beneficial for improving the electrochemical stability of the catalyst.X-ray photoelectron spectroscopy spectra show the strong electronic interaction between single-atom Rh and Ni/Fe in Rh@***,theoretical calculations confirm that the integration of single-atom Rh into the NiFeOOH successfully reduces the band gap,regulates the d-band center （εd）,accelerates the charge transfer,and optimizes the adsorption behavior of oxygen-containing intermediates,thereby lowering the energy barrier of rate-determining ***,the optimized Rh@NiFe-MIL-53 exhibits excellent OER activity （240 mV） with a small Tafel slope of 48.2 mV dec-1and long-term durability (over1270 h at 10 m A cm-2and 110 h at 200 mA cm-2).This work presents a new perspective on designing highly efficient OER electrocatalysts.
