Using e^(+)e^(−)annihilation data corresponding to an integrated luminosity of 2.93 fb^(−1)taken at the center-of-mass energy√s=3.773 GeV with the BESIII detector,a joint amplitude analysis is performed on the decays D^(0)→π^(+)π^(−)π^(+)π^(−)and D^(0)→π^(+)π^(−)π^(0)π^(0)(non-η).The fit fractions of individual components are obtained,and large interferences among the dominant components of the decays D^(0)→a_(1)(1260)π,D^(0)→π(1300)π,D^(0)→ρ(770)ρ(770),and D^(0)→2(ππ)_(S)are observed in both *** the obtained amplitude model,the CP-even fractions of D^(0)→π^(+)π^(−)π^(+)π^(−)and D^(0)→π^(+)π^(−)π^(0)π^(0)(non-η)are determined to be(75.2±1.1_(stat).±1.5_(syst.))%and(68.9±1.5_(stat).±2.4_(syst.))%,*** branching fractions of D^(0)→π^(+)π^(−)π^(+)π^(−)and D^(0)→π^(+)π^(−)π^(0)π^(0)(non-η)are measured to be(0.688±0.010_(stat.)±0.010_(syst.))%and(0.951±0.025_(stat.)±0.021_(syst.))%,*** amplitude analysis provides an important model for the binning strategy in measuring the strong phase parameters of D^(0)→4πwhen used to determine the CKM angleγ(ϕ_(3))via the B^(−)→DK^(−)decay.

Rice(Oryza sativa L.) is one of the most important food crops worldwide and a model monocot plant for gene function analysis, so it is an ideal system for studying flower development. This study reports a mutant, named multi-floret spikelet 3(mfs3), which is related to the spikelet development in rice and derived from the ethylmethane sulfonate(EMS)-treated rice cultivar XIDA 1 B. In mfs3, the main body of palea(bop) was degenerated severely and only glume-like marginal regions of palea(mrp) remained, while other floral organs developed normally, indicating that the palea identity was seriously influenced by the mutation. It was also observed that the number of floral organs was increased in some spikelets, including 2 lemmas, 4 mrp, 4 lodicules, 8–10 stamens, and 2 pistils, which meant that the spikelet determinacy was lost to some degree in mfs3. Furthermore, genetic analysis demonstrated that the mfs3 trait was controlled by a single recessive gene. Using 426 F2 mutants derived from the cross between sterile line 56 S and mfs3, the MULTI-FLORET SPIKELET 3(MFS3) gene was mapped between the molecular markers RM19347 and RM19352 on Chr.6, with a physical distance of 106.3 kb. Sequencing of candidate genes revealed that an 83-bp fragment loss and a base substitution occurred in the LOC_Os06 g04540 gene in the mutant, confirming preliminarily that the LOC_Os06 g04540 gene was the MFS3 candidate gene. Subsequent q PCR analysis showed that the mutation caused the down-regulation of Os MADS1 and FON1 genes, and the up-regulation of Os IDS1 and SNB genes, which are all involved in the regulation of spikelet development. The MFS3 mutation also significantly reduced the transcription of the REP gene, which is involved in palea development. These results indicated that the MFS3 gene might be involved in the spikelet meristem determinacy and palea identity by regulating the expression of these related genes.

AIM: To investigate the role of Gadd45a in hepatic fibrosis and the transforming growth factor (TGF)-beta/ Smad signaling pathway. METHODS: Wild-type male BALB/c mice were treated with CCl4 to induce a model of chronic liver injury. Hepatic stellate cells (HSCs) were isolated from the liver of BALB/c mice and were treated with small interfering RNAs (siRNAs) targeting Gadd45a or the pcDNA3.1-Gadd45a recombinant plasmid. Cellular alpha-smooth muscle actin (alpha-SMA), beta-actin, type I collagen, phospho-Smad2, phospho-Smad3, Smad2, Smad3, and Smad4 were detected by Western blots. The mRNA levels of alpha-SMA, beta-actin, and type I collagen were determined by quantitative real-time (qRT)-PCR analyses. Reactive oxygen species production was monitored by flow cytometry using 2,7-dichlorodihydrofluorescein diacetate. Gadd45a, Gadd45b, anti-Gadd45g, type I collagen, and SMA local expression in liver tissue were measured by histologic and immunohistochemical analyses. RESULTS: Significant downregulation of Gadd45a, but not Gadd45b or Gadd45g, accompanied by activation of the TGF-beta/Smad signaling pathways was detected in fibrotic liver tissues of mice and isolated HSCs with chronic liver injury induced by CCl4 treatment. Overexpression of Gadd45a reduced the expression of extracellular matrix proteins and alpha-SMA in HSCs, whereas transient knockdown of Gadd45a with siRNA reversed this process. Gadd45a inhibited the activity of a plasminogen activator inhibitor-1 promoter construct and (CAGA)(9) MLP-luc, an artificial Smad3/4-specific reporter, as well as reduced the phosphorylation and nuclear translocation of Smad3. Gadd45a showed protective effects by scavenging reactive oxygen species and upregulating antioxidant enzymes. CONCluSION: Gadd45a may counteract hepatic fibrosis by regulating the activation of HSCs via the inhibition of TGF-beta/Smad signaling.

Auditory neuropathy spectrum disorder(ANSD)represents a variety of sensorineural deafness conditions characterized by abnormal inner hair cells and/or auditory nerve function,but with the preservation of outer hair cell *** represents up to 15%of individuals with hearing *** mutation screening,bioinformatic analysis and expression studies,we have previously identified several apoptosis-inducing factor(AIF)mitochondria-associated 1(AIFM1)variants in ANSD families and in some other sporadic ***,to elucidate the pathogenic mechanisms underlying each AIFM1 variant,we generated AIF-null cells using the clustered regularly interspersed short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)system and constructed AIF-wild type(WT)and AIF-mutant(mut)(p.T260A,p.R422W,and p.R451q)stable transfection cell *** then analyzed AIF structure,coenzyme-binding affinity,apoptosis,and other *** revealed that these variants resulted in impaired dimerization,compromising AIF *** reduction reaction of AIF variants had proceeded slower than that of *** average levels of AIF dimerization in AIF variant cells were only 34.5%-49.7%of that of AIF-WT cells,resulting in caspase-independent *** average percentage of apoptotic cells in the variants was 12.3%-17.9%,which was significantly higher than that(6.9%-7.4%)in ***,nicotinamide adenine dinucleotide(NADH)treatment promoted the reduction of apoptosis by rescuing AIF dimerization in AIF variant *** findings show that the impairment of AIF dimerization by AIFM1 variants causes apoptosis contributing to ANSD,and introduce NADH as a potential drug for ANSD *** results help elucidate the mechanisms of ANSD and may lead to the provision of novel therapies.

本文报告了1例足月妊娠因重度子痫前期行剖宫产,术后出现产后溶血性尿毒综合征(postpartum hemolytic uremic syndrome,pHUS)的病例。患者孕39周因重度子痫前期在北京大学第一医院宁夏妇女儿童医院行急诊剖宫产,术后即刻发生微血管病性溶血性贫血、血小板减少、乳酸脱氢酶升高、肝功能异常(转氨酶及胆红素升高)、肾功能异常(少尿、血尿及肌酐进行性升高)。经多学科会诊,考虑重度子痫前期并发pHUS,需行血液透析及血浆置换治疗,转诊至宁夏医科大学总医院。经血液透析及血浆置换治疗,患者病情好转出院。出院后定期肾内科随诊,肾功能术后1个月恢复正常。

In this paper,the implantation-decay method is introduced to study the β-delayed charged particle decay.A silicon detector array was used for the implantation of the incident beams and for the detection of the emitted *** experimental measurement on the β-delayed particle emission from ^(22)Al was used to demonstrate the *** half-life value,charged particle spectroscopy,γ ray spectrum,and γ particle coincidence for the decay process were obtained and compared with previous experimental results for ^(22)*** results show that the implantation-decay method,using a silicon detector array,is a suitable experimental method to study the β-delayed charged particle decay for proton-rich nuclei.

目的构建串联式幽门螺杆菌UreB抗原优势表位UreB322和UreB527原核重组表达系统，制备UreB322和UreB527表位与多肽载体Poly—***的多抗原肽（MAP）疫苗并确定其免疫原性和免疫保护性。方法采用连接引物PCR构建含肠激酶（EK）位点的串联式UreB322和UreB527表位基因及其原核表达系统。表达的目的重组融合蛋白8×[rEK-UreB322-EK-UreB527-EK]经EK水解后用SephadexG-25柱分离rUreB322-EK和rUreB527-EK片段。采用碳二亚胺法将rUreB322-EK、rUreB527-EK与***—Lys载体分子交联，制备出多抗原肽疫苗MAP—rUreB322／B527。采用ELISA和Westernblot分别检测各重组表位肽及MAP-rUreB322／527的抗原性和免疫反应性。采用幽门螺杆菌SS1株感染BALB／c小鼠模型检测MAP—rUreB322／527免疫保护效果。结果获得了8次重复串联的UreB322和UreB527编码基因及其原核表达系统，目的融合蛋白8x[rEK—UreB322-EK—rUreB527-EK]表达量可达细菌总蛋白的48％。肠激酶可将目的融合蛋白完全水解为rUreB322-EK和rUreB527-EK片段。rUreB322-EK和rUreB527-EK与Poly—Asp-Lys交联率高达92．5％。幽门螺杆菌全菌抗体及***均能识别***、***和***322／527并与之结合。***322／527免疫小鼠血清抗体水平明显高于rUreB（P〈0．05）。50或100p,gMAP—rUreB322／527对幽门螺杆菌SS1株感染小鼠的保护率（83．3％和91．7％）明显高于等量rUreB（41．7％和50．0％）（P〈0．05）。结论本研究成功地构建了幽门螺杆菌UreB抗原优势表位UreB322和UreB527重复串联式编码基因及其原核表达系统，基于UreB优势抗原表位的多抗原肽疫苗MAP—rUreB322／527能显著提高免疫保护效果。
