Proteomic analysis of primary colon cancer-associated fibroblasts using the SELDI-ProteinChip platform

Objective： Cancer-associated fibroblasts （CAFs） are one of the hallmarks of the cancer microenvironment. Recent evidence has indicated that CAFs are more competent in enhancing cancer cell growth and migration than normal fibroblasts. However, the unique protein expression of CAFs has not been fully elucidated. This study aims to investigate the characterizations of colon CAFs by comparing the differential protein expression between CAFs and normal fibroblasts. Methods： Primary fibroblasts were isolated from surgical specimen of human colon cancer and matched normal colonic tissue. Purity of the cell population was verified through immunostain analysis. Total cell lysates and conditioned media from each group of cells were extracted, and protein expression analysis was con- ducted using the surface-enhanced laser desorption/ionization time-of-flight mass spectrometry （SELDI-TOF-MS） ProteinChip platform. Results： Most primary cells showed typical fibroblast-like features after two weeks. Increased proportion of a-smooth muscle actin-positive myofibroblasts was detected within the CAFs in four of the six pairs of primary cells. Fibroblast activation protein was weakly expressed in most cells without differences. Using SELDI-TOF-MS ProteinChip platform, four protein peaks mass over charge ratio （m/z） 1142, 3011, 4035, and 4945 were detected in the total cell lysates, and two protein peaks m/z 1368 and 1389 were detected in the conditioned media. The potential candidate proteins found in the Swiss-Prot database include morphogenetic neuropeptides, FMRFamide-related peptides, insulin-like growth factor II, thymosin 13-4-like protein 3, and tight junction-associated protein 1. Conclusions： Using the SELDI-ProteinChip platform, differential protein expressions were identified in colon CAFs compared with normal colonic stromal fibroblasts. The complex proteomic alternations in colon CAFs may play important roles related to the colon cancer microenvironment.

With tremendous growing interests in Big Data, the performance improvement of Big Data systems becomes more and more important. Among many steps, the first one is to analyze and diagnose performance bottlenecks of the Big Data systems. Currently, there are two major solutions. One is the pure data-driven diagnosis approach, which may be very time-consuming;the other is the rule-based analysis method, which usually requires prior knowledge. For Big Data applications like Spark workloads, we observe that the tasks in the same stages normally execute the same or similar codes on each data partition. On basis of the stage similarity and distributed characteristics of Big Data systems, we analyze the behaviors of the Big Data applications in terms of both system and micro-architectural metrics of each stage. Furthermore, for different performance problems, we propose a hybrid approach that combines prior rules and machine learning algorithms to detect performance anomalies, such as straggler tasks, task assignment imbalance, data skew, abnormal nodes and outlier metrics. Following this methodology, we design and implement a lightweight, extensible tool, named HybridTune, and measure the overhead and anomaly detection effectiveness of HybridTune using the BigDataBench benchmarks. Our experiments show that the overhead of HybridTune is only 5%, and the accuracy of outlier detection algorithm reaches up to 93%. Finally, we report several use cases diagnosing Spark and Hadoop workloads using BigDataBench, which demonstrates the potential use of HybridTune.

To assess the responses of the soil microbial community to chronic ozone（O3）, wheat seedlings（Triticum aestivum Linn.） were planted in the field and exposed to elevated O3（e O3）concentration. Three treatments were employed：（1） Control treatment（CK）, AOT40 = 0;（2） O3-1, AOT40 = 1.59 ppm·h;（3） O3-2, AOT40 = 9.17 ppm·h. Soil samples were collected for the assessment of microbial biomass C, community-level physiological profiles（CLPPs）, and phospholipid fatty acids（PLFAs）. EO3 concentration significantly reduced soil microbial carbon and changed microbial CLPPs in rhizosphere soil, but not in non-rhizosphere *** results of the PLFAs showed that e O3 concentrations had significant effects on soil community structure in both rhizosphere and non-rhizosphere soils. The relative abundances of fungal and actinomycetous indicator PLFAs decreased in both rhizosphere and non-rhizosphere soils, while those of bacterial PLFAs increased. Thus the results proved that e O3 concentration significantly changed the soil microbial community function and composition, which would influence the soil nutrient supply and carbon dynamics under O3 exposure.

To understand the effect of precipitation pH and coexisting Mg^(2+) on phosphate adsorption onto zirconium oxide(ZrO_2), ZrO_2 particles precipitated at pH 5.3, 7.1 and 10.5, i.e., ZrO_2(5.3), ZrO_2(7.1)and ZrO_2(10.5), respectively were prepared and characterized, then their adsorption performance and mechanism in the absence and presence of Mg^(2+) were comparatively investigated in this study. The results showed that the Elovich, pseudo-second-order and Langmuir isotherm models correlated with the experimental data well. The adsorption mechanism involved the complexation between phosphate and zirconium. Coexisting Mg^(2+) slightly inhibited the adsorption of phosphate on ZrO_2(5.3), including the adsorption capacity and rate, but coexisting Mg^(2+) greatly increased the adsorption capacity and rate for ZrO_2(7.1)and ZrO_2(10.5). The enhanced adsorption of phosphate on ZrO_2(7.1) and ZrO_2(10.5) in the presence of Mg^(2+) was mainly due to the formation of Mg^(2+)-HPO_4^(2-) ion pair(MgHPO_4~0) in the solution and then the adsorption of MgHPO_4~0 on the adsorbent surface, forming the phosphatebridged ternary complex Zr(OPO_3H)Mg. In the absence of Mg^(2+) , the maximum phosphate adsorption capacity at pH 7 calculated from the Langmuir isotherm model decreased in the order of ZrO 2(7.1)(67.3 mg/g) > ZrO_2(5.3)(53.6 mg/g) ≈ ZrO_2(10.5)(53.1 mg/g), but it followed the order of Zr O2(7.1)(97.0 mg/g) > ZrO_2(10.5)(79.7 mg/g) > ZrO_2(5.3)(51.3 mg/g) in the presence of Mg^(2+) . The results of this work suggest that ZrO_2(7.1) is more suitable for use as an adsorbent for the effective removal of phosphate from municipal wastewater than ZrO_2(5.3) and ZrO_2(10.5),because Mg^(2+) is generally present in this wastewater.

A numerical study to a generalized Korteweg-de Vries (KdV) equation is adopted to model the propagation and disintegration of large-amplitude internal solitary waves (ISWs) in the South China Sea (SCS). Based on theoretical analysis and in situ measurements, the drag coefficient of the Chezy friction is regarded as inversely proportional to the initial amplitude of an ISW, rather than a constant as assumed in the previous studies. Numerical simulations of ISWs propagating from a deep basin to a continental shelf are performed with the generalized KdV model. It is found that the depression waves are disintegrated into several solitons on the continental shelf due to the variable topography. It turns out that the amplitude of the leading ISW reaches a maximum at the shelf break, which is consistent with the field observation in the ***, a dimensionless parameter defining the relative importance of the variable topography and friction is presented.

Erect milkvetch （Astragalus adsurgens Pall.）, a leguminous grass, is a major source of fuel and forage, and has an important role in the restoration of the degraded ecosystems in central and northeastern China. The objective of this work was to investigate how erect milkvetch planting would affect the physical and chemical properties of soil in degraded arable lands. Soil samples at the depths of 0-20 and 20-40 cm were collected from erect milkvetch planting fileds at ages of 0, 1, 2 and 3 yr. Changes in soil bulk density, soil porosity, total N and P, organic matter content, available P, hydrolysable N and available K were measured. The results showed that root biomass and above-ground plant biomass were both significantly increased with plantation age. The significant increase in root nodule biomass was not observed in the first two years. However, it was significantly increased after three years. Root growth of erect milkvetch improved soil structure, and hence, decreased soil bulk density and increased soil porosity. Furthermore, the nitrogen fixation by erect milkvetch and return of erect milkvetch plant to soil increased the soil total N, hydrolysable N and organic matter content of the soil. Low concentrations of P in the soil with erect milkvetch planting could be ascribed to high plant uptakes and possibly to high sequestrations of P in plant biomass. Concentrations of K significantly increased during the first two years of erect milkvetch planting. The high accumulation of K under erect milkvetch cultivation in the first two years could partly be attributed to low plant uptake, and partly to relatively quick recycling within plant-soil systems. Three years after erect milkvetch plantingr, K accumulation at 0-20 cm soil layer was significantly lower than that from non-vegetated field sites, which could be attributed to high plant uptake. To improve the positioning accuracy in GPS point positioning, the geometric dilution of precision （GDOP） including HDOP, VDOP, TDOP, PDOP is commonly considered. The properties of the DOP for the GPS satellite navigation system are studied and the coordinate system is improved in order to decrease the amount of variables. In the end, by simulation and discussing the results, the corresponding conclusions are presented.

A new approach is provided to resolve the large-scale applications of coal tar pitch. Carbon foams with uniform pore size are prepared at the foaming pressure of normal pressure using coal tar pitch as raw materials. The physical and chemical performance of high softening point pitch(HSPP) can be regulated by vacuumizing owing to the cooperation of vacuumizing and polycondensation. Results indicate that the optimum softening point and weight ratio of quinoline insoluble are about 292℃ and 65.7%, respectively. And the optimum viscosity of HSPP during the foaming process is distributed in the range of 1000-10000 Pa·s. The resultant carbon foam exhibits excellent performance, such as uniform pore structure, high compressive strength(4.7 MPa), low thermal conductivity(0.07 W·m^(-1) ·K^(-1)), specially, it cannot be fired under the high temperature of 1200 ℃.Thus, this kind of carbon foam is a potential candidate for thermal insulation material applied in energy saving building.
