Decline in tree-ring growth of Picea mongolica and its intra-annual eco-physiological responses to drought and CO_(2) enrichment in semi-arid China

Episodes of drought-induced decline in tree growth and mortality are becoming more frequent as a result of climate warming and enhanced water stress in semi-arid ***,the ecophysiological mechanisms under-lying the impact of drought on tree growth remains *** this study,earlywood and latewood tree-ring growth,δ^(13)C,andδ^(18)O chronologies of Picea mongolica from 1900 to 2013 were developed to clarify the intra-and inter-annual tree-ring growth responses to increasingly fre-quent *** results indicate that annual basal area increment residuals(BAI_(res)),which removed tree age and size effects,have significantly decreased since ***-ever,the decreasing trend of earlywood BAI_(res) was higher than that of *** response analysis suggests that the dominant parameters for earlywood and latewood proxies(BAI_(res),δ^(13)C andδ^(18)O)were drought-related climate variables(Palmer drought severity index,temperature,rela-tive humidity,and vapor pressure deficit).The most signifi-cant period of earlywood and latewood proxies’responses to climate variables were focused on June-July and July-August,***_(res),andδ^(13)C were significantly affected by temperature and moisture conditions,whereasδ^(18)O was slightly *** stomatal conduct-ance due to drought outweighed the influence of increasing CO_(2) on intrinsic water use efficiency(iWUE),and ultimately led to a decline in BAI_(res).Compared to latewood,the faster decreasing BAI_(res) and smaller increasing iWUE of early-wood suggested trees were more vulnerable to water stress in the early growing *** study provides insights into the inter-and intra-annual mechanisms of tree-ring growth in semi-arid regions under rising CO_(2) and climate change.

A Federated Learning Framework with Blockchain-Based Auditable Participant Selection

Federated learning is an important distributed model training technique in Internet of Things(IoT),in which participant selection is a key component that plays a role in improving training efficiency and model *** module enables a central server to select a subset of participants to performmodel training based on data and device *** doing so,selected participants are rewarded and actively perform model training,while participants that are detrimental to training efficiency and model accuracy are ***,in practice,participants may suspect that the central server may have miscalculated and thus not made the selection *** lack of trustworthiness problem,which can demotivate participants,has received little *** problem that has received little attention is the leakage of participants’private information during the selection *** will therefore propose a federated learning framework with auditable participant *** supports smart contracts in selecting a set of suitable participants based on their training loss without compromising the *** the possibility of malicious campaigning and impersonation of participants,the framework employs commitment schemes and zero-knowledge proofs to counteract these malicious ***,we analyze the security of the framework and conduct a series of experiments to demonstrate that the framework can effectively improve the efficiency of federated learning.

Biodistribution and preparation of technetium-99m-labeled D-D3 monoclonal antibody against pro-gastrin-releasing peptide ~31-98） in mice

Background We previously reported that iodine-131（131^I）-Iabeled anti-pro-gastrin-releasing peptide （ProGRP（31-98）） monoclonal antibody D-D3 could selectively accumulate in the tumor sites of nude mice bearing small cell lung cancer （SCLC） xenografts. However, 1311-D-D3 was cleared slowly from the body, and the best radioimmunoimaging time for SCLC was 72-96 hours after injection. The aims of this study were to radiolabel anti-ProGRP（31-98） D-D3 monoclonal antibody with technetium-99m（99m^Tc） and to investigate the biodistribution of this antibody in healthy ICR mice. Methods D-D3was labeled with 99m^Tc via the 2-mercaptoethanol reduction method. 99m^Tc-D-g3 was purified by the gel column separation method. The labeling efficiency and radiochemical purity were measured by thin-layer chromatography. The immunological activity of 99m^Tc-D-O3 was determined with cell conjugation assays. 99m^Tc-D-D3 was injected into healthy ICR mice via a tail vein, and all the healthy ICR mice were sacrificed by cervical dislocation at a designated time. Then, the blood and major organs were removed and weighed, and counted in a gamma scintillation counter to determine the percentage of the injected dose per gram （%ID/g）. Results The labeling rate and the radiochemical purity of 99m^TC-D-D3 were （73.87±2.89）% and （94.13±4.49）%, respectively. The immunobinding rates of 99m^Tc-O-D3 to the human small cell lung cancer NCI-H446 cell line and lung adenocarcinoma A549 cell line were （81.2±2.37）% and （24.3±1.46）%, respectively. The distribution data of normal ICR mice demonstrated that 99m^TC-D-D3was mainly distributed in the liver, kidney and lung, and less in the brain tissue and muscle. Conclusions 99m^Tc-D-D3 antibody not only had high radiochemical purity, but also had good stability both in vitro and in vivo, and maintained good immunological activity. 99m^Tc-D-D3 was metabolized mainly in the kidney and liver, and the blood radioactivity decreased rapidly. Study on the mechanism of apoptosis of myelocytic leukemia cell lines induced by curcumin

Objective: To investigate the mechanism of apoptosis of myelocytic leukemia lines NB4, K562 and THP-1 cells induced by curcumin. Methods: After the cells were treated with curcumin at different concentration (25, 12.5, 6.25, 3.125, 0 μmol/L) for various times (0, 12, 24, 48 h), flow cytometry (FCM) was used to determine the rate of apoptosis of cells. After the cells were treated with curcumin at 25 μmol/L for 24 h, flow cytometry was used to determine the expression level of Fas, and Western blot was performed to determine the expression of Caspase-8 and Caspase-9. Results: (1) Curcumin could induced the apoptosis of NB4, K562 and THP-1 cells in a time- and dose-dependent manner. After the cells were treated with curcumin at 25 μmol/L for 48 h, the rate of apoptosis of cells was over fifty percent. (2) Fas level showed remarkable increase (P < 0.01) after above cells were treated with 25 μmol/L curcumin for 24 h. (3) The apoptosis proteins of Caspase-8 and Cas- pase-9 were also increased obviously (P < 0.01) after the cells were treated with 25 μmol/L curcumin for 24 h. Conclusion: The molecular pathway of apoptosis of myelocytic leukemia lines induced by curcumin are concerned with death receptor and mitochondria.

Achieving single-cell-resolution lineage tracing in zebrafish by continuous barcoding mutations during embryogenesis

Unraveling the lineage relationships of all descendants from a zygote is fundamental to advancing our understanding of developmental and stem cell ***,existing cell barcoding technologies in zebrafish lack the resolution to capture the majority of cell divisions during embryogenesis.A recently developed method,a substitution mutation-aided lineage-tracing system(SMALT),successfully reconstructed high-resolution cell phylogenetic trees for Drosophila ***,we implement the SMALT system in zebrafish,recording a median of 14 substitution mutations on a one-kilobase-pair barcoding sequence for one-day post-fertilization *** this system,we reconstruct four cell lineage trees for zebrafish fin cells,encompassing both original and regenerated *** tree consists of hundreds of internal nodes with a median bootstrap support of 99%.Analysis of the obtained cell lineage trees reveals that regenerated fin cells mainly originate from cells in the same part of the *** multiple times sampling germ cells from the same individual,we show the stability of the germ cell pool and the early separation of germ cell and somatic cell *** system offers the potential for reconstructing high-quality cell phylogenies across diverse tissues,providing valuable insights into development and disease in zebrafish.

Antibody-drug conjugates (ADCs) are biologically targeted drugs composed of antibodies and cytotoxic drugs connected by linkers. These innovative compounds enable precise drug delivery to tumor cells, minimizing harm to normal tissues and offering excellent prospects for cancer treatment. However, monoclonal antibody-based ADCs still present challenges, especially in terms of balancing efficacy and safety. Bispecific antibodies are alternatives to monoclonal antibodies and exhibit superior internalization and selectivity, producing ADCs with increased safety and therapeutic efficacy. In this review, we present available evidence and future prospects regarding the use of bispecific ADCs for cancer treatment, including a comprehensive overview of bispecific ADCs that are currently in clinical trials. We offer insights into the future development of bispecific ADCs to provide novel strategies for cancer treatment.

Emotion classification in textual conversations focuses on classifying the emotion of each utterance from textual *** is becoming one of the most important tasks for natural language processing in recent ***,it is a challenging task for machines to conduct emotion classification in textual conversations because emotions rely heavily on textual *** address the challenge,we propose a method to classify emotion in textual conversations,by integrating the advantages of deep learning and broad learning,namely *** aims to provide a more effective solution to capture local contextual information(i.e.,utterance-level)in an utterance,as well as global contextual information(i.e.,speaker-level)in a conversation,based on Convolutional Neural Network(CNN),Bidirectional Long Short-Term Memory(Bi-LSTM),and broad *** experiments have been conducted on three public textual conversation datasets,which show that the context in both utterance-level and speaker-level is consistently beneficial to the performance of emotion *** addition,the results show that our proposed method outperforms the baseline methods on most of the testing datasets in weighted-average F1.

Efficient oxidation of monosaccharides to sugar acids under neutral condition in flow reactors with gold-supported activated carbon catalysts

A significant reaction in the synthesis of biomass-based chemicals is the catalyst-based and targeted oxidation of monosaccharides into valuable sugar acids. In this study, an activated carbon supported gold catalyst was used to oxidize glucose and xylose to gluconic acid and xylonic acid under neutral condition. Optimization of reaction conditions for the catalysts was performed using both a batch reactor and a flow-through reactor. In a batch reactor, the yields of gluconic and xylonic acid reached 93% and 92%, respectively, at 90 degrees C within 180 min. In a flow reactor, both reactions reached a similar yield at 80 degrees C with the weight hourly space velocity of 47.1 h-1. The reaction kinetics were explored in the flow reactor. The oxidation of glucose and xylose to gluconic and xylonic acid followed a first-order kinetics and the turnover frequency was 0.195 and 0.161 s-1, respectively. The activation energy was evaluated to be 60.58 and 59.30 kJmol-1, respectively. This study presents an environmentally friendly and feasible method for the selective oxidation of monosaccharides using an activated carbon supported gold catalyst, benefiting the high-value application of carbohydrates.

Evolution of High-energy Electron Distribution in Pulsar Wind Nebulae

In this paper,we analyze the spectral energy distributions of 17 powerful(with a spin-down luminosity greater than10~(35)erg s~(-1))young(with an age less than 15,000 yr)pulsar wind nebulae(PWNe)using a simple timein-dependent one-zone emission *** aim is to investigate correlations between model parameters and the ages of the corresponding PWNe,thereby revealing the evolution of high-energy electron distributions within *** findings are as follows:(1)The electron distributions in PWNe can be characterized by a double power-law with a super-exponential cutoff.(2)As PWNe evolve,the high-energy end of the electron distribution spectrum becomes harder with the index decreasing from approximately 3.5 to 2.5,while the low-energy end spectrum index remains constant near 1.5.(3)There is no apparent correlation between the break energy or cutoff energy and the age of PWNe.(4)The average magnetic field within PWNe decreases with age,leading to a positive correlation between the energy loss timescale of electrons at the break energy or the high-energy cutoff and the age of the PWN.(5)The total electron energy within PWNe remains constant near 2×10~(48)erg,while the total magnetic energy decreases with age.
