BACKGROUND Gastric signet ring cell carcinoma(GSRC)represents a specific subtype of gastric cancer renowned for its contentious epidemiological features,treatment principles,and prognostic *** To investigate the epidemiology of GSRC and establish an improved model for predicting the prognosis of patients with locally advanced GSRC(LAGSRC)after *** The annual rates of GSRC incidence and mortality,covering the years 1975 to 2019,were extracted from the Surveillance,Epidemiology,and End Results(SEER)database to explore the temporal trends in both disease incidence and mortality rates using Joinpoint *** clinical data of 3793 postoperative LAGSRC patients were collected from the SEER database for the analysis of survival *** Cox regression model was used to explore the independent prognostic factors for overall survival(OS).The risk factors extracted were used to establish a prognostic *** The overall incidence of GSRC increased dramatically between 1975 and 1998,followed by a significant downward trend in incidence after *** recent years,there has been a similarly optimistic trend in GSRC mortality *** trend in GSRC showed discrepancies based on age and *** operating characteristic curves,calibration curves,and decision curve analysis for 1-year,3-year,and 5-year OS demonstrated the high discriminative ability and clinical utility of this *** area under the curve indicated that the performance of the new model outperformed that of the pathological staging *** The model we established can aid clinicians in the early prognostication of LAGSRC patients,resulting in improved clinical outcomes by modifying management strategies and patient health care.

Local anesthetic lidocaine induces apoptosis in human corneal stromal cells in vitro

AIM:To demonstrate the apoptosis-inducing effect of iidocalne on human corneal stromal(HCS)cells fn vitm,and provide experimental basis for safety anesthetic usage In clinic of ***:In vitro cultured HCS cells were treated with lidocaine at different doses and times,and their morphology was monitored successively with inverted phase contrast *** membrane permeability of them was detected by acridine orange/ethidium bromide(AO/EB)double *** DNA fragmentation of them was examined by agarose gel electrophoresis,and their ultrastructure was observed by transmission electron microscopy(TEM),***:Exposure to lidocaine at doses from0.3125g/L to 20g/L induced morphological changes of HCS cells such as cytoplasmic vacuolation,cellular shrinkage,and turning round,and elevated membrane permeability of these cells in AO/EB *** change of morphology and membrane permeability was doseand time-dependent,while lidocaine at dose below0.15625g/L could not induce these ***,lidocaine induced DNA fragmentation and ultrastructural changes such as cytoplasmic vacuolation,structural disorganization,chromatin condensation,and apoptotic body appearance of the ***:Lidocaine has significant cytotoxicity on human corneal stromal cells in vitro in a dose-and time-dependent manner by inducing apoptosis of these *** established experimental model and findingsbased on this model here help provide new insight into the apoptosis-inducing effect of local anesthetics in eye clinic.

Transplantation of tissue-engineered human corneal epithelium in limbal stem cell deficiency rabbit models

AIM: To evaluate the biological functions of tissue-engineered human corneal epithelium (TE-HCEP) by corneal transplantation in limbal stem cell deficiency (LSCD) rabbit models. METHODS: TE-HCEPs were reconstructed with DiI-labeled untransfected HCEP cells and denuded amniotic membrane (dAM) in air-liquid interface culture, and their morphology and structure were characterized by hematoxylin-eosin (HE) staining of paraffin-sections, immunohistochemistry and electron microscopy. LSCD models were established by mechanical and alcohol treatment of the left eyes of New zealand white rabbits, and their eyes were transplanted with TE-HCEPs with dAM surface outside by lamellar keratoplasty (LKP). Corneal transparency, neovascularization, thickness, and epithelial integrality of both traumatic and post transplantation eyes were checked once a week by slit-lamp corneal microscopy, a corneal pachymeter, and periodic acid-Schiff (PAS) staining. At day 120 post surgery, the rabbits in each group were sacrificed and their corneas were examined by DiI label observation, HE staining, immunohistochemistry and electron microscopy. RESULTS: After cultured for 5 days on dAM, HCEP cells, maintaining keratin 3 expression, reconstructed a 6-7 layer TE-HCEP with normal morphology and structure. The traumatic rabbit corneas, entirely opaque, conjunctivalized and with invaded blood vessels, were used as LSCD models for TE-HCEP transplantation. After transplantation, obvious edema was not found in TE-HCEP-transplanted corneas which became more and more transparent, the invaded blood vessels reduced gradually throughout the monitoring period. The corneas decreased to normal thickness on day 25, while those of dAM eyes were over 575 mu m in thickness during the monitoring period. A 45 layer of epithelium consisting of TE-HCEP originated cells attached tightly to the anterior surface of stroma was reconstructed 120 days after TE-HCEP transplantation, which was similar to the normal control eye

Machine learning-based predictions of current and future susceptibility to retrogressive thaw slumps across the Northern Hemisphere

Retrogressive thaw slumps(RTSs)caused by the thawing of ground ice on permafrost slopes have dramatically increased and become a common permafrost hazard across the Northern Hemisphere during previous ***,a gap remains in our comprehensive understanding of the spatial controlling factors,including the climate and terrain,that are conducive to these RTSs at a global *** machine learning methodologies,we mapped the current and future RTSs susceptibility distributions by incorporating a range of environmental factors and RTSs *** identified freezing-degree days and maximum summer rainfall as the primary environmental factors affecting RTSs *** final ensemble susceptibility map suggests that regions with high to very high susceptibility could constitute(11.6±0.78)%of the Northern Hemisphere's permafrost *** juxtaposed with the current(2000-2020)RTSs susceptibility map,the total area with high to very high susceptibility could witness an increase ranging from(31.7±0.65)%(SSP585)to(51.9±0.73)%(SSP126)by the *** insights gleaned from this study not only offer valuable implications for engineering applications across the Northern Hemisphere,but also provide a long-term insight into the potential change of RTSs in permafrost regions in response to climate change.

DLF-YOLOF:an improved YOLOF-based surface defect detection for steel plate

Surface defects can affect the quality of steel *** methods based on computer vision are currently applied to surface defect detection of steel ***,their real-time performance and object detection of small defect are still *** improved object detection network based on You Only Look One-level Feature(YOLOF)is proposed to show excellent performance in surface defect detection of steel plate,called ***,the anchor-free detector is used to reduce the network ***,deformable convolution network and local spatial attention module are introduced into the feature extraction network to increase the contextual information in the feature ***,the soft non-maximum suppression is used to improve detection accuracy ***,data augmentation is performed for small defect objects during training to improve detection *** show the average precision and average precision for small objects are 42.7%and 33.5%at a detection speed of 62 frames per second on a single GPU,*** shows that DLF-YOLOF has excellent performance to meet the needs of industrial real-time detection.

In this study,a novel equivalent damping ratio model that is suitable for reinforced concrete(RC)structures considering cyclic degradation behavior is developed,and a new equivalent linearization analysis method for implementing the proposed equivalent damping ratio model for use in seismic damage evaluation is *** this end,Ibarra’s peak-oriented model,which incorporates an energy-based degradation rule,is selected for representing hysteretic behavior of RC structure,and the optimized equivalent damping for predicting the maximum displacement response is presented by using the empirical method,in which the effect of cyclic degradation is ***,the relationship between the hysteretic energy dissipation of the inelastic system and the elastic strain energy of the equivalent linear system is established so that the proposed equivalent linear system can be directly integrated with the Park-Ang seismic model to implement seismic damage *** to the simplicity of the equivalent linearization method,the proposed method provides an efficient and reliable way of obtaining comprehensive insight into the seismic performance of RC *** verification demonstrates the validity of the proposed method.

The two-stage electrostatic precipitator is widely used to purify oil mist ***,there is limited research on the influences of relative humidity,particle deposition characteristics,and the generation of gaseous ***,this paper established a numerical model of the electrostatic oil mist purifier and applied it to a two-stage electrostatic *** the model was used to investigate the corona discharge characteristics under different relative humidity conditions in the charged zone,the particle deposition characteristics,the purification efficiency,the ozone concentration distribution,and the oil vapor concentration distribution in the collection *** results indicate that,with a constant temperature,the corona current decreases as relative humidity increase,and there is a quadratic relationship between relative humidity and *** variation in relative humidity has little impact on the purification *** maximum ozone concentration occurs near the electrode line,and its concentration is influenced by the discharge current and inlet airflow *** oil vapor concentration reaches its maximum value at the side plates,with a value of 19 ppb,while it reaches the minimum value at the collecting zone electrode plate,with a value of 2 *** temperature is the main factor affecting the volatilization of the oil film,with higher temperatures resulting in higher oil vapor.
