Polyunsaturated fatty acids （PUFAs） are essential for normal growth in mammals, especially the ω-3 PUFAs, which play important roles in preventing several life-threatening diseases, such as coronary heart disease and diabetes. In this study, we aimed to investigate whether the sFat-1 gene from Caenorhabditis briggsae could be functionally expressed in transgenic pigs, and whether the transgenic could synthesize high quality ω-3 PUFAs endogenously. In this study, a gene construct consisting of CMV promoter and 1.9 kb cDNA of ω-3 fatty acid desaturase gene （sFat-1） from C. briggsae was injected into the male pronucleus of pig embryos by microinjection. The piglets were screened for the transgene by PCR, Southern blot and reverse transcription-PCR analysis. Pigs that give positive results were mated with wild-type pigs to produce the next generation and the transmission of transgene was examined by PCR analysis. Fatty acids compositions of various tissues in the transgenic pigs were then analyzed by gas chromatograph. A total of 878 embryos were transferred into 42 recipients, among which 29 successfully got pregnant and gave birth to a total of 162 piglets, and 8 of them were identified to be transgenic. Fatty acid compositions in the transgenic pigs were altered, and the levels of ω-6：ω-3 ratios were decreased from 14.53 in the control to 2.62 in Fat-1 transgenic pigs. A number of primary sFat-1-transgenic pigs were bred in this study, which lays the foundation for cultivation of new varieties of transgenic pigs.

Association between Ambient Air Pollution and Outpatient Visits for Acute Bronchitis in a Chinese City

Objective To investigate the short-term association between outdoor air pollution and outpatient visits for acute bronchitis,which is a rare subject of research in the mainland of *** A time-series analysis was conducted to examine the association of outdoor air pollutants with hospital outpatient visits in Shanghai by using two-year daily data（2010-2011）.Results Outdoor air pollution was found to be associated with an increased risk of outpatient visits for acute bronchitis in *** effect estimates of air pollutants varied with the lag structures of the concentrations of the *** lag06,a 10 μg/m3 increase in the concentrations of PM10,SO2,and NO2 corresponded to 0.94%（95% CI：0.83%,1.05%）,11.12%（95% CI：10.76%,11.48%）,and 4.84%（95% CI：4.49%,5.18%） increases in hospital visits for acute bronchitis,*** associations appeared to be stronger in females（P〈0.05）.Between-age differences were significant for SO2（P〈0.05）,and between-season differences were also significant for SO2（P〈0.05）.Conclusion Our analyses have provided the first evidence that the current air pollution level in China has an effect on acute bronchitis and that the rationale for further limiting air pollution levels in Shanghai should be strengthened.

Pulmonary hypertension (PH) is a fatal disorder characterized by pulmonary vascular remodeling and obstruction. The phosphodiesterase 4 (PDE4) family hydrolyzes cyclic AMP (cAMP) and is comprised of four subtypes (PDE4A–D). Previous studies have shown the beneficial effects of pan-PDE4 inhibitors in rodent PH;however, this class of drugs is associated with side effects owing to the broad inhibition of all four PDE4 isozymes. Here, we demonstrate that PDE4B is the predominant PDE isozyme in lungs and that it was upregulated in rodent and human PH lung tissues. We also confirmed that PDE4B is mainly expressed in the lung endothelial cells (ECs). Evaluation of PH in Pde4b wild type and knockout mice confirmed that Pde4b is important for the vascular remodeling associated with PH. In vivo EC lineage tracing demonstrated that Pde4b induces PH development by driving endothelial-to-mesenchymal transition (EndMT), and mechanistic studies showed that Pde4b regulates EndMT by antagonizing the cAMP-dependent PKA–CREB–BMPRII axis. Finally, treating PH rats with a PDE4B-specific inhibitor validated that PDE4B inhibition has a significant pharmacological effect in the alleviation of PH. Collectively, our findings indicate a critical role for PDE4B in EndMT and PH, prompting further studies of PDE4B-specific inhibitors as a therapeutic strategy for PH.

Small interference RNA targeting vascular endothelial growth factor gene effectively attenuates retinal neovascularization in mice model

Background The mechanism of retinal neovascularization is not understood completely. Many growth factors are involved in the process of retinal neovascularization, such as vascular endothelial growth factor （VEGF） and pigment epithelium-deprived factor （PEDF）, which are the representatives of angiogenic and antiangiogenic molecules respectively. Oxygen induced retinopathy （OIR） is a useful model to investigate retinal neovascularization. The present study was conducted to investigate the feasibility of small interference RNA （siRNA） targeting VEGF gene in attenuating oxygen induced retinopathy （OIR） by regulating VEGF to PEDF ratio （VEGF/PEDF）. Methods In vitro, cultured EOMA cells were transfected with VEGF-siRNA （psi-HITM/EGFPNEGF siRNA） and LipofectamineTM 2000 for 24, 48, and 72 hours, respectively. Expression of VEGF mRNA was evaluated by real time polymerase chain reaction （PCR） and the level of VEGF protein was analyzed by Western blotting. In vivo, OIR model mice were established, the mice （C57BL/6J） received an intra-vitreal injection of μ1 of mixture of psi-HITM/EGFPNEGF siRNA and Lipofectamine 2000. Expressions of retinal VEGF and PEDF protein were measured by Western blotting, retinal neovascularization was observed by fluorescein angiography, and quantified. Results In vitro psi-HITM/EGFPNEGF siRNA treatment significantly reduced VEGF mRNA and protein expression. In vivo, with decreased VEGF and VEGF-PEDF ratio, significant attenuation of neovascular tufts, avascular regions, tortuous, and dilated blood vessels were observed in the interfered animals. Conclusions VEGF plays an important role in OIR, and the transfection of VEGF-siRNA can effectively downregulate VEGF expression in vivo, accompanied by the downregulation of VEGF-PEDF ratio, and simultaneous attenuation of retinal neovascularization was also observed. 进入21世纪后,美国自由民主制度呈现出深度的自我困惑,美国之乱成为政治顽疾。美国政治是一个“两面神”的设计,是自由主义美国与保守主义美国的混合体,具有一定韧性和适应力。理解美国政治,需确立利益政治与身份政治相结合的分析框架,分析在不同生态环境下美国政治周期的治乱转化。不同于利益政治造就的充斥着物质性与既定性的混乱,美国的身份政治充斥着认知的消极性、情感的负面性与行为的激进性,爆发出更强的破坏效力。随着身份政治的强势崛起,美国政治运行基础正在发生从利益政治主导向身份政治主导的位移,而逐渐崛起的z世代成为推动身份政治演进和变革的主力军,在这一位移过程中正在且将会发挥着重大作用。

大孔柚子型微结构光纤探针的表面增强拉曼散射性能研究

随着光纤制备工艺以及纳米材料制备技术的发展,光纤探针已成为一种新型的表面增强拉曼散射(SERS)基底,通过在普通单模光纤或多模光纤上制备不同的结构并修饰相应的纳米材料,可以得到多种类型的光纤表面增强拉曼散射探针,并实现较好的检测效果。但受限于光纤本身的结构,普通光纤仅能利用端面或侧表面提供拉曼检测的“热点”区域,限制了其SERS性能的进一步提高。因此制备了大孔柚子型微结构光纤(MSF)表面增强拉曼散射(SERS)探针,其中大孔柚子型MSF SERS探针结构通过一段阶跃多模光纤与柚子型微结构光纤熔接制得。实验分别对自制的纳米银溶胶基底以及大孔柚子型MSF SERS探针的SERS性能进行检测。采用溶胶自组装法制备负载银纳米颗粒的MSF SERS探针,通过控制自组装时间制备不同光纤SERS探针(Ag/MSF-x,其中x为自组装时间,分别为15、30、45、60 min)。采用溶液检测方法,利用Ag/MSF-x探针对10^(-3) mol·L^(-1)的亚甲基蓝(MB)探针分子进行检测,通过比较相同条件下的增强效果筛选得到Ag/MSF-45探针。为进一步检测Ag/MSF-45探针的SERS性能,制备不同浓度的MB溶液,分别利用纳米银溶胶基底和Ag/MSF-45探针对其进行检测。实验结果表明,纳米银溶胶基底对MB的检测限(LOD)为10^(-6) mol·L^(-1),Ag/MSF-45探针对MB的检测限(LOD)为10^(-7) mol·L^(-1);拉曼信号的再现性结果表明纳米银溶胶基底以及Ag/MSF-45探针在各个特征峰处的RSD值均在合理范围内;在1619 cm^(-1)拉曼位移处对纳米银溶胶基底以及Ag/MSF-45探针检测MB的拉曼强度和浓度进行对数转换拟合,纳米银溶胶基底的拟合优度R2达0.91628,Ag/MSF-45探针的拟合优度R2达0.98848;纳米银溶胶基底和Ag/MSF-45探针的再现性结果表明,其在各个特征峰处的RSD值均处于合理范围,但Ag/MSF-45探针的各特征峰RSD值均小于纳米银溶胶基底,最大为13.89%;利用10^(-6) mol·L^(-1)的MB对Ag/MSF-45探针的增强因子(AEF)进行计算,Ag/MSF-45探针的AEF达到6.09×10^(6),表现出良好的增强效果。因此,基于大孔柚子型MSF SERS探针凭借独特的空气孔结构使其具有较高的灵敏度以及良好的再现性,且其SERS性能均优于纳米银溶胶基底,在农业、化学分析、生物检测等领域及大分子物质检测等方面应用前景广泛。

However, because of the ill-posedness of the D A -based hverse problem and the instability of reconstruction algorithm s, the location accuracy of the reconstucted sources is low. Radiative transfer equation （RTE）, which considers the direction of the photon m igration and the effect of absorption and scattering in tissues, can accurately express the transmission ofbiolum hescent photns through the tissues. In this paper, we studied the biolum hescence inaging based on the RTE. 2D sinuiations were performed, and quantitative evaluation was given by the absolute source position error, the relative source area error and the m h in um bound hg box. The resu Its of the experin ent showed that the in aging quality based on R TE was beer than thatone based on D A.
