High-dose dexamethasone regulates microglial polarization via the GR/JAK1/STAT3 signaling pathway after traumatic brain injury

Although microglial polarization and neuroinflammation are crucial cellular responses after traumatic brain injury,the fundamental regulatory and functional mechanisms remain insufficiently *** potent anti-inflammato ry agents,the use of glucoco rticoids in traumatic brain injury is still controversial,and their regulatory effects on microglial polarization are not yet *** the present study,we sought to determine whether exacerbation of traumatic brain injury caused by high-dose dexamethasone is related to its regulatory effects on microglial polarization and its mechanisms of *** vitro cultured BV2 cells and primary microglia and a controlled cortical impact mouse model were used to investigate the effects of dexamethasone on microglial ***,dexamethasone,RU486(a glucocorticoid receptor antagonist),and ruxolitinib(a Janus kinase 1 antagonist)were ***-sequencing data obtained from a C57BL/6 mouse model of traumatic brain injury were used to identify potential targets of *** Morris water maze,quantitative reverse transcription-polymerase chain reaction,western blotting,immunofluorescence and confocal microscopy analysis,and TUNEL,Nissl,and Golgi staining were performed to investigate our ***-throughput sequencing results showed that arginase 1,a marker of M2 microglia,was significantly downregulated in the dexamethasone group compared with the traumatic brain injury group at3 days post-traumatic brain *** dexamethasone inhibited M1 and M2 microglia,with a more pronounced inhibitory effect on M2microglia in vitro and in *** receptor plays an indispensable role in microglial polarization after dexamethasone treatment following traumatic brain ***,glucocorticoid receptor activation increased the number of apoptotic cells and neuronal death,and also decreased the density of dendritic spines.A possible downstream receptor signaling mechanism is th

At present,most researches on the critical rainfall threshold of debris flow initiation use a linear model obtained through *** relatively weak fault tolerance,this method not only ignores nonlinear effects but also is susceptible to singular noise samples,which makes it difficult to characterize the true quantization relationship of the rainfall ***,the early warning threshold determined by statistical parameters is susceptible to negative samples(samples where no debris flow has occurred),which leads to uncertainty in the reliability of the early warning results by the regression *** overcome the above limitations,this study develops a data-driven multiobjective evolutionary optimization method that combines an artificial neural network(ANN)and a multiobjective evolutionary optimization implemented by particle swarm optimization(PSO).Firstly,the Pareto optimality method is used to represent the nonlinear and conflicting critical thresholds for the rainfall intensity I and the rainfall duration *** ANN is used to construct a dual-target(dual-task)predictive surrogate model,and then a PSO-based multiobjective evolutionary optimization algorithm is applied to train the ANN and stochastically search the trained ANN for obtaining the Pareto front of the I-D surrogate prediction model,which is intended to overcome the limitations of the existing linear regression-based threshold ***,a double early warning curve model that can effectively control the false alarm rate and negative alarm rate of hazard warnings are proposed based on the decision space and target space *** study provides theoretical guidance for the early warning and forecasting of debris flows and has strong applicability.

Numerical calculations of creep damage development and life behavior of circular notched specimens of nickel-base single crystal had been performed. The creep stress distributions depend on the specimen geometry. For a small notch radius, von Mises stress has an especial distribution. The damage distribution is greatly influenced by the notch depth, notch radius as well as notch type. The creep crack initiation place is different for each notched specimen. The characteristics of notch strengthening and notch weakening depend on the notch radius and notch type. For the same notch type, the creep rupture lives decrease with the decreasing of notch radius. A creep life model has been presented for the multiaxial stress states based on the crystallographic slip system theory.

Decentralized methods for targeting poverty are widely adopted in developing countries to improve the performance of various poverty alleviation programs.A common challenge for implementing successful decentralized targeting is the existence of elite *** has recently implemented a nationwide decentralized poverty targeting program,the targeted poverty alleviation(TPA)policy,to achieve the national goal of eliminating absolute poverty by the end of *** the largest decentralized poverty targeting program in the world,TPA's successful implementation was believed to be threatened by elite capture in some earlier *** 2015,a targeting correction mechanism,called"follow-up checks"policy,has been *** the"follow-up checks"policy,the elites and other ineligible households who receive benefits under TPA were removed from the *** paper investigates the elite capture phenomenon in TPA using village census data from a poverty-stricken county in 2017-two years after implementing the"follow-up checks"*** find no evidence of elite capture in *** elites are unlikely to become beneficiaries or receive more benefits than *** results contradict earlier findings that reported elite capture in *** argue that the reason is the accountability emphasized by the central government in the"follow-up checks"*** findings imply that having proper accountability is critical for improving targeting performance by global antipoverty initiatives.

MICROSTRUCTURE ANALYSIS OF AN ION-NITRIDINGLAER ON A NO.45 STEEL

The cross-sectional microstructure of an ion-nitrided layer on a No.45 steel was studied by transmission electron microscopy (TEM). The results show that the columnar crystals constitute the compound layer. Most of them have the microstructure of alternating ε-Fe2-3N and γ-Fe4N and others are single ε-Fe2-3N phase. There are abnormal strip ferrites between the columnar crystals. The transition layer mainly consists of spherical γ-Fe4N and cementite (Fe3C) particles in the pearlite are dissolved in this layer. In diffusion layer, besides the equilibrium phase γ-Fe4N, there are α -Fe16N2 and the satellite spots around main spots of α-Fe16N2 demonstrate its modulation structure. A metastable ordered cluster zone of the nitropen atoms was found in the division layer.

Understanding the structural origin of the competition between oxygen 2p and transition-metal 3d orbitals in oxygen-redox(OR)layered oxides is eminently desirable for exploring reversible and high-energy-density Li/Na-ion ***,we reveal the correlation between cationic ordering transition and OR degradation in ribbon-ordered P3-Na_(0.6)Li_(0.2)Mn_(0.8)O_(2) via in situ structural *** two different voltage windows,the OR capacity can be improved approximately twofold when suppressing the in-plane cationic ordering *** find that the intralayer cationic migration is promoted by electrochemical reduction from Mn^(4+)to Jahn–Teller Mn^(3+)and the concomitant NaO_(6) stacking transformation from triangular prisms to octahedra,resulting in the loss of ribbon ordering and electrochemical ***-principles calculations reveal that Mn^(4+)/Mn^(3+)charge ordering and alignment of the degenerate eg orbital induce lattice-level collective Jahn–Teller distortion,which favors intralayer Mn-ion migration and thereby accelerates OR *** findings unravel the relationship between in-plane cationic ordering and OR reversibility and highlight the importance of superstructure protection for the rational design of reversible OR-active layered oxide cathodes.

Improving the Input of Classified Neural Networks Through Feature Construction

A general classification algorithm of neural networks is unable to obtain satisfied results because of the uncertain problems existing among the features in moot classification programs, such as interaction. With new features constructed by optimizing decision trees of examples, the input of neural networks is improved and an optimized classification algorithm based on neural networks is presented. A concept of dispersion of a classification program is also introduced too in this paper. At the end of the paper, an analysis is made with an example.

Preparation and Microstructure Analysis of High Strength Cementitious Materials Containing Metakaolin

The preparation and microstructure analysis of high strength cementitious materials containing metakaolin (MK) was studied in this paper. The MK was prepared firstly,and then was mixed with fly ash,ground blast furnace slag,quartz powder and cement with different percentage to produce high strength cementitious materials. After cured under different environment,the compressive strength of such materials was tested. Techniques of X-ray diffraction (XRD) and scanning electron microscopy (SEM) were further employed to identify the composition and microstructure. The results obtained reveal that the compressive strength of such materials mainly effected by component of raw materials,the water binder ratio (W/B) and the curing regime. When the three factors above are optimized,the compressive strength of such materials can get to 156 MPa in maximum. The X-ray diffraction analysis and the scanning electron microscopic images indicated that under optimal curing condition,more raw materials can take chemical reaction and the microstructure is dense to yield good mechanical properties.

Finite element simulations on the mechanical properties of MHS materials

Finite element simulations are carried out to examine the mechanical behavior of the metallic hollow sphere （MHS） material during their large plastic deformation and to estimate the energy absorbing capacity of these materials under uniaxial compression. A simplified model is proposed from experimental observations to describe the connection between the neighboring spheres, which greatly improves the computation efficiency. The effects of the governing physical and geometrical parameters are evaluated; whilst a special attention is paid to the plateau stress, which is directly related to the energy absorbing capacity. Finally, the empirical functions of the relative material density are proposed for the elastic modulus, yield strength and plateau stress for FCC packing arrangement of hollow spheres, showing a good agreement with the experimental results obtained in our previous study.

The rare-earth nickelates(RENiO_(3)) exhibit an exceptional complex electronic phase diagram and multiple electronic phase transitions that enrich promising applications in correlated electronic devices beyond conventional ***,the practical applications of RENiO_(3) are challenged by their intrinsic thermodynamic metastability in material synthesis and high material ***,developing an economical strategy to achieve the batch synthesis of RENiO_(3) is of vital *** this work,we enlarged the synthesis amount of RENiO_(3) up to 20 g per batch using chloride(KCI) assisted molten salt *** optimizing the reaction conditions,the powder of RENiO_(3) with the cubic shape and average size of ~2μm was effectively synthesized,while their phase purity exceeded 95%.In addition,the cost to synthesize RENiO_(3) was further reduced by using rare-earth extraction intermediate products as the raw materials,instead of using the pure rare-earth *** also achieved wide adjustments in the metal-to-insulator-transition temperature from160 to 420 K without significantly reducing the transition *** enlarging the synthesis amount and the reducing the cost,it paves the way to the device application of RENiO_(3).
