从药效相关性和成分差异阐释青翘与老翘改善博来霉素(BLM)诱导肺纤维化小鼠的作用差异。小鼠随机分为正常组,模型组,吡非尼酮组(50 mg·kg^(-1)),青翘低、高剂量组(1.3、2.6 g·kg^(-1)),老翘低、高剂量组(1.3、2.6 g·kg^(-1))。采用气管滴注BLM制备肺纤维化模型,统计小鼠存活率及体质量变化;给药完成后计算肺指数,HE染色、Masson染色和天狼星红染色评价肺组织病理变化;透射电子显微镜观察肺组织超微结构;生化法测定肺组织转化生长因子-β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)、E钙黏着蛋白(E-cadherin)、羟脯氨酸(HYP)及肺泡灌洗液中白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平;RT-qPCR、Western blot法检测肺组织中基质金属蛋白酶7(MMP7)、胶原蛋白Ⅰ型(collagenⅠ)、E-cadherin、TNF-α、波形蛋白(vimentin)、TGF-β1、α-SMA表达;免疫荧光检测肺组织中α-SMA的表达;主成分分析法评价老翘与青翘改善肺纤维化的作用差异;分子对接技术分析老翘中含量高的化合物与TGF-β1受体之间的相互作用,CCK-8法检测其对TGF-β1诱导BEAS-2B和HFL1细胞模型的影响。结果显示,青翘、老翘可提高肺纤维化小鼠存活率,降低肺指数,改善肺组织病理损伤与胶原纤维沉积水平,改善小鼠Ⅱ型肺泡上皮细胞板层小体受损情况,降低肺泡灌洗液中IL-6、TNF-α水平,下调肺组织中HYP、MMP7、vi-mentin、collagenⅠ、TGF-β1、α-SMA的表达,上调E-cadherin的表达。通过主成分分析综合评价胶原纤维沉积水平,其改善程度为老翘高剂量>老翘低剂量>青翘高剂量>青翘低剂量。分子对接显示羟基酪醇、咖啡酸、连翘脂素、落叶松脂素与TGF-β1受体有较好的结合活性,且可显著减轻TGF-β1诱导BEAS-2B细胞损伤,抑制TGF-β1诱导HFL1细胞增殖。综上,青翘、老翘均可有效改善BLM诱导的肺纤维化小鼠损伤,老翘在减少胶原沉积方面优于青翘,可能与老翘中羟基酪醇、咖啡酸、连翘脂素、落叶松脂素含量高于青翘有关。

Among oncolytic viruses, the vesicular stomatitis virus(VSV) is especially potent and a highly promising agent for the treatment of cancer. But, even though effective against multiple tumor entities in preclinical animal models, replicationcompetent VSV exhibits inherent neurovirulence, which has so far hindered clinical development. To overcome this limitation,replication-defective VSV vectors for cancer gene therapy have been tested and proven to be safe. However, gene delivery was inefficient and only minor antitumor efficacy was observed. Here, we present semireplicationcompetent vector systems for VSV(sr VSV), composed of two trans-complementing, propagation-deficient VSV vectors. The de novo generated deletion mutants of the two VSV polymerase proteins P(phosphoprotein) and L(large catalytic subunit), VSVΔP and VSVΔL respectively, were used mutually or in combination with VSVΔG vectors. These sr VSV systems copropagated in vitro and in vivo without recombinatory reversion to replication-competent virus. The sr VSV systems were highly lytic for human glioblastoma cell lines,spheroids, and subcutaneous xenografts. Especially the combination of VSVΔG/VSVΔL vectors was as potent as wild-type VSV(VSV-WT) in vitro and induced long-term tumor regression in vivo without any associated adverse effects. In contrast, 90% of VSV-WT-treated animals succumbed to neurological disease shortly after tumor clearance. Most importantly, even when injected into the brain, VSVΔG/VSVΔL did not show any neurotoxicity. In conclusion, sr VSVis a promising platform for virotherapeutic approaches and also for VSV-based vector vaccines, combining improved safety with an increased coding capacity for therapeutic transgenes, potentially allowing for multipronged approaches.

Sand-fflled splash cups were used to study the erosive power of rainfall and throughfall in the humid subtropics of Southeast China. The splash cup measurements yielded precise and reproducible results under both open field conditions and forest vegetation. The splash cups were exposed to specific forest stands of different ages and to selected species （Schima superba, Castanopsis eyrei, Daphniphyllum oldhamii, Lithocarpus glaber） in the Gutianshan （古田山） National Nature Reserve （GNNR）. The results of the measurements under forest vegetation show that the erosive power of throughfall drops to be 2.59 times higher compared to the open field. This accentuates the importance of shrub, herb and litter layers in forest ecosystems to protect the soil against erosion. Coalescing drops from leaves and branches （drips） are responsible for this notable gain in erosive power. Moreover, differences in sandloss between the investigated tree species （deciduous, evergreen） revealed that the erosion potential and the spatial heterogeneity of throughfall are species-specific. This highlights the importance of selecting specific species for afforestation projects considering the prevention of soil erosion.

μC/OS-Ⅱ is an open source real-time kernel adopting priority preemptive schedule strategy. Aiming at the problem of μC/OS-Ⅱ failing to support homology priority tasks scheduling, an approach for solution is proposed. The basic idea is adding round-robin scheduling strategy in its original scheduler in order to schedule homology priority tasks through time slice roundrobin. Implementation approach is given in detail. Firstly, the Task Control Block （TCB） is extended. And then, a new priority index table is created, in which each index pointer points to a set of homology priority tasks. Eventually, on the basis of reconstructing μC/OS-Ⅱ real-time kernel, task scheduling module is rewritten. Otherwise, schedulability of homology task supported by modified kernel had been analyzed, and deadline formula of created homology tasks is given. By theoretical analysis and experiment verification, the modified kernel can support homology priority tasks scheduling, meanwhile, it also remains preemptive property of original μC/OS-Ⅱ.

在电网电压不平衡情况下,并网逆变器的输出有功功率和电流将存在二次脉动和畸变。为了提高电网不平衡时并网逆变器并网电流质量,有效降低并网逆变器对电网的冲击,研究不平衡电网情况下并网逆变器控制策略成为了迫切的需求。根据不同控制目标,从并网同步算法、正负序分离、电流跟踪控制、谐波补偿和滤波器技术等方面对现有并网逆变器控制策略进行综述。最后,对不平衡电网情况下并网逆变器发展趋势进行探讨。

分析各因素对农村土地流转价格的影响方向与作用强度可为确定土地承包经营权流转价格提供依据。该文以黑龙江省农村固定观察点14个村的转入方农户为样本,采用相关性分析和多元线性回归方法分别确定单因素与多因素对流转价格的影响,并对比分析各因素对高于均价组和低于均价组中流转价格的影响差异。结果表明:1)单因素对流转价格的影响分析中,耕地粮食产量对流转价格产生显著正向影响,是影响流转价格最重要的因素;2)多因素对流转价格的影响分析中,耕地粮食产量、村域经济发达程度、县域交通通达性、户主年龄对流转价格产生显著正向影响,县域人均GDP、与市中心距离对流转价格产生显著负向影响;3)对比各因素对高于均价组和低于均价组的影响,发现交通通达性、与市中心距离对高于均价组流转价格的影响方向与总样本一致,这些因素对低于均价组流转价格的影响方向与总样本相反。需明确耕地信息、了解地区经济与交通情况,为确定土地流转价格、完善农村土地流转市场创造条件。
