Effects of daily light integral on tomato(Solanum lycopersicon L.)grafting and quality in a controlled environment

As the source of energy and biological signals,light can influence the healing process of grafted seedlings by regulating the synthesis of the endogenous hormone,regeneration of wound-healing tissue,and connection of vascular tissue in grafted seedlings.The effect of daily light integral(Dli)on the healing process and seedling quality of tomato(Solanum lycopersicum L.)was analyzed in this study,with the comparison of grafted seedlings treated in dark for 7 d after grafting.The results showed that the height increment of scion and rootstock,adhesion of graft union,stem flow,total chlorophyll content,and net photosynthesis rate increased gradually with increasing light intensity,and no longer increased significantly when the Dli was higher than 5.04 mol/m^(2)·d.The contents of auxin(IAA)and gibberellin(GA)in tomato leaves increased and abscisic acid(ABA)decreased with the increase of Dli.However,there was no significant difference between the treatments with Dli higher than 6.48 mol/m^(2)·d.Both the biomass and energy use efficiency(EUE)of grafted seedlings increased with Dli in a certain range and then decreased.The biomass was the largest when Dli was 5.04 mol/m^(2)·d.However,EUE was highest when Dli was 7.46 mol/m^(2)·d.In conclusion,a suitable Dli is beneficial to cultivate high-quality grafted tomato seedlings,and increasing Dli within a certain range can promote biomass accumulation,connection of vascular tissue,and endogenous hormone biosynthesis in tomato grafted seedlings during the healing period.The lighting environment with Dli of 5.04 mol/m^(2)·d(light intensity of 100μmol/m^(2)·s and light time of 14 h/d)is recommended for the healing treatment in high-quality production,which also improves EUE during the healing period of tomato grafted seedlings.

Because of their large volume variation and inferior electrical conductivity,Mn_(3)O_(4)-based oxide anode materials have short cyclic lives and poor rate capability,which obstructs their development.In this study,we successfully prepared a Mn_(3)O_(4)/N-doped honeycomb carbon composite using a smart and facile synthetic method.The Mn_(3)O_(4)nanopolyhedra are grown on N-doped honeycomb carbon,which evidently mitigates the volume change in the charging and discharging processes but also improves the electrochemical reaction kinetics.More importantly,the Mn-O-C bond in the Mn_(3)O_(4)/N-doped honeycomb carbon composite benefits electrochemical reversibility.These features of the Mn_(3)O_(4)/N-doped honeycomb carbon(NHC)composite are responsible for its superior electrochemical performance.When used for li-ion batteries,the Mn_(3)O_(4)/N-doped honeycomb carbon anode exhibits a high reversible capacity of 598 mAh·g^(−1)after 350 cycles at 1 A·g^(−1).Even at 2 A·g^(−1),the Mn_(3)O_(4)/NHC anode still delivers a high capacity of 472 mAh·g^(−1).This work provides a new prospect for synthesizing and developing manganese-based oxide materials for energy storage.

Histological staining is the gold standard for tissue examination in clinical pathology and life-science research,which visualizes the tissue and cellular structures using chromatic dyes or fluorescence labels to aid the microscopic assessment of tissue.However,the current histological staining workflow requires tedious sample preparation steps,specialized laboratory infrastructure,and trained histotechnologists,making it expensive,time-consuming,and not accessible in resource-limited settings.Deep learning techniques created new opportunities to revolutionize staining methods by digitally generating histological stains using trained neural networks,providing rapid,cost-effective,and accurate alternatives to standard chemical staining methods.These techniques,broadly referred to as virtual staining,were extensively explored by multiple research groups and demonstrated to be successful in generating various types of histological stains from label-free microscopic images of unstained samples;similar approaches were also used for transforming images of an already stained tissue sample into another type of stain,performing virtual stain-to-stain transformations.In this Review,we provide a comprehensive overview of the recent research advances in deep learning-enabled virtual histological staining techniques.The basic concepts and the typical workflow of virtual staining are introduced,followed by a discussion of representative works and their technical innovations.We also share our perspectives on the future of this emerging field,aiming to inspire readers from diverse scientifc fields to further expand the scope of deep learning-enabled virtual histological staining techniques and their applications.

Applying hot-embossing technology,a simple and cost-effective method for the fabrication of microstructured High Density Polyethylene(HDPE)surfaces with a robust superhydrophobic wetting state is proposed.Micro-meshes and micro-grooves in the flexible template are filled by the PE melt in the hot embossing process.Subsequently,a two-stage microstructure on the PE film surface is formed.This PE film exhibits a contact angle of 151.8°±2°and roll-offangle of>90~when a 5μL water droplet is dropped on its surface.Water pinning ability on the surface is figured out and roll-off angles are as a quadratic function of specified water droplet volume.Specifically,a 356μN water pinng force appears on the HDPE film due to the solid-vapor composite interface on its surface.Meanwhile,self-cleaning and immersion tests reveal that the HDPE surface with micro-pillars exhibit robust Cassie impregnating wetting state against external pressure.The proposed method for facial fabrication ofmicrostructured surfaces is an appropriate candidate for the development of droplet manipulation and functional biomimetic polymer surfaces.

Mathematical calculation usually requires sustained attention to manipulate numbers in the mind,while listening to light music has a relaxing effect on the brain.The differences in the corresponding brain functional network topologies underlying these behaviors remain rarely known.Here,we systematically examined the brain dynamics of four behaviors(resting with eyes closed and eyes open,tasks of music listening and mental calculation)using 64-channel electroencephalogram(EEG)recordings and graph theory analysis.We developed static and dynamic minimum spanning tree(MST)analysis method and demonstrated that the brain network topology under mental calculation is a more line-like structure with less tree hierarchy and leaf fraction;however,the hub regions,which are mainly located in the frontal,temporal and parietal regions,grow more stable over time.In contrast,music-listening drives the brain to exhibit a highly rich network of star structure,and the hub regions are mainly located in the posterior regions.We then adopted the dynamic dissimilarity of different MSTs over time based on the graph Laplacian and revealed low dissimilarity during mental calculation.These results suggest that the human brain functional connectivity of individuals has unique dynamic diversity and flexibility under various behaviors.

Single molecular localization microscopy(SMLM)is a useful tool in biological observation with sub-10-nm resolution.However,SMLM is incapable of discerning two molecules within the diffraction-limited region unless with the help of a stochastic on–off switching scheme which yet entails time-consuming processes.Here,we produce a novel kind of focal spot pattern,called sub-diffraction dark spot(SDS),to localize molecules within the sub-diffraction region of interest.In our proposed technique nominated as sub-diffracted dark spot localization microscopy(SDLM),multiple molecules within the diffraction-limited region could be distinguished without the requirement of stochastic fluorescent switches.We have numerically investigated some related impacts of SDLM,such as detection circle diameter,collected photon number,background noise,and SDS size.Simulative localization framework has been implemented on randomly distributed and specifically structured samples.In either two-or three-dimensional case,SDLM is evidenced to have2 nm localization accuracy.

L-Arginine is the precursor of nitric oxide(NO),a host immune effector against intracellular pathogens including Mycobacterium tuberculosis(M.tb).Pathogens including M.tb have evolved various strategies targeting arginine to block the production of NO for better survival and proliferation.However,L-arginine metabolism and regulation in Mycobacterium are poorly understood.Here,we report the identification of M.smegmatis MSMEG_1415(homolog of M.tb Rv2324)as an arginine-responsive transcriptional factor regulating the arginase pathway.In the absence of L-arginine,MSMEG_1415 acts as a repressor to inhibit the transcription of the roc(for arginine,ornithine catabolism)gene cluster,thereby switching off the arginase pathway.Treatment with L-arginine relieves the transcriptional inhibition of MSMEG_1415 on the roc gene cluster to activate the arginase pathway.Moreover,the L-arginine-MSMEG_1415 complex activates the transcription of the roc gene cluster by recognizing and binding a 15-bp palindrome motif,thereby preventing the excess accumulation of L-arginine in M.smegmatis.Physiologically,MSMEG_1415 confers mycobacteria resistance to starvation and fluoroquinolones exposure,suggestive of its important role in M.smegmatis persistence.The results uncover a unique regulatory mechanism of arginine metabolism in mycobacteria and identify M.tb Rv2324 as an attractive candidate target for the design of drugs against tuberculosis.
