Mechanisms of autophagy activ.tion in endothelial cell and their targeting during normothermic machine liv.r perfusion

Ischaemia-reperfusion injury(IRI) is the leading cause of injury seen in the liv.r following transplantation. IRI also causes injury following liv.r surgery and haemodynamic shock. The first cells within the liv.r to be injured by IRI are the liv.r sinusoidal endothelial cells(LSEC). Recent ev.dence suggests that LSEC coordinate and regulates the liv.rs response to a v.riety of injuries. It is becoming increasingly apparent that the cyto-protectiv. cellular process of autophagy is a key regulator of IRI. In particular LSEC autophagy may be an essential gatekeeper to the dev.lopment of IRI. The recent av.ilability of liv.r perfusion dev.ces has allowed for the therapeutic targeting of autophagy to reduce IRI. In particular normothermic machine liv.r perfusion(NMP-L) allow the deliv.ry of pharmacological agents to donor liv.rs whilst maintaining physiological temperature and hepatic flow rates. In this rev.ew we summarise the current understanding of endothelial autophagy and how this may be manipulated during NMP-L to reduce liv.r IRI.

Risk Assessment of Aircraft’s Lateral v.er-off the Runway Surface during Landing

This article presents a methodology to determine the risk of aircrafts lateral runway excursion during landing v.a mathematical risk modeling. In addition, the methodology is demonstrated by means of detailed calculation of the lateral runway excursion risk v.lue during the landing of the aircraft Airbus A310-200, in v.ew of the maximum landing weight and the appropriate range of landing v.locities according to the International Civ.l Av.ation Organization specification. Obv.ously, the calculation demonstrates that the dev.loped math solutions and equations presented herein are powerful tools to ev.luate the risk of lateral runway excursion of the majority of aircrafts and for any airport. The method is also applicable to assess the residual lev.l of risk at any specific airport and its dev.ation compared to the recommended safety lev.l. Consequently, the presented mathematical solutions to determine the risk rate of lateral runway excursion during landing offers airports’ operational and safety management departments a v.able tool so that appropriate measurements could be adopted. Finally, it is a methodology not only to assess the risk but also to determine the appropriate runway width.

Changes of Reef Community near Ku Lao Cham Islands (South China Sea) after Sangshen Typhoon

Coral reefs of Ku Lao Cham Islands in the Central v.etnam were studied ov.r a quarter of a century. It was established that the reefs of these Islands were lost after the Sangshen typhoon and a heav. flood, which happened in 2006. As a result of these natural phenomena a significant part of coral populations was physically destroyed, and a powerful export of clay substances from the nearest v.etnamese coast and from the Bung Riv.r, flowing into the sea opposite the Islands, causing abundant sedimentation and death of almost all the remaining scleractinian corals. At present, where formerly there were coral reefs, now there is algal-alcyonarian stage of succession of reef communities. This is typical, as a rule, for restoration of reefs after a destructiv. typhoon effect.

Association of the lev.l of heteroplasmy of the 15059G>A mutation in the MT-CYB mitochondrial gene with essential hypertension

AIM: To examine whether the heteroplasmy lev.l for 15059G>A mutation in the mitochondrial genome might be associated with essential hypertension. METHODS: This cross-sectional study inv.lv.d 196 unrelated participants randomly selected from general population (90 males and 106 females) who underwent a regular medical check-up at the Institute for Ath-erosclerosis Research (Moscow, Russia). One hundred and twenty of them (61%) had essential hypertension, and 76 (39%) were apparently healthy normotensiv. persons. The lev.l of heteroplasmy for 15059G>A mutation occurring in the coding region of cytochrome b gene (MT-CYB) of mtDNA isolated from the blood leukocytes, was quantified using DNA pyrosequencing method. RESULTS: The 15059G>A heteroplasmy lev.l ranged between 4% and 83%, with a median lev.l of 31%. Between the upper and lower quartiles of 15059G>A heteroplasmy distribution, significant differences were observ.d for patients’ age, systolic blood pressure, and triglyceride lev.ls. 15059G>A heteroplasmy correlated both with age (r = 0.331, P v.aled that the age explains 12% v.riability of 15059G>A heteroplasmy, and hypertension independently explains more 5% v.riability. The 15059G>A heteroplasmy exceeding 31% was found to be significantly associated with a higher risk of essential hypertension (odds ratio 2.76; P (Fisher) 0.019]. The study participants with high 15059G>A heteroplasmy lev.l were found to hav. significantly higher age (P v.lence of essential hypertension (P = 0.033), as compared to those with low 15059G>A heteroplasmy lev.l. These observ.tions suggested a positiv. correlation between the lev.l of 15059G>A heteroplasmy and essential hypertension. CONCLUSION: This study prov.des the ev.dence of association of mtDNA 15059G>A mutation heteroplasmy with essential hypertension.
