Enterogenic Stenotrophomonas maltophilia migrates to the mammary gland to induce mastitis by activating the calcium-ROS-AMPK-mTOR-autophagy pathway

Background Mastitis is an inflammatory disease of the mammary gland that has serious economic impacts on the dairy industry and endangers food *** previous study found that the body has a gut/rumen-mammary gland axis and that disturbance of the gut/rumen microbiota could result in‘gastroenterogenic mastitis'.However,the mechanism has not been fully ***,we found that long-term feeding of a high-concentrate diet induced mastitis in dairy cows,and the abundance of Stenotrophomonas maltophilia(***)was significantly increased in both the rumen and milk ***,we hypothesized that‘gastroenterogenic mastitis'can be induced by the migration of endogenous gut bacteria to the mammary ***,this study investigated the mechanism by which enterogenic *** induces *** First,*** was labelled with superfolder GFP and administered to mice via *** results showed that treatment with *** promoted the occurrence of mastitis and increased the permeability of the blood-milk barrier,leading to intestinal inflammation and intestinal ***,tracking of ingested *** revealed that *** could migrate from the gut to the mammary gland and induce ***,mammary gland transcriptome analysis showed that the calcium and AMPK signalling pathways were significantly upregulated in mice treated with ***,using mouse mammary epithelial cells(MMECs),we verified that *** induces mastitis through activation of the calcium-ROS-AMPK-mTOR-autophagy *** In conclusion,the results showed that enterogenic *** could migrate from the gut to the mammary gland via the gut-mammary axis and activate the calcium-ROS-AMPK-mTOR-autophagy pathway to induce *** the gut-mammary gland axis may also be an effective method to treat mastitis.

Mannogalactoglucan from mushrooms protects pancreatic islets via restoring UPR and promotes insulin secretion in TIDM mice

Type 1 diabetes mellitus(T1DM) lacks insulin secretion due to autoimmune deficiency of pancreaticβ-*** pancreatic islets and enhancing insulin secretion has been therapeutic *** is the main type of polysaccharide from natural mushroom,which has potential medicinal ***,the antidiabetic property of mannogalactoglucan in T1DM has not been fully *** this study,we obtained the neutral fraction of alkali-soluble Armillaria mellea polysaccharide(AAMP-N) with the structure of mannogalactoglucan from the fruiting body of *** and investigated the potential therapeutic value of AAMP-N in *** demonstrated that AAMP-N lowered blood glucose and improved diabetes symptoms in T1DM ***-N activated unfolded protein response(UPR) signaling pathway to maintain ER protein folding homeostasis and promote insulin secretion in *** that,AAMP-N promoted insulin synthesis via upregulating the expression of transcription factors,increased Ca^(2+) signals to stimulate intracellular insulin secretory vesicle transport via activating calcium/calmodulin-dependent kinase Ⅱ(CamkⅡ) and cAMP/PKA signals,and enhanced insulin secretory vesicle fusion with the plasma membrane via vesicle-associated membrane protein 2(VAMP2).Collectively,these studies demonstrated that the therapeutic potential of AAMP-N on pancreatic islets function,indicating that mannogalactoglucan could be natural nutraceutical used for the treatment of T1DM.

Oxalate regulates crystal-cell adhesion and macrophage metabolism via JPT2/PI3K/AKT signaling to promote the progression of kidney stones

Oxalate is an organic dicarboxylic acid that is a common component of plant *** kidneys are essential organs for oxalate excretion,but excessive oxalates may induce kidney *** microtubule associated homolog 2(JPT2)is a critical molecule in Ca^(2+)mobilization,and its intrinsic mechanism in oxalate exposure and kidney stones remains *** study aimed to reveal the mechanism of JPT2 in oxalate exposure and kidney *** approaches were used to control JPT2 expression in cells and mice,and the JPT2 mechanism of action was analyzed using transcriptomics and untargeted *** results showed that oxalate exposure triggered the upregulation of JPT2,which is involved in nicotinic acid adenine dinucleotide phosphate(NAADP)-mediated Ca^(2+)*** analysis revealed that cell adhesion and macrophage inflammatory polarization were inhibited by JPT2 knockdown,and these were dominated by phosphatidylinositol 3-kinase(PI3K)/AKT signaling,*** metabolomics indicated that JPT2 knockdown inhibited the production of succinic acid semialdehyde(SSA)in ***,JPT2 deficiency in mice inhibited kidney stones *** conclusion,this study demonstrates that oxalate exposure facilitates kidney stones by promoting crystal-cell adhesion,and modulating macrophage metabolism and inflammatory polarization via JPT2/PI3K/AKT signaling.

UAV-based transient electromagnetic 3D forward modeling and inversion and analysis of exploration capability

Unmanned aerial vehicle transient electromagnetic(UAV-TEM)is a novel airborne exploration method that offers advantages such as low cost,simple operation,high exploration efficiency and suitability for near-surface exploration in complex terrain *** improve the accuracy of data interpretation in this method,the authors conducted a systematic three-dimensional(3D)forward modeling and inversion of the *** study utilized the finite element method based on unstructured tetrahedral elements and employed the second-order backward Euler method for time *** allowed for accurate 3D modeling and accounted for the effects of complex *** on these,the influence characteristics of flight altitudes and the sizes,burial depths,and resistivities of anomalies are compared and analyzed to explore the UAV-TEM systems’exploration ***,four typical geoelectrical models of landslides are designed,and the inversion method based on the Gauss-Newton optimization method is used to image the landslide models and analyze the imaging effect of the UAV-TEM method on landslide *** results showed that UAV-TEM could have better exploration resolution and fine imaging of nearsurface structures,providing important technical support for monitoring,early warning,and preventing landslides and other geological hazards.

High qualityβ-Ga_(2)O_(3)single crystal nanobelts with length of 2−3 mm and width from tens of microns to 132μm were synthesized by carbothermal reduction *** on the grown nanobelt with the length of 600μm,the dual-Schottky-junctions coupling device(DSCD)was *** to the electrically floating Ga_(2)O_(3)nanobelt region coupling with the double Schottky-junctions,the current I_(S2)increases firstly and rapidly reaches into saturation as increase the voltage V_(S2).The saturation current is about 10 pA,which is two orders of magnitude lower than that of a single *** the case of solar-blind ultraviolet(UV)light irradiation,the photogenerated electrons further aggravate the coupling physical mechanism in device.I_(S2)increases as the intensity of UV light *** the UV light of 1820μW/cm^(2),I_(S2)quickly enters the saturation *** V_(S2)=10 V,photo-to-dark current ratio(PDCR)of the device reaches more than 104,the external quantum efficiency(EQE)is 1.6×10^(3)%,and the detectivity(D*)is 7.5×10^(12)*** addition,the device has a very short rise and decay times of 25−54 ms under different positive and negative *** shows unique electrical and optical control characteristics,which will open a new way for the application of nanobelt-based devices.

3D time-domain forward modeling of airborne transient electromagnetism considering superparamagnetic effect

The superparamagnetic effect arises from the superparamagnetism exhibited by a multitude of nano-sized magnetic mineral particles under an external electric field. This phenomenon manifests in transient electromagnetic data primarily as a deceleration in the attenuation rate of late-stage signals， a characteristic difficult to discern directly from airborne transient electromagnetic signals， consequently leading to significant misinterpretations of subterranean electrical structures. This study embarks on 3D forward modeling of airborne electromagnetic responses in the frequency domain， accounting for the superparamagnetic effect， utilizing an unstructured finite element method. Superparamagnetic responses in the time domain were obtained through frequency-time conversion. This investigation explores the influence of various parameters—such as magnetic susceptibility， time constants， and flight altitude—on the superparamagnetic effect by examining the response characteristics of typical targets. Findings indicate that in its late stages， the superparamagnetic effect can induce a relative anomaly of up to 300%. There is a positive correlation between magnetic susceptibility and the strength of the superparamagnetic effect. The influence of the time constant’s upper and lower limits on the superparamagnetic effect is minimal; however， the range between these limits significantly affects the effect， showing a negative correlation with its intensity. Higher flight altitudes weaken the superparamagnetic signal. The impact is most pronounced when superparamagnetic minerals are shallowly buried， effectively shielding the underlying geology with the characteristics of a good conductivity anomaly， but this effect diminishes with greater depth. The insights from this study provide a theoretical framework for a deeper understanding of the superparamagnetic effect in transient electromagnetic signals and for more accurate interpretations of subterranean geological and electrica

Calycosin improves atherosclerosis by reshaping the interaction between the gut microbiome and bile acid metabolism

Calycosin， Astragali Radix most prominent ingredient， has drawn more attention as a result of its ability to treat atherosclerosis(AS). However， the mechanism of action has not been fully elucidated. We investigated the effects of calycosin on bile acid (BA) metabolism and gut microbiome in ApoE-/-mice fed a high-fat diet (HFD). The data showed that the aorta of ApoE-/-mice treated with HFD showed significant atheromatous plaque formation and lipid accumulation， and the levels of TC， TG and LDL-C were significantly increased， while the levels of HDL-C were significantly decreased. Calycosin can substantially regulate lipid levels， thereby alleviating liver lipid deposition induced by atherosclerosis. In addition， 16S rRNA sequencing showed that calycosin treatment has reshaped the gut microbiota disturbed by HFD， in particular， increasing the ratio of Bacteroidetes/Firmicutes， and improving the relative abundance of Bilophila， Desulfovibrio， Bacteroides， Lactobacillus， etc. Meanwhile， targeted metabolomics analysis showed that calycosin treatment significantly modulated CDCA， TCA， LCA， DCA， TDCA and bile acid pool composition， which were associated with atherosclerotic plaque areas. In addition， calycosin treatment also down-regulated FXR protein levels and up-regulated CYP7A1 protein levels in the hepatic. At the same time， calycosin inhibits the ileum FXR/TGR5 signaling pathway， inhibits BA reabsorption， promotes BA excretion， and reduces hepatic cholesterol accumulation by enterohepatic circulation. In addition， we found that calycosin significantly promoted the expression of hepatic ABCA1 and ABCG1 to mediate cholesterol efflux. Meanwhile， calycosin regulates gut microbiota， and Bacteroides， Alistipes， Desulfovibrio， Lactobacillus， Bilophila and Odoribacter are closely related to specific BAs. 宁夏固原地区汉墓随葬的单耳罐和双耳罐与中原地区陶器区别明显,与当地战国陶器也有较大差异。扩大对比范围后可知,固原汉代的附耳罐主要和关中西部至陇山一带的发现最为接近,应当存在区域间的文化交流。此外,在陕西中北部也存在类似的战国陶器,被称为“辛庄类遗存”。结合历史背景,推测秦人很可能在扩张的过程中吸收了北方民族具有“辛庄类遗存”特征的附耳罐,而后将其带入固原地区,当地战国时期的文化因素随之消亡。类似的文化交融持续至西汉,最终呈现出固原墓葬的文化因素与中原高度一致的局面。

Asymmetric expression of CA2 and CA13 linked to calcification in the bilateral mandibular condyles cause crossed beaks in chickens

Crossed beak is a complex mode of inheritance with prevalence ranging from 0.2 to 7.4% in at least 12 chicken strains *** reveal the intrinsic factors causing crossed beaks,genes expression patterns in bilateral mandibular condyle between affected and normal birds were characterized by RNA sequencing analysis in the present *** beak was induced by short length of unilateral mandibular ramus,and a total of 110differentially expressed genes were up-or down-regulated in the affected(short)mandibular condyle side as compared to the normal *** anhydrase 2(CA2)and Carbonic anhydrase 13(CA13)were enriched in the carbonate dehydratase activity,and high-expressed in mandibular condyle and osteoblasts(P<0.05).However,both were low-expressed in short mandibular condyle side of affected birds(P<0.05).The carbonate dehydratase inhibitor experiments confirmed that there is positive association between the calcification and carbonic anhydrase *** analysis with cetylpyridinium chloride showed a decrease in calcification when the cells were transfected with an anti-CA13 *** research suggested that CA2 and CA13 are down-calcified in shortside mandibular condyle,and caused mandibular ramus to grow ***2 and CA13 have the critical role in crossed beaks by regulating calcification of mandibular condyle.
