The clinical translation of stem cells and their extracellular vesicles(EVs)-based therapy for central nervous system(CNS) diseases is booming. Nevertheless, the insufficient CNS delivery and retention together with the invasiveness of current administration routes prevent stem cells or EVs from fully exerting their clinical therapeutic potential. Intranasal(IN) delivery is a possible strategy to solve problems as IN route could circumvent the brain-blood barrier non-invasively and fit repeated dosage regimens. Herein, we gave an overview of studies and clinical trials involved with IN route and discussed the possibility of employing IN delivery to solve problems in stem cells or EVs-based therapy. We reviewed relevant researches that combining stem cells or EVs-based therapy with IN administration and analyzed benefits brought by IN route. Finally, we proposed possible suggestions to facilitate the development of IN delivery of stem cells or EVs.

Natural disaster risk monitoring is an important task for disaster prevention and reduction.In the case of immovable cultural relics,however,the feedback mechanism,risk factors,monitoring logic,and monitoring indicators of natural disaster risk monitoring are complex.How to achieve intelligent perception and monitoring of natural disaster risk for immovable cultural relics has always been a focus and a challenge for researchers.Based on the analysis of the concepts and issues related to the natural disaster risk of immovable cultural relics,this paper proposes a framework for natural disaster risk monitoring for immovable cultural relics based on the digital twin.This framework focuses on risk monitoring,including the physical entities of natural disaster risk for immovable cultural relics,monitoring indicators,and virtual entity construction.A platform for monitoring the natural disaster risk of immovable cultural relics is proposed.Using the Puzhou Ancient City Site as a test bed,the proposed concept can be used for monitoring the natural disaster risk of immovable cultural relics at different scales.

Patients with refractory immune thrombocytopenia(ITP)frequently encounter substantial bleeding risks and demonstrate limited responsiveness to existing therapies.Umbilical cord-derived mesenchymal stem cells(UC-MSCs)present a promising alternative,capitalizing on their low immunogenicity and potent immunomodulatory effects for treating diverse autoimmune disorders.This prospective phase I trial enrolled eighteen eligible patients to explore the safety and efficacy of UC-MSCs in treating refractory ITP.The research design included administering UC-MSCs at escalating doses of 0.5×10^(6)cells/kg,1.0×10^(6)cells/kg,and 2.0×10^(6)cells/kg weekly for four consecutive weeks across three cohorts during the dose-escalation phase,followed by a dose of 2.0×10^(6)cells/kg weekly for the dose-expansion phase.Adverse events,platelet counts,and changes in peripheral blood immunity were monitored and recorded throughout the administration and follow-up period.Ultimately,12(with an addition of three patients in the 2.0×10^(6)cells/kg group due to dose-limiting toxicity)and six patients were enrolled in the dose-escalation and dose-expansion phase,respectively.Thirteen patients(13/18,72.2%)experienced one or more treatment emergent adverse events.Serious adverse events occurred in four patients(4/18,22.2%),including gastrointestinal hemorrhage(2/4),profuse menstruation(1/4),and acute myocardial infarction(1/4).The response rates were 41.7%in the dose-escalation phase(5/12,two received 1.0×10^(6)cells/kg per week,and three received 2.0×10^(6)cells/kg per week)and 50.0%(3/6)in the dose-expansion phase.The overall response rate was 44.4%(8/18)among all enrolled patients.To sum up,UC-MSCs are effective and well tolerated in treating refractory ITP(ClinicalTrials.gov ID:NCT04014166).

目的针对当下烟草物流中心条烟分拣机及人工分拣时会产生错烟等问题。从兼顾实时性、识别精度出发,基于YOLOv5s算法提出一种收敛速度更快、准确率更高的条烟识别模型。方法首先在YOLOv5s网络架构中融入CA注意力模块来更好地提取特征,提高模型获取目标位置的准确度;其次将原网络中的最近邻插值上采样算子改为轻量级通用上采样算子CARAFE,获得更大的感受野;然后在骨干网络中嵌入Ghost模块,对网络进行轻量化处理;最后在烟草物流中心搭建条烟图像采集系统,建立条烟图像数据集。结果相较于YOLOv5s,本文提出的优化算法计算量减少了45.8%,mAP@0.5值达到了99.3%,在条烟纠错系统上识别率约为99.9%。结论本文提出的优化算法能够高精度满足高速条烟分拣识别需求。

电动汽车(EV)作为连接交通电气化和电网清洁化的纽带和桥梁,可实现电力-交通-信息之间的深度耦合,形成电力-交通一体化的规划及运行架构。配电-交通融合系统(DTIS)中,EV充放电行为多时空动态交织、电能流-交通流-信息流深度融合、多利益主体动态博弈等大量不确定性因素使配电网的规划及运行优化的边界都将发生重大转变,但同时也给挖掘利用EV的移动储能特性和提升配电网灵活性带来了契机。为此,在分析“人-车-桩-路-网”深度耦合下配电网的形态演化特征基础上,剖析了新形态下配电网协同规划与运行优化所面临的新挑战,进而针对“人-车-桩-路-网”耦合下的EV灵活性建模、灵活域高效构建及预测、协同规划、运行优化4个方面关键技术展开讨论,并对相关技术问题的研究方向进行了展望。

热流计是量化热能转移的仪器。为解决非对称平面热流计、热流传感器校准的难题,基于绝对法原理,对保护板法进行改进,建立一种单侧热绝缘的热流计校准装置,由主加热器、绝热层、保护加热器、功率输出系统、温度反馈调节系统和数据采集系统组成,装置可溯源至标准功率计。在400~1 000 W/m2范围内对装置进行验证,测试结果与传统保护板法热流计校准装置标定结果基本一致。根据测试结果评估电流源偏差、背向和侧向热损失、蓄热损失、测量重复性、面积不一致、装载方式对测量结果的影响。经过实验和分析,该装置的不确定度可以达到3%以内。

水介质中冰-桥墩碰撞动力响应分析

为准确模拟冰-桥墩碰撞过程中墩体结构的动力响应,基于流固耦合(fluid-solid interaction, FSI)的计算方法,运用ANSYS/LS-DYNA软件数值模拟了水介质中冰排与桥墩在不同碰撞影响参数下的动力响应。结果表明:恒定附加质量(constant added mass, CAM)模型相比FSI模型冰载荷计算结果偏大,是因为CAM模型忽略了降低冰排速度的“水垫效应”,但其计算效率高,计算时间是FSI模型的1/10,更有利于桥墩防撞设计,而FSI模型能够更真实地模拟冰-桥墩碰撞场景。综合分析该研究所模拟的不同冰排工况,发现桥墩在冰排撞击作用下呈现明显的冰激结构振动特征,桥墩侧向位移云图随着应力波的传递呈现明显的层状分布,其顶部侧向位移幅度最大,因此在工程设计时应充分考虑其对桥梁整体安全的影响;随着冰排速度、冰排厚度和冰排压缩强度的增大,冰载荷平均值均呈现近似多项式函数关系。此外,研究发现当冰排压缩强度小于2.668 MPa(环境温度高于-15℃)时,冰载荷平均值明显减小,表明环境温度的变化对于桥墩受冰排撞击挤压作用所受的冰害问题具有显著的差异。其仿真研究成果拟为桥梁及桥墩防撞设施设计提供参考。

Hazard risk assessment of tropical cyclones based on joint probability theory

The main hazard-causing factors of tropical cyclones are strong wind,heavy rainfall,and storm surge.Evaluation of the hazard-causing degree of a tropical cyclone requires a joint intensity analysis of these hazard-causing factors.According to the maximum hourly mean wind speed,total rainfall,and maximum tide level at various observation stations in Hong Kong during these tropical cyclones,three hazard-causing indices for tropical cyclones are introduced:the strong-wind index(VI),total-rainfall index(RI),and tide-level index(LI).Through a joint probability analysis of VI,RI,and LI for a tropical cyclone affecting Hong Kong,the joint return period is calculated to evaluate its joint hazard-causing intensity.A limit state function of Hong Kong’s resistance to tropical cyclones is developed and used to evaluate the regional risk of tropical cyclones affecting Hong Kong.The results indicate that the joint return period of VI,RI,and LI can reflect the joint hazard-causing intensity of strong wind,heavy rain,and storm surge caused by tropical cyclones;if the overall design return periods of the regional structures decrease,the regional ability to defend against tropical cyclone disasters is degraded.
