Comparative transcriptomic analysis of rat versus mouse cerebral cortex after traumatic brain injury

The heterogeneity of traumatic brain injury(TBI)-induced secondary injury has greatly hampered the development of effective treatments for TBI *** common processes across species may be an innovative strategy to combat debilitating *** the present study, a cross-species transcriptome comparison was performed for the first time to determine the fundamental processes of secondary brain injury in Sprague-Dawley rat and C57/BL6 mouse models of TBI, caused by acute controlled cortical *** RNA sequencing data from the mouse model of TBI were downloaded from the Gene Expression Omnibus(ID: GSE79441) at the National Center for Biotechnology *** the rat data, peri-injury cerebral cortex samples were collected for transcriptomic analysis 24 hours after *** expressed gene-based functional analysis revealed that common features between the two species were mainly involved in the regulation and activation of the innate immune response, including complement cascades as well as Toll-like and nucleotide oligomerization domain-like receptor *** findings were further corroborated by gene set enrichment ***, transcription factor analysis revealed that the families of signal transducers and activators of transcription(STAT), basic leucine zipper(BZIP), Rel homology domain(RHD), and interferon regulatory factor(IRF) transcription factors play vital regulatory roles in the pathophysiological processes of TBI, and are also largely associated with *** findings suggest that targeting the common innate immune response might be a promising therapeutic approach for *** animal experimental procedures were approved by the Beijing Neurosurgical Institute Animal Care and Use Committee(approval No.201802001) on June 6, 2018.

We theoretically investigate the orientation of the cold KRb molecules in a weak electrostatic field assisted by a shaped laser *** results show that the slow turn-on and rapid turn-off laser pulse can enhance the molecular orientation of KRb significantly,and a high degree of molecular orientation of more than 0.9 can be obtained when the electrostatic field is as low as 3 kV/*** addition,the influences of the electrostatic field strength and the parameters of the laser pulse are studied in detail.

Transition metal sulfides(TMS)hold great promise as anode materials for Li^(+)/Na^(+)***,their practical application still faces several challenges,such as inadequate electrical conductivity,substantial volume changes and a propensity for *** tackle these challenges,a 3D composite structure composed of graphene nanosheets crosslinked core−shell FeS_(2)@N,S co−doped porous carbon(FeS_(2)@NSC/GNs)is created by combining self−template polymerization with the graphene encapsulation *** characterization and analysis demonstrate the effectiveness of the self−template polymerization strategy in generating a porous core−shell structure,which facilitates the uniform dispersion and optimal contact of the FeS_(2) core within the carbon ***,the integration of graphene,alongside the porous carbon shell,introduces a sophisticated dual−protection mechanism against volume expansion and undesirable FeS_(2) ***,the resulting 3D architecture enables efficient electron/ion transport and provides abundant sites for Li^(+)/Na^(+)*** these inherent benefits,the FeS_(2)@NSC/GNs composite exhibits significantly improved lithium/sodium storage performance in comparison to the ***,our proposed approach offers valuable guidance for the construction of advanced anodes for lithium/sodium−ion batteries.

Current opinion and mechanistic interpretation of combination therapy for castration-resistant prostate cancer

Recent advances in genomics technology have led to the massive discovery of new drug targets for prostate cancer;however, none of the currently available therapeutics is curative. One of the greatest challenges is drug resistance. Combinations of therapies with distinct mechanisms of action represent a promising strategy that has received renewed attention in recent years. Combination therapies exert cancer killing functions through either concomitant targeting of multiple pro-cancer factors or more effective inhibition of a single pathway. Theoretically, the combination therapy can improve efficacy and efficiency compared with monotherapy. Although increasing numbers of drug combinations are currently being tested in clinical trials, the mechanisms by which these combinations can overcome drug resistance have yet to be fully understood. The purpose of this review is to summarize recent work on therapeutic combinations in the treatment of castration-resistant prostate cancer and discuss emerging mechanisms underlying drug resistance. In addition, we provide an overview of the current preclinical mechanistic studies on potential therapeutic combinations to overcome drug resistance.

Added value of shear-wave elastography in the prediction of extracapsular extension and seminal vesicle invasion before radical prostatectomy

The purpose of this study was to analyze the value of transrectal shear-wave elastography(SWE)in combination with multivariable tools for predicting adverse pathological features before radical prostatectomy(RP).Preoperative clinicopathological variables,multiparametric magnetic resonance imaging(mp-MRI)manifestations,and the maximum elastic value of the prostate(Emax)on SWE were retrospectively *** accuracy of SWE for predicting adverse pathological features was evaluated based on postoperative pathology,and parameters with statistical significance were *** diagnostic performance of various models,including preoperative clinicopathological variables(model 1),preoperative clinicopathological variables+mp-MRI(model 2),and preoperative clinicopathological variables+mp-MRI+SWE(model 3),was evaluated with area under the receiver operator characteristic curve(AUC)*** was significantly higher in prostate cancer with extracapsular extension(ECE)or seminal vesicle invasion(SVI)with both P<*** optimal cutoff Emax values for ECE and SVI were 60.45 kPa and 81.55 kPa,*** of mp-MRI and SWE improved discrimination by clinical models for ECE(model 2 vs model 1,P=0.031;model 3 vs model 1,P=0.002;model 3 vs model 2,P=0.018)and SVI(model 2 vs model 1,P=0.147;model 3 vs model 1,P=0.037;model 3 vs model 2,P=0.134).SWE is valuable for identifying patients at high risk of adverse pathology.

Synthesis of Iron-containing Carbon Microparticles from Deoiled Asphalt and Ferrocene

The deoiled asphalt as the carbon source and the ferrocene as the metal source and the catalyst precursor were chosen to synthesize iron-containing carbon microparticles through co-carbonization at the temperature of about 450℃ for 3 h. The resulting products were treated at 2 000 ℃ for 2 h. All samples were examined by high resolution transmission electron microscopy （HRTEM） and X-ray diffraction （XRD）. The results show that the iron particles in the heat-treated material are completely coated by carbon. In addition to the fully filled carbon microparticles as well as hollow carbon ones, also form carbon fibers with hollow centers. The formation mechanism of the as-prepared products was discussed briefly.

Prognostic values of apoptosis-stimulating P53-binding protein 1 and 2 and their relationships with clinical characteristics of esophageal squamous cell carcinoma patients:a retrospective study

Background: Esophageal squamous cell carcinoma(ESCC) is a leading cause of cancer?related death, and new prognostic biomarkers are urgently needed. Apoptosis?stimulating P53?binding protein 1(ASPP1) and 2(ASPP2) have been reported to play important roles in the development, progression, metastasis, and prognosis of cancers, but their roles in ESCC have not been elucidated. In this study, we examined the expression of ASPP1 and ASPP2 in ESCC to evaluate their prognostic ***: The protein expression of ASPP1, ASPP2, and P53 in 175 specimens of ESCC was detected using immuno?histochemical staining; their expression in cancerous and noncancerous tissues was scored according to the stain?ing intensity and the percentage of stained cells. The associations of ASPP1, ASPP2, and P53 with clinicopathologic parameters, overall survival(OS), and disease?free survival(DFS) were ***: The protein expression levels of ASPP2 and P53 were significantly higher in cancerous tissues than in paired noncancerous tissues(P < 0.001), whereas the expression levels of ASPP1 in the two groups were similar. In ESCCs, ASPP1 expression was significantly associated with histological differentiation(P = 0.002) and invasive depth(P = 0.014); ASPP2 expression was associated with age(P = 0.029) and histological differentiation(P < 0.001); and P53 expression was associated with age(P and P53 expression. Survival an= 0.021) and tumor size(P alysis revealed that high AS= 0.040). No correlations were found between ASPP1, ASPP2,PP2 expression was significantly associated with increased 5?year OS(P = 0.001) and DFS rates(P ate of ESCC patients(= 0.010) and that high P53 expression was significantly associated with a reduced 5?year DFS rP atio(HR): 0.541, 9= 0.015). Multivariate Cox analysis indicated that ASPP2 was an inde?pendent predictor of OS [hazard r5% confidence interval(CI) 0.363–0.804] and DFS(HR: 0.599, 95% CI 0.404–0.888) of ESCC patients and that P53 was an independent pre
