Murine hepatocellular carcinoma derived stem cells reveal epithelial-to-mesenchymal plasticity

AIM To establish a model to enrich and characterize stemlike cells from murine normal liver and hepatocellular carcinoma(HCC) cell lines and to further investigate stem-like cell association with epithelial-to-mesenchymal transition(EMT).METHODS In this study,we utilized a stem cell conditioned serumfree medium to enrich stem-like cells from mouse HCC and normal liver cell lines,Hepa 1-6 and AML12,*** isolated the 3-dimensional spheres and assessed their stemness characteristics by evaluating theRNA levels of stemness genes and a cell surface stem cell marker by quantitative reverse transcriptase-PCR(q RTPCR).Next,we examined the relationship between stem cells and EMT using q *** Three-dimensional spheres were enriched by culturing murine HCC and normal hepatocyte cell lines in stem cell conditioned serum-free medium supplemented with epidermal growth factor,basic fibroblast growth factor and heparin *** 3-dimensional spheres had enhanced stemness markers such as Klf4 and Bmi1 and hepatic cancer stem cell(CSC) marker Cd44 compared to parental cells grown as adherent *** report that epithelial markers E-cadherin and ZO-1 were downregulated,while mesenchymal markers Vimentin and Fibronectin were upregulated in 3-dimensional *** 3-dimensional spheres also exhibited changes in expression of Snai,Zeb and Twist family of EMT transcription *** Our novel method successfully enriched stem-like cells which possessed an EMT *** isolation and characterization of murine hepatic CSCs could establish a precise target for the development of more effective therapies for HCC.

＜正＞This paper describes a Japanese-to-Chinese spoken language translation （SLT） method based on simple expression and presents the experimental results. The method is aimed at developing a compact speech translation system, which is robust for spontaneous spoken language phenomena, including the recognition errors and different expression from various speakers, The idea of translation method based on simple expression is that the mechanism interprets speech-act rather than the direct translation of the speaker’s words. The method is realized by mapping the simple expression instead of deep parsing. In this method, only keywords in speech recognition results are extracted, and the corresponding target sentences are extracted from the database by selecting similar example sentences with the keywords. All training sentences in the bilingual corpus are rewritten into the simple expression and grouped by rules. The rules are specially designed to develop the compact size example database for translation. Comparing with other example-based approaches to SLT, the method based on simple expression is easy to realize, and especially it is practical to develop the SLT systems limited in specific domains.

Endoscopic submucosal dissection of a rectal carcinoid tumor using grasping type scissors forceps

Endoscopic submucosal dissection （ESD） with a knife is a technically demanding procedure a ssociated with ahigh complication rate. The shortcomings of this meth-od are the inability to fix the knife to the target lesion,and compression of the lesion. These can lead to major complications such as perforation and bleeding. To reduce the risk of complications related to ESD, we developed a new grasping type scissors forceps （GSF）,which can grasp and incise the targeted tissue using electrosurgical current. Colonoscopy on a 55-year-old woman revealed a 10-ram rectal submucosal *** histological diagnosis of the specimen obtained by biopsy was carcinoid tumor. Endoscopic ultrasonog-raphy demonstrated a hypoechoic solid tumor limitedto the submucosa without lymph node involvement. Itwas safely and accurately resected without unexpectedincision by ESD using a GSF. No delayed hemorrhage or perforation occurred. Histological examination confirmed the carcinoid tumor was completely excisedwith negative resection margin.

Augmentation of tumor necrosis factor family-induced apoptosis by E3330 in human hepatocellular carcinoma cell lines via inhibition of NFκB

AIM- To investigate the reduction of cell viability in human hepatocellular carcinoma （HCC） cell lines induced by inhibition of nuclear factor kB （NFkB）. METIIOI）S： HLE, SKHep1, and HepG2 were incubated and E3330 was used to compare the stimulation of some chemotherapeutic drugs with that of TNF family, Fas ligand, TNF（x and TNF-related apoptosis-inducing ligand （TRAIL） at the point of the reduction of cell viability by inhibiting NFkB. RESULTS： E3330 decreased NFKB levels in HLE cells stimulated by TNF and TRAIL. The cytotoxicity of the combination of TRAIL, TNFa, Fas ligand, and E3330 increased synergistically in a dose-dependent manner compared to either E3330 alone in all HCC cell lines by MTT assay. However, the combination of some chemotherapeutic drugs and E3330 did not decrease the cell viability. CONCLUSION： Inhibition of NFd3 sensitizes human HCC cell lines to TNF-mediated apoptosis including TRAIL, and TRAIL-based tumor therapy might be a powerful potential therapeutic tool in the treatment of human HCC.

Risk factors for the recurrence of hepatocellular carcinoma after radiofrequency ablation of hepatocellular carcinoma in patients with hepatitis C

AIM: To analyze the risk factors of hepatocellular carcinoma (HCC) recurrence after radiofrequency ablation (RFA) treatment with HCV-associated hepatitis. METHODS: Twenty-six patients with HCV-associated HCC who were followed-up for more than 12 mo were selected for this study. Risk factors for distant intrahepatic recurrences of HCC were evaluated for patients in whom complete coagulation was achieved without recurrence in the same subsegment as the primary nodule. Twelve clinical and tumoral factors were examined: Age, gender, nodule diameter, number of primary HCC nodule, Child-Pugh classification, serum platelet, serum albumin, serum AST, post RFA AST, serum ALT, post RFA ALT, post RFA ***: Distant recurrences of HCC in remnant liver after RFA were observed in 14 cases and in the number of primary HCC nodules (P = 0.047), and the serum platelets (P = 0.030), the clear difference came out by the recurrence group and the non-recurrence group. The cumulative recurrence rates after 1 and 2 years were30.8% and 86.8%, respectively for primary multinodular HCC, and 15.4% and 29.5% respectively, for primary uninodular HCC. In addition the 1-year recurrence rates for patients with serum albumin more than 3.4 g/dL and less than 3.4 g/dL were 23.1% for both, but the 2-years recurrence rates were 89.0% and 23.1%, respectively. The number of primary HCC nodules (relative risk, 6.970; P = 0.016) were found to be a statistically significant predictor for poor distant intrahepatic recurrence by univariate ***: Patients who have multiple HCC nodules, low serum platelets and low serum albumin accompanied by HCV infection, should be carefully followed because of the high incidence of new HCC lesions in the remnant liver, even if coagulation RFA is complete.

Analgesic Efficacy of the Erector Spinae Plane (ESP) Block for Pneumothorax Surgery: A Retrospective Study

Background: Poor perioperative pain management during pneumothorax surgery leads to respiratory complications in the post-operative period. The erector spinae plane (ESP) block technique has been shown to be able to block the thoracic spinal nerves. Therefore, the ESP block may provide effective analgesic during thoracic surgery. We have retrospectively investigated the effectiveness of the ESP block for postoperative pain management in pneumothorax surgery. Patients and Methods: Patients who underwent pneumothorax surgery in 2017 were selected for the study. The primary outcome was assessed using the numeric pain rating (NRS) scales until the morning of the second post-operative day. The secondary outcomes were the cumulative amount of additional intravenous fentanyl administration until the morning of the second post-operative day. Results: This retrospective study included 29 patients who underwent pneumothorax surgery. Of these patients, 13 patients received only general anaesthesia (control group), while the other 16 patients received the ESP block in addition to general anaesthesia (study group). Compared to the control group, the study group did not show lower NRS scores at 1, 2, 4, 6, 12, and 24 hours post-surgery (P = 0.09, 0.17, 0.06, 0.36, 0.47, and 0.71). As for the cumulative amount of additional fentanyl, there were also no significant differences between the both groups. Conclusions: The ESP block could not provide effective analgesia for the 24 hours post-surgery period in patients undergoing pneumothorax surgery.

Selective changes in the <i>α</i>-adrenoceptor-mediated contraction in the senescent rat urinary bladder

The urinary bladder is innervated and functionally regulated by the autonomic nervous system. In order to elucidate the mechanism of functional changes in aged rat urinary bladder, we studied the influence of senescence on, 1) the α-adrenergic contractile response to phenylephrine in the urinary bladder body and trigone, 2) the muscarinic contractile response to carbachol in the body and trigone. The binding characteristics of [3H]quinuclidinyl benzilate (QNB) to muscarinic cholinoceptors were compared in young and aged bladder. Bladders from young (2 - 3 month-old) and aged (27 month-old) male Fischer 344 rats were isolated, cut into strips and mounted in the organ bath, then the developed tension was recorded. Histologically, the aged bladder did not show pathologic changes such as inflammation and hypertrophy. Carbachol-induced contraction in aged rat bladder was identical to that obtained in young rat. In the receptor binding assay, [3H]QNB maximal binding capacity and Kd value were not significantly changed in aged bladder. In contrast, a selective α-adrenergic agonist phenylephrine, elicited greater contractions both in the aged body and trigone than those in young rats. The augmentation of α-adrenoceptor-mediated contractions in aged bladder may induce urinary dysfunction such as voiding difficulty.

White-opaque Switching in Different Mating Type-like Locus Gene Types of Clinical Candida albicans Isolates

Background：Candida albicans （***） can become a pathogen causing superficial as well as life-threatening systemic infections,especially in immunocompromised *** phenotypic attributes contribute to its capacity to colonize human *** our study,93 *** isolates from patients of various candidiasis in a hospital of China were *** aimed to investigate the white-opaque （WO） switching competence,drug sensitivity,and virulence of mating type-like （MTL） a/α ***：Internal transcribed spacer （ITS） gene and the MTL configuration were detected in all the isolates by reverse transcription-polymerase chain ***/opaque phenotype and doubling time of cell growth were *** minimum inhibitory concentrations of antifungal agent were measured using broth microdilution ***：Sixty-four isolates （69.6%） were classified to serotype A,19 （20.6%） to serotype B,and 9 （9.8%） to serotype ***,phylogenetic analysis showed that these isolates were divided into four different subgroups of ITS *** of our clinical isolates were MTLa/α type,while 6.8% remained MTLa or MTLα *** frequency of opaque phenotype was 71.0% （66 isolates）.Following the guidelines of Clinical and Laboratory Standards Institute M27-A3,all isolates were susceptible to caspofungin and a few （0.6-3.2%） of them showed resistance against amphotericin B,flucytosine,fluconazole,itraconazole,and ***：From these analyses,there were comparatively more *** strains classified into serotype B,and the frequency of opaque phase strains was significant in the clinical isolates from ***,phenotypic,or drug susceptibility patterns were not significantly different from previous ***/α isolates could also undergo WO switching which facilitates their survival.

The University-City Interface: Plazas and Boulevards

It has become an increased challenge for designers to define the boundaries between the university and its surrounding city. The amount of space serving as a nexus between universities and urban areas is gradually increasing. This study defines such intermediate spaces as “the university-city interface”—areas that influence the university’s physical and functional connection to the surrounding city. The research presents comparative case studies of three universities in urban contexts—Harvard University, University of Pennsylvania, and Université Catholique de Louvain— by analyzing plazas and boulevards. These representative open spaces provide interfaces for both the university campuses and their surrounding cities. This paper analyzes design elements from the perspective of campus spatial structure, as well as locations and functions, to develop a comparative checklist for plazas and boulevards. The results offer a set of urban design principles for university plazas and boulevards that could significantly improve the quality of the university-city relationship. While these checklists and principles may vary depending on circumstances, they can be useful starting points for initializing design processes.
