BACKGROUND Single nucleotide polymorphisms(SNPs)are universally present in nucleotide excision repair(NER)pathway genes,which could make impacts on colorectal carcinogenesis and *** To explore the association of all tagSNPs in NER pathway genes with colorectal cancer(CRC)risk and prognosis in a northern Chinese population by a two-stage case-control design composed of a discovery and validation *** Genotyping for NER SNPs was performed using kompetitive allele specific *** the discovery stage,39 tagSNPs in eight genes were genotyped in 368 subjects,including 184 CRC cases and 184 individual-matched *** the validation stage,13 SNPs in six genes were analyzed in a total of 1712 subjects,including 854 CRC cases and 858 CRC-free *** Two SNPs(XPA rs10817938 and XPC rs2607775)were associated with an increased CRC risk in overall and stratification *** cumulative and interaction effects were also demonstrated in the studied SNPs on CRC *** two SNPs(ERCC2 rs1052555 and ERCC5 rs2228959)were newly found to be associated with a poor overall survival of CRC *** Our findings suggest novel SNPs in NER pathway genes that can be predictive for CRC risk and prognosis in a large-scale Chinese *** present study has referential values for the identification of all-round NER-based genetic biomarkers in predicting the susceptibility and clinical outcome of CRC.

AIM: To explore the essential characteristics of serum pepsinogen (PG) levels in Chinese people, by analyzing the population-based data on the serum levels of PG Ⅰ and Ⅱ and the PGⅠ/Ⅱ ratio, and their influencing factors in Chinese from North China. METHODS: A total of 6990 subjects, who underwent a gastric cancer screening in North China from 1997 to 2002, were collected in this study. Serum pepsinogen levels were measured by enzyme-linked immunosorbent assay (ELISA). H pylori status was determined by histological examination and H pylori-IgG ELISA. The cut-off point was calculated by using receiving operator characteristics (ROC) curves. Factors linked to serum PG Ⅰ/Ⅱ ratio were identified using a multivariate logistic regression. RESULTS: The serum PGⅠ and PGⅡ levels were significantly higher in males than in females (95.2 μg/L vs 79.7 μg/L, P < 0.01; 12.1 μg/L vs 9.4 μg/L, P < 0.01), PGⅠ/Ⅱ ratio was significantly lower in males than in females (7.9 vs 8.3, P < 0.01). The PG Ⅰ/Ⅱ ratio decreased significantly in the aged groups following the progression of gastric mucosa from normal to non-atrophic and atrophic lesions (10.4, 8.8, and 6.6, respectively). The serum PGⅠand Ⅱ levels were significantly higher in patients with H pylori infection than in those without H pylori infection (88.7 μg/L vs 81.4 μg/L, P < 0.01; 11.4 μg/L vs 8.4 μg/L, P < 0.01), while the PGⅠ/Ⅱ ratio was significantly lower in patients with H pylori infection than in those without H pylori infection (7.7 vs 9.6, P < 0.01). For patients with atrophic lesions, the area under the PGⅠ/Ⅱ ROC curve was 0.622. The best cut-off point for PGⅠ/Ⅱ was 6.9, with a sensitivity of 53.2%, and a specificity of 67.5%. Factors linked to PGⅠ/Ⅱ were sensitive to identified PG using a multinomial logistic regression relying on the following inputs: males (OR: 1.151, 95% CI: 1.042-1.272, P = 0.006), age ≥ 61 years (OR: 1.358, 95% CI: 1.188-1.553, P = 0.000), atrophic lesion (OR: 2.075, 95% CI: 1.870-2.302, P = 0.000)

Objective： In cancer prevention, the targeting of precancerous lesions has been recognized as the most promising method. However, little attention has been paid to the risk factors of precancerous gastric lesions, especially in rural China where there is high prevalence of precancerous gastric lesions. We therefore conducted a cross-sectional study in Liaoning province, China, to investigate the potential risk and protective factors of these precancerous gastric lesions. Methods： A total of 1,179 subjects with high risk of gastric cancer from Zhuanghe County were included in this study. Standard questionnaires were used in collecting epidemiological factors and the data were then analyzed by the unconditional logistic regression model. Results： Smoking and drinking were the risk factors for the precancerous gastric lesions among rural subjects, and the association of smoking or drinking and the precancerous gastric lesions increased in strength with the daily consumption and duration. As the factors such as age, gender, smoking, alcohol were controlled, a multivariable analysis revealed that there was a significant correlation between the deep-fry food intake and the gastric epithelial dysplasia with the odds ratio （OR） of 1.78 [95% confidence interval （CI）： 1.01-3.12]. Garlic eating was shown to confer protection against the development of gastric ulcer （OR=0.55, 95% CI： 0.33-0.92）. Conclusion： Smoking and drinking were the risk factors for the precancerous gastric lesions among rural subjects. Deep-fry food intake might be one of the risk factors for the precancerous gastric lesions and garlic eating was shown to confer protection against the development of gastric ulcer among rural Chinese population.

Objective： To investigate the expressions of E-cadherin, β-catenin, transcription factor 4 （TCF4）, which are related with Wnt/β-catenin signaling pathway, and Caudal-type Homeobox Protein 2 （CDX2） originated from intestine, in various gastric diseases and its influence on gastric carcinogenesis and differentiation. Methods： A total of 319 various gastric diseases samples from gastroscopy were detected in situ using immunohistochemistry, and data were analyzed with Х^2 test and Kendall＇s tau-b rank correlation coefficient analysis. Results： Along with the progression from superficial gastritis （SG） to atrophic gastritis （AG） to gastric cancer （GC）, the expressions of E-cadherin and β-catenin appeared a decreasing tendency in total, while β-catenin emerged a decreasing tendency in the cell membrane and an increasing in the nucleus, and the highly positive expression rate of intranuclear transcription factor TCF4 appeared an increasing tendency. The transcription factor CDX2 had a higher expression in AG and GC than that in SG. The expression rates of the four proteins above in intestinal type adenocarcinoma were all higher than those in diffuse type carcinoma. There were positive correlations between E-cadherin and β-catenin expressed in cytoplasm, between β-catenin expressed in cell membrane and β-catenin in cytoplasm, between β-catenin expressed in cytoplasm and β-catenin in nucleus, between β-catenin and TCF4 in nucleus, between TCF4 and CDX2 in nucleus, respectively （P〈0.05）. And β-catenin and CDX2 had no relevance in nucleus （P〉0.05）. Conclusion： We found that the four proteins were involved in gastric carcinogenesis and the differentiation to intestinal type adenocarcinoma.

AIM： To determine the effect of allitridi on cell cycle of human gastric cancer （HGC） cell lines MGC803 and SGC7901 and its possible mechanism. METHODS： Trypan blue dye exclusion was used to evaluate the proliferation, inhibition of cells and damages of these ceils were detected with electron microscope, Flow cytometry and cell mitotic index were used to analyze the change of cell cycle, immunohistochemistry, and RT- PCR was used to examine expression of the p21^WAF1 gene, RESULTS： MGC803 cell growth was inhibited by allitridi with 24 h ICso being 6.4μg/mL, SGC7901 cell growth was also inhibited by allitridi with 24 h IC50 being 7.3μg/mL, After being treated with allitridi at the concentration of 12μg/mL for 24 h, cells were found to have direct cytotoxic effects, including broken cellular membrane, swollen and vesiculated mitochondria and rough endoplasmic reticula, and mass lipid droplet, When cells were treated with allitridi at the concentration of 3, 6, and 9μg/mL for 24 h, the percentage of Go/G1 phase cells was decreased and that of G2/M phase cells was significantly increased （P = 0.002） compared with those in the group, When cells were treated with allitridi at the concentration of 6μg/mL, cell mitotic index was much higher （P = 0.003） than that of control group, indicating that allitridi could cause gastric cancer cell arrest in M phase, Besides, the expression levels of p21^WAF1 gene of MGC803 cells and p21^WAF1 gene of SGC7901 cells were remarkably upregulated after treatment, CONCLUSION： Allitridi can cause gastric cancer cell arrest in M phase, and this may be one of the mechanisms for inhibiting cell proliferation, Effect of allitridi on cells in M phase may be associated with the upregulation of p21^WAF1 genes. This study provides experimental data for clinical use of allitridi in the treatment of gastric carcinoma.

AIM: To study the relationship between Helicobacter pylori (H. pylori) and gastric carcinoma and its possible pathogenesis by H. pylori. METHODS: DNEL technique and immunohistochemical technique were used to study the state of apoptosis, proliferation and p53 gene expression. A total of 100 gastric mucosal biopsy specimens, including 20 normal mucosa, 30 H. pylori-negative and 30 H. pylori-positive gastric precancerous lesions along with 20 gastric carcinomas were studied. RESULTS: There were several apoptotic cells in the superficial epithelium and a few proliferative cells within the neck of gastric glands, and no p53 protein expression in normal mucosa. In gastric carcinoma, there were few apoptotic cells, while there were a large number of proliferative cells, and expression of p53 protein significantly was increased. In the phase of metaplasia, the apoptotic index (AI, 4.36%+/-1.95%), proliferative index (PI, 19.11%+/-6.79%) and positivity of p53 expression (46.7%) in H. pylori-positive group were higher than those in normal mucosa (P<0.01). AI in H. pylori-positive group was higher than that in H. pylori-negative group (3.81%+/-1.76%), PI in H. pylori-positive group was higher than that in H. pylori-negative group (12.25%+/-5.63%, P<0.01). In the phase of dysplasia, AI (2.31%+/-1.10%) in H. pylori-positive group was lower (3.05%+/-1.29%) than that in H. pylori-negative group, but PI (33.89%+/-11.65%) was significantly higher (22.09+/-8018%, P<0.01). In phases of metaplasia, dysplasia and gastric cancer in the H. pylori-positive group, AIs had an evidently graduall decreasing trend (P<0.01), while PIs had an evidently gradual increasing trend (P<0.05 or P<0.01), and there was also a trend of gradual increase in the expression of p53 gene. CONCLUSION: In the course of the formation of gastric carcinoma, proliferation of gastric mucosa can be greatly increased by H. pylori, and H. pylori can induce apoptosis in the phase of metaplasia, but in th

Non-obstructive azoospermia(NOA),which is defined as the absence of spermatozoa in the ejaculate secondary to impaired spermatogenesis within the testis,may be caused by a variety of etiologies,including varicocele-induced testicular damage,cryptorchidism,prior testicular torsion,post-pubertal mumps orchitis,gonadotoxic effects from medications,genetic abnormalities,chemotherapy/radiation,and other unknown causes currently classified as idiopathic(Cocuzza et al.,2013).The microdissection testicular sperm extraction(micro-TESE)technique involves a meticulous microsurgical exploration of the testicular parenchyma to identify and selectively extract larger seminiferous tubules that carry a higher probability of complete spermatogenesis(Schlegel,1999).
