Cellular strategies to induce immune tolerance after liver transplantation:Clinical perspectives

liver transplantation(LT)has become the most efficient treatment for pediatric and adult end-stage liver disease and the survival time after transplantation is becoming longer due to the development of surgical techniques and perioperative ***,long-term side-effects of immunosuppressants,like infection,metabolic disorders and malignant tumor are gaining more *** tolerance is the status in which LT recipients no longer need to take any immunosuppressants,but the liver function and intrahepatic histology maintain *** approaches to achieve immune tolerance after transplantation include spontaneous,operational and induced *** first two means require no specific intervention but withdrawing immunosuppressant gradually during *** clinical factors or biomarkers so far could accurately predict who are suitable for immunosuppressant withdraw after *** the understanding to the underlying mechanisms of immune tolerance,many strategies have been developed to induce tolerance in LT *** strategy is one of the most promising methods for immune tolerance induction,including chimerism induced by hematopoietic stem cells and adoptive transfer of regulatory immune *** safety and efficacy of various cell products have been evaluated by prospective preclinical and clinical trials,while obstacles still exist before translating into clinical ***,we will summarize the latest perspectives and concerns on the clinical application of cellular strategies in LT recipients.

Effect of wall-disruption on nutrient composition and in vitro digestion of camellia and lotus bee pollens

The nutrient digestion,absorption and biological activity of bee pollen may be limited due to the complex pollen ***,the effect of superfine grinding technology on the release of nutrients from bee pollen were investigated,and their antioxidant activities and in vitro digestion were explored in this *** showed that the content of nutrients in bee pollen increased after wall *** them,fat content increased by 22.55%-8.31%,protein content increased by 0.54%-4.91%,starch content increased by 36.31%-48.64%,soluble sugar content increased by 20.57%-29.67%,total phenolic acid content increased by 11.73%-86.98%and total flavonoids content increased by 14.29%-24.79%.At the same time,the antioxidant activity increased by 14.84%-46.00%.Furthermore,the active components such as phenolic compounds in the wall-disruption bee pollen were more readily to be released during the in vitro digestion,and easier to be absorbed because of their higher *** activities during in vitro digestion were also improved in walldisruption bee *** findings provide evidence that bee pollen wall disruption was suggested,thus,it is more conducive to exerting the value of bee pollen in functional foods.

Elaidic acid-induced intestinal barrier damage led to gut-liver axis derangement and triggered NLRP3 inflammasome in the liver of SD rats

Previous studies have shown that trans fatty acids(TFA) are associated with several chronic diseases,the gut microbiota is directly influenced by dietary components and linked to chronic *** research investigated the effects of elaidic acid(EA),a typical TFA,on the gut microbiota to understand the underlying mechanisms of TFA-related chronic diseases.16S rDNA gene sequencing on faecal samples from Sprague-Dawley rats were performed to explore the composition change of the gut microbiota by EA gavage for 4 *** results showed that the intake of EA increased the abundance of well-documented harmful bacteria,such as Proteobacteria,Anaerotruncus,Oscillibacter and ***,EA induced translocation of lipopolysaccharides(LPS) and the above pathogenic bacteria,disrupted the intestinal barrier,led to gut-liver axis derangement and TLR4 pathway activation in the ***,EA induced intestinal barrier damage and regulated TLR4-MyD88-NF-κB/MAPK pathways in the liver of SD rats,leading to the activation of NLRP3 inflammasome and inflammatory liver damage.

Macrocyclic trichothecenes from Myrothecium verrucaria PA 57 and their cytotoxic activity

Four novel macrocyclic trichothecenes,termed mytoxins D-G(1-4),along with four known analogs(5-8),were isolated from the ethyl acetate extract of fermented rice inoculated with the fungus Myrothecium verrucaria *** compound features a tricyclic 12,13-epoxytrichothec-9-ene(EPT)***,mytoxin G(4)represents the first instance of a macrocyclic trichothecene incorporating a glucosyl unit within the trichothecene *** structures of the newly identified compounds were elucidated through comprehensive spectroscopic analysis combined with quantum chemical *** isolated compounds demonstrated cytotoxic activity against the CAL27 and HCT116 cell lines,which are models for human oral squamous cell carcinoma and colorectal cancer,***,mytoxin D(1)and mytoxin F(3)exhibited pronounced cytotoxic effects against both cancer cell lines,with IC_(50)values ranging from 3 to 6 nmol·L^(-1).Moreover,compounds 1 and 3 were found to induce apoptosis in HCT116 cells by activating caspase-3.

Predicting submerged vegetation drag with a machine learning-based method

Accurate estimation of the drag forces generated by vegetation stems is crucial for the comprehensive assessment of the impact of aquatic vegetation on hydrodynamic processes in aquatic *** coupling relationship between vegetation layer flow velocity and vegetation drag makes precise prediction of submerged vegetation drag forces particularly *** present study utilized published data on submerged vegetation drag force measurements and employed a genetic programming(GP)algorithm,a machine learning technique,to establish the connection between submerged vegetation drag forces and flow and vegetation *** using the bulk velocity,U,as the reference velocity scale to define the drag coefficient,C_(d),and stem Reynolds number,the GP runs revealed that the drag coefficient of submerged vegetation is related to submergence ratio(H^(*)),aspect ratio(d^(*)),blockage ratio(ψ^(*)),and vegetation density(λ).The relation between vegetation stem drag forces and flow velocity is implicitly embedded in the definition of C_(d).Comparisons with experimental drag force measurements indicate that using the bulk velocity as the reference velocity,as opposed to using the vegetation layer average velocity,U_(v),eliminates the need for complex iterative processes to estimate U_(v)and avoids introducing additional errors associated with U_(v)*** approach significantly enhances the model’s predictive capabilities and results in a simpler and more user-friendly formula expression.

A fast and high-accurate commutation failure identification method for LCC-HVDC system

Commutation failure (CF) is one of the most common issues in line-commuted converter-based high voltage direct current systems (LCC-HVDC), leading to critical power system security and stability problems. Accurate and rapid identification of CF is crucial to prevent subsequent CF in HVDC systems. However, the existing CF identification methods are lack of the required accuracy and speed. The relationship between bridge arm current, commutation voltage, and trigger pulse based on the commutation progress mechanism after the AC fault is analysed in this paper. A novel CF identification method is presented, which utilises the CF identification factor, enabling fast and high-accurate identification without relying on the value of the extinction angle. The proposed method offers a simpler and more efficient implementation in engineering practice. Finally, the effectiveness of the proposed method is verified in both the standard International Council on Large Electric systems HVDC model and the Hardware in Loop test using practical project parameters. The results demonstrate that the proposed method can accurately and fast identify CF.

On Robust Cross-view Consistency in Self-supervised Monocular Depth Estimation

Remarkable progress has been made in self-supervised monocular depth estimation (SS-MDE) by exploring cross-view consistency, e.g., photometric consistency and 3D point cloud consistency. However, they are very vulnerable to illumination variance, occlusions, texture-less regions, as well as moving objects, making them not robust enough to deal with various scenes. To address this challenge, we study two kinds of robust cross-view consistency in this paper. Firstly, the spatial offset field between adjacent frames is obtained by reconstructing the reference frame from its neighbors via deformable alignment, which is used to align the temporal depth features via a depth feature alignment (DFA) loss. Secondly, the 3D point clouds of each reference frame and its nearby frames are calculated and transformed into voxel space, where the point density in each voxel is calculated and aligned via a voxel density alignment (VDA) loss. In this way, we exploit the temporal coherence in both depth feature space and 3D voxel space for SS-MDE, shifting the “point-to-point” alignment paradigm to the “region-to-region” one. Compared with the photometric consistency loss as well as the rigid point cloud alignment loss, the proposed DFA and VDA losses are more robust owing to the strong representation power of deep features as well as the high tolerance of voxel density to the aforementioned challenges. Experimental results on several outdoor benchmarks show that our method outperforms current state-of-the-art techniques. Extensive ablation study and analysis validate the effectiveness of the proposed losses, especially in challenging scenes. The code and models are available at https://***/sunnyHelen/RCVC-depth.
