Targeting the adenosine A2A receptor for neuroprotection and cognitive improvement in traumatic brain injury and Parkinson's disease

Adenosine exerts its dual functions of homeostasis and neuromodulation in the brain by acting at mainly 2 G-protein coupled receptors,called A1 and A2A *** adenosine A2A receptor(A2AR)antagonists have been clinically pursued for the last 2 decades,leading to final approval of the istradefylline,an A2AR antagonist,for the treatment of OFF-Parkinson's disease(PD)*** approval paves the way to develop novel therapeutic methods for A2AR antagonists to address 2 major unmet medical needs in PD and traumatic brain injury(TBI),namely neuroprotection or improving *** this review,we first consider the evidence for aberrantly increased adenosine signaling in PD and TBI and the sufficiency of the increased A2AR signaling to trigger neurotoxicity and cognitive *** further discuss the increasing preclinical data on the reversal of cognitive deficits in PD and TBI by A2AR antagonists through control of degenerative proteins and synaptotoxicity,and on protection against TBI and PD pathologies by A2AR antagonists through control of ***,we provide the supporting evidence from multiple human prospective epidemiological studies which revealed an inverse relation between the consumption of caffeine and the risk of developing PD and cognitive decline in aging population and Alzheimer's disease ***,the convergence of clinical,epidemiological and experimental evidence supports the validity of A2AR as a new therapeutic target and facilitates the design of A2AR antagonists in clinical trials for disease-modifying and cognitive benefit in PD and TBI patients.

Adequate vascularization is a critical determinant for the successful construction and clinical implementation of complex organotypic tissue models. Currently, low cell and vessel density and insufficient vascular maturation make vascularized organotypic tissue construction difficult,greatly limiting its use in tissue engineering and regenerative medicine. To address these limitations, recent studies have adopted pre-vascularized microtissue assembly for the rapid generation of functional tissue analogs with dense vascular networks and high cell density. In this article, we summarize the development of module assembly-based vascularized organotypic tissue construction and its application in tissue repair and regeneration, organ-scale tissue biomanufacturing, as well as advanced tissue modeling.

Deep genome skimming reveals the hybrid origin of Pseudosasa gracilis (Poaceae: Bambusoideae)

Pseudosasa gracilis(Poaceae:Bambusoideae)is a temperate woody bamboo species endemic to Southcentral China with a narrow *** phylogenetic studies revealed an unexpected,isolated phylogenetic position of *** we conducted phylogenomic analysis by sampling populations of *** and its sympatric species *** and Sinosasa polytricha reflecting different genomic signals,by deep genome *** molecular evidence from chloroplast genes and genome-wide SNPs,we deciphered the phylogenetic relationships of *** plastid and nuclear data indicate that *** is more closely related to Sinosasa,which is discordant with the taxonomic *** further explore this molecular-morphological conflict,we screened 411“perfect-copy”syntenic genes to reconstruct phylogenies using both the concatenation and coalescent *** observed extensive discordance between gene trees and the putative species tree.A significant hybridization event was detected based on 411 genes from the D subgenome,showing *** was a hybrid descendant between Sinosasa longiligulata and ***,with 63.56%and 36.44%inheritance probabilities of each ***,introgression events were detected in the C subgenome between *** and *** in the same distribution *** findings suggest that sympatric hybridization and introgression play a crucial role in the origin of *** providing an empirical example of bamboo of hybrid origin using comprehensive analyses based on genomic data from different inheritance systems and morphological characters,our study represents a step forward in understanding of reticulate evolution of bamboos.

Effects of diamond particle size on microstructure and properties of diamond/Al-12Si composites prepared by vacuum-assisted pressure infiltration

Diamond/aluminium composites have attracted attention in the field of thermal management of electronic packaging for their excellent *** order to solve the interfacial problem between diamond and aluminium,a novel process combining pressure infiltration with vacuum-assisted technology was proposed to prepare diamond/aluminum *** effect of diamond particle size on the microstructure and properties of the diamond/Al-12Si composites was *** results show that the diamond/Al-12Si composites exhibit high relative density and a uniform *** thermal conductivity and coefficient of thermal expansion increase with increasing particle size,while the bending strength exhibits the opposite *** the average diamond particle size increases from 45μm to 425μm,the thermal conductivity of the composites increases from 455 W·m^(-1)·K^(-1)to 713 W·m^(-1)·K^(-1)and the coefficient of thermal expansion increases from 4.97×10^(-6)K^(-1)to 6.72×10^(-6)K^(-1),while the bending strength decreases from 353 MPa to 246 *** research demonstrates that high-quality composites can be prepared by the vacuum-assisted pressure infiltration process and the thermal conductivity of the composites can be effectively improved by increasing the diamond particle size.

Reinforcement corrosion is the main cause of performance deterioration of reinforced concrete(RC)*** research has been performed to investigate the life-cycle cost(LCC)of coastal bridge piers with nonuniform corrosion using different *** this study,a reliability-based design optimization(RBDO)procedure is improved for the design of coastal bridge piers using six groups of commonly used materials,i.e.,normal performance concrete(NPC)with black steel(BS)rebar,high strength steel(HSS)rebar,epoxy coated(EC)rebar,and stainless steel(SS)rebar(named NPC-BS,NPC-HSS,NPC-EC,and NPC-SS,respectively),NPC with BS with silane soakage on the pier surface(named NPC-Silane),and high-performance concrete(HPC)with BS rebar(named HPC-BS).First,the RBDO procedure is improved for the design optimization of coastal bridge piers,and a bridge is selected to illustrate the ***,reliability analysis of the pier designed with each group of materials is carried out to obtain the time-dependent reliability in terms of the ultimate and serviceability ***,the repair time of the pier is predicted based on the time-dependent reliability ***,the time-dependent LCCs for the pier are obtained for the selection of the optimal design.

Development and validation of a prognostic immunoinflammatory index for patients with gastric cancer

Discovery potential of the Glashow resonance in an air shower neutrino telescope

The in-ice or in-water Cherenkov neutrino telescope,such as IceCube,has already proved its power in measuring the Glashow resonance by searching for the bump around E_(ν)=6.3PeV arising from the W-boson *** are many proposals for the next few decades for observations of cosmic tau neutrinos with extensive air showers,also known as tau neutrino *** air shower telescope is,in principle,sensitive to the Glashow resonance via the channel W→τν_(τ)followed by the tau decay in the air(e.g.,TAMBO,which has a geometric area of approximately 500km^(2)).Using a thorough numerical analysis,we find that the discovery significance can be up to 90%with a TAMBO-like setup if PeV neutrinos primarily originate from neutron decays,considering the flux parameters measured by IceCube as the *** presence of new physics affecting the neutrino flavor composition can also increase the ***,if ultrahigh-energy neutrinos are dominantly produced from meson decays,it will be statistically difficult for an advanced proposal such as TAMBO to discriminate the Glashow resonance induced byν¯e from the intrinsicντ/ν¯_(τ)*** have identified several limitations for such advanced telescopes,in comparison with the in-ice or in-water telescope,when measuring resonances:(i)a suppressed branching ratio of 11%for the decay W→τν_(τ);(ii)the smearing effect and reduced acceptance because the daughter neutrino takes away(y)∼75%of the energy from the W decay;and(iii)a large attenuation effect for Earth-skimming neutrinos with the resonance.

Thermogravimetric characteristics of corn straw and bituminous coal copyrolysis based the ilmenite oxygen carriers

Herein,the co-pyrolysis reaction characteristics of corn straw(CS)and bituminous coal in the presence of ilmenite oxygen carriers(OCs)are investigated via thermogravimetry coupled with mass *** results reveal that the participation of OCs weakens the devolatilization intensity of *** the CS blending ratio is<50%,the mixed fuel exhibits positive synergistic *** fitting results according to the Coats-Redfern integral method show that the solid-solid interaction between OCs and coke changes the reaction kinetics,enhancing the co-pyrolysis reactivity at the high-temperature zone(750-950C).The synergistic effect is most prominent at a 30%CS blending ratio,with copyrolysis activation energy in the range of 26.35-40.57 kJ·mol^(-1).

Paired Immunoglobulin-like Receptor B Inhibition in Müller Cells Promotes Neurite Regeneration After Retinal Ganglion Cell Injury in vitro

In the central nervous system(CNS),three types of myelin-associated inhibitors(MAIs) have major inhibitory effects on nerve *** include Nogo-A,myelin-associated glycoprotein,and oligodendrocyte-myelin *** possess two co-receptors,Nogo receptor(NgR) and paired immunoglobulin-like receptor B(PirB).Previous studies have confirmed that the inhibition of NgR only results in a modest increase in regeneration in the CNS;however,the inhibitory effects of PirB with regard to nerve regeneration after binding to MAIs remain *** this study,we demonstrated that PirB is expressed in primary cultures of retinal ganglion cells(RGCs),and the inhibitory effects of the three MAIs on the growth of RGC neurites are not significantly decreased after direct PirB knockdown using adenovirus PirB ***,we found that retinal Müller cells expressed PirB and that its knockdown enhanced the regeneration of co-cultured RGC *** knockdown also activated the JAK/Stat3 signaling pathway in Müller cells and upregulated ciliary neurotrophic factor *** findings indicate that PirB plays a novel role in retinal Müller cells and that its action in these cells may indirectly affect the growth of RGC *** results also reveal that PirB in Müller cells affects RGC neurite *** findings provide a novel basis for the use of PirB as a target molecule to promote nerve regeneration.

The past and present of blast injury research in China

With the increasing incidence of blast injury, the research on its mechanisms and protective measures draws more and more attention. Blast injury has many characteristics different from general war injuries or trauma. For example, soldiers often have various degrees of visceral injury without significant surface damage, combined injuries and arterial air embolism. Researchers in China began to investigate blast injury later than the United States and Sweden, but the development is so fast that lots of achievements have been gained, including the development of biological shock tube, the mechanisms and characteristics of blast injury in various organs, as well as protective measures under special environments. This article reviews the past and current situation of blast injury research in China.
