Computed tomography radiogenomics:A potential tool for prediction of molecular subtypes in gastric stromal tumor

BACKGROUND Preoperative knowledge of mutational status of gastrointestinal stromal tumors(GISTs)is essential to guide the individualized precision *** To develop a combined model that integrates clinical and contrast-enhanced computed tomography(CE-CT)features to predict gastric GISTs with specific genetic mutations,namely KIT exon 11 mutations or KIT exon 11 codons 557-558 *** A total of 2.1 GIST patients with definitive genetic phenotypes were divided into a training dataset and a validation dataset in a 7:3 *** models were constructed using selected clinical features,conventional CT features,and radiomics features extracted from abdominal CE-CT *** models were developed:ModelCT sign,modelCT sign+rad,and model CTsign+rad+*** diagnostic performance of these models was evaluated using receiver operating characteristic(ROC)curve analysis and the Delong *** The ROC analyses revealed that in the training cohort,the area under the curve(AUC)values for model_(CT sign),model_(CT sign+rad),and modelCT_(sign+rad+clinic)for predicting KIT exon 11 mutation were 0.743,0.818,and 0.915,*** the validation cohort,the AUC values for the same models were 0.670,0.781,and 0.811,*** predicting KIT exon 11 codons 557-558 deletions,the AUC values in the training cohort were 0.667,0.842.and 0.72. for model_(CT sign),model_(CT sign+rad),and modelCT_(sign+rad+clinic),*** the validation cohort,the AUC values for the same models were 0.610,0.782.and 0.795,*** on the decision curve analysis,it was determined that the model_(CT sign+rad+clinic)had clinical significance and *** Our findings demonstrate that the combined modelCT_(sign+rad+clinic)effectively distinguishes GISTs with KIT exon 11 mutation and KIT exon 11 codons 557-558 *** combined model has the potential to be valuable in assessing the genotype of GISTs.

Significance of consensus CYC-binding sites found in the promoters of both ChCYC and ChRAD genes in Chirita heterotricha (Gesneriaceae)

CYC-like genes are widely conserved in controlling floral dorsoventral asymmetry(zygomorphy)through persistent expression in corresponding domains in core *** understand how CYC-like gene expression is maintained during flower development,we selected Chirita heterotricha as a material and isolated the promoter sequences of the ChCYC1C and ChCYC1D genes,homologs of CYC,by inverse polymerase chain *** promoter analyses led to the identification of a putative cis-regulatory element in each promoter matching the consensus DNA binding site for Antirrhinum CYC protein:GGCCCCTC at-165 for ChCYC1C,and GGCCCCCC at-163 for *** indicates that both the ChCYC1C and ChCYC1D genes have probably evolved autoregulatory loops to sustain their expression in developing *** also isolated the coding and promoter sequences of the ChRAD gene,a homolog of Antirrhinum *** analysis showed that the ChRAD gene promoter also contained a putative CYC-binding site(GGCCCAC at-134).Therefore,ChRAD is likely a direct target of the ChCYC1 genes,which is similar to Antirrhinum *** results imply that the establishment of floral zygomorphy in Chirita may have been achieved by the evolution of an autoregulatory loop for CYC-like genes,which was probably accompanied by simultaneous co-option of the RAD-like gene into their regulatory network.

Soil Fertility Indices of Citrus Orchard Land Along Topographic Gradients in the Three Gorges Area of china

In the Three Gorges Area of china, soil erosion and the resultant non-point source pollution and ecological degradation have endangered agricultural ecosystems and fresh water reservoirs. Although efforts have been undertaken to reduce soil and water losses from slope land used for citrus production, information on the effects of management practices on soil fertility indices is either limited or nonexistent. This study was conducted to compare the effects of 10 years of various management practices, citrus intercropped with white clover (WC), citrus mulched with straw (SM), citrus intercropped with contour hedgerows (CH), citrus orchard land with impermeable membrane (IM), and citrus intercropped with wheat (Triticum aestivum) and peanut (Arachris hypogaea) (WP), as treatments on soil fertility indices with that of the conventional citrus management (CM). Results showed that the soil organic carbon, total and available nitrogen, available potassium, and water-stable aggregate (> 0.2. mm) contents at the 0–5 cm depth were higher for the WC and SM treatments than the CM treatment. There was also spatial variation in soil fertility along slopes of the WC and SM treatments. The soil total and available nitrogen, phosphorus, and potassium, and water-stable aggregate (> 0.2. mm) contents at both the 0–5 and 5–2. cm depths were higher for the CH and IM treatments than the CM treatment. The average soil available nitrogen and available potassium contents were higher for the WP treatment than the CM treatment, but the WP treatment had little effect on the soil organic carbon, total nitrogen, and water-stable aggregate (> 0.2. mm) contents. These suggested that white clover intercropping and straw mulching were the most effective approaches to improve soil fertility in citrus orchard land of the Three Gorges area.

Photodynamic therapy(PDT)has emerged as a significant cancer therapy ***,cation-based organic small molecule aggregation-induced emission(AIE)photosensitizers(PSs)attract the wide atten-tion of many scientists,due to improved reactive oxygen species(ROS)production after ***,such PSs tend to localize only the mitochondria,limiting the death way of tumor cells(usu-ally apoptosis)during PDT process,which may affect the therapeutic effect under some ***,we designed a novel water-soluble three positive charge PS,TPAN-18F,which could be distributed uniformly in cell cytoplasm and had distribution in different sub-organelles(mitochondria,endoplasmic reticulum,lysosome).The experimental results showed that TPAN-18F-based PDT process can not only disrupt mitochondrial functions(reducing ATP production and destroying mitochondrial membrane po-tential),but also elevate the intracellular lipid peroxides(LPOs)level,which evoke the non-apoptotic death manner of tumor ***,in vivo studies showed that TPAN-18F-based PDT could effectively inhibit tumor ***,we believe that the construction of TPAN-18F is suggestive for tumor non-apoptotic therapy.

Sea-level change in coastal areas of china:Status in 2.2.
