Maraviroc promotes recovery from traumatic brain injury in mice by suppression of neuroinflammation and activation of neurotoxic reactive astrocytes

Neuroinflammation and the NACHT,LRR,and PYD domains-containing protein 3 inflammasome play crucial roles in secondary tissue damage following an initial insult in patients with traumatic brain injury(TBI).Maraviroc,a C-C chemokine receptor type 5 antagonist,has been viewed as a new therapeutic strategy for many neuroinflammatory diseases.We studied the effect of maraviroc on TBI-induced neuroinflammation.A moderate-TBI mouse model was subjected to a controlled cortical impact device.Maraviroc or vehicle was injected intraperitoneally 1 hour after TBI and then once per day for 3 consecutive days.Western blot,immunohistochemistry,and TUNEL(terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling)analyses were performed to evaluate the molecular mechanisms of maraviroc at 3 days post-TBI.Our results suggest that maraviroc administration reduced NACHT,LRR,and PYD domains-containing protein 3 inflammasome activation,modulated microglial polarization from M1 to M2,decreased neutrophil and macrophage infiltration,and inhibited the release of inflammatory factors after TBI.Moreover,maraviroc treatment decreased the activation of neurotoxic reactive astrocytes,which,in turn,exacerbated neuronal cell death.Additionally,we confirmed the neuroprotective effect of maraviroc using the modified neurological severity score,rotarod test,Morris water maze test,and lesion volume measurements.In summary,our findings indicate that maraviroc might be a desirable pharmacotherapeutic strategy for TBI,and C-C chemokine receptor type 5 might be a promising pharmacotherapeutic target to improve recovery after TBI.

Feasible and effective cell models for hepatitis B virus(HBV) infection are required for investigating the complete lifecycle of this virus, including the early steps of viral entry. Resistance to dimethyl sulfoxide/polyethylene glycol(DMSO/PEG), h NTCP expression, and a differentiated state are the limiting factors for successful HBV infection models. In the present study, we used a hepatoma cell line(Hu7^(hDNTCPh)) to overcome these limiting factors so that it exhibits excellent susceptibility to HBV infection. To achieve this goal, different hepatoma cell lines were tested with 2.5% DMSO/4%PEG8000, and one resistant cell line(Huh7 D) was used to construct a stable h NTCP-expressing cell line(Hu7^(hDNTCPh)) using a recombinant lentivirus system. Then, the morphological characteristics and differentiation molecular markers of Hu7^(hDNTCPh) cells with or without DMSO treatment were characterized. Finally, the susceptibility of Hu7^(hDNTCPh) cells to HBV infection was assessed. Our results showed that Huh7 D cells were resistant to 2.5% DMSO/4% PEG8000, whereas the others were not. Hu7^(hDNTCPh) cells were established to express a high level of h NTCP compared to liver extracts, and Hu7^(hDNTCPh) cells rapidly transformed into a non-dividing, well-differentiated polarized phenotype under DMSO treatment. Hu7^(hDNTCPh) cells fully supported the entire lifecycle of HBV infection. This cell culture system will be useful for the analysis of host-virus interactions, which should facilitate the discovery of antiviral drugs and vaccines.

Upgrading Pathways of Intelligent Manufacturing in China:Transitioning across Technological Paradigms

Intelligent technologies are leading to the next wave of industrial revolution in manufacturing.In developed economies,firms are embracing these advanced technologies following a sequential upgrading strategy-from digital manufacturing to smart manufacturing(digital-networked),and then to newgeneration intelligent manufacturing paradigms.However,Chinese firms face a different scenario.On the one hand,they have diverse technological bases that vary from low-end electrified machinery to leading-edge digital-network technologies;thus,they may not follow an identical upgrading pathway.On the other hand,Chinese firms aim to rapidly catch up and transition from technology followers to probable frontrunners;thus,the turbulences in the transitioning phase may trigger a precious opportunity for leapfrogging,if Chinese manufacturers can swiftly acquire domain expertise through the adoption of intelligent manufacturing technologies.This study addresses the following question by conducting multiple case studies:Can Chinese firms upgrade intelligent manufacturing through different pathways than the sequential one followed in developed economies?The data sources include semistructured interviews and archival data.This study finds that Chinese manufacturing firms have a variety of pathways to transition across the three technological paradigms of intelligent manufacturing in nonconsecutive ways.This finding implies that Chinese firms may strategize their own upgrading pathways toward intelligent manufacturing according to their capabilities and industrial specifics;furthermore,this finding can be extended to other catching-up economies.This paper provides a strategic roadmap as an explanatory guide to manufacturing firms,policymakers,and investors.

Joint DOA and channel estimation with data detection based on 2D unitary ESPRIT in massive MIMO systems

We propose a novel method for joint two-dimensional (2D) direction-of-arrival (DOA) and channel estimation with data detection for uniform rectangular arrays (URAs) for the massive multiple-input multiple-output (MIMO) systems. The conventional DOA estimation algorithms usually assume that the channel impulse responses are known exactly. However, the large number of antennas in a massive MIMO system can lead to a challenge in estimating accurate corresponding channel impulse responses. In contrast, a joint DOA and channel estimation scheme is proposed, which first estimates the channel impulse responses for the links between the transmitters and antenna elements using training sequences. After that, the DOAs of the waves are estimated based on a unitary ESPRIT algorithm using previous channel impulse response estimates instead of accurate channel impulse responses and then, the enhanced channel impulse response estimates can be obtained. The proposed estimator enjoys closedform expressions, and thus it bypasses the search and pairing processes. In addition, a low-complexity approach toward data detection is presented by reducing the dimension of the inversion matrix in massive MIMO systems.Different cases for the proposed method are analyzed by changing the number of antennas. Experimental results demonstrate the validity of the proposed method.

Chiral single-photon switch-assisted quantum logic gate with a nitrogen-vacancy center in a hybrid system

We propose what we believe is a novel proposal for realizing a quantum C-NOT logic gate, through fabricating an interesting hybrid device with a chiral photon-pulse switch, a single nitrogen-vacancy(NV) center, and an optical microcavity. Three major different practical routes on realizing a chiral photon emitter are discussed, which can implement a chiral control unit via the nonreciprocal emitter–photon interactions, so-called "propagationdirection-dependent" emission. With the assistance of dichromatic microwave driving fields, we carry out the relevant C-NOT operations by engineering the interactions on a single NV spin in a cavity. We note that this logic gate is robust against practical noise and experimental imperfection, and this attempt may evoke wide and fruitful applications in quantum information processing.

Separate Neural Networks for Gains and Losses in Intertemporal Choice

An important and unresolved question is how human brain regions process information and interact with each other in intertemporal choice related to gains and losses. Using psychophysiological interaction and dynamic causal modeling analyses, we investigated the functional interactions between regions involved in the decision- making process while participants performed temporal discounting tasks in both the gains and losses domains. We found two distinct intrinsic valuation systems underlying temporal discounting in the gains and losses domains： gains were specifically evaluated in the medial regions, including the medial prefrontal and orbitofrontal cortices, and losses were evaluated in the lateral dorsolateral prefrontal cortex. In addition, immediate reward or pun- ishment was found to modulate the functional interactions between the dorsolateral prefrontal cortex and distinct regions in both the gains and losses domains： in the gains domain, the mesolimbic regions; in the losses domain, the medial prefrontal cortex, anterior cingulate cortex, and insula. These findings suggest that intertemporal choice of gains and losses might involve distinct valuation systems, and more importantly, separate neural interactions may implement the intertemporal choices of gains and losses. These findings may provide a new biological perspective for understanding the neural mechanisms underlying intertemporal choice of gains and losses.

Salivary proteins and microbiota as biomarkers for early childhood caries risk assessment

Early childhood caries （ECC） is a term used to describe dental caries in children aged 6 years or younger. Oral streptococci, such as Streptococcus mutans and Streptococcus sorbrinus, are considered to be the main etiological agents of tooth decay in children. Other bacteria, such as Prevotella spp. and Lactobacillus spp., and fungus, that is, Candida albicans, are related to the development and progression of ECC. Biomolecules in saliva, mainly proteins, affect the survival of oral microorganisms by multiple innate defensive mechanisms, thus modulating the oral microflora. Therefore, the protein composition of saliva can be a sensitive indicator for dental health. Resistance or susceptibility to caries may be significantly correlated with alterations in salivary protein components. Some oral microorganisms and saliva proteins may serve as useful biomarkers in predicting the risk and prognosis of caries. Current research has generated abundant information that contributes to a better understanding of the roles of microorganisms and salivary proteins in ECC occurrence and prevention. This review summarizes the microorganisms that cause caries and tooth-protective salivary proteins with their potential as functional biomarkers for ECC risk assessment. The identification of biomarkers for children at high risk of ECC is not only critical for early diagnosis but also important for preventing and treating the disease.

Protein-Protein Interactions in the Regulation of WRKY Transcription Factors

It has been almost 20 years since the first report of a WRKY transcription factor, SPF1, from sweet potato. Great progress has been made since then in establishing the diverse biological roles of WRKY transcription factors in plant growth, development, and responses to biotic and abiotic stress. Despite the functional diversity, almost all ana-lyzed WRKY proteins recognize the TrGACC/T W-box sequences and, therefore, mechanisms other than mere recognition of the core W-box promoter elements are necessary to achieve the regulatory specificity of WRKY transcription factors. Research over the past several years has revealed that WRKY transcription factors physically interact with a wide range of proteins with roles in signaling, transcription, and chromatin remodeling. Studies of WRKY-interacting proteins have provided important insights into the regulation and mode of action of members of the important family of transcrip-tion factors. It has also emerged that the slightly varied WRKY domains and other protein motifs conserved within each of the seven WRKY subfamilies participate in protein-protein interactions and mediate complex functional interactions between WRKY proteins and between WRKY and other regulatory proteins in the modulation of important biologi- cal processes. In this review, we summarize studies of protein-protein interactions for WRKY transcription factors and discuss how the interacting partners contribute, at different levels, to the establishment of the complex regulatory and functional network of WRKY transcription factors.

Formamidinium(FA)-based Sn-Pb perovskite solar cells(FAPb_(0.5)Sn_(0.5)I_(3) PSCs)with ideal bandgap and impressive thermal stability have caught enormous attention recently.However,it still suffers from the challenge of realizing high efficiency due to the surface imperfections of the transport materials and the energy-level mismatch between functional contacts.Herein,it is demonstrated that the modification on buried interface with alkali metal salts is a viable strategy to alleviate these issues.We systematically investigate the role of three alkali metal bromide salts(NaBr,KBr,CsBr)by burying them between the NiOx hole transport layer(HTL)and the perovskite light-absorbing layer,which can effectively passivate interface defects,improve energy-level matching and release the internal residual strain in perovskite layers.The device with CsBr buffer layer exhibits the best power conversion efficiency(PCE)approaching 20%,which is one of the highest efficiencies for FA-based Sn-Pb PSCs employing NiO_(x) HTLs.Impressively,the long-term storage stability of the unencapsulated device is also greatly boosted.Our work provides an efficient strategy to prepare desired FA-based ideal-bandgap Sn-Pb PSCs which could be applied in tandem solar cells.

Dear Editor, Hepatitis B Virus(HBV)infection remains a major health problem worldwide despite the availability of a highly ef- fective preventive vaccine.HBV genome codes several distinct proteins,among which the non-particulate e-antigen(HBeAg)is one of the important markers in HBV infection. Sera containing HBeAg are highly infectious but those with anti-HBeAg have little infectivity.
