Heterogeneous expression of stearoyl-acyl carrier protein desaturase genes SAD1 and SAD2 from Linum usitatissimum enhances seed oleic acid accumulation and seedling cold and drought tolerance in Brassica napus

Flax(Linum usitatissimum L.)is a versatile crop and its seeds are a major source of unsaturated fatty acids.Stearoyl-acyl carrier protein desaturase(SAD)is a dehydrogenase enzyme that plays a key role in oleic acid biosynthesis as well as responses to biotic and abiotic stresses.However,the function of SAD orthologs from L.usitatissimum has not been assessed.Here,we found that two LuSAD genes,LuSAD1 and LuSAD2,are present in the genome of L.usitatissimum cultivar‘Longya 10’.Heterogeneous expression of either LuSAD1 or LuSAD2 in Arabidopsis thaliana resulted in higher contents of total fatty acids and oleic acid in the seeds.Interestingly,ectopic expression of LuSAD2 in A.thaliana caused altered plant architecture.Similarly,the overexpression of either LuSAD1 or LuSAD2 in Brassica napus also resulted in increased contents of total fatty acids and oleic acid in the seeds.Furthermore,we demonstrated that either LuSAD1 or LuSAD2 enhances seedling resistance to cold and drought stresses by improving antioxidant enzyme activity and nonenzymatic antioxidant levels,as well as reducing membrane damage.These findings not only broaden our knowledge of the LuSAD functions in plants,but also offer promising targets for improving the quantity and quality of oil,and the abiotic stress tolerance of oil-producing crops,through molecular manipulation.

Transcriptional regulation is essential for balancing multiple metabolic pathways that influence oil accumulation in seeds.Thus far,the transcriptional regulatory mechanisms that govern seed oil accumulation remain largely unknown.Here,we identified the transcriptional regulatory network composed of MADS-box transcription factors SEEDSTICK(STK)and SEPALLATA3(SEP3),which bridges several key genes to regulate oil accumulation in seeds.We found that STK,highly expressed in the developing embryo,positively regulates seed oil accumulation in Arabidopsis(Arabidopsis thaliana).Furthermore,we discovered that SEP3 physically interacts with STK in vivo and in vitro.Seed oil content is increased by the SEP3 mutation,while it is decreased by SEP3 overexpression.The chromatin immunoprecipitation,electrophoretic mobility shift assay,and transient dual-luciferase reporter assays showed that STK positively regulates seed oil accumulation by directly repressing the expression of MYB5,SEP3,and SEED FATTY ACID REDUCER 4(SFAR4).Moreover,genetic and molecular analyses demonstrated that STK and SEP3 antagonistically regulate seed oil production and that SEP3 weakens the binding ability of STK to MYB5,SEP3,and SFAR4.Additionally,we demonstrated that TRANSPARENT TESTA 8(TT8)and ACYL-ACYL CARRIER PROTEIN DESATURASE 3(AAD3)are direct targets of MYB5 during seed oil accumulation in Arabidopsis.Together,our findings provide the transcriptional regulatory network antagonistically orchestrated by STK and SEP3,which fine tunes oil accumulation in seeds.

Adenosine exerts its dual functions of homeostasis and neuromodulation in the brain by acting at mainly 2 G-protein coupled receptors,called A1 and A2A receptors.The adenosine A2A receptor(A2AR)antagonists have been clinically pursued for the last 2 decades,leading to final approval of the istradefylline,an A2AR antagonist,for the treatment of OFF-Parkinson's disease(PD)patients.The approval paves the way to develop novel therapeutic methods for A2AR antagonists to address 2 major unmet medical needs in PD and traumatic brain injury(TBI),namely neuroprotection or improving cognition.In this review,we first consider the evidence for aberrantly increased adenosine signaling in PD and TBI and the sufficiency of the increased A2AR signaling to trigger neurotoxicity and cognitive impairment.We further discuss the increasing preclinical data on the reversal of cognitive deficits in PD and TBI by A2AR antagonists through control of degenerative proteins and synaptotoxicity,and on protection against TBI and PD pathologies by A2AR antagonists through control of neuroinflammation.Moreover,we provide the supporting evidence from multiple human prospective epidemiological studies which revealed an inverse relation between the consumption of caffeine and the risk of developing PD and cognitive decline in aging population and Alzheimer's disease patients.Collectively,the convergence of clinical,epidemiological and experimental evidence supports the validity of A2AR as a new therapeutic target and facilitates the design of A2AR antagonists in clinical trials for disease-modifying and cognitive benefit in PD and TBI patients.

Creep property and microstructural evolution of DD11 alloy under high temperature and low stress

In this paper,the interrupted and ruptured creep tests were carried out in a novel second generation single crystal superalloy named DD11 at 1100℃/130 MPa.The alloy exhibited typical creep curve including primary,steady,and tertiary three creep stages.The microstructural evolution at different stages of the creep were analyzed by scanning electron microscopy(SEM)and transmission electron microscopy(TEM).The results show that theγ′phases transform into rafted structure at the early stage of the steady creep and keep stable during the steady creep stage.As the creep goes on,the rafted structure further coarsens and the topological inversion occurs.In addition,at the primary creep,the dislocations mainly move in theγmatrix and pile up in theγ/γ′interface since the matrix channels widen slightly.The formation of the regular interfacial dislocation networks occurs at the early stage of the steady creep.Under the low stress,the dominated deformation mechanism during steady creep stage is the climbing of the〈010〉type edge dislocation.Furthermore,the effect of the deformation mechanism on creep property was discussed in detail.

A novel method for road network mining from floating car data

Vehicles have been increasingly equipped with GPS receivers to record their trajectories,which we call floating car data.Compared with other data sources,these data are characterized by low cost,wide coverage,and rapid updating.The data have become an important source for road network extraction.In this paper,we propose a novel approach for mining road networks from floating car data.First,a Gaussian model is used to transform the data into bitmap,and the Otsu algorithm is utilized to detect road intersections.Then,a clothoid-based method is used to resample the GPS points to improve the clustering accuracy,and the data are clustered based on a distance-direction algorithm.Last,road centerlines are extracted with a weighted least squares algorithm.We report on experiments that were conducted on floating car data from Wuhan,China.To conclude,existing methods are compared with our method to prove that the proposed method is practical and effective.

The tumor microenvironment is a complex network of cells,extracellular matrix,and signaling molecules that plays a critical role in tumor progression and metastasis.Lymphatic and blood vessels are major routes for solid tumor metastasis and essential parts of tumor drainage conduits.However,recent studies have shown that lymphatic endothelial cells(LECs)and blood endothelial cells(BECs)also play multifaceted roles in the tumor microenvironment beyond their structural functions,particularly in hepatocellular carcinoma(HCC).This comprehensive review summarizes the diverse roles played by LECs and BECs in HCC,including their involvement in angiogenesis,immune modulation,lymphangiogenesis,and metastasis.By providing a detailed account of the complex interplay between LECs,BECs,and tumor cells,this review aims to shed light on future research directions regarding the immune regulatory function of LECs and potential therapeutic targets for HCC.

Microstructure Design of High-Entropy Alloys Through a Multistage Mechanical Alloying Strategy for Temperature-Stable Megahertz Electromagnetic Absorption

Developing megahertz(MHz)electromagnetic wave(EMW)absorption materials with broadband absorption,multi-temperature adaptability,and facile preparation method remains a challenge.Herein,nanocrystalline FeCoNiCr_(0.4)Cu_(0.2) high-entropy alloy powders(HEAs)with both large aspect ratios and thin intergranular amorphous layers are constructed by a multistage mechanical alloying strategy,aiming to achieve excellent and temperature-stable permeability and EMW absorption.A single-phase face-centered cubic structure with good ductility and high crystallinity is obtained as wet milling precursors,via precisely controlling dry milling time.Then,HEAs are flattened to improve aspect ratios by synergistically regulating wet milling time.FeCoNiCr_(0.4)Cu_(0.2) HEAs with dry milling 20 h and wet milling 5 h(D20)exhibit higher and more stable permeability because of larger aspect ratios and thinner intergranular amorphous layers.The maximum reflection loss(RL)of D20/SiO_(2) composites is greater than-7 dB with 5 mm thickness,and EMW absorption bandwidth(RL<-7 dB)can maintain between 523 and 600 MHz from-50 to 150℃.Furthermore,relying on the“cocktail effect”of HEAs,D20 sample also exhibits excellent corrosion resistance and high Curie temperature.This work provides a facile and tunable strategy to design MHz electromagnetic absorbers with temperature stability,broadband,and resistance to harsh environments.

Association of acute glycemic parameters at admission with cardiovascular mortality in the oldest old with acute myocardial infarction

OBJECTIVES Stress-related glycemic indicators,including admission blood glucose(ABG),stress-hyperglycemia ratio(SHR),and glycemic gap(GG),have been associated with worse outcomes after acute myocardial infarction(AMI).However,data regarding their prognostic value in the oldest old with AMI are unavailable.Therefore,this study aimed to investigate the association of stress-related glycemic indicators with short-and long-term cardiovascular mortality(CVM)in the oldest old(≥80 years)with AMI.METHODS In this prospective study,a total of 933 consecutive old patients with AMI admitted to FuWai hospital(Beijing,China)were enrolled.On admission,ABG,SHR,and GG were assessed and all participants were classified according to their quartiles.Kaplan-Meier,restricted cubic splines(RCS),and multivariate Cox regression analyses were performed to evaluate the association between these glycemic indicators and CVM within 30 days and long-term follow-up.RESULTS During an average of 1954 patient-years of follow-up,a total of 250 cardiovascular deaths were recorded.Kaplan-Meier analyses showed the lowest CVM in quartile 1 of ABG and in quartile 2 of SHR and GG.After adjusting for potential covariates,patients in quartile 4 of ABG,SHR,and GG had a respective 1.67-fold(95%CI:1.03-2.69;P=0.036),1.80-fold(95%CI:1.16-2.79;P=0.009),and 1.78-fold(95%CI:1.14-2.79;P=0.011)higher risk of long-term CVM risk compared to those in the reference groups(quartile 1 of ABG and quartile 2 of SHR and GG).Furthermore,RCS suggested a J-shaped relationship of ABG and a Ushaped association of SHR and GG with long-term CVM.Additionally,we observed similar associations of these acute glycemic parameters with 30-day CVM.CONCLUSIONS Our data first indicated that SHR and GG consistently had a U-shaped association with both 30-day and longterm CVM among the oldest old with AMI,suggesting that they may be useful for risk stratification in this special population.

Objective To describe the treatment patterns and survival status of advanced gastric cancer(AGC)in China in the past two decades,and objectively evaluate the impact of standardized Chinese medicine(CM)treatment on the survival of AGC patients.Methods This multicenter registry designed and propensity score analysis study described the diagnosis characteristics,treatment-pattern development and survival status of AGC from 10 hospitals in China between January 1,2000 and July 31,2021.Overall survival(OS)was evaluated between non-CM cohort(standard medical treatment)and CM cohort(integrated standard CM treatment≥3 months).Propensity score matching(PSM)and inverse probability of treatment weighting(IPTW)were performed to adjust any difference in average outcomes for bias.Results A total of 2,001 patients histologically confirmed locally advanced and/or metastasis stomach and gastroesophageal junction adenocarcinoma were enrolled.Among them,1,607 received systemic chemotherapy,215(10.74%)accepted molecular targeted therapy,44(2.2%)received checkpoint inhibitor therapy,and 769(38.43%)received CM.Two-drug regimen was the main choice for first-line treatment,with fluoropyrimidine plus platinum as the most common regimen(530 cases,60.09%).While 45.71%(16 cases)of patients with HER2 amplification received trastuzumab in first-line.The application of apatinib increased(33.33%)in third-line.The application of checkpoint inhibitors has increased since 2020.COX analysis showed that Lauren mixed type(P=0.017),cycles of first-line treatment>6(P=0.000),CM(P=0.000),palliative gastrectomy(P=0.000),trastuzumab(P=0.011),and apatinib(P=0.008)were independent prognostic factors for the OS of AGC.After PSM and IPTW,the median OS of CM cohort and non-CM cohort was 18.17 and 12.45 months,respectively(P<0.001).Conclusions In real-world practice for AGC in China,therapy choices consisted with guidelines.Two-drug regimen was the main first-line choice.Standardized CM treatment was an independent prognostic factor and could prolong the OS of C

滹沱河流域横跨山西和河北两省,自2000年以来由于经济发展,城市扩建,滹沱河一度断流,但随着国家对生态文明建设的重视和南水北调工程的实施,滹沱河流域生态保护成效显著提升.基于GEE平台选取MODIS数据、Landsat数据和夜光遥感数据等,利用生态环境指数(EI)中的生物丰富度指数、植被覆盖指数、土地胁迫指数和污染负荷指数以及遥感生态指数(RSEI)中的湿度指数相结合生成新的评价指标体系,利用变异系数法和熵权法对指标赋权并构建生态环境评价模型,对2000~2020年滹沱河流域进行生态环境质量评价和分级,并运用地理探测器对其驱动因子进行解释.结果表明:①时间尺度上,2000~2015年滹沱河流域的生态环境处于“衰退期”,2015~2020年滹沱河处于“恢复期”.从格网尺度上看,流域内中部地区生态环境质量呈现逐年提高的状态;流域内西部和东部地区,“衰退期”生态环境质量逐年降低,“恢复期”生态环境质量有所提高.②热点分析表明,滹沱河流域的生态环境质量呈现“中间高两侧低”的空间分布状态.冷点区域集中分布于东部和南部的城市乡镇地区,零星分布于西侧河谷地区.③地理探测分析表明,单因子探测驱动因子主要为人口密度、植被净初级生产力(NPP)、植被覆盖度(FVC)和地貌类型.交互探测的主导因子为“地貌类型+FVC”.随着生态文明建设不断深入和《滹沱河保护条例》的颁布实施,结合流域内不同的自然环境和社会特征等因子,滹沱河流域生态环境评价研究可为因地制宜地改善生态环境政策提供数据支持.
