Chronic ingestion of high concentrations of hexavalent chromium[Cr(VI)]in drinking water induces intestinal tumors in mice;however,information on its toxicity on intestinal smooth muscle cells is *** present study aimed to assess the in vitro and in vivo toxicological effects of Cr(VI)on intestinal smooth muscle *** intestinal smooth muscle cells(HISM cells)were cultured with different concentrations of Cr(VI)to evaluate effects on cell proliferation ability,oxidative stress levels,and antioxidant ***,tissue sections in Cr(VI)exposed rabbits were analyzed to evaluate toxicity on intestinal muscle cells in *** chips were utilized to assess differential gene expression profiles at the genome-wide level in 1μmol/L Cr(VI)treated *** tissue biopsy results showed that Cr(VI)increased the incidences of diffuse epithelial hyperplasia in intestinal jejunum but caused no obvious damage to the structure of the *** proliferation analysis revealed that high concentrations(≥64μmol/L)but not low concentrations of Cr(VI)(≤16μmol/L)significantly inhibited the growth of HISM *** oxidative stress levels,the expression of reactive oxygen species(ROS)and nitric oxide(NO)was elevated at high concentrations(≥64μmol/L)but not at low concentrations of Cr(VI)(≤16μmol/L).In addition,dose-dependent increases in the activity of oxidized glutathione(GSSH)/total-glutathione(T-GSH)were also *** chip screened 491 differentially expressed genes including genes associated with cell apoptosis,oxidations,and *** of these differentially expressed genes may be unique to smooth muscle cells in response to Cr(VI)induction.

In this study, the formation of iodinated trihalomethanes(I-THMs) was systematically evaluated and compared for three treatment processes-(i) chlorination,(ii) monochloramine, and(iii) dichloramination-under different p H conditions. The results demonstrated that ITHM formation decreased in the order of monochloramination > dichloramination > chlorination in acidic and neutral p H. However, the generation of I-THMs increased in the dichloramination < chlorination < monochloramination order in alkaline condition. Specifically, the formation of I-THMs increased as p H increased from 5 to 9 during chlorination and monochloramination processes, while the maximum I-THM formation occurred at pH 7 during dichloramination. The discrepancy could be mainly related to the stability of the three chlor(am) ine disinfectants at different p H conditions. Moreover, in order to gain a thorough insight into the mechanisms of I-THM formation during dichloramination, further investigation was conducted on the influencing factors of DOC concentration and Br/Imolar ratio. I-THM formation exhibited an increasing and then decreasing trend as the concentration of DOC increased from 1 to 7 mg-C/L, while the yield of I-THMs increased with increasing Br/Imolar ratio from 5:0 to 5:10. During the three processes mentioned above, similar I-THM formation results were also obtained in real water, which indicates that the excessive generation of I-THMs should be paid special attention during the disinfection of iodide-containing water.

Some barley mutants can synthesize neither anthocyanins nor proanthocyanidins in the seed coat, which is related to several genes in locus Ant13, but the exact model of action remains unknown. We used the cDNA microarray technology with barley transcription-deficient mutant (ant13-152) that does not synthesize proanthocyanidins as the tester, and its wild type genotype (Triumph) as the driver, to study this question. Six-thousand and forty-eight clones from the wild type Morex testa+pericarp cDNA library were amplified using PCR, and the DNA fragments were spotted on commercial amino-modified glass slide as microarray. The mRNAs from the developing seed coat (8–15 days) of both the mutant and the wild-type barley plants were isolated, and labeled respectively with Cy3-dUTP and Cy5-dUTP when reversely trans- cribed to cDNAs. The labeled cDNAs were used as probes, mixed at the same molar concentration, and hybridized with the DNA fragments on the slide. Seventy clones exhibiting marked differential expression (ratio>4) were identified from the microarray. All the 25 cDNA clones that showed an over-expression in wild type in comparison to the mutant ant13-152 were sequenced. It was found that most of these over- expressing clones were transcription/translation and hordein-associated genes. These results have laid a solid material basis for further elucidation of the metabolic pathway in proanthocyanidin synthesis in barley and likely other plants.

Modeling of turbidity dynamics caused by wind-induced waves and current in the Taihu Lake

A simple turbidity model was developed with a sound physical basis based on in situ high-frequency observations of short-term,strong wind-induced sediment suspension in Taihu Lake,*** validation results show that the model could successfully simulate turbidity caused by strong wind events,despite the relatively poor simulation accuracy for high values of turbidity caused by the entrainment of cyanobacteria by turbulence. The in situ observations and model simulation results indicate that the wind waves were within a narrow spectral band,with spectral energy mainly distributed within the 0.28-0.75 Hz band on opposite sides of the peak *** high-frequency and low-energy wind waves are sensitive to depth ***,the average depth of the lake is only 1.9 m,and wind waves still represent the main force of sediment suspension at the sediment-water ***,lake currents were of significance to the maintenance of background turbidity in calm waves or ripples and in the determination of critical shear *** considering the spatial distribution of hydrodynamics and sediment,the model can be used to simulate the turbidity of the entire lake as well as boundary conditions for three-dimensional numerical models.

Recent studies have shown that global translational reprogramming is an early activation event in pattern-triggered immunity,when plants recognize microbe-associated molecular ***,it is not fully known whether translational regulation also occurs in subsequent immune responses,such as effector-triggered immunity(ETI).In this study,we performed genome-wide ribosome profiling in Arabidopsis upon RPS2-mediated ETI activation and discovered that specific groups of genes were translationally regulated,mostly in coordination with *** genes encode enzymes involved in aromatic amino acid,phenylpropanoid,camalexin,and sphingolipid *** functional significance of these components in ETI was confirmed by genetic and biochemical *** findings provide new insights into diverse translational regulation of plant immune responses and demonstrate that translational coordination of metabolic gene expression is an important strategy for ETI.
