Prognostic prediction model of colorectal cancer based on preoperative serum tumor markers

BACKGROUND Preoperative serum tumor markers not only play a role in the auxiliary diagnosis and postoperative monitoring in colorectal cancer(CRC),but also have been found to have potential prognostic value.AIM To analyze whether preoperative serum tumor markers,including carcinoembryonic antigen(CEA)and carbohydrate antigen 19-9(CA19-9),affect the prognosis of CRC.METHODS This was a retrospective study conducted in a single center.Patients with nonmetastatic CRC who underwent initial surgery between January 2011 and January 2020 were enrolled and divided into development site and validation site groups at a ratio of 7:3.The independent prognostic factors were screened by Cox regression analysis,and finally,a prognostic nomogram model was established.The newly developed model was tested by internal validation.RESULTS Eventually,3526 postoperative patients with nonmetastatic CRC were included in the study.There were 2473 patients at the development site and 1056 patients at the validation site.Age(P<0.01,HR=1.042,95%CI=1.033-1.051),tumor node metastasis(TNM)classification(P<0.01,HR=1.938,95%CI=1.665-2.255),preoperative CEA(P=0.001,HR=1.393,95%CI=1.137-1.707)and CA19-9(P<0.01,HR=1.948,95%CI=1.614-2.438)levels were considered independent prognostic factors for patients with nonmetastatic CRC and were used as variables in the nomogram model.The areas under the curve of the development and validation sites were 0.655 and 0.658,respectively.The calibration plot also showed the significant performance of the newly established nomogram.CONCLUSION We successfully constructed a nomogram model based on age,TNM stage,preoperative CEA,and CA19-9 levels to evaluate the overall survival of patients with nonmetastatic CRC.

Preoperative albumin-bilirubin score predicts short-term outcomes and long-term prognosis in colorectal cancer patients undergoing radical surgery

BACKGROUND The albumin-bilirubin(ALBI)score is a serum biochemical indicator of liver function and has been proven to have prognostic value in a variety of cancers.In colorectal cancer(CRC),a high ALBI score tends to be associated with poorer survival.AIM To investigate the correlation between the preoperative ALBI score and outcomes in CRC patients who underwent radical surgery.METHODS Patients who underwent radical CRC surgery between January 2011 and January 2020 at a single clinical center were included.The ALBI score was calculated by the formula(log10 bilirubin×0.66)+(albumin×-0.085),and the cutoff value for grouping patients was-2.8.The short-term outcomes,overall survival(OS),and disease-free survival(DFS)were calculated.RESULTS A total of 4025 CRC patients who underwent radical surgery were enrolled in this study,and there were 1908 patients in the low ALBI group and 2117 patients in the high ALBI group.Cox regression analysis revealed that age,tumor size,tumor stage,ALBI score,and overall complications were independent risk factors for OS;age,tumor stage,ALBI score,and overall complications were identified as independent risk factors for DFS.CONCLUSION A high preoperative ALBI score is correlated with adverse short-term outcomes,and the ALBI score is an independent risk factor for OS and DFS in patients with CRC undergoing radical surgery.

Endovascular coiling versus microsurgical clipping for ruptured intracranial aneurysms:a meta-analysis and systematic review

Background:The purpose of this analysis is to evaluate the current evidence with regard to the effectiveness and safety between coiling and clipping in patients with ruptured intracranial aneurysms(RIAs).Methods:We performed a meta-analysis that compared clipping with coiling between July 2000 and September 2021.PubMed,EMBASE,and the Cochrane Library were searched for related articles systematically.And the treatment efficacy and postoperative complications were analyzed.Results:We identified three randomized controlled trials and thirty-seven observational studies involving 60,875 patients with ruptured cerebral aneurysms.The summary results showed that coiling was related a better quality of life(mRS0-2;OR=1.327;CI=1.093-1.612;p<0.05),a higher risk of mortality(OR=1.116;CI=1.054-1.180;p<0.05),higher rate of rebleeding(RR=1.410;CI=1.092-1.822;p<0.05),lower incidence of vasospasm(OR=0.787;CI=0.649-0.954;p<0.05),higher risk of hydrocephalous(RR=1.143;CI=1.043-1.252;p<0.05),lower risk of cerebral infarction(RR=0.669;CI=0.596-0.751;p<0.05),lower risk of neuro deficits(RR=0.720;CI=0.582-0.892;p<0.05),and a lower rate of complete occlusion(OR=0.495;CI=0.280-0.876;p<0.05).Conclusion:Coiling was significantly associated with a better life quality(mRS0-2),a lower incidence of postoperative complications,and a higher rate of mortality,rebleeding,hydrocephalous,and a lower rate of complete occlusion than clipping.

Intraspecific floral colour variation in three Pedicularis species

Flower constancy describes the phenomenon that pollinators tend to successively visit flowers of a single species during foraging,reducing reproductive interference in natural communities.The extent of flower constancy is largely determined by the floral traits of co-flowering species.Both higher inter-specific and lower intraspecific differences of floral traits should contribute to a higher level of flower constancy.However,previous studies mainly focused on interspecific difference,and the intraspecific variation(consistency)of floral traits received much less attention.We hypothesise that selection may favour lower intraspecific floral trait variation in communities composed of multiple co-flowering congeners.We investigated the floral colour variation of three focal Pedicularis species that share pollinators in 19communities composed of either single or multiple Pedicularis species.Colour was quantified using image-based colour analysis as perceived by pollinators.We found that most of the intrapopulation floral colour variation was below the colour discrimination threshold of bumblebees,implying strongly constrained by the visual selection by pollinators.Contrary to the hypothesis,there is no significant difference in intraspecific floral colour variation between different community contexts.It may be due to the relatively large interspecific floral colour differences of most co-flowering species.The influence of community context on intraspecific variation may be reflected in floral traits other than colours.

Epilepsy frequently leads to cognitive dysfunction and approaches to treatment remain limited.Although regular exercise effectively improves learning and memory functions across multiple neurological diseases,its application in patients with epilepsy remains controversial.Here,we adopted a 14-day treadmill-exercise paradigm in a pilocarpine injection-induced mouse model of epilepsy.Cognitive assays confirmed the improvement of object and spatial memory after endurance training,and electrophysiological studies revealed the maintenance of hippocampal plasticity as a result of physical exercise.Investigations of the mechanisms underlying this effect revealed that exercise protected parvalbumin interneurons,probably via the suppression of neuroinflammation and improved integrity of blood-brain barrier.In summary,this work identified a previously unknown mechanism through which exercise improves cognitive rehabilitation in epilepsy.

Effect of Cr_(2)O_(3) on properties of CaO-SiO_(2)-Fe_(t)O-MgO system and dissolution behavior of lime

The productivity of the chrome-containing semi-steel converter smelting process is directly affected by the slag-forming speed during the converter preliminary stage.The effects of Cr_(2)O_(3) content on the physicochemical properties of the CaO-SiO_(2)-Fe_(t)O-MgO system,such as melting temperature,solidification behavior,mineral composition,and lime dissolution rate,were studied.The results showed that the slag was an amorphous phase at 1500℃.When Cr_(2)O_(3) was added,Ca(Fe,Mg)Si_(2)O_(6) and spinel were formed in the slag.With the increase in Cr_(2)O_(3) content,the amount of spinel precipitation increased,and the dendritic FegO_(4) crystal gradually changed into the granular(Fe,Mg)(Fe,Cr)2O_(4) crystal.As the Cr_(2)O_(3) content increased from O to 2.91 wt.%,the melting temperature of the slag rose,and the melting range of the slag expanded slightly,but the lime dissolution rate did not change.When the Cr_(2)O_(3) content further increased to 9.09 wt.%,the melting temperature continued to rise,the melting range rapidly expanded,and the lime dissolution rate decreased.

Experimental observations on the nonproportional multiaxial ratchetting of cast AZ91 magnesium alloy at room temperature

The nonproportional multiaxial ratchetting of cast AZ91 magnesium (Mg) alloy was examined by performing a sequence of axial-torsional cyclic tests controlled by stress with various loading paths at room temperature (RT).The evolutionary characteristics and path dependence of multiaxial ratchetting were discussed.Results illustrate that the cast AZ91 Mg alloy exhibits considerable nonproportional additional softening during cyclic loading with multiple nonproportional multiaxial loading paths;multiaxial ratchetting presents strong path dependence,and axial ratchetting strains are larger under nonproportional loading paths than under uniaxial and proportional45°linear loading paths;multiaxial ratchetting becomes increasingly pronounced as the applied stress amplitude and axial mean stress increase.Moreover,stress-strain curves show a convex and symmetrical shape in axial/torsional directions.Multiaxial ratchetting exhibits quasi-shakedown after certain loading cycles.The abundant experimental data obtained in this work can be used to develop a cyclic plasticity model of cast Mg alloys.

Numerical investigations of the interactions between bubble induced shock waves and particle based on OpenFOAM

The presence of particles and the shock waves generated by the cavitation bubbles can significantly affect the safety and the performance of hydrodynamic machineries.In the present paper,the shock waves generated by cavitation bubble collapsing near the particle are numerically investigated based on the OpenFOAM together with the numerical schlieren for the shock wave identifications.The numerical results reveal that the stand-off distance is one of the paramount factors affecting the interactions between the particle and the shock waves.Several different kinds of shock waves(e.g.,bubble-inception,jet formation,particle reflected and jet-split shock waves)are observed during the bubble collapsing near the particle.For stand-off distance smaller than 0.5 or larger than 1.1,the maximum pressure at particle surface generated by the bubble growth can surpass those of the collapse stage.

Role of micro-Ca/In alloying in tailoring the microstructural characteristics and discharge performance of dilute Mg-Bi-Sn-based alloys as anodes for Mg-air batteries

The influence of micro-Ca/In alloying on the microstructural charac teristics,electrochemical behaviors and discharge properties of extruded dilute Mg-0.5Bi-0.5Sn-based(wt.%)alloys as anodes for Mg-air batteries are evaluated.The grain size and texture intensity of the Mg-Bi-Sn-based alloys are significantly decreased after the Ca/In alloying,particularly for the In-containing alloy.Note that,in addition to nanoscale Mg_(3)Bi_(2)phase,a new microscale Mg_(2)Bi_(2)Ca phase forms in the Ca-containing alloy.The electrochemical test results demonstrate that Ca/In micro-alloying can enhance the electrochemical activity.Using In to alloy the Mg-Bi-Sn-based alloy is effective in restricting the cathodic hydrogen evolution(CHE)kinetics,leading to a low self-corrosion rate,while severe CHE occurred after Ca alloying.The micro-alloying of Ca/In to Mg-Bi-Sn-based alloy strongly deteriorates the compactness of discharge products film and mitigates the"chunk effect"(CE),hence the cell voltage,anodic efficiency as well as discharge capacity are greatly improved.The In-containing alloy exhibits outstanding discharge performance under the combined effect of the modified microstructure and discharge products,thus making it a potential anode material for primary Mg-air battery.

Patient-derived induced pluripotent stem cells with a MERTK mutation exhibit cell junction abnormalities and aberrant cellular differentiation potential

BACKGROUND Human induced pluripotent stem cell(hiPSC)technology is a valuable tool for generating patient-specific stem cells,facilitating disease modeling,and invest-igating disease mechanisms.However,iPSCs carrying specific mutations may limit their clinical applications due to certain inherent characteristics.AIM To investigate the impact of MERTK mutations on hiPSCs and determine whether hiPSC-derived extracellular vesicles(EVs)influence anomalous cell junction and differentiation potential.METHODS We employed a non-integrating reprogramming technique to generate peripheral blood-derived hiPSCs with and hiPSCs without a MERTK mutation.Chromo-somal karyotype analysis,flow cytometry,and immunofluorescent staining were utilized for hiPSC identification.Transcriptomics and proteomics were employed to elucidate the expression patterns associated with cell junction abnormalities and cellular differentiation potential.Additionally,EVs were isolated from the supernatant,and their RNA and protein cargos were examined to investigate the involvement of hiPSC-derived EVs in stem cell junction and differentiation.RESULTS The generated hiPSCs,both with and without a MERTK mutation,exhibited normal karyotype and expressed pluripotency markers;however,hiPSCs with a MERTK mutation demonstrated anomalous adhesion capability and differentiation potential,as confirmed by transcriptomic and proteomic profiling.Furthermore,hiPSC-derived EVs were involved in various biological processes,including cell junction and differentiation.CONCLUSION HiPSCs with a MERTK mutation displayed altered junction characteristics and aberrant differentiation potential.Furthermore,hiPSC-derived EVs played a regulatory role in various biological processes,including cell junction and differentiation.
