G protein-coupled receptors(GPCRs),the largest family of human membrane proteins and an important class of drug targets,play a role in maintaining numerous physiological *** or antagonist,orthosteric effects or allosteric effects,and biased signaling or balanced signaling,characterize the complexity of GPCR dynamic *** this study,we first review the structural advancements,activation mechanisms,and functional diversity of *** then focus on GPCR drug discovery by revealing the detailed drug-target interactions and the underlying mechanisms of orthosteric drugs approved by the US Food and Drug Administration in the past five ***,an up-to-date analysis is performed on available GPCR structures complexed with synthetic small-molecule allosteric modulators to elucidate key receptor-ligand interactions and allosteric ***,we highlight how the widespread GPCR-druggable allosteric sites can guide structure-or mechanism-based drug design and propose prospects of designing bitopic ligands for the future therapeutic potential of targeting this receptor family.

Micro(nano)plastics(MNPs)have become a significant environmental concern due to their widespread presence in the biosphere and potential harm to ecosystems and human ***,we propose for the first time a MNPs capture,utilization,and storage(PCUS)concept to achieve MNPs remediation from water while meeting economically productive upcycling and environmentally sustainable plastic waste management.A highly efficient capturing material derived from surface-modified woody biomass waste(M-Basswood)is developed to remove a broad spectrum of multidimensional and compositional MNPs from *** M-Basswood delivered a high and stable capture efficiency of>99.1%at different pH or salinity *** exceptional capture performance is driven by multiscale interactions between M-Basswood and MNPs,involving physical trapping,strong electrostatic attractions,and triggered MNPs cluster-like aggregation ***,the in vivo biodistribution of MNPs shows low ingestion and accumulation of MNPs in the mice *** MNPs remediation from water,the M-Basswood,together with captured MNPs,is further processed into a high-performance composite board product where MNPs serve as the glue for utilization and ***,the life cycle assessment(LCA)and techno-economic analysis(TEA)results demonstrate the environmental friendliness and economic viability of our proposed full-chain PCUS strategy,promising to drive positive change in plastic pollution and foster a circular economy.

Cancer immunotherapy has achieved tremendous successful clinical results and obtained historic victories in tumor ***,great limitations associated with feeble immune responses and serious adverse effects still cannot be neglected due to the complicated multifactorial etiology and pathologic microenvironment in *** rapid development of nanomedical science and material science has facilitated the advanced progress of engineering biomaterials to tackle critical *** supramolecular biomaterials with flexible and modular structures have exhibited unparalleled advantages of high cargo-loading efficiency,excellent biocompatibility,and diversiform immunomodulatory activity,thereby providing a powerful weapon for cancer *** past decades,supramolecular biomaterials were extensively explored as versatile delivery platforms for immunotherapeutic agents or designed to interact with the key moleculars in immune system in a precise and controllable *** this review,we focused on the crucial role of supramolecular biomaterials in the modulation of pivotal steps during tumor immunotherapy,including antigen delivery and presentation,T lymphocyte activation,tumor-associated macrophage elimination and repolarization,and myeloid-derived suppressor cell *** on extensive research,we explored the current limitations and development prospects of supramolecular biomaterials in cancer immunotherapy.

Cyanoethylation of phenylamine is one of the important steps for the production of dicyanoethyl-based disperse ***,the exothermic nature of this reaction and the inherent instability of intermittent dynamic operation pose challenges in achieving both high safety and reaction *** this study,a continuous cyanoethylation of phenylamine for synthesizing N,N-dicyanoethylaniline in a microreactor system has been *** optimizing the reaction conditions,the reaction time was significantly reduced from over 2 h in batch operation to approximately 14 min in the microreactor,while high conversion and selectivity were *** on the reaction network constructed,the reaction kinetics was established,and the kinetic parameters were then *** findings provide valuable insights into a controllable cyanoethylation reaction,which would be helpful for the design of efficient processes and optimization of reactors.

Polycyclic aromatic hydrocarbons(PAHs),PANHs,and peptoids dust spectral calculations from the interstellar medium(ISM)are important for dust observations and *** goal is to calculate the radiation spectrum of spherical PAHs dust clusters in a vacuum containing ionized and applied in the presence of an electric *** propose a new simple computational model to calculate the size of three-dimensional spherical dust clusters formed by different initial dust *** the Vienna Ab-initio Simulation Package code,the density functional theory with the generalized approximation was used to calculate the electron density gradient and obtain the radiation spectrum of *** the radius of spherical dust clusters is~[0.009-0.042]μm,the dust radiation spectrum agrees well with the Z=0.02 mMMP stellar spectra,and the PAHs radiation spectrum of NGC 4676 at wavelengths of(0-5]μm and(5-10]μm,*** the ionized state,the N-PAH,C_(10)H_(9)N,2(C_(4)H_(4))^(1+),and peptoids 4(CHON),(C_(8)H_(10)N_(2)O_(5))^(1+)dust clusters at 3.3μm,while the 2(C_(22)H_(21)N_(3)O_(2))^(1+),4(CHON)dust clusters at 5.2μm have obvious *** is a characteristic of part of PAHs and peptoids clusters radiation at the nearinfrared wavelength of 2μ***,especially after applying an electric field to the dust,the emission spectrum of the dust increases significantly in the radiation wavelength range[3-10]μ***,the dust clusters of PAHs,PANHs,and peptoids of the radius size~[0.009-0.042]μm are likely to exist in the ISM.

Spatially resolved transcriptomics(SRT)is capable of comprehensively characterizing gene expression patterns and providing an unbiased image of spatial *** fully understand the organizational complexity and tumor immune escape mechanism,we propose stMGATF,a multiview graph attention fusion model that integrates gene expression,histological images,spatial location,and gene *** better extract information,stMGATF exploits SimCLRv2 for visual feature exaction and employs edge feature enhanced graph attention networks for the learning potential embedding of each view.A global attention mechanism is used to adaptively integrate 3 views to obtain low-dimensional *** to diverse SRT datasets,stMGATF is robust and outperforms other methods in detecting spatial domains and denoising data even with different resolutions and *** particular,stMGATF contributes to the elucidation of tissue heterogeneity and extraction of 3-dimensional expression ***,considering the associations between genes in tumors,stMGATF can identify the spatial dark genes ignored by traditional methods,which can be used to predict tumor-driving transcription factors and reveal tumor immune escape mechanisms,providing theoretical evidence for the development of new immunotherapeutic strategies.

Owing to their low flexibility,poor processability and a lack of responsiveness,inorganic materials are usually non-ideal for constructing a living ***,to date,lifelike materials with structural hierarchies and adaptive properties usually rely on light and soft organic molecules,although few exceptions have been acquired using two-dimensional(2D)inorganic ***,with a systematic study on the gelation behavior of carbon-based 0D quantum dots,1D nanotubes,and 3D fullerenes,we find that acidified 1D carbon nanotubes(CNTs)can serve as an alternative building block for fabricating purely inorganic biomimetic soft *** as-prepared CNT gels exhibit not only a pH-or photothermal-triggered mechanical and tribological adaptivity,which allows them to simulate the behavior of sea cucumbers,peacock mantis shrimps,and mammalian muscles or cortical bones,but also a unique damping property that is similar to spider’s cuticular *** high elasticity,effective lubrication,excellent biocompatibility,and controllable friction and wear also allow them to function as a new type of smart lubricants,whose tribological properties can be regulated either by its internal pH changes or spatiotemporally by near-infrared(NIR)light irradiations,free of any toxic and flammable base oils or additives.

Drought stress limits agricultural productivity *** and characterizing genetic components of drought stress-tolerance networks may improve crop resistance to drought *** show that the regulatory module formed by miR166 and its target gene,ATHB14-LIKE,functions in the regulation of drought tolerance in soybean(Glycine max).Drought stress represses the accumulation of miR166,leading to upregulation of its target *** knockdown of miR166 in the stable transgenic line GmSTTM166 conferred drought tolerance without affecting *** of ABA signaling pathway genes was regulated by the miR166-mediated regulatory pathway,and ATHB14-LIKE directly activates some of these *** is a feedback regulation between ATHB14-LIKE and MIR166 genes,and ATHB14-LIKE inhibits MIR166 *** findings reveal that drought-triggered regulation of the miR166-mediated regulatory pathway increases plants drought resistance,providing new insights into drought stress regulatory network in soybean.

As a byproduct of water treatment,drinking water treatment aluminum sludge(DWTAS)has challenges related to imperfect treatment and disposal,which has caused potential harm to human health and the *** this paper,heat treatment DWTAS as a supplement cementitious material was used to prepare a green cementing *** results show that the 800℃ is considered as the optimum heat treatment temperature for ***-800℃ is fully activated after thermal decomposition to form incompletely crystallized highly activeγ-Al2O3 and active SiO_(2).The addition of DWTAS promoted the formation of ettringite and C-(A)-S-H gel,which could make up for the low early compressive strength of cementing materials to a certain *** cured for 90 days,the compressive strength of the mortar with 30% DWTAS-800℃ reached 44.86 *** dynamic process was well simulated by Krstulovi′c-Dabi′c hydration kinetics *** study provided a methodology for the fabrication of environmentally friendly and cost-effective compound cementitiousmaterials and proposed a“waste-to-resource”strategy for the sustainable management of typical solid wastes.
