Corilagin alleviates podocyte injury in diabetic nephropathy by regulating autophagy via the SIRT1-AMPK pathway

BACKGROUND Diabetic nephropathy(DN)is the most frequent chronic microvascular consequence of diabetes,and podocyte injury and malfunction are closely related to the development of *** have shown that corilagin(Cor)has hepatoprotective,anti-inflammatory,antibacterial,antioxidant,anti-hypertensive,antidiabetic,and anti-tumor *** To explore the protective effect of Cor against podocyte injury in DN mice and the underlying *** Streptozotocin and a high-fat diet were combined to generate DN mice models,which were then divided into either a Cor group or a DN group(n=8 in each group).Mice in the Cor group were intraperitoneally injected with Cor(30 mg/kg/d)for 12 wk,and mice in the DN group were treated with *** analysis was used to measure the blood lipid *** and eosin staining was used to detect pathological changes in kidney *** and Western blotting were used to assess the protein expression of nephrin and *** podocyte cells(MPC5)were cultured and treated with glucose(5 mmol/L),Cor(50μM),high glucose(HG)(30 mmol/L),and HG(30 mmol/L)plus Cor(50μM).Real-time quantitative PCR and Western blotting RESULTS Compared with the control group,the DN mice models had increased fasting blood glucose,glycosylated hemoglobin,triglycerides,and total cholesterol,decreased nephrin and podocin expression,increased apoptosis rate,elevated inflammatory cytokines,and enhanced oxidative *** of the conditions mentioned above were alleviated after intervention with *** addition,Cor therapy improved SIRT1 and AMPK expression(Pting autophagy via the SIRT1-AMPK pathway,thereby exerting its protective impact on renal function in DN mice.

Postnatal development of rat retina:a continuous observation and comparison between the organotypic retinal explant model and in vivo development

The organotypic retinal explant culture has been established for more than a decade and offers a range of unique advantages compared with in vivo experiments and cell ***,the lack of systematic and continuous comparison between in vivo retinal development and the organotypic retinal explant culture makes this model controversial in postnatal retinal development ***,we aimed to verify the feasibility of using this model for postnatal retinal development studies by comparing it with the in vivo *** this study,we showed that postnatal retinal explants undergo normal development,and exhibit a consistent structure and timeline with retinas in ***,we used SOX2 and PAX6 immunostaining to identify retinal progenitor *** then examined cell proliferation and migration by immunostaining with Ki-67 and doublecortin,***-67-and doublecortin-positive cells decreased in both in vivo and explants during postnatal retinogenesis,and exhibited a high degree of similarity in abundance and distribution between ***,we used Ceh-10 homeodomain-containing homolog,glutamate-ammonia ligase(glutamine synthetase),neuronal nuclei,and ionized calcium-binding adapter molecule 1 immunostaining to examine the emergence of bipolar cells,Müller glia,mature neurons,and microglia,*** timing and spatial patterns of the emergence of these cell types were remarkably consistent between in vivo and explant *** study showed that the organotypic retinal explant culture model had a high degree of consistency with the progression of in vivo early postnatal retina *** findings confirm the accuracy and credibility of this model and support its use for long-term,systematic,and continuous observation.

Encapsulating perovskite quantum dots into 3D COF for visible light-driven CO2 reduction

Lead halide perovskite quantum dots（LHP QDs） have been revealed to possess great potential in photocatalytic applications including CO2reduction, which however suffer from poor stability. Herein, a high crystalline hydrazine-linked three-dimensional（3D） covalent organic framework, USTB-17, was fabricated from the reaction between 12-connected building block and 4-connected 3,5,7-tetrakis（4-aldophenyl）-adamantane. Post-modification with Ni2+affords the metallic framework USTB-17（Ni）followed by sequential deposition of the CH3NH2PbI3（MAPbI3） perovskite QDs into its pores, generating the USTB-17（Ni）@MAPbI3composite. Powder X-ray diffraction analysis together with theoretical simulations and transmission electron microscopy discloses the crystalline nature of USTB-17, USTB-17（Ni）, and USTB-17（Ni）@MAPbI3with an unprecedented noninterpenetrated hpt topology. The close contact of QDs inside the COF pores with the Ni catalytic site locating at the pore surface of COF allows a rapid transfer of the photogenerated electrons in QDs to the Ni catalytic sites, enhancing the photocatalytic activity for CO2reduction. This endows USTB-17（Ni）@MAPbI3with efficient photocatalysis performance for photocatalytic CO2reduction with CO generation rate of 365 μmol g-1h-1and CO selectivity up to 96% under visible-light irradiation, 7 times higher than that of USTB-17（Ni）. After four cycles of reactions, the photocatalytic CO generation rate remains almost unchanged, demonstrating its excellent cycle stability.

Background This study aimed to explore the relationship of 25-hydroxyvitamin D[25(OH)D]in three trimesters and at birth with neurodevelopment at 24 months of *** From 2013 to 2016,pregnant women from the Shanghai Birth Cohort in China were recruited for the ***,649 mother-infant pairs were *** 25(OH)D was measured with mass spectrometry in three trimesters,and cord blood was divided into deficiency(ting for confounding factors,cord blood 25(OH)D in the sufficient group was positively correlated with cognitive[β=11.43,95%confidence interval(CI)=5.65–17.22],language(β=6.01,95%CI=1.67–10.3),and motor scores(β=6.43,95%CI=1.73–11.1);cord blood 25(OH)D in the insufficient group was also positively correlated with cognitive scores(β=9.42,95%CI=3.74–15.11).Additionally,sufficient vitamin D status in the four periods and persistent 25(OH)D≥30 ng/mL throughout pregnancy were associated with a lower risk of suboptimal cognitive development in adjusted models,although the effects were attenuated after applying the false discovery rate *** Cord blood 25(OH)D≥12 ng/mL has a significant positive association with cognitive,language,and motor development at 24 months of *** vitamin D status in pregnancy might be a protective factor for suboptimal neurocognition development at 24 months of age.

Insight into the formation mechanism of levoglucosenone in phosphoric acid-catalyzed fast pyrolysis of cellulose

The catalytic fast pyrolysis of cellulose impregnated with phosphoric acid (H3PO4) offers a promising method for the selective production of levoglucosenone (LGO),a valuable anhydrosugar ***,the fundamental mechanism for selective LGO formation is ***,quantum chemistry calculations and catalytic fast pyrolysis experiments were performed to reveal the formation mechanism of LGO in H3PO4-catalyzed cellulose pyrolysis.H3PO4 significantly decreased the energy barriers of the pyrolytic reactions and altered the competitiveness of the LGO formation pathways,promoting LGO *** different pathways in the non-catalytic and H3P04-catalyzed conditions,LGO is mainly produced from the primary decomposition of glucose units of cellulose and secondary conversion of *** major catalytic formation pathways of LGO comprise similar reactions,with dehydration at the 3-OH+2-H site as the rate-determining ***,secondary conversion of 1,4;3,6-dianhydro-α-D-glucopyranose is not feasible for LGO formation,in contrast to previous *** addition,a high degree of polymerization is beneficial for the selectivity of LGO formation in the catalytic process,because the glycosidic bond is important for the formation of the bicyclic structure (1,5-and1,6-acetal rings).

Bimetallic anchoring catalysis for C–H and C–C activation

Catalytic Characteristics of Lipase Immobilized in CTAB/TX-100 MBG

CTAB/TX-100 microemtdsion-based gel（MBG）, which could be soaked in aqueous solution for a long time without mechanical strength and shape changes, was successfully prepared. Lipase immobilized in this gel had a higher activity than that in CTAB MBG when catalyzed the esterification reaction between n-hexanoic acid and n-octanol and its enantioselectivity was slightly higher when catalyzed the stereoselective esterification reaction between racemic ibuprofen and n-octanol. In aqueous solution, lipase in CTAB/TX-100 MBG could catalyze smoothly the hydrolysis reaction of olive oil and could be reused many times without the evident change of MBG mechanical strength although its activity decreased 41.8% after nine-time recycles.
