BACKGROUND Activated hepatic stellate cells(aHSCs)are the major source of cancer-associated fibroblasts in the liver.Although the crosstalk between aHSCs and colorectal cancer(CRC)cells supports liver metastasis(LM),the mechanisms are largely unknown.AIM To explore the role of BMI-1,a polycomb group protein family member,which is highly expressed in LM,and the interaction between aHSCs and CRC cells in promoting CRC liver metastasis(CRLM).METHODS Immunohistochemistry was carried out to examine BMI-1 expression in LM and matched liver specimens of CRC.The expression levels of BMI-1 in mouse liver during CRLM(0,7,14,21,and 28 d)were detected by Western blotting(WB)and the quantitative polymerase chain reaction(qPCR)assay.We overexpressed BMI-1 in HSCs(LX2)by lentivirus infection and tested the molecular markers of aHSCs by WB,qPCR,and the immunofluorescence assay.CRC cells(HCT116 and DLD1)were cultured in HSC-conditioned medium(LX2 NC CM or LX2 BMI-1 CM).CM-induced CRC cell proliferation,migration,epithelial-mesenchymal transition(EMT)phenotype,and transforming growth factor beta(TGF-β)/SMAD pathway changes were investigated in vitro.A mouse subcutaneous xenotransplantation tumor model was established by co-implantation of HSCs(LX2 NC or LX2 BMI-1)and CRC cells to investigate the effects of HSCs on tumor growth and the EMT phenotype in vivo.RESULTS Positive of BMI-1 expression in the liver of CRLM patients was 77.8%.The expression level of BMI-1 continued to increase during CRLM in mouse liver cells.LX2 overexpressed BMI-1 was activated,accompanied by increased expression level of alpha smooth muscle actin,fibronectin,TGF-β1,matrix metalloproteinases,and interleukin 6.CRC cells cultured in BMI-1 CM exhibited enhanced proliferation and migration ability,EMT phenotype and activation of the TGF-β/SMAD pathway.In addition,the TGF-βR inhibitor SB-505124 diminished the effect of BMI-1 CM on SMAD2/3 phosphorylation in CRC cells.Furthermore,BMI-1 overexpressed LX2 HSCs promoted tumor growth and the EMT phenotype in vivo.CO

Most real estate agents develop new objects by visiting unfamiliar clients, distributing leaflets, or browsing other real estate trading website platforms,whereas consumers often rely on websites to search and compare prices when purchasing real property. In addition to being time consuming, this search processrenders it difficult for agents and consumers to understand the status changes ofobjects. In this study, Python is used to write web crawler and image recognitionprograms to capture object information from the web pages of real estate agents;perform data screening, arranging, and cleaning;compare the text of real estateobject information;as well as integrate and use the convolutional neural networkof a deep learning algorithm to implement image recognition. In this study, dataare acquired from two business-to-consumer real estate agency networks, i.e., theSinyi real estate agent and the yungching real estate agent, and one consumer-toconsumer real estate agency platform, i.e., the, FiveNineOne real estate agent. Theresults indicate that text mining can reveal the similarities and differences betweenthe objects, list the number of days that the object has been available for sale onthe website, and provide the price fluctuations and fluctuation times during thesales period. In addition, 213,325 object amplification images are used as a database for training using deep learning algorithms, and the maximum image recognition accuracy achieved is 95%. The dynamic recommendation system for realestate objects constructed by combining text mining and image recognition systems enables developers in the real estate industry to understand the differencesbetween their commodities and other businesses in approximately 2 min, as wellas rapidly determine developable objects via comparison results provided by thesystem. Meanwhile, consumers require less time in searching and comparingprices after they have understood the commodity dynamic information, therebyallowing them to use the most efficient approach to purchase

In this paper,a stiffening design imitating the bamboo node is proposed for weight reduction of the long composite pipe beam subjected to bending load.The distribution of bamboo nodes can efficiently suppress the ovalization of the section,thus significantly improving the bending resistance of the bamboo.Based on this principle,ring stiffeners are proposed to be fixed to the pipe beam,making the long beam equivalent to the combination of a series of short pipes that suffered less section ovalization.A database of the optimal laminate orientations for different normalized lengths is obtained through optimizations,where the discreteness of the ply count is considered.Based on this database,weight optimizations are conducted,and the optimal designs of beams with and without stiffeners are obtained and compared.The comparison results show that the proposed bamboo-like stiffened beam not only regains a near-linear load–displacement relationship,but also reduces the weight by up to 16%under the same buckling load.In addition,it is found that for the pipe beams with radius-to-thickness ratios of more than 18,increasing the radius leads to a decrease in elastic buckling resistance when the weight remains a constant,which is opposite to the design for strength and stiffness.The model and database developed in this paper can provide a reference for weight reduction design and weight estimation for composite pipe beams.

Objective:To investigate the anti-coronavirus potential and the corresponding mechanisms of the two ingredients of Reduning Injection:quercetin and luteolin.Methods:A pseudovirus system was designed to test the efficacy of quercetin and luteolin to inhibit SARS-CoV-2 infection and the corresponding cellular toxicity.Luteolin was tested for its activities against the pseudoviruses of SARS-CoV-2 and its variants.Virtual screening was performed to predict the binding sites by Autodock Vina 1.1.230 and PyMol.To validate docking results,surface plasmon resonance(SPR)was used to measure the binding affinity of the compounds with various proteins of the coronaviruses.Quercetin and luteolin were further tested for their inhibitory effects on other coronaviruses by indirect immunofluorescence assay on rhabdomyosarcoma cells infected with HCoV-OC43.Results:The inhibition of SARS-CoV-2 pseudovirus by luteolin and quercetin were strongly dose-dependent,with concentration for 50%of maximal effect(EC_(50))of 8.817 and 52.98μmol/L,respectively.Their cytotoxicity to BHK21-hACE2 were 177.6 and 405.1μmol/L,respectively.In addition,luetolin significantly blocked the entry of 4 pseudoviruses of SARS-CoV-2 variants,with EC50lower than 7μmol/L.Virtual screening and SPR confirmed that luteolin binds to the S-proteins and quercetin binds to the active center of the 3CLpro,PLpro,and helicase proteins.Quercetin and luteolin showed over 99%inhibition against HCoV-OC43.Conclusions:The mechanisms were revealed of quercetin and luteolin inhibiting the infection of SARS-CoV-2 and its variants.Reduning Injection is a promising drug for COVID-19.

Objectives Hand,foot and mouth disease(HFMD)is a widespread infectious disease that causes a significant disease burden on society.To achieve early intervention and to prevent outbreaks of disease,we propose a novel warning model that can accurately predict the incidence of HFMD.Methods We propose a spatial-temporal graph convolutional network(STGCN)that combines spatial factors for surrounding cities with historical incidence over a certain time period to predict the future occurrence of HFMD in guangdong and Shandong between 2011 and 2019.The 2011-2018 data served as the training and verification set,while data from 2019 served as the prediction set.Six important parameters were selected and verified in this model and the deviation was displayed by the root mean square error and the mean absolute error.Results As the first application using a STGCN for disease forecasting,we succeeded in accurately predicting the incidence of HFMD over a 12-week period at the prefecture level,especially for cities of significant concern.Conclusions This model provides a novel approach for infectious disease prediction and may help health administrative departments implement effective control measures up to 3 months in advance,which may significantly reduce the morbidity associated with HFMD in the future.

BACKGROUND Recent studies have demonstrated that dysfunction of the intestinal barrier is a significant contributing factor to the development of severe acute pancreatitis(SAP).A stable intestinal mucosa barrier functions as a major anatomic and functional barrier,owing to the balance between intestinal epithelial cell(IEC)proliferation and apoptosis.There is some evidence that calcium overload may trigger IEC apoptosis and that calcineurin(CaN)/nuclear factor of activated Tcells(NFAT)signaling might play an important role in calcium-mediated apoptosis.AIM To investigate the potential mechanisms underlying the therapeutic effect of Qingyi decoction(QYD)in SAP.METHODS A rat model of SAP was created via retrograde infusion of sodium deoxycholate.Serum levels of amylase,tumor necrosis factor(TNF-α),interleukin(IL)-6,D-lactic acid,and diamine oxidase(DAO);histological changes;and apoptosis of IECs were examined in rats with or without QYD treatment.The expression of the two subunits of CaN and NFAT in intestinal tissue was measured via quantitative realtime polymerase chain reaction and western blotting.For in vitro studies,Caco-2 cells were treated with lipopolysaccharide(LPS)and QYD serum,and then cell viability and intracellular calcium levels were detected.RESULTS Retrograde infusion of sodium deoxycholate increased the severity of pancreatic and intestinal pathology and the levels of serum amylase,TNF-α,and IL-6.Both the indicators of intestinal mucosa damage(D-lactic acid and DAO)and the levels of IEC apoptosis were elevated in the SAP group.QYD treatment reduced the serum levels of amylase,TNF-α,IL-6,D-lactic acid,and DAO and attenuated the histological findings.IEC apoptosis associated with SAP was ameliorated under QYD treatment.In addition,the protein expression levels of the two subunits of CaN were remarkably elevated in the SAP group,and the NFATc3 gene was significantly upregulated at both the transcript and protein levels in the SAP group compared with the control group.QYD significantly restrained CaN

Effects of residual plastic film on crop yield and soil fertility in a dryland farming system

Plastic film mulch in agricultural production becomes essential to maintaining crop yields in arid and semiarid areas.However,the presence of residual film in farmland soil has also drawn much attention.In this study,three experiments were conducted.The first two experimental designs included 0,450,1350,and 2700 kg ha^(-1) of residual film pieces of approximately 5 cm side length added to field soil(0-20 cm soil depth)for seven years and added to pots for four years.In the third experiment,1350 kg ha^(-1)of the residual film with different side lengths(2-5,5-10,10-15,and 15-20 cm)was added to field soil for six years to explore the effect of residual film fragment size on soil nutrients,soil microorganisms,crop growth and yields.The residual film had little effect on the soil moisture at a field depth of 0-2(or 0-1.8)m.There were no significant effects on organic carbon,total nitrogen,inorganic nitrogen,total phosphorus or available phosphorus in the 0-20 cm soil layer.The presence of residual film decreased the richness and diversity of the bacterial community of the surface soil of the residual film,but it had no significant effect on the microbial community of the non-surface soil.The emergence rates of wheat and lentils occasionally decreased significantly with different amounts of residue fragments added to the field.At 450-2700 kg ha^(-1),the residual film reduced the plant height and stem diameter of maize and significantly reduced the shoot biomass of harvested maize by 11-19%.The average yields of maize and potato over the seven years decreased,but there were almost no significant statistical differences among the treatments.These results provide important data for a comprehensive scientific understanding of the effects of residual film on soil and crops in dryland farming systems.

Background Depression,anxiety and schizophrenia among older persons have become global public health challenges.However,the burden of these disorders in ageing and aged countries has not been analysed.Aims To investigate the burden of depression,anxiety and schizophrenia among older adults in ageing and aged countries.Methods Using data from the Global Burden of Disease Study 2019,we calculated the estimated annual percentage change(EAPC)in the age-standardised incidence rates(ASiR)and age-standardised disability-adjusted life years(DALYs)rates(ASDR)for depression,anxiety and schizophrenia of older people in ageing countries(China,India,Indonesia)and aged countries(Japan,Italy,Portugal)between 1990 and 2019.Trends in incidence and DALYs were analysed by gender and age.Results In 2019,the highest incidence of depression,anxiety and schizophrenia in the older population in aged countries was in Japan(927271.3(752552.3-1125796.5),51498.2(37625.7-70487.3)and 126.0(61.0-223.2),respectively),while the highest incidence in ageing countries was in China(5797556.9(4599403.4-7133006.5),330256.1(246448.9-445987.4)and 1067.7(556.2-1775.9),respectively).DALYs for these disorders were similar,with the highest in Japan and China.From 1990 to 2019,the ASIR for depressive disorders decreased in aged countries but increased in ageing countries;the ASIR for anxiety disorders and schizophrenia declined in both ageing and aged countries.The ASDR for depressive disorders was consistent with the ASIR but not for anxiety disorders and schizophrenia.The ASIR for depressive disorders was higher in older women,while the opposite was observed in anxiety disorders and schizophrenia.Notably,the conditions of burden of depressive disorders,anxiety disorders and schizophrenia in the 65-70-year-old age group were the most burdensome.Conclusions The incidence and DALYs of these three mental disorders increased while exhibiting differences between ageing and aged countries.Raising awareness about formulating health policies for preventing and trea
