Objective: a.dehyde dehydrogena.e (a.DH) enzyma.ic a.tivity identifies ova.ia. ca.cer stem-like cells. We investiga.ed the a.tineopla.tic a.tivity of the a.DH inhibitor Disulfira. on bulk ova.ia. ca.cer cells a.d CD133+/a.DH+ ca.cer stem-like cells. Study Design: Ova.ia. ca.cer cell lines, huma. ova.ia. surfa.e epithelia. cells, a.d mesenchyma. stem cells were trea.ed with increa.ing concentra.ions of Disulfira. a.d/or Cispla.in in vitro. Trea.ed cells were a.sessed for via.ility or Fa.S-a.a.yzed for either percenta.e of ova.ia. ca.cer stem-like cells or induction of a.optosis. Disulfira.’s impa.t on ca.cer stem-like cells wa. tested in vitro using tumor sphere forma.ion a.sa.s a.d in vivo using tumor initia.ion a.sa.s with in vitro-trea.ed a.780 cells in NSG mice. Fina.ly, Disulfira.’s in vivo a.tivity wa. a.sessed versus CD133+/a.DH+ cell-initia.ed tumor xenogra.ts. Results: Disulfira. demonstra.ed a.tineopla.tic a.tivity a.a.nst multiple ova.ia. ca.cer cell lines. While Disulfira. ha. limited in vitro toxicity a.a.nst huma. ova.ia. surfa.e epithelia. cells or mesenchyma. stem cells (IC50 of ~15 μM a.d >30 μM, respectively), its a.tineopla.tic a.tivity a.a.nst cell lines wa. compa.a.le to Cispla.in (IC50 ~1.5 μM). Disulfira.-media.ed cell dea.h wa. due, a. lea.t in pa.t, to induction of a.optosis. Disulfira. a.tivity wa. a.ditive with chemothera.y. Disulfira. demonstra.ed selective depletion of CD44+ cells but not the CD133+ ca.cer stem-like cells. Disulfira. ha. no thera.eutic impa.t on tumor initia.ion studies or in vivo thera.y of whole cell line or stem cell-initia.ed tumor xenogra.ts. Conclusions: In biologica.ly releva.t concentra.ions, Disulfira. ha. clea. a.tineopla.tic a.tivity a.a.nst ova.ia. ca.cer cells in vitro. Disulfira. selectively depleted CD44+ but not CD133+ ova.ia. ca.cer stem-like cells in vitro. However, Disulfira. ha. no significa.t a.tivity in vivo.
