IL-1β activates p44/42 and p38 mitogen-activated protein kinases via different pathways in cat esophageal smooth muscle cells

AIM： To examine the pathway related to the IL-1β induced activation of mitogen-activated protein （MAP） kinases in cat esophageal smooth muscle cells. METHODS： Culture of the esophageal smooth muscle cells from cat was prepared. Specific inhibitors were treated before applying the IL-β3. Western blot analysis was performed to detect the expressions of COX, iNOS and MAP kinases. RESULTS： In the primary cultured cells, although IL-β3 failed to upregulate the COX and iNOS levels, the levels of the phosphorylated forms of 1344142 HAP kinase and p38 MAP kinase increased in both concentration- and time-dependent manner, of which the level of activation reached a maximum within 3 and 18 h, respectively. The pertussis toxin reduced the level of p44/42 MAP kinase phosphorylation. Tyrphostin 51 and genistein also inhibited this activation. Neomycin decreased the density of the p44/42 HAP kinase band to the basal level. Phosphokinase C （PKC） was found to play a mediating role in the IL-1β-induced p44/42 MAP kinase activity. In contrast, the activation of p38 MAP kinase was inhibited only by a pretreatment with forskolin, and was unaffected by the other compounds. CONCLUSION： Based on these results, IL-1β-induced p44/42 MAP kinase activation is mediated by the Gi protein, tyrosine kinase, phospholipase C （PLC） and PKC. The pathway for p38 MAP kinase phosphorylation is different from that of p44/42 MAP kinase, suggesting that it plays a different role in the cellular response to IL- 1β.

AIM： To investigate the mechanism of bombesin-induced circular smooth muscle cell contraction in cat esophagus. METHODS： Specific G protein or phospholipase C involved in cat esophagus contraction was identified, muscle cells were permeabilized with saponin. After per- meabilization of muscle cells, the Gi3 antibody inhibited bombesin-induced smooth muscle cell contraction. RESULTS： Incubation of permeabilized circular muscle cells with PLC-β3 antibody could inhibit bombesin-induced contraction. H-7, chelerythrine （PKC inhibitor） and genistein （protein tyrosine kinase inhibitor） inhibited bombesin-induced contraction, but DAG kinase inhibitor, R59949, could not inhibit it. To examine which mitogen-activated protein kinase （MAPK） was involved in bombesin-induced contTaction, the specific MAPK inhibitors （MEK inhibitor, PD98059 and p38 MAPK inhibitor, SB202190） were used. Preincubation of PD98059 blocked the contraction induced by bombesin in a concentration-dependent manner. However, SB202190 had no effects on contraction. CONCLUSION： Bombesin-induced circular muscle cell contraction in cat esophagus is mediated via a PKC or a PTK-dependent pathway or p44/p42 HAPK pathway.

To investigate the characteristic radiologic findings of cryptogenic multifocal ulcerous stenosing enteritis (CMUSE) which can be differentiated from other similar bowel disease and to assess their clinical behavior. METHODSTwenty pathologically and clinically confirmed CMUSE patients (males:females = 8:12; mean age: 40.4 years) between March 2002 and August 2015 from seven academic centers in South Korea were retrospectively reviewed. We evaluated small bowel series (SBS; n = 25), computed tomography (CT) enterography (n = 21), magnetic resonance (MR) enterography (n = 2), and abdominopelvic CT (n = 18) images, focusing on enteric and perienteric manifestations. Any change in radiologic features during follow-up period was recorded. We evaluated clinical data including presenting symptoms, laboratory finding and presence of relapse from electronic medical records. Histopathologic findings were also evaluated. RESULTSThe main symptoms were abdominal pain (n = 12) and anemia (n = 10). All patients showed small bowel strictures (n = 52, mean: 2.6 per patient) on initial CT/MR, located in the ileum (n = 47) or jejunum (n = 5). Strictures showed short-length (mean: 10.44 mm) and circumferential bowel wall thickening (mean: 5.56 mm) with layered enhancement (n = 48) that were also noted on initial SBS (n = 36) with shallow ulcers (n = 10). Some ulcerative lesions or wall thickening progressed into strictures on follow-up SBS/CT, and some strictures revealed recurrent ulceration on follow-up SBS. There were no penetrating disease features like fistula or abscess and no gastrointestinal tract involvement except the small bowel. Nine patients experienced disease recurrence (median relapse-free period: 32 mo) even post-operatively. Histopathologic features of surgically resected specimens were characterized as multiple superficial ulcerations confined to mucosa or submucosa and multiple strictures. Red ginseng is a well-known alternative medicine with anti-inflammatory *** exerts pharmacological effects through the transformation of saponin into metabolites by intestinal *** that intestinal microflora vary among individuals,the pharmacological effects of red ginseng likely vary among *** order to produce homogeneously effective red ginseng,we prepared probiotic-fermented red ginseng and evaluated its activity using a dextran sulfate sodium(DSS)-induced colitis model in *** analysis of intestinal damage indicated that the administration of probiotic-fermented red ginseng significantly decreased the severity of colitis,compared with the control and the activity was higher than that induced by oral administration of ginseng powder or probiotics *** analysis of the levels of serum IL-6 and TNF-α,inflammatory biomarkers that are increased at the initiation stage of colitis,were significantly decreased in probiotic-fermented red ginseng-treated groups in comparison to the control *** levels of inflammatory cytokines and mR NAs for inflammatory factors in colorectal tissues were also significantly decreased in probiotic-fermented red ginseng-treated ***,oral administration of probiotic-fermented red ginseng reduced the severity of colitis in a mouse model,suggesting that it can be used as a uniformly effective red ginseng product.

Task scheduling scheme by checkpoint sharing and task duplication in P2P-based desktop grids

A scheduling scheme is proposed to reduce execution time by means of both checkpoint sharing and task duplication under a peer-to-peer(P2P) architecture. In the scheme, the checkpoint executed by each peer(i.e., a resource) is used as an intermediate result and executed in other peers via its duplication and transmission. As the checkpoint is close to a final result, the reduction of execution time for each task becomes higher, leading to reducing turnaround time. To evaluate the performance of our scheduling scheme in terms of transmission cost and execution time, an analytical model with an embedded Markov chain is presented. We also conduct simulations with a failure rate of tasks and compare the performance of our scheduling scheme with that of the existing scheme based on client-server architecture. Performance results show that our scheduling scheme is superior to the existing scheme with respect to the reduction of execution time and turnaround time.

In this paper, a convex feasibility problem is considered. We construct an iterative method to approximate a common element of the solution set of classical variational inequalities and of the fixed point set of a strict pseudocontraction. Strong convergence theorems for the common element are established in the framework of Hilbert spaces.

Deformation prediction of porcelain-enameled steels with strain history by press forming and high-temperature behavior of coating layer

Porcelain enamel coatings were widely applied for the protection of steel products because they offered high corrosion protection,resistance to heat and abrasion,high hardness,hygiene and ease of *** typical process to produce enameled steels is roughly divided into two stages:the first stage consists of a forming process to give the desired shape to a steel substrate,and the second stage consists of a firing process to bond enamel frits on the *** firing process requires a high temperature above 800 °C,which may lead to austenitic transformation and severe thermal deformation of the steel *** aim of this study is to develop a finite element analysis (FE analysis) technique to predict the mechanical and thermal deformations of the enameled steels during forming and any further enameling process,including *** FE analysis involves analyzing the strain history of the steel substrate,which comprises the stress and thickness distributions of the substrate and its deformed shape,and the high-temperature behavior of the enamel coating *** validity of the FE analysis is verified through the U-bending test and firing test with various numbers and positions of enamel coating layers on the *** results reveal that the FE analysis results agree well with the experimental results with 8% error.

These systems span a large number of machines and store huge amount of data for a lot of *** the distributed storage systems,a row can be directly accessed using a row *** concentrate on a problem of efficient processing of queries whose predicate is on a column but not a row *** this paper,we present a cache management technique,called DICE which maintains query results of range queries to support the next range *** accelerate the search time of the cached query results,we use modified Interval Ski *** addition,we devise a novel cache replacement policy since DICE maintains an interval rather than a data *** our cache replacement policy considers the properties of intervals,our proposed technique is more efficient than traditional buffer replacement *** experimental result demonstrates the efficiency of our proposed technique.
