Extract from Zanthoxylum piperitum Induces Apoptosis of AGS Gastric Cancer Cells through Akt/MDM2/p53 Signaling Pathway

Objective:To determine the effect of Zanthoxylum piperitum extracet(ZPE)on apoptosis and analyze anticancer substances in ZPE,changes in proteins related to apoptosis,and pathological changes in tumors in ***:Fifteen 4-week-old female BALB/c nu/nu mice were divided into 3 groups depending on ZPE dose,with 5 in each *** gastric carcinoma cells(1 x 10^(6) cells/200 jxL)were subcutaneously injected into the flank of each *** week after the injection of AGS cells,ZPE was administered to the skin tissue[10 or 50 mg/(kg-d)]in the low-and high-dose groups,respectively for 20 *** animals were injected with vehicle *** 3 weeks,the tumor was extracted and carried out for immunohistochemistry,the tendency of apoptosis and p53 in the body was checked using TdT-mediated dUTP nick-end labeling(TUNEL)*** 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay,annexin V dead cell staining,cell cycle arrest and Western blotting,AGS gastric carcinoma cells were incubated with various concentrations of ZPE for 24 *** survival rates were analyzed by MTT *** was analyzed using annexin V dead cell staining and cell cycle arrest and measured using Muse cell ***:High performance liquid chromatography(HPLC)analysis showed that ZPE contained organic sulfur compounds such as alliin and *** assay results revealed that ZPE(10-85»xg/mL)could effectively inhibit the growth of AGS gastric cancer cells at higher concentrations(P<0.05,P<0.01).The annexin V&dead cell staining assay and cell cycle arrest assay confirmed a dose-dependent increase in the apoptosis rate and G!phase in ZPE(10-70 jig/mL)*** decreased the expression of anti-apoptotic proteins(p-Akt,p-MDM2,Bcl-2),while increased pro-apoptotic proteins(cleaved PARP,p53,pro-Caspase 3,Bax).TUNEL assays revealed an increase in cell *** staining confirmed the involvement of ***:ZPE decreases AGS cell

Clinical significance of MET gene amplification in metastatic or locally advanced gastric cancer treated with first-line fluoropyrimidine and platinum combination chemotherapy

Objective: To investigate the clinical significance of MET gene amplification in patients with gastric cancer in the palliative ***: MET amplification was assessed using fluorescence in situ hybridization(FISH) in 50 patients and quantitative polymerase chain reaction(qPCR) in 326 patients;259 patients treated with first-line fluoropyrimidine and platinum were included for survival ***: The results of FISH and qPCR indicated that the c-MET/CEP7 ratio was correlated with gene copy number. The optimal cutoff value for the copy number using qPCR to detect MET gene amplification with FISH was 5(κ=0.778, P5 detected by qPCR. MET-amplified gastric cancer was associated with an Eastern Cooperative Oncology Group(ECOG) performance status(PS) score of ≥2(33.3% vs. 10.5% P=0.007), peritoneal metastasis(76.2% vs. 46.2%, P=0.008), and elevated bilirubin levels(28.6% vs. 7.3%, P=0.006). The median overall survival(OS) and progression-free survival(PFS) were 11.9 and 5.6 months, respectively. MET-amplified gastric cancer was not associated with survival outcomes [hazard ratio(HR)=0.68, 95% confidence interval(95% CI): 0.35-1.32,P=0.254 for PFS;HR=0.68, 95% CI: 0.35-1.32, P=0.251 for OS].Conclusions: qPCR can be used to detect MET gene amplification. MET amplification was not a predictor of poor prognosis in patients with metastatic or unresectable gastric cancer.

Diagnostic accuracy of administrative database for bile duct cancer by ICD-10 code in a tertiary institute in Korea

Background:Administrative database provides valuable information for large cohort studies,especially when tissue diagnosis is rather difficult such as the diagnosis for bile duct cancer(BDC).The aim of this study was to evaluate the diagnostic accuracy of administrative database for BDC by International Classification of Diseases(ICD)-10 codes in a tertiary ***:BDC and control groups were collected from 2003 to 2016 at Seoul National University Bundang *** of BDC were identified in the National Health Insurance Service(NHIS)database by ICD 10-code supported by V *** control group was selected from cases without ICD-10 codes for BDC.A definite or possible diagnosis was defined according to pathologic *** records,images,and pathology reports were analyzed to evaluate ICD-10 codes for ***,specificity,positive predictive value,and negative predictive value for BDC were analyzed according to diagnostic criteria and cancer ***:A total of 1707 patients with BDC and 1707 controls were *** those with BDC,1320(77.3%)were diagnosed by definite ***(99.4%)of them had *** of definite diagnosis was the highest for ampulla of Vater(88.9%),followed by that for extrahepatic(84.9%)and intrahepatic(68.3%)*** positive cases commonly had hepatocellular *** overall diagnosis of BDC,sensitivity,specificity,positive predictive value,and negative predictive value were 99.94%,98.33%,98.30%,and 99.94%,*** accuracies were similar regardless of diagnostic criteria or tumor ***:Administrative database for BDC collected according to ICD-10 code with V code shows good accuracy.

Selexipag as Add-on Therapy for Patients with Pulmonary Arterial Hypertension Associated with Congenital Heart Disease:A Single-Center Retrospective Study

Purpose:This study examined the efficacy and safety of selexipag in treating pulmonary arterial hypertension(PAH)associated with congenital heart disease(CHD).Materials and Methods:We conducted a retrospective study of patients with CHD-associated PAH,treated with selexipag since December *** adult patients(mean age,45.4 years;women,77%)were treated with selexipag as add-on *** characteristics,World Health Organization functional class,6-minute walking distance(6MWD)test results,N-terminal pro-B-type natriuretic peptide levels,echocardiographic data,and incidence of side effects were ***:The majority of patients(12/13,92.3%)experienced more than one treatment-associated complication;one patient dropped out of the study due to intolerable *** results of 6MWD test(from 299.2±56.2 m to 363.8±86.5 m,p=0.039)and tricuspid regurgitation(TR)pressure gradient(from 84.7±20.5 mmHg to 61.6±24.0 mmHg,p=0.018)improved and remained improved after selexipag treatment in 12 *** on the results of a non-invasive risk assessment,8(66.7%)patients showed improvement,3(25.0%)showed no interval change,and the status of one patient(8.3%)***,compared to patients treated with a low dosage,patients treated with a medium-to-high dosage showed a greater increase in 6MWD results(88.3±26.4 m vs.55.3±27.6 m,p=0.043)and a greater reduction in the TR pressure gradient(-33.7±10.9 mmHg vs.-12.5±12.0 mmHg,p=0.015).Conclusion:Selexipag is an efficient pulmonary vasodilator as add-on therapy in treating CHD-associated PAH.

Fabrication of high-strength duplex nanoporous Cu by dealloying a dual-phase Mg-Cu precursor alloy

Duplex nanoporous Cu was successfully fabricated by dealloying a dual-phase Mg-Cu precursor alloy consisting of intermetallic Mg2Cu and *** duplex nanoporous Cu with embedded nanoporous struts exhibited highly enhanced strength compared to the typical monolithic nanoporous Cu under both compressive and flexural test conditions at room temperature;the duplex np-Cu sample exhibited a 12 times higher compressive strength and a 40 times greater flexural strength than the monolithic np-Cu *** responsible for the strength enhancement in the duplex nanoporous Cu are discussed.

We introduce a new function space that is much finer than the classical Lebesgue spaces, and investigate the continuity and the discontinuity of the Calderon-Zygmund operators on the new weighted spaces. In order to identify the continuity of singular integrals, we prove an interpolation theorem on the new spaces and propose a concept of the spectrum of base functions.

Smooth Schubert varieties in rational homogeneous manifolds of Picard number 1 are horospherical varieties. We characterize standard embeddings of smooth Schubert varieties in rational homogeneous manifolds of Picard number 1 by means of varieties of minimal rational tangents. In particular, we mainly consider nonhomogeneous smooth Schubert varieties in symplectic Grassmannians and in the 20-dimensional F_4- homogeneous manifold associated to a short simple root.

Primordial germ cell-mediated transgenesis and genome editing in birds

Transgenesis and genome editing in birds are based on a unique germline transmission system using primordial germ cells(PGCs), which is quite different from the mammalian transgenic and genome editing system. PGCs are progenitor cells of gametes that can deliver genetic information to the next generation. Since avian PGCs were first discovered in nineteenth century, there have been numerous efforts to reveal their origin, specification, and unique migration pattern, and to improve germline transmission efficiency. Recent advances in the isolation and in vitro culture of avian PGCs with genetic manipulation and genome editing tools enable the development of valuable avian models that were unavailable before. However, many challenges remain in the production of transgenic and genome-edited birds,including the precise control of germline transmission, introduction of exogenous genes, and genome editing in ***, establishing reliable germline-competent PGCs and applying precise genome editing systems are critical current issues in the production of avian models. Here, we introduce a historical overview of avian PGCs and their application, including improved techniques and methodologies in the production of transgenic and genome-edited birds, and we discuss the future potential applications of transgenic and genome-edited birds to provide opportunities and benefits for humans.

Dispersal patterns of exotic forest pests in South Korea

Invasive species have potentially devastating effects on ecological communi- ties and ecosystems. To understand the invasion process of exotic forest pests in South Korea, we reviewed four major species of exotic forest pests： the pine needle gall midge （Thecodiplosis japonensis）, pine wilt disease caused by the pine wood nematode （Bur- saphelenchus xylophilus）, the fall webworm （Hyphantria cunea） and the black pine bast scale （Matsucoccus thunbergianae）. We consider their biology, ecology, invasion history, dispersal patterns and related traits, and management as exotic species. Among these species, the dispersal process of fall webworm was linear, showing a constant range expan- sion as a function of time, whereas the other three species showed biphasic patterns, rapidly increasing dispersal speed after slow dispersal at the early invasion stage. Moreover, human activities accelerated their expansion, suggesting that prevention of the artificial movement of damaged trees would be useful to slow expansion of exotic species. We believe that this information would be useful to establish management strategies for invasion species.

Influence of dehydrating agents on the oxidative carbonylation of methanol for dimethyl carbonate synthesis over a Cu/Y-zeolite catalyst

The influence of the dehydration by metal oxides on the synthesis of dimethyl carbonate(DMC) via oxidative carbonylation of methanol was studied. A Cu/Y-zeolite catalyst was prepared by the ion exchange method from CuCl_2·2 H_2O and the commercial NH_4-form of the Y type zeolite. The catalyst was characterized by X-ray fluorescence(XRF), N_2 adsorption(BET method), X-ray diffraction(XRD), and temperature-programmed desorption of ammonia(NH_3-TPD) to evaluate its Cu and Cl content, surface area, structure, and acidity. Reaction tests were carried out using an autoclave(batch reactor) for 18 h at 403 K and 5.5 MPa(2CH_3OH + 1/2O_2+CO?(CH_3O)_2CO + H_2O). The influence of various dehydrating agents(ZnO, MgO, and CaO) was examined with the aim of increasing the methanol conversion(X_(MeOH), MeOH conversion). The MeOH conversion increased upon addition of metal oxides in the order CaO >> MgO > ZnO, with the DMC selectivity(SDMC) following the order MgO >CaO > ZnO. The catalysts and dehydrating agents were characterized before and after the oxidative carbonylation of methanol by thermogravimetric and differential thermogravimetric(TG/DTG), and XRD to confirm that the dehydration reaction occurred via the metal oxide(MO + H_2O → M(OH)_2). The MeOH conversion increased from 8.7% to 14.6% and DMC selectivity increased from 39.0% to 53.1%, when using the dehydrating agent CaO.
