Fine-tuning the structure-tolerance-antitumor efficacy axis of prodrug nanoassemblies via branched aliphatic functionalization

Small-molecule prodrug nanoassemblies have emerged as efficient antitumor drug delivery ***,in the case of camptothecins-based prodrug nanoassemblies,linear aliphatic side chain modification often results in rod-shaped or irregularly shaped nanoassemblies,which are highly unfavorable for sterilization through filtration,and may cause capillary blockage upon intravenous *** rational design of camptothecins-based prodrug nanoassemblies remains a ***,we propose that branched aliphatic alcohol(BAA)functionalization could fine-tune the structure-tolerance-antitumor efficacy axis of prodrug ***,four SN38-BAA prodrugs were synthesized by conjugating 7-ethyl-10-hydroxycamptothecin(SN38)with BAAs of varying lengths via a tumor redox-responsive disulfide bond,which self-assemble into uniform spherical *** length of BAA was found to significant impact the multiple drug delivery process,including colloidal stability,drug release profiles and ***,SN38-C21 NPs(SN38-11-heneicosanol nanoparticles),featuring the longest BAA,showcased multiple therapeutic advantages,ultimately culminating the optimal antitumor efficacy and *** findings underscore the potential of BAA functionalization in strengthening the therapeutic outcomes of prodrug nanoassemblies,and provide valuable insights for developing translational camptothecins-based nanomedicines.

Pitting is a common type of localized corrosion in passive alloys that can cause rapid failure of material or *** the case of stainless steels,non-metallic inclusions have been identified as the most susceptible sites for pitting,and have therefore garnered significant *** review critically ex-amines the issue of how inclusions induce pitting,with a particular focus on three mechanisms:sponta-neous dissolution of inclusions,active dissolution of Cr-depleted regions,and propagation of microcracks at the inclusion-matrix *** researchers have made significant strides in understanding these mechanisms over the past few decades,many gaps and controversies *** such as the ini-tial driving force of inclusion dissolution and factors affecting Cr-depleted regions require further ***,some old concepts and methods need to be revised to arrive at more credible *** review aims to delve deeply into these important issues and provide inspiration for future research.

Relationship between Authoritative Parenting Style and Preschool Children’s Emotion Regulation:A Moderated Mediation Model

An authoritative parenting style has been shown to promote children’s emotion regulation in European-American family ***,little is known about how sleep problems and the child’s sibling status in Chinese families affect this *** on family system theory,this study attempts to better understand the relationship between authoritative parenting style and emotion *** of preschool children in Chinese kindergartens completed questionnaires about their children’s sleep habits,their authoritative parenting styles,and children’s emotion regulation.A total of 531 children participated in this *** showed that authoritative parenting was positively associated with emotional *** problems mediated the effects of authoritative parenting style on emotion *** child’s sibling status moderated the mediating effects of sleep problems in authoritative parenting and emotion regulation ***,the relationship between the authoritative parenting style and sleep problems was significant for only children,while birth order had no significant influence on the authoritative parenting style and sleep problems in two-child *** findings suggest that a lowauthoritative parenting style predicts low emotion regulation through sleep problems,and this depends on the child’s sibling status,indicating that children without siblings may impair emotion regulation due to increased sleep problems.

Web Layout Design of Large Cavity Structures Based on Topology Optimization

Large cavity structures are widely employed in aerospace engineering, such as thin-walled cylinders, blades andwings. Enhancing performance of aerial vehicles while reducing manufacturing costs and fuel consumptionhas become a focal point for contemporary researchers. Therefore, this paper aims to investigate the topologyoptimization of large cavity structures as a means to enhance their performance, safety, and efficiency. By usingthe variable density method, lightweight design is achieved without compromising structural strength. Theoptimization model considers both concentrated and distributed loads, and utilizes techniques like sensitivityfiltering and projection to obtain a robust optimized configuration. The mechanical properties are checked bycomparing the stress distribution and displacement of the unoptimized and optimized structures under the sameload. The results confirm that the optimized structures exhibit improved mechanical properties, thus offering keyinsights for engineering lightweight, high-strength large cavity structures.

Enhanced Temporal Correlation for Universal Lesion Detection

Universal lesion detection(ULD)methods for computed tomography(CT)images play a vital role in the modern clinical medicine and intelligent *** is well known that single 2D CT slices lack spatial-temporal characteristics and contextual information compared to 3D CT ***,3D CT blocks necessitate significantly higher hardware resources during the learning ***,efficiently exploiting temporal correlation and spatial-temporal features of 2D CT slices is crucial for ULD *** this paper,we propose a ULD network with the enhanced temporal correlation for this purpose,named *** designed TCE module is applied to enrich the discriminate feature representation of multiple sequential CT ***,we employ multi-scale feature maps to facilitate the localization and detection of lesions in various *** experiments are conducted on the DeepLesion benchmark demonstrate that thismethod achieves 66.84%and 78.18%for FS@0.5 and FS@1.0,respectively,outperforming compared state-of-the-art methods.

Higher palaeoelevation in the Baoshan Basin:Implications for landscape evolution at the southeastern margin of the Tibetan Plateau

Surface uplift at the southeastern margin of the Tibetan Plateau has been widely studied,but more palaeoaltimetry data are required to better understand the elevation history of this geologically complex *** this study,fossil leaves of Abies(Pinaceae),a cool-temperate element,recovered from the latest Miocene-Pliocene Yangyi Formation of the southern Baoshan Basin,were used as a proxy to estimate the local *** on the regional modern altitude range(2100-4280 m)of the genus as well as regional temperature discrepancy(1.5℃)between the past and present,the palaeoelevation of the study area was calculated to be>2360 m above sea level as compared to 1670 m at *** result suggests that the southern Baoshan Basin experienced pronounced uplift prior to the time of fossil deposition,probably as a result of crustal shortening and thickening of the northern Baoshan Terrane during the *** infer that surface growth in areas south of the Dali Basin may have been greater than previously interpreted,and that a widespread plateau or plateau patches higher than 2000 m probably extended southwards into at least the Baoshan Basin by the latest *** also infer that the elevation of the southern Baoshan Basin has decreased by at least 690 m since then,in contrast to most other scenarios in which the elevation of the southeastern margin of the Tibetan Plateau has increased or remained close to modern levels since the late *** major cause of the inferred altitude decline is likely tectonic *** a transtensional graben basin,the Baoshan Basin has experienced pull-apart and base-fall movement since the late Miocene,which would reduce the altitude of its southern part located on the hanging *** erosion associated with the increased summer rainfall might also have played a role especially in reducing the local relief,although its contribution can be *** study provides one of the few palaeoelevation

Perturbation of autophagy pathways in murine alveolar macrophage by 2D TMDCs is chalcogen-dependent

Increasing risks of incidental and occupational exposures to two-dimensional transition metal dichalcogenides(2D TMDCs)due to their broad application in various areas raised their public health *** the composition-dependent cytotoxicity of 2D TMDCs has been well-recognized,how the outer chalcogenide atoms and inner transition metal atoms differentially contribute to their perturbation on cell homeostasis at non-lethal doses remains to be *** the present work,we compared the autophagy induction and related mechanisms in response to WS_(2),NbS_(2),WSe_(2)and Nb Se_(2)nanosheets exposures in MH-S murine alveolar *** these 2D TMDCs had comparable physicochemical properties,overall cytotoxicity and capability in triggering autophagy in MH-S cells,but showed outer chalcogen-dependent subcellular localization and activation of autophagy ***,WS_(2)and NbS_(2)nanosheets adhered on the cell surface and internalized in the lysosomes,and triggered m TOR-dependent activation of ***,WSe_(2)and Nb Se_(2)nanosheets had extensive distribution in cytoplasm of MH-S cells and induced autophagy in an m TOR-independent ***,the 2D TMDCs-induced perturbation on autophagy aggravated the cytotoxicity of respirable benzo[a]*** findings provide a deeper insight into the potential health risk of environmental 2D TMDCs from the perspective of homeostasis perturbation.

SDCC: software-defined collective communication for distributed training

Communication is crucial to the performance of distributed training. Today's solutions tightly couple the control and data planes and lack flexibility, generality, and performance. In this study, we present SDCC, a software-defined collective communication framework for distributed training. SDCC is based on the principle of modern systems design to effectively decouple the control plane from the data *** abstracts the operations for collective communication in distributed training with dataflow operations and unifies computing and communication with a single dataflow graph. The abstraction, together with the unification, is powerful: it enables users to easily express new and existing collective communication algorithms and optimizations, simplifies the integration with different computing engines(e.g., Py Torch and Tensor Flow) and network transports(e.g., Linux TCP and kernel bypass), and allows the system to improve performance by exploiting parallelism exposed by the dataflow graph. We further demonstrate the benefits of SDCC in four use cases.

Polymer donors with hydrophilic side-chains enabling efficient and thermally-stable polymer solar cells by non-halogenated solvent processing

Polymer solar cells(PSCs)with high power conversion efficiency(PCE)and environment-friendly fabrication are the main requirements enabling their production in industrial *** the use of non-halogenated solvent processing is inevitable for the PSC fabrication,it significantly reduces the processability of polymer donors(PDS)and small-molecule acceptors(SMAs).This often results in unoptimized blend morphology and limits the device *** address this issue,hydrophilic oligoethylene glycol(OEG)side-chains are introduced into a PD(2EG)to enhance the molecular compatibility between the PD and L8-BO *** 2EG PD induces higher crystallinity and alleviates phase separation with the SMA compared to the reference PD(PM7)with hydrocarbon ***,the 2EG-based PSCs exhibit a higher PCE(15.8%)than the PM7-based PSCs(PCE=14.4%)in the ortho-xylene based ***,benefitted from the reduced phase separation and increased crystallinity of 2EG PDS,the 2EG-based PSCs show enhanced thermal stability(84%of initial PCE after 120 h heating)compared to that of the PM7-based PSCs(60%of initial PCE after 120 h heating).This study demonstrates the potential of OEG side-chain-incorporated materials in developing efficient,stable,and eco-friendly PSCs.
