Effects of drought stress on root morphology and spatial distribution of soybean and adzuki bean

Due to global climate change,Korea is facing severe droughts that affect the planting and early vegetative periods of upland *** and adzuki bean are important legume crops in Korea,so it is critical to understand their adaptations to water *** study investigated the changes in root morphological properties in soybean and adzuki bean and quantified the findings using fractal *** experiment was performed at the National Institute of Crop Science in Miryang,*** and adzuki beans were planted in test boxes and grown for 30 *** boxes were filled with bed soil with various soil moisture *** images were obtained and scanned every two days,and the root properties were characterized by root length,depth and surface area,number of roots,and fractal parameters(fractal dimension and lacunarity).Root depth,length and surface area and the number of roots increased in both crops as the soil moisture content *** fractal dimension and lacunarity values increased as the soil moisture content *** results indicated that the greater the soil moisture,the more heterogeneous the root *** analysis of the morphological properties and fractal parameters indicated that soybean and adzuki bean had different root structure *** soybean and adzuki bean were sensitive to the amount of soil moisture in the early vegetative *** required a soil moisture content greater than 70%of the field capacity to develop a full root structure,while adzuki bean required 100%of the field *** results would be useful in understanding the responses of soybean and adzuki bean to water stress and managing irrigation during cultivation.

Mitochondria-targeting self-assembled nanoparticles derived from triphenylphosphonium-conjugated cyanostilbene enable site-specific imaging and anticancer drug delivery

Subcellular organelle-specific nanoparticles for simultaneous tumor targeting, imaging, and drug delivery are of enormous interest in cancer therapy. Herein, we report a selective mitochondria-targeting probe 1, which was synthesized by incorporating a triphenyl phosphine with a cyanostilbene and a long alkyl chain moiety. Probe 1 was found to display fluorescence via aggregation-induced emission （AIE）. The low molecular-weight cyanostilbene-based probe 1, with and without an anticancer drug, formed a narrow homogeneous nanorod with ca. 110 nm of length or nanopartides with ca. 20 nm diameter in aqueous media. The self-assembled cyanostilbene nanoparticles （N1） selectively accumulated in the mitochondria of cancer cells and emitted fluorescence. N1 was also able to deliver an anticancer drug, doxorubicin （doX）, to the mitochondria with high efficiency. More importantl~ N1 exhibited highly selective cytotoxicity for cancer cells over normal cells. The great potential applications of this self-assembled nanoparticle to biological systems result from its ability to aggregate in the mitochondria. This aggregation led to a significant increase in the generation of intraceUular reactive oxygen species and to a decrease in the mitochondrial membrane potential in cancer cells. Furthermore, tumor tissue uptake experiments in mice proposed that the self-assembled N1 had the ability to internalize and deliver the anticancer drug into tumor tissues effectively. Moreover, both N1 and Nl-encapsulated doxorubicin （N1-doX） effectively suppressed tumor growth in a xenograft model in vivo. Taken together, our findings indicate that applications of N1 as a mitochondrial targeting probe, drug delivery platform, and chemotherapeutic agent provide a unique strategy for potential image-guided therapy as well as a site-specific delivery system to cancer cells.

Discrepancies between primary physician practice and treatment guidelines for Helicobacterpylori infection in Korea

AIM： To evaluate the attitude of primary care physicians in the diagnosis and treatment of Helicobacter pylori （fl pylori） infection. METHODS： Primary care physicians in the Seoul metropolitan area answered self-administered questionnaire from January to March 2003. RESULTS： One hundred and eight doctors responded to the questionnaire. The most frequent reasons for testing H pylori infection were gastric and duodenal ulcers （93.5% and 88.9%, respectively）. For patients with Hplori positive dyspepsia, 28.7% of doctors always tried to eradicate the worm and 34.4% treated selectively. A large proportion （28.7%） of primary care physicians treated H pylori on a patient＇s request basis. Only 9.3% of primary care physicians always conducted follow-up testing after treating H pylori infection. When H pylori was not cleared by the first treatment, 40.7% of doctors reused the same regimen, 16.7% changed to another triple regimen and 25% to a quadruple regimen. CONCLUSION： It has been well documented that the issuance of guidelines alone has little impact on practice. Communication between primary care physicians and gastroenterologists in the form of continuous medical education is required.

Dioscorea batatas Decne(DBD)has been used to heal various illnesses of the kidney and intestine as an herbal medicine in *** a source of therapeutic agents,many glycoproteins have been isolated from mushrooms and plants,but the functional role of glycoprotein in intestinal epithelial wound healing has not been understood *** the present study,we investigated the wound healing potentials of the 30 kDa glycoprotein(DBD glycoprotein)isolated from DBD in human intestinal epithelial(INT-407)*** found that DBD glycoprotein(100μg·mL^-1)significantly increased the motility of INT-407 cells for 24 h by activating protein kinase C(PKC).DBD glycoprotein stimulated the activation of p38 mitogen-activated protein kinase(MAPK),which is responsible for the phosphorylation of NF-κB inhibitorα(IκBα).DBD glycoprotein increased the level of profilin-1(PFN1),α-actinin and F-actin expression via activation of transcription factor,nuclear factor-kappa B(NF-κB)during its promotion of cell *** mouse colitis was induced by adding dextran sulfate sodium(DSS)to the drinking water at a concentration of 4%(W/V)for 7 *** figured out that administration of DBD glycoprotein(10 and 20 mg·kg-1)lowers the levels of disease activity index and histological inflammation in DSS-treated ICR *** this regard,we suggest that DBD glycoprotein has ability to promote the F-actin-related migration signaling events via activation of PKC and NF-κB in intestinal epithelial cells and prevent inflammatory bowel disease.

Artemisia capillaris Thunberg is a medicinal plant used as a traditional medicine in many cultures. It is an effective remedy for liver problems including hepatitis. Recent pharmacological reports have indicated that Artemisia species can exert various neurological effects. Previously, we reported a memory-enhancing effect of Artemisia species. However, the mechanisms underlying the neuroprotective effect of A. capillaris(AC) are still unknown. In the present study, we investigated the effect of an ethanol extract of AC on ischemic brain injury in a mouse model of transient forebrain ischemia. The mice were treated with AC for seven days, beginning one day before induction of transient forebrain ischemia. Behavioral deficits were investigated using the Y-maze. Nissl and Fluoro-jade B staining were used to indicate the site of injury. To determine the underlying mechanisms for the drug, we measured acetylcholinesterase activity. AC(200 mg·kg-1) treatment reduced transient forebrain ischemia-induced neuronal cell death in the hippocampal CA1 region. The AC-treated group also showed significant amelioration in the spontaneous alternation of the Y-maze test performance, compared to that in the untreated transient forebrain ischemia group. Moreover, AC treatment showed a concentration-dependent inhibitory effect on acetylcholinesterase activity in vitro. Finally, the effect of AC on forebrain ischemia was blocked by mecamylamine, a nonselective nicotinic acetylcholine receptor antagonist. Our results suggested that in a model of forebrain ischemia, AC protected against neuronal death through the activation of nicotinic acetylcholine receptors.

AIM： To examine whether trans-10,cis-12 CLA （t10C12） or cis-9,trans-11 CLA （c9t11） inhibits heregulin （HRG）-β- stimulated cell growth and HRG-β-ErbB3 signaling in HT-29 cells. METHODS： We cultured HT-29 cells in the absence or presence of the CLA isomers and/or the ErbB3 ligand HRG-β. MTT assay, [^3H]thymidine incorporation, Annexin V staining, RT-PCR, Western blotting, immunoprecipitation, and in vitro kinase assay were performed. RESULTS： HRG-β increased cell growth, but did not prevent DNA t10c12-induced growth inhibition. T10C12 inhibited DNA synthesis and induced apoptosis of HT-29 cells, whereas c9t11 had no effect. T10c12 decreased the levels of ErbB1, ErbB2, and ErbB3 proteins and transcripts in a dose-dependent manner, whereas c9t11 had no effect. Immunoprecipitation/ Western blot studies revealed that t10c12 inhibited HRG- β-stimulated phosphorylation of ErbB3, recruitment of the p85 subunit of phosphoinositide 3-kinase （PI3K） to ErbB3, ErbB3-associated PI3K activities, and phosphorylation of Akt. However, c9t11 had no effect on phospho Aid： levels. Neither t10c12 nor c9t11 had any effect on HRG-β-induced phosphorylation of ERK-1/2. CONCLUSION： These results indicate that the inhibition of HT-29 cell growth by t10c12 may be induced via its modulation of ErbB3 signaling leading to inhibition of Akt activation.

Chikungunya fever is a vector-borne viral disease transmitted to humans by chikungunya virus(CHIKV)-infected *** have been many outbreaks of CHIKV infection worldwide,and the virus poses ongoing risks to global *** prevent and control CHIKV infection,it is important to improve the current CHIKV diagnostic approaches to allow for the detection of low CHIKV concentrations and to correctly distinguish CHIKV infections from those due to other mosquito-transmitted viruses,including dengue virus(DENV),Japanese encephalitis virus(JEV),and Zika virus(ZIKV).Here,we produced monoclonal antibodies(mAbs)against the CHIKV envelope 2 protein(CHIKV-E2)and compared their sensitivity and specificity with commercially available mAbs using enzyme-linked immunosorbent assays(ELISA).Two anti-CHIKV-E2 mAbs,19-1 and 21-1,showed higher binding affinities to CHIKV-E2 protein than the commercial mAbs *** particular,the 19-1 mAb had the strongest binding affinity to inactivated ***,the 19-1 mAb had very little cross-reactivity with other mosquito-borne viruses,such as ZIKV,JEV,and *** results suggest that the newly produced anti-CHIKV-E2 mAb,19-1,could he used for CHIKV diagnostic approaches.

Is mannitol combined with furosemide a new treatment for refractory lymphedema?A case report

BACKGROUND Mannitol is a hyperosmolar agent and the combination of mannitol and furosemide is a widely used treatment for intracranial pressure *** the hypertonic properties of mannitol to move water out of intracellular spaces,we hypothesized that mannitol combined with furosemide could relieve focal tissue swelling in refractory *** SUMMARY A 90-year-old female had been diagnosed with intracranial hemorrhage and received a combination of mannitol and furosemide for intracranial pressure *** of the intracranial hemorrhage,she had refractory lymphedema of the left lower extremity since ***,after receiving the mannitol and furosemide,the patient’s lower extremity lymphedema improved *** the mannitol and furosemide were discontinued,the lymphedema worsened in spite of complete decongestive therapy(CDT)and intermittent pneumatic compression treatment(IPC).To identify the presumed effect of mannitol and furosemide on the lymphedema,these agents were resumed,and the lymphedema improved *** The present case raises the possibility that a combination of mannitol and furosemide might be considered another effective therapeutic option for refractory lymphedema when CDT and IPC are ineffective.

BACKGROUND Herpes zoster is a painful infectious disease caused by the varicella zoster *** zoster radiculopathy,which is a type of segmental zoster paresis,can complicate the disease and cause motor *** complication should be considered when a patient with a rash complains of acute-onset motor weakness,and the diagnosis can be verified via electrodiagnostic *** SUMMARY A 64-year-old female with a history of asthma presented to the emergency department with stabbing pain,an itching sensation,and a rash on the right anterior shoulder that had begun 5 d *** examination revealed multiple erythematous grouped vesicles in the right C4-5 and T1 dermatome *** herpes zoster was suspected,the patient immediately received intravenous *** the third hospital day,she complained of motor weakness in the right upper *** resonance imaging of the cervical spine revealed mild intervertebral disc herniation at C4-C5 without evidence of nerve root *** the 12th hospital day,electrodiagnostic study revealed right cervical radiculopathy,mainly in the C5/6 *** months later,monoparesis resolved,and follow-up electrodiagnostic study was *** This case emphasizes that clinicians should consider the possibility of postherpetic paresis,such as herpes zoster radiculopathy,and that electrodiagnostic study is useful for diagnosis and follow-up.
