Plasma metabolites and risk of myocardial infarction:a bidirectional Mendelian randomization study

BACKGROUND Myocardial infarction(MI)is a critical cardiovascular event with multifaceted etiology,involving several genetic and environmental *** is essential to understand the function of plasma metabolites in the development of MI and unravel its complex *** This study employed a bidirectional Mendelian randomization(MR)approach to investigate the causal relationships between plasma metabolites and MI *** used genetic instruments as proxies for plasma metabolites and MI and conducted MR analyses in both directions to assess the impact of metabolites on MI risk and vice *** addition,the large-scale genome-wide association studies datasets was used to identify genetic variants associated with plasma metabolite(1400 metabolites)and MI(20,917 individuals with MI and 440,906 individuals without MI)*** variance weighted was the primary method for estimating causal *** estimates are expressed as beta coefficients or odds ratio(OR)with 95%*** We identified 14 plasma metabolites associated with the occurrence of MI(P<0.05),among which 8 plasma metabolites[propionylglycine levels(OR=0.922,95%CI:0.881–0.965,P<0.001),gamma-glutamylglycine levels(OR=0.903,95%CI:0.861–0.948,P<0.001),hexadecanedioate(C16-DC)levels(OR=0.941,95%CI:0.911–0.973,P<0.001),pentose acid levels(OR=0.923,95%CI:0.877–0.972,P=0.002),X-24546 levels(OR=0.936,95%CI:0.902–0.971,P<0.001),glycine levels(OR=0.936,95%CI:0.909–0.964,P<0.001),glycine to serine ratio(OR=0.930,95%CI:0.888–0.974,P=0.002),and mannose to trans-4-hydroxyproline ratio(OR=0.912,95%CI:0.869–0.958,P<0.001)]were correlated with a decreased risk of MI,whereas the remaining 6 plasma metabolites[1-palmitoyl-2-arachidonoyl-GPE(16:0/20:4)levels(OR=1.051,95%CI:1.018–1.084,P=0.002),behenoyl dihydrosphingomyelin(d18:0/22:0)levels(OR=1.076,95%CI:1.027–1.128,P=0.002),1-stearoyl-2-docosahexaenoyl-GPE(18:0/22:6)levels(OR=1.067,95%CI:1.027–1.109,P=0.001),alpha-ketobutyrate levels(OR=1.108,95%

Scaffold hopping refers to computer-aided screening for active compounds with different structures against the same receptor to enrich privileged scaffolds,which is a topic of high interest in organic and medicinal ***,most approaches cannot efficiently predict the potency level of candidates after scaffold ***,we identified potent PDE5 inhibitors with a novel scaffold via a free energy perturbation(FEP)-guided scaffold-hopping strategy,and FEP shows great advantages to precisely predict the theoretical binding potenciesΔGFEPbetween ligands and their target,which were more consistent with the experimental binding potenciesΔGEXP(the mean absolute deviations|ΔGFEP-ΔGEXP|ying that it will have a variety of future applications in rational molecular design and drug discovery.

Discovery of highly selective and orally available benzimidazole-based phosphodiesterase 10 inhibitors with improved solubility and pharmacokinetic properties for treatment of pulmonary arterial hypertension

Optimization efforts were devoted to discover novel PDE10 A inhibitors in order to improve solubility and pharmacokinetics properties for a long-term therapy against pulmonary arterial hypertension(PAH)starting from the previously synthesized inhibitor *** a result,a potent and highly selective PDE10 A inhibitor,14·3 HC1(half maximal inhibitory concentration,IC50=2.8 nmol/L and>3500-fold selectivity)exhibiting desirable solubility and metabolic stability with a remarkable bioavailability of50%was identified with the aid of efficient methods of binding free energy *** PAH studies showed that the improvement offered by 14·3 HCl[2.5 mg/kg,oral administration(p.o.)]was comparable to tadalafil(5.0 mg/kg,p.o.),verifying the feasibility of PDE10 A inhibitors for the antiPAH *** crystal structure of the PDE10 A-14 complex illustrates their binding pattern,which provided a guideline for rational design of highly selective PDE10 A inhibitors.

The Spatial Equivalence Between Wavelet Decomposition and Phase Space Embedding of EEG

Using both the wavelet decomposition and the phase space embedding, the phase trajectories of electroencephalogram (EEG) is described. It is illustrated based on the present work,that is,the wavelet decomposition of EEG is essentially a projection of EEG chaotic attractor onto the wavelet space opened by wavelet filter vectors, which is in correspondence with the phase space embedding of the same EEG. In other words, wavelet decomposition and phase space embedding are equivalent in methodology. Our experimental results show that in both the wavelet space and the embedded space the structure of phase trajectory of EEG is similar to each other. These results demonstrate that wavelet decomposition is effective on characterizing EEG time series.

Furfural(FF),an important bio-based platform molecule,can be valorized to high-value-added chemicals(e.g.,furfuryl alcohol(FA)or 2-methylfuran(MF))through catalytic hydrogenation[1].The traditional thermocatalytic transformation of FF always requires high-temperature and highpressure conditions[2].Accordingly,electrocatalytic hydrogenation is emerging as a promising approach for the selective reduction of FF to FA or MF under environmentally friendly and benign conditions.

Optimal Cooperative Secondary Control for Islanded DC Microgrids via a Fully Actuated Approach

DC-DC converter-based multi-bus DC microgrids(MGs) in series have received much attention, where the conflict between voltage recovery and current balancing has been a hot topic. The lack of models that accurately portray the electrical characteristics of actual MGs while is controller design-friendly has kept the issue active. To this end, this paper establishes a large-signal model containing the comprehensive dynamical behavior of the DC MGs based on the theory of high-order fully actuated systems, and proposes distributed optimal control based on this. The proposed secondary control method can achieve the two goals of voltage recovery and current sharing for multi-bus DC MGs. Additionally, the simple structure of the proposed approach is similar to one based on droop control, which allows this control technique to be easily implemented in a variety of modern microgrids with different configurations. In contrast to existing studies, the process of controller design in this paper is closely tied to the actual dynamics of the MGs. It is a prominent feature that enables engineers to customize the performance metrics of the system. In addition, the analysis of the stability of the closed-loop DC microgrid system, as well as the optimality and consensus of current sharing are given. Finally, a scaled-down solar and battery-based microgrid prototype with maximum power point tracking controller is developed in the laboratory to experimentally test the efficacy of the proposed control method.

Betulinic acid protects against ovarian impairment by decreasing F-2 toxin-induced oxidative stress and inflammation associated with the downregulation of p38 expression in mice

F-2 toxin is an estrogenic mycotoxin that causes reproductive disorders in *** acid(BA)is a natural pentacyclic lupane-structure triterpenoid that has diverse pharmacological *** this study,the antioxidative and anti-inflammatory effects of BA and its underlying mechanism are explored in F-2 toxin-triggered mouse ovarian *** found that BA alleviated the F-2 toxin-induced ovarian impairment by stimulating follicle growth,reducing inflammatory cell infiltration,repairing damaged mitochondria and endoplasmic ***,BA not only reversed F-2 toxin-induced reduction of follicle stimulating hormone(FSH)and luteinizing hormone(LH)levels in the serum,but also restrained the protein expression of the estrogen receptors a(ERa)and ERβ.Moreover,BA restored the balance of F-2 toxin-induced ovarian redox system ***,we found that 0.25 mg/kg BA played an anti-inflammatory role in the F-2 toxin-induced ovarian impairment by decreasing interleukin-1β(IL-1β).IL-6,and tumor necrosis factor-α(TNF-α)mRNA expression,as well as inhibiting p38 protein *** data demonstrated that BA exerts its protective effect on F-2 toxin-induced ovarian oxidative impairment and inflammation by inhibiting p38 expression,which implies a natural product-based medicine to ameliorate F-2 toxin-caused female reproductive toxicity and provides a detoxifying method for food contaminated by mycotoxin.

Effect of the yaw angle on turbulent flow structures around the submarine model

The objective of this paper is to investigate the turbulent flow structures around the submarine model and evaluate the effect of the yaw angle on the turbulent flow *** large eddy simulation based on the boundary data immersion method is used to *** computational domain consists of 1.2×10^(8)uniformly distributed Cartesian orthogonal grid nodes to capture the basic flow characteristics around the *** pressure coefficient,friction coefficient and wake velocity distribution are in good agreement with the experimental *** different types of vortex structures were mainly captured around the model,including horseshoe vortex,sail tip vortex and crossflow separation *** the increase of the yaw angle,the asymmetry of the horseshoe vortex and the tip vortex gradually increases,and the vortex strength of the vortex leg on the windward of the horseshoe vortex and the vortex strength of the tip vortex also increase *** the crossflow separation vortex,the flow separation zone gradually expands and migrates downstream with the increase of the yaw angle.

In recent years,lithium-sulfur batteries(LSBs)are considered as one of the most promising new generation energies with the advantages of high theoretical speeific capacity of sulfur(1675 mAh·g^(-1)),abundant sulfur resources,and environmental friendliness storage technologies,and they are receiving wide attention from the ***,the problems of the shuttle effect and lithium dendrite growth in LSBs have limited their practical application,so there is a need to find ways to solve these *** is an excellent choice to use novel materials to modify battery *** those novel materials,the metal-organic framework(MOF)has the properties of regular pores and controllable *** applied as a positive electrode and diaphragm,it can restrain the shuttle effect and lithium dendrite growth,especially since it shows excellent performance in diaphragm ***,various design strategies and synthesis methods of MOF-modified separators are reviewed in this paper,and the applications of MOF in LSBs separators in different forms are introduced,including the composite of MOF and carbon-based materials,the compounding of MOF and polymer,self-carbonization to form MOF-derived *** the same time,different characterization techniques are systematically reviewed to obtain the physical and chemical properties of MOF particles and the working mechanism of MOF-modified diaphragm,which provides a basis for further research in this ***,some future research trends and directions are put forward to fully tap the future commercial potential of MOF-modified diaphragm in LSBs.
