Ectopic porcine spermatogenesis in murine subcutis： tissue grafting versus cell-injection methods

Fragments of testis tissue from immature animals grow subcutaneous areas of immunodeficient mice. The same results testes of rodents are injected into the subcutis of nude mice. and develop spermatogenesis when grafted onto are obtained when dissociated cells from immature Those cells reconstitute seminiferous tubules and facilitate spermatogenesis. We compared these two methods, tissue grafting and cell-injection methods, in terms of the efficiency of spermatogenesis in the backs of three strains of immunodeficient mice, using neonatal porcine testicular tissues and cells as donor material. Nude, severe combined immunodeficient （SCID） and NOD/Shi- SCID, IL-2Rγc^null （NOG） mice were used as recipients. At 10 months after surgery, the transplants were examined histologically. Both grafting and cell-injection methods resulted in porcine spermatogenesis on the backs of recipient mice; the percentage of spermatids present in the transplants was 67% and 22%, respectively. Using the grafting method, all three strains of mice supported the same extent of spermatogenesis. As for the cell- injection method, although SCID mice were the best host for supporting reconstitution and spermatogenesis, any difference from the other strains was not signifcant. As NOG mice did not show any better results, the severity of immunodeficiency seemed to be irrelevant for supporting xeno-ectopic spermatogenesis. Our results confirmed that tubular reconstitution is applicable to porcine testicular cells. This method as well as the grafting method would be useful for studying spermatogenesis in different kinds of animals.

Synergistic Antibacterial Performance of a Cu/WO<sub>3</sub>-Added PTFE Particulate Superhydrophobic Composite under Visible-Light Exposure

Addition of TiO2 to a polytetrafluoroethylene (PTFE) particle-dispersed composite contributes to the self-cleaning properties of the water-repellent composite. However, its application is limited to outdoor usage or under ultraviolet (UV) irradiation. In this study, a novel visible-light-sensitive photocatalytic and superhydrophobic material was developed by adding Cu/WO3 to a PTFE particulate composite material to overcome this deficit. A remarkable property of this novel composite material is the synergistic antibacterial performance against Escherichia coli (E. coli), Staphylo-coccus aureus, and methicillin-resistant Staphylococcus aureus compared with the addition of Cu/WO3 without PTFE particles material. During 24-h exposure in visible light at 2000 lx, the number of viable cells of the three strains on the surface of the 8wt% Cu/WO3-added PTFE particulate composite decreased from 2 – 4 × 105 colony-formation units (CFUs) to less than 10, the limit of detection. This bactericidal rate is four times higher than that of 8wt% Cu/WO3 without PTFE particles material, which is attributed to the air trapped in the rough surface of the novel material providing additional oxygen to the photocatalytic reaction. Even for exposure to visible light at 100 lx, the decrease in CFUs of E. coli on the 12wt% Cu/WO3-added PTFE particulate composite reached nearly 2.0 logs. The characterization of the Cu/WO3-added PTFE particulate composite indicated that the composite material containing 80wt% PTFE maintained a superhydrophobic or water-repellent property with a water contact angle >150。, although the Cu/WO3 in the composite material remained hydrophilic under visible light. The Cu/WO3-added PTFE particulate composite displayed photo-catalytic reactions to decompose oleic acid adsorbed on its surface and gaseous acetaldehyde under UV-A and visible-light illumination. Land and Water Use in Rice-upland Crop Rotation Areas in Lower Ili River Basin, Kazakhstan

The lower Ili River Basin is located in semi-arid area, and the annual rainfall is 177mm. Therefore, the irrigation is inevitable for agriculture. Large-scale irrigated agriculture had been developed since 1960＇s in the lower parts of the river and the total irrigated area is about 32 000 hm2. In the project area, the paddy rice-upland crop rotation has been practiced. Due to the domestic water use for hydropower and agriculture as well as water use among riparian countries, the deficit of water for agriculture in the lower part has been concerned. The authors, therefore, conducted the field survey and water balance analysis of the Akdara irrigation project in the lower Ill River Basin in order to assess the land and water uses. Moreover, the impact of the water use on water environment to the basin was analyzed. The following results were obtained as following （1） the groundwater level in the irrigated district varied from 1.5 m to 3.5 m through year. （2） 1970＇s groundwater level was drastically raised from 8 m to 3 m and the groundwater had been recharged in this period. （3） Water use efficiency of agriculture, which is the ratio of total evapotranspiration to the total water withdrawal was as low as 0.23.
