Pathogenesis of Demyelinating Encephalopathy in Dogs with Spontaneous Acute Canine Distemper

So far, the pathogenesis of demyelination caused by canine distemper virus （CDV） in the central nervous system has remained unclear, although a lot of studies have been done extensively. To further investigate the relation of variety cells in brain to demyelination, this study was performed on 15 dogs with spontaneous acute canine distemper and 2 controls. According to anatomical relation, the brain was divided into cerebrum, cerebral stem and cerebellum. The sections with no, mild, moderate, or severe demyelinating lesions were selected respectively and stained by HE and immunohistochemistry. Immuno-localisation of CDV antigen was used to conftrm CDV infection. The brain was examined for co-localisation of the CDV antigen with either an astrocyte-specific marker, glial fibrillary acidic protein （GFAP）, or an oligodendrocyte-specific marker, galactocerebroside （GalC）. Apoptotic cell was detected by TdT-mediated nick end-labeling assay （TUNEL）. The results demonstrated that the local disturbance of blood circulation mainly included congestion, edema, thrombosis, and disseminated intravascular coagulation （DIC）. The CDV neucleocapsid protein positive reaction, metabolic disorder and apoptosis of oligodendrocytes were observed in demyelinating areas. Lots of astrocytes displayed CDV antigen-positive, especially in their process. Some of them became apoptotic cell confirmed by TUNEL staining. Fibrous astrocytes showed more intense GFAP-positive in mild and moderate demyelinating area. Some of nervous cells located in pyramidal cell layers and nucleus nervi were in degeneration, necrosis. Satellitosis, neuronophagia and apoptotic neurons were examined by hematoxylin and eosin （HE） and TUNEL staining. The results suggested that the demyelinating changes in brain tissues infected with CDV mainly related to the metabolic disorder and apoptosis of ogliodendrocytes and astrocytes; also involved with the local disturbance of blood circulation and some neuron lost.

Up to now, the pathogenesis of dimyelination in brain tissues has remained unclear, although a lot of studies about neuron and glia changes have been done extensively. To further investigate the relation of variety cell in brain tissue to demyelination, this experiment was performed on 12 dogs with canine distemper. To detect the lesions carefully the brain tissue was divided into three portions and eleven slices that were including cerebrum (four slices) , cerebral stem (five slices) and cerebellum (two slices) according to anatomical relation. The sections stained by HE were observed under light microscope. The sections (no, mild, moderate and severe demyelinatig tissues) were selected respectively and stained by immunohistochemistry. The results demonstrated that the disturbance of blood circulation mainly included congestion, edema, thrombus formation, and the occurrence of diffusive intravascular coagulation (DIC). Oli-godendrocytes were occurred metabolic disorders and apoptosis in demyelinating areas. With anti - CDV antigen staining astrocytes were often displayed as intense positive reaction. With anti - GFAP staining, the fibrous astrocytes were more intense positive reaction in acute, mild and moderate demyelinating area, apoptotic astrocytes were detected in lesion foci . Some ependymal cells were infected by CDV. A lot of inclusion bodies and apoptotic cells were revealed. A few of nervous cells were degeneration and shrunken with pyknotic nuclei. According to the results, it was considered that the demyelinating changes in brain tissues related mainly to the disturbance of circulation and metabolic disorder and apoptosis of ogliodendrocyles. On the whole, the causation of demyelination of brain tissues was a common active result by many factors, but not only one.

In order to investigate the changes of T, B cells subsets and the pathogenesis of lymphocytic depletion in lymphoid tissues this study was done in dogs naturally infected with canine distemper virus. CD3, CD45RO antigen expression of T cells and IgG, IgM antibody expression of B cells, and distribution of canine distemper virus CDV antigen were detected by immunohistochemistry. The results displayed that CDV antigen often occurred in lymphocytes, foilicular dendritic cells, lymphoid dendritic cells and macrophages of lymphoid tissues. The early myelocytes of CDV antigen positive reaction and with eosinophilic intracytoplasmic inclusion bodies were revealed in bone marrow. Compared with control group, CD3 and CD45RO - positive cells mainly presented in T cell areas. The numbers of CD3 and CD45RO - positive cells were reduced in the region of T cell. Macrophages lied in lymphoid tissue were stained by CD45RO. IgG, IgM -positive cells reduced in the zone of B cell. Some of plasma cell distributed in lymphoid tissues stained by IgG or IgM respectively. The orders of lymphocytic depletion in lymphoid tissues were, sharply IgG, moderate IgM and CD45RO, mild CD3 - positive cells. It is considered that the lymphocytic depletion in lymphoid tissues was mainly caused by B cells loss;the lower lymphocytic multiplication is significant reason to bring about lymphocytic depletion in lymphoid tissues.

Comparison between colorectal low- and high-grade mucinous adenocarcinoma with MUC1 and MUC5AC

AIM:To explore useful prognostic factors for mucinous adenocarcinoma(MAC) in the colon and ***:MAC was divided into low-and high-grade types based on the degree of structural differentiation;low-grade MAC arisen from well to moderately differentiated adenocarcinoma and papillary carcinoma,and high-grade MAC from poorly differentiated adenocarcinoma and signet ring cell ***,the expression of 2 types of MUC1(MUC1/DF and MUC1/CORE),MUC2,2 types of MUC5AC(MUC5AC/CHL2 and HGM),MUC6,CDX2,and CD10 was examined in 16 cases of MAC consisting of 6 low-and 10 high-grade ***:MUC1/DF3 was expressed in 3 of 6 low-grade MAC(50) and 10 of 10 high-grade MAC(100).MUC1/CORE was expressed in 1 of 6 lowgrade MAC(16.7) and 7 of 10 high-grade MAC(70).MUC2 was expressed in all MAC regardless of the ***5AC was expressed in 6 of 6 low-grade MAC(100) and 4 of 10 high-grade MAC(40).HGM was expressed in 5 of 6 low-grade MAC(83.3) and 6 of 10 high-grade MAC(60).Expression of MUC6 and CD10 was undetected in all MAC regardless of the ***2 was expressed in 5 of 6 low-grade MAC(83.3) and 7 of 10 high-grade MAC(70).Taken together,MUC1/DF3 was expressed significantly more frequently in high-grade MAC than in low-grade,and MUC5AC/CHL2 was expressed significantly more frequently in low-grade MAC than in ***:It is proposed that MUC1/DF3 and MUC5AC/CHL2 immunostaining is useful to discriminate high-grade MAC from low-grade MAC.

Colorectal neuroendocrine carcinoma: A case report and review of the literature

BACKGROUND Colorectal neuroendocrine carcinoma (NEC) is a rare tumor that demonstrates aggressive growth pattern with ingrowth into the tract, metastasis to the other organs, and invasion to the surrounding organs;these clinical characteristics result in poor prognosis. Surgical resection appears as an effective approach;however, because it is difficult to accurately diagnose NEC during the early stage and owing to its aggressive growth pattern, development of a reliable standard chemotherapy regimen and management strategies are essential. CASE SUMMARY Here, we report the case of patient with NEC showing an aggressive growth pattern that resulted in the rupture of the tumor to the outside the colon after stenting of the internal colonic stenosis. In addition, the tumor invaded into the duodenum, thereby causing duodenal stenosis that required an additional stent in the duodenum. This aggressive growth pattern is one of the main features of the NEC that is different from adenocarcinoma. To clarify the clinical characteristics, we reviewed 60 recently reported cases, including data on tumor location, size, treatment, and prognosis. CONCLUSION We consider that the information presented here is of great significance for the diagnosis, treatment, and management of symptoms of the patients with NEC.

Association of depressive symptoms with dietary habits among Japanese physicians

The aim of this study was to determine the association of depressive symptoms with dietary habits among physicians working in hospitals in Japan. We mailed an anonymous questionnaire with the Quick Inventory of Depressive Symptomatology and items about dietary habits to 10,000 randomly selected physicians. Logistic regression analysis was used to explore the association of depressive symptoms with dietary habits. The participants comprised 3862 physicians (3025 men and 837 women). Among the respondents, 252 (8.3%) men and 88 (10.5%) women were determined to be in depressive symptoms. For men, “never eat a balanced diet” (Odds ratio;95% confidence interval, 2.54;1.70 - 3.80), and for women “eat quite small meals” (3.81;1.14 - 13.1), “always eat until full” (4.40;1.48 - 13.1), and “never eat a balanced diet” (3.05;1.49 - 6.23) were associated with depressive symptoms. For men, “do not eat too much” (0.57;0.39 - 0.83) was associated with prevention of depression. Physicians should recognize the important role of healthy dietary habits in good mental health. Hospitals should intervene to help physicians address depressive symptoms and dietary habits.

目前治疗晚期胃癌的方法为开腹胃切除及D2淋巴结清扫并在术后行S-1或卡倍他滨加奥沙利铂辅助化疗。然而III期胃癌患者的预后并不理想。为了提高生存率,需要加大化疗药物的剂量。鉴于患者接受二联或三联化疗的顺应性差,新辅助化疗将是一个充满前景的方法。目前在东亚国家,正进行多项III期临床试验用于评估新辅助化疗效果。另一方面,手术治疗方法已倾向于行腹腔镜手术。在日本和韩国已经进行了多项III期临床试验评估腹腔镜辅助远端胃切除(LADG)在早期和晚期患者中的疗效。因此,未来标准治疗肿瘤位于中、下1/3晚期胃癌的候选方案由综合治疗组成,包括新辅助化疗及随后的LADG。只要新辅助化疗是按标准实施,随后的腹腔镜胃切除术的可行性、安全性及长期生存率是有保障的。在这个背景下,我们实施了一项随机II期临床试验比较新辅助化疗后LADG和开腹远端胃切除术(ODG)治疗胃癌的疗效。

Effects of 4-(3-Chloro-Benzyl)-6,7-Dimethoxy-Quinazoline on Kinetics of P120-Catenin and Periplakin in Human Buccal Mucosa Squamous Carcinoma Cell Line

In order to detect molecular markers for the epidermal growth factor inhibitor 4-(3-chloro-benzyl)- 6,7-dimethoxy-quinazoline (tyrphostin), we investigated the kinetics of p120-catenin and periplakin in the human buccal mucosa squamous cancer cell line BICR 10 treated with 3 nM tyrphostin. Growth of BICR 10 cells was inhibited by treatment with tyrphostin. Although changes were not observed in the expression of EGFR and p120-catenin, expression of Akt, Src and periplakin in BICR 10 treated with 3 nM tyrphostin tended to decrease. In addition, phosphorylation of EGFR, Akt and Src was inhibited by treatment with tyrphostin. On immunocytochemical staining, immunoreactions with phosphorylated EGFR, phosphorylated Akt and phosphorylated p120-catenin were weak in BICR 10 treated with tyrphostin. There was a slight immunocy to chemical reaction to periplakin in BICR 10 cells induced by tyrphostin. In conclusion, the decrease in phosphorylation in EGFR and p120-catenin by tyrphostin, following the decrease in Src or Akt phosphorylation, may inhibit expression of several growth factors associated with the proliferation and migration of cancer cells.
