Depletion of CD25^+CD4^+T cells (Tregs) enhances the HBV-specific CD8^+ T cell response primed by DNA immunization

AIM: Persistent hepatitis B virus (HBV) infection is characterized by a weak CD8+ T cell response to HBV. Immunotherapeutic strategies that overcome tolerance and boost these suboptimal responses may facilitate viral clearance in chronically infected individuals. Therefore, we examined whether CD25+CD4+ regulatory T (Treg) cells might be involved in a inhibition of CD8+T cell priming or in the modulation of the magnitude of the 'peak' antiviral CD8+ T cell response primed by DNA immunization. METHODS: B10.D2 mice were immunized once with plasmid pCMV-S. Mice received 500 μg of anti-CD25 mAb injected intraperitoneally 3 d before DNA immunization to deplete CD25+ cells. Induction of HBV-specific CD8+ T cells in peripheral blood mononuclear cells (PBMCs) was measured by S28-39 peptide loaded DimerX staining and their function was analyzed by intracellular IFN-γ staining. RESULTS: DNA immunization induced HBV-specific CD8+ T cells. At the peak T cell response (d 10), 7.1±2.0% of CD8+ T cells were HBV-specific after DNA immunization, whereas 12.7±3.2% of CD8+ T cells were HBV-specific in Treg-depleted mice, suggesting that DNA immunization induced more antigen-specific CD8+ T cells in the absence of CD25+ Treg cells (n = 6, P<0.05). Similarly, fewer HBV specific memory T cells were detected in the presence of these cells (1.3±0.4%) in comparison to Treg-depleted mice (2.6±0.9%) on d 30 after DNA immunization (n - 6, P<0.01). Both IFN-γ production and the avidity of the HBV-specific CD8+ T cell response to antigen were higher in HBV-specific CD8+ T cells induced in the absence of Treg cells. CONCLUSION: CD25+ Treg cells suppress priming and/or expansion of antigen-specific CD8+ T cells during DNA immunization and the peak CD8+ T cell response is enhanced by depleting this cell population. Furthermore, Treg cells appear to be involved in the contraction phase of the CD8+ T cell response and may affect the quality of memory T cell pools. The elimination of Treg cells or the

Quasi-stationary flow structure in turbidity currents

A turbidity current is a particle-laden current driven by density differences due to suspended sediment *** currents can transport large amounts of sediment down slopes over great distances,and play a significant role in fluvial,lake and submarine *** better understand the sediment transport process,the flow system of an experimentally produced turbidity current in an inclined flume was investigated using video *** observed that the current progresses with constant frontal velocity and maintains an unchanged global interface *** addition,the spatio-temporal profiles of the inner mean and turbulence velocity obtained by ultrasound velocity profiler(UVP) showed that similar distributions were maintained,with low *** results indicate that the turbidity current progressed in a quasi-stationary state,which enabled long-distance sediment *** understand the mechanisms behind the quasi-stationary flow,we analyzed the forces acting on the turbidity *** found that under particular densities of suspended particles,the gravitational force is balanced by the viscous forces along the slope *** conclude that this specific force balance induces the quasi-stationary flow structure,enabling the long-distance transport of a substantial amount of sediment downstream with low dissipation.

Spatio-temporal deposition profile of an experimentally produced turbidity current with a continuous suspension supply

A turbidity current is a turbulent, particle-laden gravity current that is driven by density differences resulting from the presence of suspended sediment particles. The current travels downslope, bearing a large amount of sediment over a great distance, and forms fluvial and submarine bedforms. Knowledge of the spatio-temporal deposition profile of turbidity-deposited sediment is important for a better understanding of sediment transport by turbidity currents. In the current study, the depositional process of experimentally produced quasi-steady turbidity currents in an inclined flume was investigated using an electrical-resistance-based depositmeter. It was found that the amount of sediment deposited along the flume bottom increases linearly with time at a specific rate, and decreases exponentially in the downstream direction. Inspired by this observation, the mass conservation law was modified for the suspension layer and a simple theoretical model was proposed involving two parameters, the initial sedimentation rate and the decay constant. The proposed model reproduces the observed data from the flume experiment with high accuracy, and is consistent with the sediment transport distance of a natural example, the Bengal Fan. It was concluded that the two parameters that govern the sedimentation process are the key to estimating the deposition profile of fluvial systems.

A Case of CD34-Negative Superficial Acral Fibromyxoma

Superficial acral fibromyxoma (SAF) is a tumor that occurs on the distal phalanges of the digits. As it does not spontaneously regress and is often associated with pain, the primary treatment is surgical resection. It is often associated with the nail component of the affected digit, and thus cosmesis is an important goal of the operation. We herein describe a case of SAF on the distal phalanges of the fifth digit of the foot beneath the nail, which was successfully resected with the nail component kept intact. Moreover, although SAF is most commonly CD34-positive, the present case was CD34-negative except for endothelial cells within the tumor. While CD34-negative SAF has been previously reported, the current case further indicates that CD34-positivity is not essential for the diagnosis of SAF.

Numerical Simulation for Remediation Planning for 1,4-Dioxane-Contaminated Groundwater at Kuwana Illegal Dumping Site in Japan Based on the Concept of Verified Follow Up

At Kuwana illegal dumping site in Japan, where hazardous waste was illegally dumped, groundwater was severely contaminated by Volatile Organic Compounds (VOCs). Groundwater was already remedied by conducting Pump-and-Treat (P&T) after containment of all the waste by vertical slurry walls from 2002 to 2007. However, 1,4-dioxane was detected in both waste and groundwater outside of slurry walls after it was newly added into Japan environmental standards in late 2009, which suggested that the walls did not contain 1,4-dioxane completely. Our previous study developed a model to predict the 1,4-dioxane distribution in groundwater after the previous remediation at the site. In this study, numerical simulation was applied for remediation planning at the site based on the concept of Verified Follow Up (VF-UP) that had been proposed as a new approach to complete remediation effectively with consideration of future risks. The amount of waste to be removed and pumping plans were discussed by numerical simulation to achieve the remedial objective in which 1,4-dioxane in groundwater outside of walls is remedied within 10 years and 1,4-dioxane spreading throughout the walls is prevented in the case where a portion of waste is remained. Firstly, the amount of waste to be removed considering pumping plans for P&T was determined by scenario analysis. As a result, at least two-third of waste should be removed by combining with P&T. However, if the waste is remained, future risks of 1,4-dioxane spreading through the slurry walls may occur. Our simulation suggested that groundwater within the remaining waste must be pumped up at least 20 m3/d for containment of 1,4-dioxane within the remaining waste. In conclusion, our numerical simulation determined the amount of waste to be removed and the pumping plans for P&T to achieve the remedial objective effectively considering future risks based on the concept of VF-UP.

Prediction of Leachate Generation in a Landfill Using Artificial Neural Networks

One of the problems encountered in the operation of a leachate treatment in a landfill is the quantity of the fluctuating leachate. Therefore, information on the precise prediction about the quantity of leachate produced in a landfill is required. This information can be obtained by using an ANN （artificial neural networks） model. In this study, a prediction on a leachate generation for a period of 15 days was made. The input for the ANN model consists of data such as rainfall, temperature, humidity, duration of solar radiation, and the landfill characteristics, while the output is the leachate landfills production in Minamiashigara, Japan. The ANN algorithm uses a BP （back propagation） with LM （Levenberg-Marquadrt） training type. By using the input-output data pairs, the training of ANN model was conducted in order to obtain the values of the weights that describe the relationship between the input-output data. Furthermore, with the trained ANN model, the prediction of leachate generation for a period of 15 days was made. The study result shows that the prediction accuracy ofleachate generation of ANN-C model, with a correlation coefficient （r） of 0.924, is quite good. Thus, the prediction of leachate generation using artificial neural network model can be recommended for predicting leachate generation in the future. In this study, a prediction on a leachate generation for a period of 15 days was made. The quantity of leachate generation in a landfill can be obtained by using ANN for future periods. By entering data for future periods （t ＋1） in ANN models, the leachate generation for the period （t ＋1） can be predicted.

A Pregnant Patient with Aortic Regurgitation and Symptoms of Acute Heart Failure Caused by Peripartum Cardiomyopathy: A Case Report

Introduction: Knowledge of the risks of pregnancy with heart disease is important because the maternal mortality is much higher than the average. Peripartum cardiomyopathy (PPCM) is rare but it is one of major causes of maternal death. We experienced a pregnant patient with severe aortic regurgitation (AR) presented symptoms of acute heart failure. Her heart failure was not better after an emergency cesarean section and aortic valve replacement (AVR) therefore we think that PPCM caused her heart failure. Case presentation: A 35-year-old woman diagnosed as having severe AR became pregnant. No changes in the AR were apparent during pregnancy. However, the patient developed symptoms of acute heart failure at 37 weeks of gestation, and an emergency cesarean section was performed under general anesthesia. Her hemodynamic status worsened after the cesarean section, and AVR was performed. She was supported with percutaneous cardiopulmonary support (PCPS) after the operation. As recovery seemed to take longer than usual, we decided to implant a ventricular assist device (VAD). Her condition improved after VAD placement, but then she died from a cerebral infarction. In this case, the heart failure was an acute-onset even though AR was stable before and after the pregnancy, and the heart failure did not improve after AVR. Therefore, we concluded that PPCM, rather than AR caused her heart failure. Conclusions: We encountered a case of a pregnant patient with severe AR who presented with symptoms of acute heart failure caused by PPCM. The effect of AR to her heart failure could not be easily denied. This delayed the diagnosis of PPCM, which in turn delayed our decision to use a VAD. Therefore, PPCM should be considered when pregnant patients with heart disease present symptoms of heart failure.
