Release characteristics of arsenic from sediments and its source or sink competition with phosphorus:A case of a great lake with grass-algae alternation

The arsenic(As)release from sediments in great lakes is affected by various *** this study,the characteristics of As release fromsediments was investigated,and the As sources and sinks with the strengths in sediments from different areas(grass-type,algae-type,and grass-algae alternation areas)in great shallow lakes(Taihu Lake,China)were analyzed,and the influence of P competition in the process of As release was also *** results showed that changing trend of the values of equilibrium As concentration in sediments were consistent with the regional changes(0 to 28.12μg/L),and the sediments from algaetype areas had the higher *** sediments from western lake and northwest lake bay were a strong As and a weak P source,and the north lake bay had the opposite trend of these two *** P source competition with As from the sediments occurred in algae-type *** grass-type areas had strong As and P retention capacities,indicating a sink role of sediment with high As and P sorption *** degree of As and P saturation had similar trend in sediments,and the grass-type areas had the higher values,18.3%-21.4%and 15.31%-20.34%,*** analysis results showed that most of As release contribution was from the bottom(30-50 cm)sediments,and the surface(0-10 cm)sediments from algae-type areas contributed more to the overlying water than other region.

Phomaketals A(1)and B(2),two tropolonic meroterpenoids with the unprecedented pentacyclic skeletons,were isolated from the solid-substrate fermentation cultures of a eupC overexpressed mutant strain of the fungus Phoma sp.,together with a biogenetically related secondary metabolite pughiinin B(3),and the known one noreupenifeldin B(4).The structures of 1–3 were elucidated primarily by nuclear magnetic resonance(NMR)*** absolute configurations of 1 and 2 were assigned by electronic circular dichroism calculations and the calculated NMR with DP4+analysis,while that of 3 was established by single-crystal X-ray diffraction analysis using Cu Kα***,phomaketals A(1)and B(2)could be derived from the hypothetical tropolonic sesquiterpene intermediates neosetophomone B(6)and 9-R-neosetophomone B(6),respectively,via different reactions *** 1 showed antiproliferative effect only against the SUPB15 cells,with an 50%inhibitory concentration(IC50)value of 4.85μmol/L,while the co-isolated known meroterpenoid 4 displayed potent effects against three tumor cell lines,SUPB15,EL4,and H9,showing IC50values of 0.36–27.08μmol/L.

Fusagerins A–F(1–6),six new alkaloids including a unique one with the rare a-(N-formyl)carboxamide moiety(1),a hydantoin(imidazolidin-2,4-dione)derivative(2),and four fungerin analogues(3–6),were isolated from the crude extract of the fungus Fusarium sp.,together with the known compound fungerin(7).Compound 2 was isolated as a racemate and further separated into two enantiomers on a chiral HPLC *** structures of 1–6 were determined mainly by NMR experiments,and the absolute configuration of 1 and 2 was assigned by electronic circular dichroism(ECD)*** 7 showed antibacterial activity against Staphylococcus aureus and Streptococcus pneumoniae,and weak cytotoxicity against the T24 cells.

Carrimycin inhibits coronavirus replication by decreasing the efficiency of programmed–1 ribosomal frameshifting through directly binding to the RNA pseudoknot of viral frameshift-stimulatory element

The pandemic of SARS-CoV-2 worldwide with successive emerging variants urgently calls for small-molecule oral drugs with broad-spectrum antiviral ***,we show that carrimycin,a new macrolide antibiotic in the clinic and an antiviral candidate for SARS-CoV-2 in phase III trials,decreases the efficiency of programmed–1 ribosomal frameshifting of coronaviruses and thus impedes viral replication in a broad-spectrum *** binds directly to the coronaviral frameshift-stimulatory element(FSE)RNA pseudoknot,interrupting the viral protein translation switch from ORF1a to ORF1b and thereby reducing the level of the core components of the viral replication and transcription *** carrimycin with known viral replicase inhibitors yielded a synergistic inhibitory effect on *** the FSE mechanism is essential in all coronaviruses,carrimycin could be a new broad-spectrum antiviral drug for human coronaviruses by directly targeting the conserved coronaviral FSE *** finding may open a new direction in antiviral drug discovery for coronavirus variants.

Many microorganisms have mechanisms that protect cells against attack from *** fermentation components of Streptomyces sp.1647 exhibit potent anti-influenza A virus(IAV)*** strain was isolated from soil in southern China in the 1970s,but the chemical nature of its antiviral substance(s)has remained unknown until *** used an integrated multi-omics strategy to identify the antiviral agents from this *** antibiotics and Secondary Metabolite Analysis Shell(antiSMASH)analysis of its genome sequence revealed 38 biosynthetic gene clusters(BGCs)for secondary metabolites,and the target BGCs possibly responsible for the production of antiviral components were narrowed down to three BGCs by bioactivity-guided comparative transcriptomics *** bioinformatics analysis and genetic manipulation of the regulators and a biosynthetic gene,cluster 36 was identified as the BGC responsible for the biosynthesis of the antiviral ***-based molecular networking analysis of mass spectrometric data from different recombinant strains illustrated that the antiviral compounds were a class of structural ***,18 pseudo-tetrapeptides with an internal ureido linkage,omicsynins A1–A6,B1–B6,and C1–C6,were identified and/or isolated from fermentation *** them,11 compounds(omicsynins A1,A2,A6,B1–B3,B5,B6,C1,C2,and C6)are new *** B1–B4 exhibited potent antiviral activity against IAV with the 50%inhibitory concentration(IC_(50))of approximately 1μmol·L^(-1)and a selectivity index(SI)ranging from 100 to *** B1–B4 also showed significant antiviral activity against human coronavirus *** integrating multi-omics data,we discovered a number of novel antiviral pseudo-tetrapeptides produced by Streptomyces sp.1647,indicating that the secondary metabolites of microorganisms are a valuable source of novel antivirals.

Dynamic event-triggered data-driven iterative learning bipartite tracking control for nonlinear MASs with prescribed performance

This article proposes a distributed dynamic event-triggered data-driven iterative learning control(DET-DDILC)scheme under a predefined performance to tackle the bipartite tracking control problem for multiagent systems(MASs). An improved dynamic linearization technique is utilized to convert the nonlinear MASs into an iterative linear data model. First,a peer-to-peer mapping function is introduced to map the constrained distributed system output homeomorphism to an unconstrained one. In addition, a DET mechanism based on a time-iteration-varying function is devised to conserve network communication resources. Based on the unconstrained transformation and the designed DET mechanism, the DET-DDILC algorithm is devised to ensure that the bipartite tracking performance of MASs can be within the preset range. Finally, the effectiveness and feasibility of the designed control scheme are demonstrated via a simulation case by a comparison.

COVID-19 pandemic caused by SARS-CoV-2 infection severely threatens global health and economic *** effective antiviral drug is currently available to treat COVID-19 and any other human coronavirus *** report herein that a macrolide antibiotic,carrimycin,potently inhibited the cytopathic effects(CPE)and reduced the levels of viral protein and RNA in multiple cell types infected by human coronavirus 229 E,OC43,and ***-of-addition and pseudotype virus infection studies indicated that carrimycin inhibited one or multiple post-entry replication events of human coronavirus *** support of this notion,metabolic labelling studies showed that carrimycin significantly inhibited the synthesis of viral *** studies thus strongly suggest that carrimycin is an antiviral agent against a broad-spectrum of human coronaviruses and its therapeutic efficacy to COVID-19 is currently under clinical investigation.

A logistic-Lasso-regression-based seismic fragility analysis method for electrical equipment considering structural and seismic parameter uncertainty

Damage to electrical equipment in an earthquake can lead to power outage of power *** fragility analysis is a common method to assess the seismic reliability of electrical *** further guarantee the efficiency of analysis,multi-source uncertainties including the structure itself and seismic excitation need to be considered.A method for seismic fragility analysis that reflects structural and seismic parameter uncertainty was developed in this *** proposed method used a random sampling method based on Latin hypercube sampling(LHS)to account for the structure parameter uncertainty and the group structure characteristics of electrical ***,logistic Lasso regression(LLR)was used to find the seismic fragility surface based on double ground motion intensity measures(IM).The seismic fragility based on the finite element model of an±1000 kV main transformer(UHVMT)was analyzed using the proposed *** results show that the seismic fragility function obtained by this method can be used to construct the relationship between the uncertainty parameters and the failure *** seismic fragility surface did not only provide the probabilities of seismic damage states under different IMs,but also had better stability than the fragility ***,the sensitivity analysis of the structural parameters revealed that the elastic module of the bushing and the height of the high-voltage bushing may have a greater influence.

Towards few-shot mixed-type dialogue generation

Building an agent capable of conducting both open-domain and task-oriented dialogues, known as mixed-type dialogues, has been an enduring challenge for the AI community. Previous approaches have focused on constructing largescale human-annotated datasets for training models. However, annotating these datasets is expensive and hinders the practical application of these models. This paper identifies a novel challenge, few-shot mixed-type dialogue generation. To address this challenge, we first present a pre-trained dialogue generation framework with modular-based architecture and prompttuning component. Additionally, we collect a mixed-type dialogue dataset that combines persona-chat with conversational recommendation or task-oriented dialogues within a single dialogue session. Specifically, the modular-based architecture allows us to easily incorporate more supervised signals and human-annotated information, thereby facilitating the learning of sessionlevel dialogue logic. We pre-train this dialogue generation framework using multiple external datasets and then fine-tune it on the mixed-type dialogue dataset we collected. Experimental results demonstrate that the three key designs — modular-based architecture, prompt-tuning component, and model pre-training — significantly enhance the performance of this framework compared to state-of-the-art baselines.
