FGF4 protects the liver from immune-mediated injury by activating CaMKKβ-PINK1 signal pathway to inhibit hepatocellular apoptosis

Immune-mediated liver injury (Ili) is a condition where an aberrant immune response due to various triggers causes the destruction of hepatocytes. Fibroblast growth factor 4 (FGF4) was recently identified as a hepatoprotective cytokine;however, its role in Ili remains unclear. In patients with autoimmune hepatitis (type of Ili) and mouse models of concanavalin A (ConA)- or S-100-induced Ili, we observed a biphasic pattern in hepatic FGF4 expression, characterized by an initial increase followed by a return to basal levels. Hepatic FGF4 deficiency activated the mitochondria-associated intrinsic apoptotic pathway, aggravating hepatocellular apoptosis. This led to intrahepatic immune hyper-reactivity, inflammation accentuation, and subsequent liver injury in both Ili models. Conversely, administration of recombinant FGF4 reduced hepatocellular apoptosis and rectified immune imbalance, thereby mitigating liver damage. The beneficial effects of FGF4 were mediated by hepatocellular FGF receptor 4, which activated the Ca^(2+)/calmodulin-dependent protein kinasekinase 2 (CaMKKβ) and its downstream phosphatase and tensin homologue-induced putative kinase 1 (PINK1)-dependent B-cell lymphoma 2-like protein 1-isoform L (Bcl-XL) signalling axis in the mitochondria. Hence, FGF4 serves as an early response factor and plays a protective role against Ili, suggesting a therapeutic potential of FGF4 and its analogue for treating clinical immune disorder-related liver injuries.

强风下百叶窗翅片风致响应的风洞试验研究

为考察建筑工程中常见百叶窗翅片结构的抗风性能,通过1:1实物模型的风洞试验研究典型测点处的风致响应,获得百叶窗表面的风致响应分布规律及其随风速的变化特性。试验结果表明:风速43 m/s,即14级强风下,百叶窗抗风性能良好,翅片结构上各测点处风致响应分布不均匀,百叶窗边缘和中心加强杆附近的响应相对较小;极端强风下中心加强杆附近的响应迅速增加;风致响应随风速增加而增大,风速65 m/s时风振出现最大值,风速76 m/s时叶片将出现塑性变形和裂纹,最大不可恢复塑性变形量达21.6 mm;翅片阵风荷载因子变化范围为1.19~1.56。

Possible Evolution of Paroxysmal Nocturnal Hemoglobinuria into Chronic Myeloid Leukemia: A Rare Transformation

PNH is a rare acquired clonal hematopoietic stem cell disorder characterized by abnormal sensitivity of red blood cells to lysis by complement. It is caused by genetic mutation resulting in deficiency of glycosyl phosphatidylinositol anchor (GPA) for cell membrane proteins including complement regulating proteins CD55 and CD59. PNH tends to be associated with Aplastic Anemia (anemia due to failure of the bone marrow to produce red and white blood cells as well as platelets), Myelodysplastic Syndrome (a group of cancers in which immature blood cells in the bone marrow do not mature or become healthy blood cells) or rarely Acute Myeloid Leukemia (AML) (also known as acute nonlymphocytic leukemia, representing a group of clonal hematopoietic stem cell disorders in which both a block in differentiation and unchecked proliferation result in the accumulation of myeloblasts at the expense of normal hematopoietic precursors). Here we report a case and assume possible evolution of PNH into CML (a myeloproliferative malignant clonal disease characterized by presence of fusion BCR/ABL fusion oncogene).

对渗碳Cr-Ni齿轮钢进行应力比为0和0.3的室温超高周疲劳实验,观测试样中诱发裂纹萌生的夹杂和疲劳断口形貌,以全面评估渗碳Cr-Ni齿轮钢疲劳性能。将疲劳失效模式分为有细颗粒区(Fine granular area,FGA)的内部疲劳失效和有表面光滑区(Surface smooth area,SSA)的表面疲劳失效,并阐明了渗碳Cr-Ni齿轮钢的超高周内部疲劳破坏机制。基于累积损伤和位错能量法并结合细颗粒区形成机理和夹杂的最大评估尺寸,分别构建了两种渗碳Cr-Ni齿轮钢内部疲劳强度的预测模型。利用FGA尺寸与夹杂尺寸的比值和夹杂应力强度因子及应力比之间的关系,修正所提出的两种疲劳强度预测模型并给出了最大夹杂尺寸下的lFGA-S-N曲线。结果表明,基于累积损伤法和位错能量法分别构建的疲劳强度预测模型都可用于预测评估渗碳Cr-Ni齿轮钢在多种应力比下的内部疲劳强度,基于位错能量法的强度预测模型精度较高。

Clinicopathological Analysis of 39 Patients with Corneal Tumors

Purpose:Corneal tumors are rarely seen in clinical *** is a paucity of data on the incidence of various corneal tumors,their clinical and pathological features and surgical management,especially on large patient ***:The medical records of 39 patients with corneal tumors referred to Zhongshan Ophthalmic Center,Sun Yat-sen University,China from January 1,1996 to December 31,2002 were reviewed ***:Of the 39 cases with corneal tumors,31 were males and 8 were *** right eye was affected in 24 cases and the left one in 15 *** mean age at diagnosis was 45.7 years,ranged from 3 to 88 *** cell carcinoma was the most common tumor in the cornea(18 cases,46.15%),followed by pigmented naevus.(12 cases,30.77%),papillary epithelioma(3 cases,7.69%),melanoma(1 case,2.56%),oncosis hyperplasia(1 case,2.56%),inverting papilloma.(1 case,2.56%),primary acquired melanosis(1 case,2.56%),amyloid degeneration(1 case,2.56%),inflammatory pseudotumor(1 case,2.56%).They arose most commonly in the limbal *** tumors were unilaterally *** cell carcinoma of the cornea usually appears as a whitish,rough,irregular lesion or a telangiectatic,gelatinous epibulbar *** in the cornea generally becomes clinically apparent in the first or second decade of life and the lesion may enlarge or more deeply pigmented *** corneal tumors were completely excised microsurgically in 22 *** cases were treated with surgical resection combined with amniotic membrane *** cases with surgical excision plus transfer of conjunctival *** cases with surgical excision plus lamellar *** cases with surgical excision plus *** cases with orbital *** case with *** case with incision biopsy and ***:Squamous cell carcinoma and nevus,the most common corneal tumors,accounted for 76.92% of all *** therapeutic outcomes depended upon early pathol

Features Associated with Recurrence of Idiopathic Orbital Inflammatory Pseudotumor

Purpose: To explore factors which lead to recurrence of idiopathic orbital inflam-matory pseudotumor (IOIP). Methods: Idiopathic orbital inflammatory pseudotumor in 209 cases between Jan 1, 1978 and Dec 31, 1999 in our hospital was evaluated retrospectively. The comparison of clinical and pathological parameters between patients with at least one episode of recurrence and those with no recurrence at all was performed and analyzed using logistic regression method. Results: Follow-up results (with a mean follow-up time of 3.4 years, ranging from 0.5 year to 21.0 years) showed that the recurrence rate of IOIP was 41%. Sex and proptosis were associated with the recurrence of IOIP. Male gender was more likely to relapse than female counterparts, with the male being 52% and female being 25%. The severer the proptosis is, the higher the rate of recurrence. Among the clinical subtypes of IOIP, the rate of recurrence (17%) in cases with dacryoadinitis was the lowest, followed by anterior local orbital mass (44%), posterior orbital mass (54%), myositis (75%) and diffuse subtype (100%). However, the clinical subtypes did not show significant relationship with the recurrence of IOIP. Conclusions: Male gender and severe proptosis are associated with a higher recurrent rate in patients with IOIP.
