背景肝细胞癌(hepatocellular carcinoma, HCC)是临床上常见的实体恶性肿瘤,预后较差.本文拟采用生物信息学技术分析PPP1R105基因在肝细胞癌中的表达相关信号通路及其与患者预后关系.同时,采用免疫组织化学法对PPP1R105生物信息数据进行验证.目的探讨PPP1R10基因(Ki-67或MKI67)在HCC中的表达及其临床意义.方法在TCGA数据中,检索PPP1R105基因在各个肿瘤组织中的相对表达情况;分析PPP1R105基因在肝细胞癌组织和癌旁组织中的相对表达水平,并比较是否存在差异.应用蛋白相互作用数据库STRING构建PPP1R105基因编码蛋白相互作用网络.比较PPP1R105高低表达组患者无疾病进展生存(disease free survival, DFS)和总生存(overall survival, OS)是否存在差异.选取我院手术治疗的原发性肝细胞癌患者81例进行回顾性分析,采用免疫组织化学法检测肝细胞癌组织中PPP1R105蛋白表达情况,并与患者的临床特征进行相关性分析.结果PPP1R105基因mRNA在不同肿瘤中表达水平差异并不显著,其表达不具备肿瘤间特异性.在肝细胞癌中,癌组织PPP1R105m RNA表达水平显著高于癌旁正常肝组织(P<0.05),且表达水平与患者临床分期有关(P <0.05).蛋白相互作用网络区域聚集指数为0.99,蛋白相互作用网络富集明显(P<0.01). PPP1R105及其相关基因信号通路主要富集于细胞周期、细胞衰老、病毒致癌及p53信号通路. KIF18B与PPP1R105正相关表达最为显著(rpearson=0.91, P <0.05);而MCELL基因与PPP1R105负相关表达最为显著(rpearson=-0.59, P <0.05);PPP1R105高表达组OS (HR=1.90, P <0.01)和DFS(HR=1.90, P <0.01)显著低于低表达组.PPP1R105蛋白主要表达于细胞核质及核仁, 81例患者中高表达者30例(37.0%).PPP1R105蛋白高表达患者肿瘤大于5cm及TNM分期Ⅲa期患者比例显著高于MIK67蛋白低表达者(P<0.05).结论肝细胞癌患者PPP1R105高表达,其与患者总生存和无疾病进展生存较短有关,可作为肝细胞癌患者预后不良分子标志物.

It was reported a facile strategy to fabricate magnetite(Fe3O4)single-crystal particles with critical single-domain size by employing graphene oxide(GO)sheets as *** this method,the small-sized Fe2 O3 nanoparticles were first synthesized,and then low-temperature annealing under H2 would convert them into large-sized Fe3 O4 single-crystal *** synthetic particles with an average size of 100 nm exhibit high saturation magnetization(Ms)of 0.085 A·m^2·g^-1,which is very close to theoretical value,being among the highest values in ever reported for Fe3O4 made from chemical *** this basis,the small-sized Fe3 O4 particles(average size of 30 nm)were also fabricated by coating with Na2 CO3 shell.

本试验主要研究了夏季发酵床养殖模式奶牛舍与拴系式养殖模式奶牛舍内环境指标的差异。测定了牛舍的温度、噪声、相对湿度、氨气浓度和细菌密度等环境指标,并分析了其相关性。结果表明,发酵床牛舍的细菌密度为0.10×10~4个/m^3,极显著低于拴系式牛舍的2.33×10~4个/m^3(P<0.01);氨气浓度为3.92mg/m^3,也极显著低于拴系式牛舍的9.68mg/m^3(P<0.01);噪声强度为57.6d B,极显著低于拴系式牛舍的65.3d B(P<0.01);但温湿度、温湿度指数以及其他指标在两种牛舍间没有显著的差异。另外,拴系式牛舍内的温度和噪声存在显著相关关系(Sig.(2-tailed)<0.05)。综上,发酵床牛舍的环境细菌浓度、噪声强度与氨气浓度较低,能够在一定程度上改善奶牛的舒适度。

The erosion behavior of a QA19-4 alloy as a ship propeller material was *** effects of the solution,the impact angles and the sand content were *** test results demonstrate that the mass loss of the alloy in 3.5 wt%NaCl solution is 1.35 times that in tap water,due to the corrosive effect of Cl^(-).The mass loss of the alloy increases as the impact angle increases up to~30°and consequently decreases as the impact angle ***,this feature is typical for the ductile metal *** the impact angle of 0°,this feature is associated with the predominant erosion mechanisms,such as shallow plowing and surface fatigue;at 30°,this feature is micro-cutting,deep plowing and surface fatigue;and at 45°,it becomes indentation accompanied by extruded *** mass loss and surface roughness of the alloy increase as the sand content increases under the testing conditions.

Background:Simiaowan(SMW),a well-known traditional Chinese medicine,has been employed to treat hyperuricemia(HUA)and gout for ***,the bioactive components and underlying mechanisms have not been *** objective of this study was to identify the active components and potential mechanisms of SMW by integrating pharmacological experimentation,serum pharmacochemistry,network pharmacology and molecular ***:HUA rats modelling by high-fat/high-sugar diet and potassium oxonate/adenine oral administration were used to evaluate the pharmacodynamic effects of ***-Q-Exactive-MS/MS was employed to detect the bioactive components present in SMW-containing *** pharmacology and molecular docking were utilized to elucidate the potential targets and underlying ***:SMW effectively ameliorated HUA rats via the inhibition of uric acid(UA)production,promotion of UA excretion,improvement of lipid and glucose metabolic abnormalities,antioxidant,anti-inflammatory and anti-insulin resistance effects.A total of 73 compounds detected in SMW-containing serum were identified as potential active components,with alkaloids,flavonoids,organic acids,and terpenoids emerging as the primary active *** 203 corresponding targets were obtained as SMW anti-HUA/gout targets,which mainly participated in apoptosis,insulin resistance,TNF,PI3K-Akt,HIF-1,NF-κB,MAPK,IL-17 and TLR signaling *** docking indicated that active compounds(***,phellodendrine,quercetin,formononetin,ferulic acid)had superior binding abilities to the key targets(*** carrier family 22 member 12(URAT1),solute carrier family 22 member 6(OAT1),ATP-binding cassette sub-family G member 2(ABCG2),solute carrier family 2,facilitated glucose transporter member 9(GLUT9),xanthine dehydrogenase/oxidase(XDH),transcription factor p65(RELA),toll-like receptor 4(TLR4),prostaglandin G/H synthase 2(PTGS2),caspase-3(CASP3),insulin(INS)).Conclusion:S

We report an efficient kinetic resolution of racemic 2-（4-chlorophenyl）-4-hydroxytetrahydro-pyran （CLP-4-HTHP） via Pseudomonas cepacia lipase （PSL）-catalyzed transesterification, where PSL is immobilized on a core-shell MnFe204@SiO2-（CH2）3-NH2 carrier and used as a magnetically separable catalyst. The as-synthesized PSL/MnFe204@SiO2-（CH2）3-NH2 catalyst exhibits enhanced catalytic activity for resolving racemic CLP-4-HTHP to the corresponding optically pure （2R,4S）-CLP-4-HTHP compared to the free PSL. The ees for the former is 2.3 times larger than that for the latter under optimized conditions （99.4% and 44.1%, respectively）, although the eep for them are same （99.2%）. Meanwhile, the PSL/MnFe204@SiO2-（CH2）3-NH2 catalyst possesses a high saturate magnetization of 59.7 emu/g and could be easily recovered by magnetic separation and reused. The catalytic activity in six recycling tests did not significantly decrease, suggesting its great potential for industrial applications.

An ultra-low emissions combustor,namely low emission stirred swirl(LESS)combustor was studied,based on a scheme of internally-staged/lean premixed and prevaporized(LPP)*** LESS combustor consists of central pilot stage and outer surrounded coaxially main stage,between which there exists aphysical isolation,namely the step *** existence of step height delayed the pilot and main jets *** and numerical studies were carried out to investigate the influence of the step height on the combustion performance.A single dome rectangular combustor was utilized to conduct the lean lightoff and blowout experiments,and pollutant emission *** experimental results showed that with the increase of step height by 38%,the lean lightoff and blowout fuel air ratio decreased by 57.4% and 37.5%,the NOx emission increased by 35.1%,and the combustion efficiency increased by 1.78%;while the CO,unburned hydrocarbons(UHC)emissions ***,the total pressure loss was kept nearly ***-reacting and reacting flow fields were numerically investigated to analyze the coupled characteristics of pilot and main jets with different step heights.A comparison of flow characteristics,spray structure,and combustion component as well as temperature field with different step heights was *** numerical results showed that the increase of the step height shifted the peak velocity *** enlargement of the primary recirculation zone(PRZ)resulted in the increase of the combustion efficiency and NOx emission,while the CO,UHC emissions decreeased.

Seven conodont biozones are recognized in the carbonate-dominated shelf-marine Middle Ordovician developed in the intracratonic sedimentary basins(Canning,Amadeus and Georgina)of central and north-western Australia,in the Lachlan and New England orogens of New South Wales,and in the Takaka Terrane of New Zealand.A separate scheme identifying seven conodont biozones spanning the Middle Ordovician has also been developed for siliciclastic sequences deposited in slopebasinal environments in the Lachlan Orogen in New South Wales and *** biozonal classification consisting of two parallel biostratigraphic schemes for the shelf-marine and deep-marine successions respectively has significantly increased precision in regional and global biostratigraphic correlation and laid a solid foundation for the Middle Ordovician chronostratigraphy of Australia and New *** of short-ranging pandemic species as the eponymous species of the biozones also supports direct correlation with the classical conodont successions established in Baltoscandia and the North American Midcontinent,and with those of the major Chinese terranes(South and North China and Tarim).The Lachlan Orogen appears to be globally unique in enabling correlation of contemporaneous conodont faunas over a considerable spectrum of water depths and biofacies ranging from carbonate shelves,slopes to deep-water basins.
