Therapeutic DC vaccination with IL-2 as a consolidation therapy for ovarian cancer patients： a phase Ⅰ/Ⅱ trial

While ovarian cancer （OvCa） responds well to surgery and conventional chemotherapy, a high recurrence rate of advanced OvCa is observed. In this phase Ⅰ/Ⅱ study, 10 OvCa patients with minimal residual disease were treated with autologous dendritic cells （DCs） and IL-2 to evaluate the safety and feasibility of this therapeutic strategy and to characterize the antigen-specific immune alterations induced through this treatment. Approximately 4 months after initial debulking and chemotherapy, patients received two subcutaneous doses of autologous monocyte-derived DCs pulsed with autologous tumor lysate and keyhole limpet hemocyanin （KLH） at 4-week intervals. After each DC inoculation, low-dose （200 mlU） IL-2 was introduced for 14 consecutive days as an immune adjuvant. The vaccination was well tolerated. In three out of 10 patients, the inclusion status after the initial therapy showed the maintenance of complete remission （CR） after DC vaccination for 83, 80.9 and 38.2 months without disease relapse. One patient with stable disease （SD） experienced the complete disappearance of tumor after DC vaccination, and this status was maintained for 50.8 months until tumor recurrence. In two patients with partial response （PR） was not responding to DC vaccination and their disease recurred. In the three patients with disease free long-term survival, significant immune alterations were observed, including increased natural killer （NK） activity, IFN-γ-secreting T cells, immune-stimulatory cytokine secretion and reduced immune-suppressive factor secretion after DC vaccination. Thus, in patients with NED status and increased overall survival, DC vaccination induced tumor-related immunity, potentially associated with long-term clinical responses against OvCa.

Ferroptosis inhibitors reduce celastrol toxicity and preserve its insulin sensitizing effects in insulin resistant HepG2 cells

Magnetic skyrmions emerge when the energy of ferromagnetic exchange interaction promoting parallel alignment of spins enters in competition with energies favoring non-collinear alignment of spins such as Dzy aloshinskii-Moriya interaction(DMI),long-rang dipole-dipole interaction(DDI),or higher-order exchange *** perform an unbiased Monte Carlo simulation to study the DMI-based skyrmion nucleation and stabilization on the surface of magnetic nanotubular monolayer controlled by tuning constants of DDI(g) and next-nearest-neighbor antiferromagnetic exchange interaction(j') with appropriate *** g and j',the loosely distributed skyrmions initially nucleate on the surface of nanotube approaching to the magnetic field(h) direction with increasing h in the intermediate ***,the skyrmion size,shape,density,distribution and crystal structure,as well as its driven field range,are tailored by g and j'.This work demonstrates the skyrmion nucleation mechanisms in three-dimensional magnetic nanostructures with curvature effect and multiple interactions,serving as a benchmark for a guide to experimentalists for preparation of samples in magnetic skyrmion states.

An integrated MINLP model for multi-party coordination in downstream oil supply chain

Cooperation among enterprises can bring overall and individual performance improvement,and a smooth coordination method is ***,due to the lack of customized coordination methods,cooperation in the downstream oil supply chain cannot be carried out *** paper intends to propose a multi-party coordination method to promote cooperation between oil shippers and pipeline operator by optimizing oil transportation,oil substitution and pipeline pricing *** integrated game-theoretic modeling and analysis approach is developed to characterize the operation behaviors of all stakeholders in the downstream oil supply *** proposed mixed integer nonlinear programming model constrains supply and demand capacity,transportation routes,oil substitution rules and pipeline freight *** transformation and price discretization are introduced for model linear *** experiments are carried out in the oil distribution system in South *** results show that compared to the business-as-usual scheme,the new scheme saves transportation cost by 3.48%,increases pipeline turnover by 5.7%,and reduces energy consumption and emissions by 7.66%and 6.77%.It is proved that the proposed method improves the revenue of the whole system,achieves fair revenue distribution,and also improves the energy and environmental benefits of the oil supply chain.

Emission wavelength selection via anomalous polarized fluorescence in ZnO-C nanowires

Defect fluorescence from high aspect ratio semiconductor nanowires typically displays a weak polarization parallel to the nanowire’s long axis due to dielectric mismatch in high aspect ratio ***,anomalous 2.2 eV defect fluorescence distinctly polarized perpendicular to the nanowire is observed and measured from carbon-incorporated zinc oxide *** observations are significant because polarized defect emissions with consistent polarization on a mesoscopic scale are *** a systematic study and comparison of experimental results with density functional theory calculations,an oriented defect complex comprising carbon substituting on an oxygen site and an oxygen vacancy(CO-VO)is deduced to be responsible for the anomalous yellow fluorescence,demonstrating a method for relating atomic-scale defect geometry to mesoscopic *** anomalous emission can appear in both green-and red-fluorescing nanowires grown with different carbon concentrations,verifying the independence and uniqueness of the 2.2 eV *** allows for polarization-dependent emission wavelength selection from a single nanowire.

Increasing concentrations of dietary threonine,tryptophan,and glycine improve growth performance and intestinal health with decreasing stress responses in broiler chickens raised under multiple stress conditions

The current study aimed to compare the effects of increasing concentrations of dietary threonine(Thr),tryptophan(Trp),and glycine(Gly)on growth performance,stress biomarkers,and intestinal function in broiler chickens under multiple stress *** hundred sixty broiler chickens at 21 d old were randomly allotted to 5 treatments with 8 *** in a positive control(PC)treatment were raised under low stock density(16.9 birds/m2 per cage)with recommended environmental conditions,whereas birds in 4 treatments were subjected to multiple stress conditions:a cyclic heat stress of 30±0.3°C for 10 h and 23±0.2°C for 14 h per day with high stock density(25.3 birds/m2 per cage).A basal diet was assigned to both PC and negative control(NC)*** additional diets were individually formulated to contain double concentrations of digestible Thr,Trp,or Gly+Ser compared with their concentrations in the basal *** experiment lasted for 14 *** showed that NC treatment had less growth performance(Pmance and intestinal health in broiler chickens with decreasing stress response under multiple stress conditions.

Lithium-sulfur(Li-S)batteries are one of the most promising modern-day energy supply systems because of their high theoretical energy density and low ***,the development of high-energy density Li-S batteries with high loading of flammable sulfur faces the challenges of electrochemical performance degradation owing to the shuttle effect and safety issues related to fire or explosion *** this work,we report a three-dimensional(3D)conductive nitrogen-doped carbon foam supported electrostatic self-assembled MXene-ammonium polyphosphate(NCF-MXene-APP)layer as a heat-resistant,thermally-insulated,flame-retardant,and freestanding host for Li-S batteries with a facile and costeffective synthesis ***,through the use of NCF-MXene-APP hosts that strongly anchor polysulfides,the Li-S batteries demonstrate outstanding electrochemical properties,including a high initial discharge capacity of 1191.6 mA h g^(-1),excellent rate capacity of 755.0 mA h g^(-1)at 1 C,and long-term cycling stability with an extremely low-capacity decay rate of 0.12%per cycle at 2 *** importantly,these batteries can continue to operate reliably under high temperature or flame attack ***,this study provides valuable insights into the design of safe high-performance Li-S batteries.

Immune checkpoint blockade(ICB)therapy targeting PD-L1 via monoclonal antibody(m Ab)has shown extensive clinical benefits in the diverse types of advanced ***,most patients are completely refractory to ICB therapy owing to the PD-L1 recycling ***,we propose photo-induced crosslinked and anti-PD-L1 peptide incorporated liposomes(immune checkpoint blockade liposomes;ICB-LPs)to promote PD-L1 multivalent binding for inducing lysosomal degradation of PD-L1 in tumor *** ICB-LPs are prepared by formulation of DC_(8,9)PC with photo-polymerized diacetylenic moiety,1,2-dipalmitoylphosphatidylcholine(DPPC)and anti-PD-L1peptide(D-form NYSKPTDRQYHF)-conjugated DSPE-PEG_(2k)(anti-PD-L1-DSPE-PEG_(2k))in a molar ratio of 45:45:10,followed by cross-linking of liposomal bilayer upon UV *** 10 mol% antiPD-L1-DSPE-PEG_(2k)incorporated ICB-LPs have a nano-sized lipid bilayer structure with an average diameter of 137.7±1.04 nm,showing a high stability in serum ***,the ICB-LPs efficiently promote the multivalent binding with PD-L1 on the tumor cell membrane,which are endocytosed with aim to deliver PD-L1 to the lysosomes,wherein the durable PD-L1 degradation is observed for72 h,in contrast to anti PD-L1 m Abs showing the rapid PD-L1 recycling within 9 *** in vitro coculture experiments with CD8^(+)T cells show that ICB-LPs effectively enhance the T cell-mediated antitumor immune responses against tumor cells by blocking the PD-L1/PD-1 *** ICB-LPs are intravenously injected into colon tumor-bearing mice,they efficiently accumulate within the targeted tumor tissues via both passive and active tumor targeting,inducing a potent T cell-mediated antitumor immune response by effective and durable PD-L1 ***,this study demonstrates the superior antitumor efficacy of crosslinked and anti-PD-L1 peptide incorporated liposome formulation that promotes PD-L1 multivalent binding for trafficking of PD-L1 toward

Background To evaluate the association between retinal artery/arteriole occlusion(RAO)and unruptured intracranial aneurysm(UIA).Methods Incident UIA patients from a nationwide cohort(n=253240)were categorised into three groups based on subsequent treatment:observation(n=208993),microsurgical clipping(n=14168)and endovascular treatment(EVT)groups(n=30079).The incidence and the incident time of RAO were *** of RAO and associated risk factors were ***,a hospital cohort comprising 2569 consecutive UIA patients treated at a tertiary hospital was analysed with detailed clinical information of *** In the nationwide cohort analysis,the incidence of RAO was significantly higher in EVT group than in observation and clipping groups,especially within 60 days(early RAO(within 60 days):HR=4.00,95%CI:2.44 to 6.56);delayed RAO(after 60 days:HR=1.74,95%CI:1.13 to 2.68).Multivariable analysis showed that the presence of chronic kidney disease(p=0.009)and use of a balloon microcatheter during the procedure(p=0.013)were associated with a higher risk of *** hospital cohort analysis,11(0.8%)cases of RAO occurred after EVT,whereas none occurred after microsurgical clipping(p<0.001).Patients with RAO were younger and received balloon microcatheters more frequently than their *** cases of RAO(90.9%)occurred in paraclinoid aneurysms,where EVT was preferred over microsurgical *** Performing EVT for UIA may increase the risk of subsequent *** should be taken when treating paraclinoid aneurysms with balloon microcatheters.
