Cyr61 Alleviates Cholangitis by Inhibiting Cytotoxic Effects of CD8^(+) T Cells on Biliary Epithelial Cells

BACKGROUND Liver failure has high mortality and poor prognosis,and establishing new reliable markers for predicting its prognosis is ***-associated invariant T(MAIT)cells are a novel population of innate-like lymphocytes involved in inflammatory liver disease,and their potential role in liver failure remains *** To investigate alteration of circulating MAIT cells and assess its prognostic value in patients with hepatitis B virus(HBV)-related liver *** We recruited 55 patients with HBV-related liver failure,48 patients with chronic hepatitis B and 40 healthy controls(HCs)from Nantong Third People’s Hospital Affiliated to Nantong *** blood mononuclear cells were isolated,and the percentage and number of circulating MAIT cells were detected by flow *** levels of interleukin(IL)-7,IL-12p70,IL-18 and interferon-αwere measured by Luminex *** Circulating MAIT cells were significantly decreased in HBV-related liver failure patients(percentage:2.00±1.22 vs 5.19±1.27%,P<0.0001;number:5.47±4.93 vs 84.43±19.59,P<0.0001)compared with *** importantly,there was a significant reduction of MAIT cells in patients with middle/late-stage compared with early-stage liver *** MAIT cells partially recovered after disease improvement,both in percentage(4.01±1.21 vs 2.04±0.95%,P<0.0001)and in cell count(17.24±8.56 vs 7.41±4.99,P<0.0001).The proportion(2.29±1.01 vs 1.58±1.38%,P<0.05)and number(7.30±5.70 vs 2.94±1.47,P<0.001)of circulating MAIT cells were significantly higher in the survival group than in the dead/liver transplantation group,and the Kaplan–Meier curve showed that lower expression of circulating MAIT cells(both percentage and cell count)predicted poor overall survival(P<0.01).Also,the levels of IL-12(20.26±5.42 pg/mL vs 17.76±2.79 pg/mL,P=0.01)and IL-18(1470.05±1525.38 pg/mL vs 362.99±109.64 pg/mL,P<0.0001)were dramatically increased in HBV-related liver failure patients compared wit

Numerical simulations for radon migration and exhalation behavior during measuring radon exhalation rate with closed-loop method

Accurate measurements of the radon exhalation rate help identify and evaluate radon risk regions in the *** these measurement methods,the closed-loop method is frequently ***,traditional experiments are insufficient or cannot analyze the radon migration and exhalation patterns at the gas–solid interface in the accumulation *** CFD-based technique was applied to predict the radon concentration distribution in a limited space,allowing radon accumulation and exhalation inside the chamber intuitively and *** this study,three radon exhalation rates were defined,and two structural ventilation tubes were designed for the *** consistency of the simulated results with the variation in the radon exhalation rate in a previous experiment or analytical solution was *** effects of the vent tube structure and flow rate on the radon uniformity in the chamber;permeability,insertion depth,and flow rate on the radon exhalation rate and the effective diffusion coefficient on back-diffusion were *** on the results,increasing the inser-tion depth from 1 to 5 cm decreased the effective decay constant by 19.55%,whereas the curve-fitted radon exhalation rate decreased(lower than the initial value)as the deviation from the initial value increased by approximately 7%.Increasing the effective diffusion coefficient from 2.77×10^(-7) to 7.77×10^(-6) m^(2) s^(-1) made the deviation expand from 2.14 to 15.96%.The conclusion is that an increased insertion depth helps reduce leakage in the chamber,subject to notable back-diffusion,and that the closed-loop method is reasonably used for porous media with a low effective diffusion coefficient in view of the back-diffusion *** CFD-based simulation is expected to provide guidance for the optimization of the radon exhalation rate measurement method and,thus,the accurate measurement of the radon exhalation rate.

The urban water system theory is an extension of the basin water system science on an urban scale, providing a new systematic solution for the unbalanced human-water relationship and severe water challenges, such as waterlogging, black and odorous water, and ecological degradation caused by urbanization. Most existing studies on urban water systems have focused on individual water cycle processes linked with water supply and sewage treatment plants, but mutual feedback between the water cycle and its associated material circulation and water ecology, as well as human processes, still needs further exploration. In this paper, the concept, theory, and technical methodology of the urban water system were developed based on the water cycle and basin water system science. The Urban Water System 5.0(UWS 5.0) model was developed by integrating the Time Variant Gain rainfall-runoff Model with Urban water system(TVGM_Urban) in different underlying surface conditions for analyzing the natural-social water cycle processes and their associated water environmental and ecological processes and the influence of multiscale sponge measures. Herein, five major simulation functions were realized: rainfall-runoff-nonpoint source pollutant load,water and pollutant transportations through the drainage network system, terminal regulation and purification, socioeconomic water cycle, and water system assessment and regulation. The location for the case study used in this paper was Wuhan City. The findings showed that the entire urban water system should consider the built-up area and its associated rivers and lakes as the research object and explore the integrations among the urban natural-social water cycle and river regulations inside and outside of the city as well as the effects of socioeconomic development and sponge measures on the water quantity-quality-ecology processes. Mechanical properties and microstructure of Mg-treated and Catreated industrial H13 steel after quenching and tempering

The effect of magnesium treatment and calcium treatment on the microstructure and mechanical properties of industrial H13 steel after quenching and tempering was *** impact toughness and tensile tests were mainly carried out,and the microstructure was observed by scanning electron microscopy,electron backscattered diffraction,and X-ray *** results show that magnesium treatment is still feasible in industrial *** is mainly manifested in the refinement of lath martensite and *** with calcium treatment,the prior austenite grains and carbides size of industrial H13 steel treated with magnesium decreased by 3.17μm after *** quenching and tempering,the carbides(especially V-rich carbides)in Mg treatment obviously spheroidized and distributed uniformly and increased in quantity *** lath martensite size is reduced from 2.45 to 2.31μ*** suggests that magnesium treatment was able to yield smaller grains and more evenly distributed ***,the impact toughness,yield strength,and ultimate tensile strength of industrial H13 steel with magnesium treatment increased by 28%,65.5 MPa and 123.7 MPa,*** increment of strength mainly comes from dislocation strengthening,grain refinement strengthening,and precipitation strengthening,among which precipitation strengthening accounts for the largest proportion.

Recently, observational hints for supermassive black holes have been accumulating, prompting the question: Can primordial black holes（PBHs） be supermassive, particularly with masses M?10～9M⊙? A supercritical bubble,containing an inflating baby universe, that nucleated during inflation can evolve into a PBH in our observable universe. We find that when the inflaton slowly transitions past a neighboring vacuum, the nucleation rate of supercritical bubbles inevitably peaks, leading to a mass distribution of multiverse PBHs with a peak mass up to M?1011M⊙. Thus, our mechanism naturally provides a primordial origin for supermassive black holes.

Clinical efficacy of the topical application of tranexamic acid in tendon release in the hand:A randomized controlled trial

Purpose:To study the clinical effectiveness of the topical application of tranexamic acid in hand tendon ***:This was a randomized controlled trial conducted after receiving approval from the local ethics committee according to guidelines from the Helsinki *** patients who underwent hand tendon release operation in our hospital from January 2021 to December 2022 were included and randomly divided into 2 *** in the tranexamic acid group(40 cases)received intraoperative topical application of 2 g of tranexamic acid after tendon release,while patients in the conventional group(40 cases)did not receive topical application of tranexamic acid during *** operation time,perioperative hemoglobin changes,total blood loss,incidence of early postoperative complications,and total active movement(TAM)before surgery and 6 months after surgery were compared between the 2 *** continuous variable which follows normal distribution expressed by mean±SD and used t-test to compare between ***,categorical variables were used by Chi-square test,and a p<0.05 indicated that the differences were statistically ***:Both groups were followed up for 7-18 months,with a mean of 10.3 *** decrease in hemoglobin was significantly less in the tranexamic acid group than in the conventional group(t=7.611,p<0.001).The total blood loss in the tranexamic acid group(74.33±20.50)mL was less than that in the conventional group(83.05±17.73)mL,and the difference was statistically significant(p<0.05).Both groups showed improvement in thumb/finger flexion and extension range of motion after surgery,and the TAM improved compared with those before surgery,and the difference was statistically significant(p<0.001).The TAM improved more significantly in the tranexamic acid group(87.68°±10.44°)than in the conventional group(80.47°±10.93°)at 6 months after surgery,with a statistically significant difference(t=3.013,p<0.0
