Ferroptosis inhibitors reduce celastrol toxicity and preserve its insulin sensitizing effects in insulin resistant HepG2 cells

Objective:Research has shown that celastrol can effectively treat a variety of diseases,yet when passing a certain dosage threshold,celastrol becomes toxic,causing complications such as liver and kidney damage and erythrocytopenia,among *** this dichotomy in mind,it is extremely important to find ways to preserve celastrol’s efficacy while reducing or preventing its ***:In this study,insulin-resistant Hep G2(IR-Hep G2)cells were prepared using palmitic acid and used for in vitro ***-Hep G2 cells were treated with celastrol alone or in combination with Nacetylcysteine(NAC)or ferrostatin-1(Fer-1)for 12,24 or 48 h,at a range of *** counting kit-8assay,Western blotting,quantitative reverse transcription-polymerase chain reaction,glucose consumption assessment,and flow cytometry were performed to measure celastrol’s cytotoxicity and whether the cell death was linked to ***:Celastrol treatment increased lipid oxidation and decreased expression of anti-ferroptosis proteins in IR-Hep G2 *** downregulated glutathione peroxidase 4(GPX4)m *** docking models predicted that solute carrier family 7 member 11(SLC7A11)and GPX4 were covalently bound by ***,we found for the first time that the application of ferroptosis inhibitors(especially NAC)was able to reduce celastrol’s toxicity while preserving its ability to improve insulin sensitivity in IR-Hep G2 ***:One potential mechanism of celastrol’s cytotoxicity is the induction of ferroptosis,which can be alleviated by treatment with ferroptosis *** findings provide a new strategy to block celastrol’s toxicity while preserving its therapeutic effects.

Molecular dynamics simulations of helium transport through inorganic mineral nanopores

Helium transport through nanoscale inorganic mineral pores and pore throats is essential for its overall migration. To elucidate helium's transport dynamics within nanopores, we employed equilibrium and non-equilibrium molecular dynamics simulations to investigate helium's static self-diffusion and pressure-driven flow in quartz slit-shaped nanopores. We also introduced water and various gases, including hydrogen, methane, ethane, nitrogen, and carbon dioxide, into the nanopores to assess their influence on helium transport. Our findings indicate minimal helium adsorption on quartz pore surfaces. Under conditions where the pore size is less than 5 nm and the pressure under 10 MPa, environmental factors markedly influence helium diffusion. Large pore sizes, high temperatures, and low gas pressures enhance helium desorption and facilitate faster diffusion. We observed a positive correlation between helium flow velocity and factors such as pore size, pressure gradient, and surface smoothness of the pores. Notably, the presence of pore water and carrier gases in quartz nanopores, which diffuse more slowly than helium, tends to reduce helium surface adsorption and slow its diffusion. Among the carrier gases studied, nitrogen showed similar adsorption capacity, diffusivity, and stability to helium, while carbon dioxide displayed the highest adsorption capacity and the slowest diffusion rate, markedly differing from helium. Based on the simulation results, we concluded that water and carrier gases primarily function as transport mediums in helium migration, moving together with helium. Nitrogen, which shares similar properties with helium, effectively assists in this co-migration process. Conversely, carbon dioxide, due to its high adsorption capacity and slow diffusion, tends to be lost during co-migration. Sporadic E responds to the 2022 Tonga volcano eruptions recorded by the Meridian Project

In this study,ionosonde observations over Fuke(19.5°N,109.1°E),Wuhan(30.5°N,114.4°E),and Mohe(53.5°N,122.3°E)were analyzed to demonstrate the responses of the sporadic E()to the severe atmospheric disturbances caused by the Tonga volcanic eruptions on January 15,*** most prominent signature was the disappearance of the layer after~10:00 UT over Wuhan and Fuke,which was attributed to the vertical drift caused by the *** occurred intermittently after 13:00 UT following the arrival of the tropospheric Lamb *** examine the causal mechanism for the intermittence,we also included data of horizontal winds in the mesosphere and lower thermosphere region recorded by the meteor radars at Wuhan and Mohe in this *** wind disturbances with periods of~20 hours contributed to the formation of the layer in the nighttime on January 15.

Nano high-entropy oxide cathode with enhanced stability for direct borohydride fuel cells

High-entropy materials have become high-activity electrocatalysis owing to their high-entropy effect and multiple active ***,we synthesize a series of carbon-supported nano high-entropy oxides（HEOs/C）,specifically （PtFeCoNiCu）O/C,using a carbothermal shock （CTS） method for application as a cathode catalyst in direct borohydride fuel cells （DBFCs）.The microstructure of the prepared catalysts was characterized by X-ray photoelectron spectroscopy,X-ray absorption fine structure,and transmission electron *** prepared （PtFeCoNiCu）O/C,with particle sizes ranging from 2 to 4 nm,demonstrates 3.94 transferred electrons towards the oxygen reduction reaction in an alkaline environment,resulting in a minimal H2O2yield of 2.6%.Additionally,it exhibits a Tafel slope of 61 mV dec-1,surpassing that of commercial Pt/C (82 mV dec-1).Furthermore,after 40,000 cycles of cyclic voltammetry（CV） testing,the half-wave potential of （PtFeCoNiCu）O/C shows a positive shift of 3 mV,with no notable decline in the limiting current *** （PtFeCoNiCu）O/C is used as a cathode catalyst in DBFCs,the DBFC achieves a maximum power density of 441 mW cm-2at 60°C and sustains a cell voltage of approximately 0.73 V after 52 h at 30°*** findings confirm that HEO/C is a promising cathode catalyst for DBFCs.

Small extracellular vesicles derived from human induced pluripotent stem cell-differentiated neural progenitor cells mitigate retinal ganglion cell degeneration in a mouse model of optic nerve injury

Preparation of magnetically separable mesoporous activated carbons from brown coal with Fe3O4

Role of competing interactions on dynamic relaxation and exchange bias in spin-glass/ferromagnet bilayer

When a ferromagnet(FM)couples to a spin glass(SG),the hidden nature of SG may imprint on the FM via their interfacial coupling,probably resulting in elongated magnetic relaxation and enhanced unidirectional and uniaxial *** parallel with the study of interfacial roles,*** coupling and matching,we focused on the influence of the competing interactions in SG(JSG)on the magnetic relaxation properties of SG/FM bilayer and the FM magnetization reversal involving exchange bias field(HE)and coercivity(HC).A fastest decay of relaxation in SG/FM bilayer is found at an intermediate JSG^5.2 erg/cm2 and the relaxation parameters related to JSG are *** the other hand,a pronounced HE of^0.26 kOe is observed at small JSG,and as JSG increases larger than 2.275 erg/cm2,HE decreases by an order of magnitude and levels off,i.e.,a net EB *** is sensitive to JSG only at JSG above 2.275 erg/cm2 and exhibits a minimum value at JSG=4.55 erg/cm2,just below 5.2 erg/cm2 where the relaxation decay is the *** analysis unravels that the net magnetization associated with unidirectional magnetic stabilization and the large-area FM-like domains in SG are favored at small JSG,while with increasing JSG,the SG spins spontaneously reverse and thus the energy barriers are smeared out.A high susceptibility and a strong interfacial coupling lead to FM magnetization reversal under weaker fields and thus a decrease in *** too large JSG(exceeding 5.2 erg/cm2),magnetic frustration extends over the domains in SG meanwhile fierce energy competition occurs frequently,resembling enhanced quantum *** a result,dynamic relaxation and statistic coercivity are both *** work tells us that the independent modulations of unidirectional and uniaxial anisotropies are possible in SG-based systems through precisely controlling JSG,which is desired for application in the read/write head in future data storage media.
