AIM:To investigate M2 isoform of pyruvate kinase(PKM2) expression in gastric cancers and evaluate its potential as a prognostic biomarker and an anticancer ***:All tissue samples were derived from gastric cancer patients underwent curative gastrectomy as a primary *** and pathological information were obtained from the medical *** expression microarray data from 60 cancer and 19 noncancer gastric tissues were analyzed to evaluate the expression level of PKM2 *** microarrays were constructed from 368 gastric cancer *** was used to measure PKM2 expression and PKM2 positivity of cancer was determined by proportion of PKM2-positive tumor cells and staining *** between PKM2 expression and the clinicopathological factors was evaluated and the correlation between PKM2 and cancer prognosis was ***:PKM2 mRNA levels were increased more than 2-fold in primary gastric cancers compared to adjacent normal tissues from the same patients(log transformed expression level:7.6 ± 0.65 vs 6.3 ± 0.51,P < 0.001).Moreover,differentiated type cancers had significantly higher PKM2 mRNA compared to undifferentiated type cancers(log transformed expression level:7.8 ± 0.70 vs 6.7 ± 0.71,P < 0.001).PKM2 protein was mainly localized in the cytoplasm of primary cancer cells and detected in 144 of 368(39.1%) human gastric cancer ***2 expression was not related with stage(P = 0.811),but strongly correlated with gastric cancer differentiation(P < 0.001).Differentiated type cancers expressed more PKM2 protein than did the undifferentiated *** differentiated adenocarcinoma showed 63.6% PKM2-positive cells;in contrast,signet-ring cell cancers showed only 17.7% PKM2-positive ***,PKM2 expression was correlated with shorter overall survival(P < 0.05) independent of stage only in signet-ring cell ***:PKM2 expression might be an adverse prognostic factor for signet-rin

Inverse design of a Si-based high-performance vertical-emitting meta-grating coupler on a220 nm silicon-on-insulator platform

Ribonucleic acid (RNA) interference (RNAi) therapies are promising cancer treatment modalities that can specifically target abnormal proto-oncogenes, thus improving the therapeutic effect. For the treatment of pancreatic cancer, targeting one mutant proto-oncogene by RNAi usually does not yield the desired therapeutic efficiency. Both K-ras gene mutations and Notch1 overexpression are common symptoms in pancreatic cancer patients, and play a crucial role in pancreatic cancer cell drug resistance. In this study, biodegradable charged polyester-based vectors (BCPVs) were synthesized for the co-delivery of K-ras and Notch1 small interfering ribonucleic acid (siRNA) into MiaPaCa-2 cells (pancreatic cancer cell line) to overcome drug resistance to gemcitabine (GEM), a first-line chemotherapeutic drug used in the clinic. BCPVs could effectively absorb negative siRNA to form a capsule-like structure, prevent siRNA from nuclease digestion in the serum, and promote effective siRNA cell internalization and endosomal escape. Through K-ras and Notch1 gene silencing in MiaPaCa-2 cells, BCPV-siRNAK-ras-siRNANotch1 nanocomplexes effectively reversed the epithelia-mesenchymal transition (EMT) in MiaPaCa-2 cells, thereby greatly enhancing the sensitivity of MiaPaCa-2 cells to GEM. MiaPaCa-2 cell proliferation, migration, and invasion were effectively inhibited, and cell apoptosis was also significantly enhanced by the synergistic antitumor effect of BCPV-siRNAK-ras-siRNANotch1 nanocomplexes and GEM. These results suggest that this combination RNAi therapy can be used to improve cancer cell sensitivity to chemotherapeutic drugs. Specifically, this newly developed strategy has a great potential for treating pancreatic cancer.

Clinicopathologic factors and molecular markers related to lymph node metastasis in early gastric cancer

AIM: To analyze predictive factors for lymph node metastasis in early gastric ***: We analyzed 1104 patients with early gastric cancer(EGC) who underwent a gastrectomy with lymph-node dissection from May 2003 through July 2011. The clinicopathologic factors and molecular markers were assessed as predictors for lymph node metastasis. Molecular markers such as microsatellite instability, human mut L homolog 1, p53, epidermal growth factor receptor(EGFR) and human epidermal growth factor receptor 2(HER2) were included. The χ2 test and logistic regression analysis were used to determine clinicopathologic ***: Lymph node metastasis was observed in 104(9.4%) of 1104 patients. Among 104 cases of lymph node positive patients, 24 patients(3.8%) were mucosal cancers and 80 patients(16.7%) were submucosal. According to histologic evaluation, the number of lymph node metastasis found was 4(1.7%) for well differentiated tubular adenocarcinoma, 45(11.3%) for moderately differentiated tubular adenocarcinoma, 36(14.8%) for poorly differentiated tubular adenocarcinoma, and 19(8.4%) for signet ring cell carcinoma. Of 690 EGC cases, 77 cases(11.2%) showed EGFR overexpression. HER2 overexpression was present in 110 cases(27.1%) of 406 EGC patients. With multivariate analysis, female gender(OR = 2.281, P = 0.009), presence of lymphovascular invasion(OR = 10.950, P < 0.0001), diameter(≥ 20 mm, OR = 3.173, P = 0.01), and EGFR overexpression(OR = 2.185, P = 0.044) were independent risk factors for lymph node ***: Female gender, tumor size, lymphovascular invasion and EGFR overexpression were predictive risk factors for lymph node metastasis in EGC.

Convergence of Stereo Vision-Based Multimodal YOLOs for FasterDetection of Potholes

Road potholes can cause serious social issues,such as unexpected damages to vehicles and traffic *** efficient road management,technologies that quickly find potholes are required,and thus researches on such technologies have been conducted *** three-dimensional(3D)reconstruction method has relatively high accuracy and can be used in practice but it has limited application owing to its long data processing time and high sensor maintenance *** two-dimensional(2D)vision method has the advantage of inexpensive and easy application of ***,although the 2D vision method using the convolutional neural network(CNN)has shown improved pothole detection performance and adaptability,large amount of data is required to sufficiently train the ***,we propose a method to improve the learning performance of CNN-based object detection model by artificially generating synthetic data similar to a pothole and enhancing the learning ***,to make the defective areas appear more contrasting,the transformed disparity map(TDM)was calculated using stereo-vision cameras,and the detection performance of the model was further improved through the late fusion with RGB(Red,Green,Blue)***,through the convergence of multimodal You Only Look Once(YOLO)frameworks trained by RGB images and TDMs respectively,the detection performance was enhanced by 10.7%compared with that when using only ***,the superiority of the proposed method was confirmed by showing that the data processing speed was two times faster than the existing 3D reconstruction method.

Aggregation Induced Emission(AIE)is an attractive,fluorescent-based phenomenon that can be used to design fluorescent imaging agents and fluorescent *** this highlight,we discuss a recently reported study that identifies the chemical scaffold of an FDA-approved therapeutic(deferasirox)as a molecule with AIE-like properties(AIEgen).Utilizing its therapeutic efficacy for bacterial biofilms,deferasiroxderivatives were shown able to treat and image bacteria biofilms and detect a bacterial *** demonstrates the potential of AIE for drug design as this allows the simultaneous ability to image the therapeutic’s location and mode of action.

We investigated the anti-angiogenic effects of the water extract of HangAmDan (WEHAD),which is a crude extract of nine Korean medicinal substances of animal and plant *** human umbilical vein endothelial cells,WEHAD significantly inhibited bFGF-induced proliferation,adhesion,migration,and capillary tube *** used an antibody array to perform an analysis of signaling proteins,which showed up-regulated expression of various proteins including RAD51,RAD52,and p73,and down-regulated expression of *** vessel formation in a chick chorioallantoic membrane (CAM) treated with WEHAD was markedly reduced in length compared with a PBS-treated control *** results suggest that inhibition of angiogenesis by WEHAD may be the mechanism of action for the anti-cancer effects of HAD.

Clinicopathologic and molecular features associated with patient age in gastric cancer

AIM: To compare characteristics and prognosis of gastric cancer based on ***: A retrospective study was conducted on clinical and molecular data from patients(n =1658) with confirmed cases of gastric cancer in Seoul National University Bundang Hospital(Seoul, South Korea) from 2003 to 2010 after exclusion of patients diagnosed with lymphoma, gastrointestinal stromal tumor, and metastatic cancer in the stomach. DNA was isolated from tumor and adjacent normal tissue,and a set of five markers was amplified by polymerase chain reaction to assess microsatellite instability(MSI). MSI was categorized as high, low, or stable if ≥ 2, 1, or 0 markers, respectively, had *** was performed on tissue sections to detect levels of expression of p53, human epidermal growth factor receptor(HER)-2, and epidermal growth factor receptor. Statistical analysis of clinical and molecular data was performed to assess prognosis based on the stratification of patients by age(≤ 45 and> 45 years).RESULTS: Among the 1658 gastric cancer patients, the number of patients with an age ≤ 45 years was 202(12.2%; 38.9 ± 0.4 years) and the number of patients> 45 years was 1456(87.8%; 64.1 ± 0.3 years).Analyses revealed that females were predominant inthe younger group(P < 0.001). Gastric cancers in the younger patients exhibited more aggressive features and were at a more advanced stage than those in older patients. Precancerous lesions, such as atrophic gastritis and intestinal metaplasia, were observed less frequently in the older than in the younger group(P < 0.001). Molecular characteristics, including overexpression of p53(P < 0.001), overexpression of HER-2(P = 0.006), and MSI(P = 0.006), were less frequent in gastric cancer of younger patients. Fabrication of Variable Optical Attenuator With Low Insertion Loss

A voltage-controlled variable optical attenuator (VOA) based on silica-based planar lightwave circuits (PLC) was fabricated by flame hydrolysis deposition (FHD). The variable attenuator with Cr heater achieves 20dB attenuation with 318mW electrical power at 1550 nm. The insertion loss and the polarization dependent loss (PDL) at 0 attenuation are 1dB and 0.23dB, respectively.
